Studies on Bovine Mastitis in Seongwhan Area 1. Epidemiological Investigation (성환지역(成歡地域)의 유우유방염(乳牛乳房炎)에 관한 연구(硏究) 1. 유방염(乳房炎)의 발생실태(發生實態)와 그 원인균(原因菌) 조사(調査))

  • A total of 210 dairy cows of Seongwhan area were epidemiologically investigated for mastitis by Milk Quality Test(MQT) method. Milk samples from cows infected with mastitis were also bacteriologically studied. Seventy four (42.5%) of 174 dairy cows were positive for mastitis and fifty nine (79.7%) among them were infected with subclinical mastitis and fifteen (20.3%) were clinical mastitis. High infective rate were shown on August (48.7%) and July (45.7%) and it was lowered on December (35.8%). The incidence rate of subclinical mastitis for herd shown tendency of increase at the barn in which located on lower place (30.7~43.9%), whereas, the barn in which was on higher place and with bucket type milking system shown tendency of decrease causative organisms. A total of 230 strains of etiological agents were isolated from 578 infected duarters which shown MQT score (+) or more. The relevant causative organisms isolated in this study were Staphylococcus aureus (18.3%), Staphylococcus epidermidis (16.9%), fungi (16.5%), Pseudomonas aeruginosa (15.2%), Escherichia coli (7.8%) and Serratia marcescens (2.27), respectively.

Studies of Infection Rate and Causative Agents of Bovine Mastitis in Chungnam Area (충남지역(忠南地域) 유우유방염(乳牛乳房炎)의 감염율(感染率) 및 원인균(原因菌)에 관한 연구(硏究))

  • A total of 820 dairy cattle were examined formastitis by California mastitis test method and 181 dairy cattle were examined bacteriologically according to the method of Connecticut mastitis control program. The results obtained were as followings: 1. Milk samples from 1.231(38.1%) of 3,225 quarters and 568(69.3%) of 820 dairy cattle were positive for mastitis by California mastitis test method. 2. Among 181 cattle examined bacteriologically by the method of Connecticut mastitis control program 159(22.9%) quarters of 102(56.4%) cattle were infected. Clinical mastitis was found in 5 cattle. 3. Important causative organisms were found to de Staphylococcus aureus(44.8%), Streptococcus agalactiae(21.5%), and non-str, agalactiae Streptococcus(26.4%). Corynebacterium ssp., Pseudomonas spp., Coliforms and Yeasts were also occasionally found to be causative organisms. 4. Strains of Staphylococcus aureus isolated from bovine udders were tested for sensitivity to antibiotics. The rates of resistant strain were 13.7%, 11.8%, 8.8%, 5.9%, 100%, 4.9% and 4.9% for penicillin G, streptomycin, terramycin, erythromycin, chloramphenicol, ampicillin and kanamycin, respectively.

Therapeutic Dose, Duration, and Efficacy of Bee Venom for Treating Clinical Mastitis in Dairy Cow

  • This study was conducted to explore the efficacy of bee venom as a treatment for mastitis and to determine the optimal dosage and treatment period. When 6 mg or 12 mg of bee venom was administered to each experimental quarter of mastitis in dairy cow, the clinical symptoms in the 12 mg quarter were noticeably improved compared to those in the 6 mg quarter. There was no significant difference in the somatic cell count (SCC) in the milk between normal and mastitis quarters between the 6 and 12 mg doses, but there was a steady decrease in the 12 mg-treated quarter (p = 0.34). To determine the treatment period, bee venom was administered for 2, 4, and 7 days. After administering 12 mg of bee venom for 7 days, the SCC in the milk was compared before 6 days and after 7 days, and the SCC was significantly decreased to less than 100,000 SC/mL after 7 days (p = 0.01). In addition, to investigate the efficacy of bee venom, the minimum inhibitory concentration for S. aureus, E. coli, and coagulase negative staphylococci was measured, and the results showed that Gram-positive bacteria were more sensitive to bee venom than Gram-negative bacteria, and treatment for Gram-negative bacteria was limited. As a result of this study, it was confirmed that a dose of 12 mg of bee venom and a treatment period of more than 7 days were required to treat mastitis, and that treatment with bee venom alone against Gram-negative bacteria was negative.

  • A total of 2,053 quarter milk samples of 518 dairy cows from 46 herds were examined for mastitis and results obtained were as follows: 1. A total of 428 quarters(20.8%) of 271 dairy cows(52.3%) from 41 herds(89%) were found to be infected with mastitis. 2. It was found that 71 quarters(3.5%) of 41 cows(7.9%) from 21 herds(45.6%) were clinical mastitis and 357 quarters(17.3%) of 230 cows(44.4%) from 20 herds (43.4%) were subclinical mastitis. 3. The main causative organisms of the mastitis were Staphylococcus aureus(26%), Streptococcus uberis(16%), and Streptococcus agalactiae(10%). 4. The majority of causative organisms were very resistant to colistin(85.9%), penicillin(67%), and streptomycin(39%). Most of the other drugs were highly effective as inhibitor for the most causative organisms in vitro. However sulfisoxazole was moderatively effective against them.

  • A total of 364 quarter milk samples of 91 dairy cattle from 4 area around Jeonju in Jeonbug area were examined for infection rate and causative abets of mastitis by the Laboratory Procedures used in the Connecticut Mastitis-Control Program. The results obtained were as follows: 1. One hundred and thirty quarters (35.7%) from 67 cows (73.6%) were found to be infected with mastitis. It was found that 5 (1.37%) of the infected quarters were clinical mastitis and all of the rest were subclinical mastitis (125 quarters). 2. The main causative agents were found to be Staphylococcus aureus (46 quarters), Streptococcus agalactiae (49 quarters), streptococcus disgalactiae (18 quarters), Steptococcus uberis (13 quarters). Corynebacteria, Coliform organisms, Pseudomonas were also occasionally found to be causative organisms. 3. Having examined the number of infection Quarter per head of mastitis dairy cattle samplings, major findings were that the average number of infection quarter to total dairy cattle examined was 1.4 quarters, and that average number of infection quarter to the infected cattle was 1.9 quarters.

  • The diagnostic effects of the Modified Whiteside Test(M.W.T.) and the California Mastitis Test(C.M.T.) for detection of bovine mastitis were studied using 372 quarter milk samples collected from six dairy herds in suburbs of Suwon city in order to compare the effectiveness of the two methods. The leucocyte count and bacteriological observation were also performed to support analysing the results. The results obtained were as follows; 1. The CMT showed more specificity than MWT for leucocytes in the abnormal milk. 2. When comparison was based on Identical ratings, agreement was obtained in 56.4% of the samples. The stronger reaction had the higher agreement and the CMT was found to be more specific than MWT in detection of the sub-clinical mastitis. 3. The CMT reaction was instantaneous and sufficiently clear-cut. 4. The results of the CMT leucocyte count and bacteriological observation were completely agreed in 69.9% of the samples. When degrees of reaction and the leucocyte count were considered, the CMT showed agreement in 93.5% of the samples.

  • A total of 2,065 quarter milk samples of 534 dair cows from 35 herds were examined for mastis and the results obtained were as follows: 1. 654 quarters (31.6%) of 301 dairy cows (56.3%) from 34 herds (97.1%) were infected with mastitis. It was found that 8 cows (1.5%) showed clinical mastitis and other cows were subclinical. 2. Low infection rate of mastis was shown in first delivery cattle and higher infection rate in more deliverable of cattle. 3. The main causative organisms of the matitis were Staphylococcus aureus (19.4%), Streptococcus agalactiae (11.3%) and Streptococcus dysgalatiae (5%). 4. The majority of causative organisms were very resistant to colistin and highly sensitive to tetracycline. Especially Streptococcus strains were highly sensitive to leukomycin, and Staphylococcus aureus were highly sensitive to tetracycline and the other bacteria were highly sensitive to tetracycline and kanamycin.

  • Mastitis is an inflammation of the mammary gland that develops as a response to pathogenic microorganisms. Somatic cell count (SCC) and bacteriological analysis are the most accepted tools to determine udder health. This study evaluated the effects of intra-mammary gland infusion of silver ion against clinical mastitis in Holstein cows. Silver ion (20 ${\mu}g$/ml) was infused through the intra-mammary gland in quarters having clinical mastitis and milk was collected to determine SCC, and levels of lactoferrin and bovine serum albumin (BSA). Silver ion infusion decreased udder inflammation, firmness and swelling, and reduced clots, BSA, lactoferrin and SCC in milk. However, milk yield and circulating blood cells remained unchanged. The silver ion-mediated reductions of BSA and SCC indicate reduced inflammation and bacterial activity in silver ion-treated mammary glands in Holsteins with mastitis, which may be exploited in a curative strategy.

  • The functional non-woven fabrics have been applied in various industry fields, such as clothing, hygiene, environment, medical and so forth. The functional non-woven fabrics for wet tissue were manufactured by meltblown and wet tissue finishing processes. The wet tissue was contained the various composition substances such as Benzalkonium Chloride(0.2292g), lodo-2-propynyl Butyl carbamate(0.0069g) and 5-Chloro-2-Methyl-Isothiazolin -3-one, 2-Methyl-Isothiazolin-3-one (0.0034g) with purified water (999.76g). In this study, the functional wet tissue based on meltblown nonwovens has been applied in dairy cattle for prevention on cow mastitis. Mastitis is the most costly disease results in lost milk production, decreased milk quality, milk discard, early culling of cows, drug costs and labor costs in dairy cattle. Therefore, this study was investigated to evaluate clinical effect of the functional wet tissue in mastitis control. The results, after experiment the functional wet tissue used group, demonstrated that the infection rate, cure rate and mean somatic cell count(SCC) were significantly decreased as compared to the control group.

Comparison of Natural Resistance-associated Macrophage Protein (NRAMP)1 Expression between Cows with High and Low Milk Somatic Cells Counts

  • Studies using natural resistance-associated macrophage protein (NRAMP) identification indicated that cattle could be selected for immunity. Several studies performed on intracellular organisms such as Mycobacterium, Salmonella, Brucella and Leishmania in human and mouse revealed that resistance against these bacteria was dependent on high activity of NRAMP1 in macrophages. However, hardly any researches have been done on Staphylococcus aureus in bovine mastitis, which is an intracellular organism and the main cause of bovine mastitis. The objectives of this study were to establish reverse transcriptase polymerase chain reaction (RT-PCR) methods, through which NRAMP1 mRNA expression could be compared and analyzed between mastitis-resistant and -susceptible cows. NRAMP1 gene and its expression were investigated using 20 cows (Holstein Friesian) in Korea. Cows were evenly split into two groups, with and without histories of clinical mastitis. Equivalent numbers of cows were randomly selected from each group. Monocytes were isolated from the bovine peripheral blood of each selected cows and activated with lipopolysaccharide (LPS). mRNA was separated from the monocytes and cDNA of NRAMP1 was synthesized and amplified using RT-PCR with amplification of $\beta$-actin as a control. The difference in NRAMP1 expressions of mastitis-resistant (n=10) and -susceptible (n=10) Holstein cows was analyzed. Results demonstrate that resistant cows produced more NRAMP1 mRNA than the susceptible ones, and ratios of NRAMP1:$\beta$-actin expression were higher in resistant cows with or without LPS activation. Therefore, this study could be applied to select bovine mastitis resistant cows before infection based on the expression of NRAMP1.
