부산지역 가공식품 중 Bacillus cereus 분포 현황 및 독소 유전자 특성 (Distribution and Toxin Gene Characteristic of Bacillus cereus Isolated from Foods in Busan)

  • 본 연구는 2018년 10월에서 2019년 9월까지 부산지역 유통 판매되고 있는 872건의 다양한 식품을 대상으로 B. cereus 오염도를 분석하였다. 총 872건 중 78건(8.9%)에서 B. cereus 검출되었으며 식품별 B. cereus 검출률은 김치·절임·조림·젓갈류에서 23.9%, 조미식품 19.4%, 바로 섭취가능한 농산물 10.5%, 조리식품 7.3%, 즉석섭취식품 5.6%, 신선편의식품 5.0%, 즉석조리식품 0.0% 순으로 분포하였으며 통계적으로 유의한 차이가 있었다. B. cereus 오염 수준은 불검출에서 최대 20,000 CFU/g로 평균 48 CFU/g였으며, 식품별 B.cereus 오염도에는 차이가 없었다. 78건의 검체에서 분리된 113주의 B. cereus의 독소 유전자 확인 시험을 수행하였다. 식품에서 분리된 113주를 대상으로 B. cereus 독소유전자 확인 결과 최소 1종류에서 최대 5종류의 독소 유전자가 검출되었으며 총 18개 profile로 분류되었다. 장독소 5종(Cytk-nheA-entFM-bceT-hblC)을 모두 보유한 경우가 34주(30.1%)로 가장 많았다.

  • 2001년부터 2003년까지 부산지역의 냉각탑수에서 분리한 L. pneumophila (serogroup 1) 39균주에 대해 제한효소 SfiI 처리한 경우 PFGE 양상은 dice coefficient $<65\%$의 유사성을 가진 band를 Awl로 10개의 pulsotype으로 나누었고, 가장 많았던 유형은 E pulsotype으로 39주중 18주로 $46.2\%$를 나타내었으며, 그 외 A pulsotype $17.9\%$, C pulsotype $15.4\%$, F pulsotype $7.7\%$ 및 B, D, G, H, I, J pulsotype이 각각 $2.6\%$로 유전적 양상이 다양하였다. 제한효소 NotI 처리한 경우 PFGE 양상은 dice coefficient $<60\%$의 유사성을 가진 band를 $a\~h$로 8개의 pulsotype으로 나누었고, 가장 많았던 유형은 f pulsotype이 $38.5\%$였으며 그 외 d pulsotype $20.5\%$, e pulsotype $17.9\%$, a pulsotype $10.3\%$, h pulsotype $7.7\%$ 및 b, c, g pulsotype이 각각 $2.6\%$를 차지하였다. 본 연구를 통하여 부산지역에서 분리되는 레지오넬라균에 대한 유전자 유형분석 결과를 데이터베이스화하여, 레지오넬라증의 집단 발생 또는 산발적인 질병 발생이 있을 경우 분자학적 측면에서 사람과 환경에서 분리된 균으로부터 감염원과 감염경로를 규명하는 역학적인 도구로써 이용 가능할 것으로 사료되었다.

  • For urban development, natural covering area such as urban green or river is more rapidly reducing than artificial surface covering area like concrete or asphalt, so thermal environment in a city is being drastically deteriorated. Recently, since people recognize the importance of parks or rivers in a city which play roles as an environmental buffer in it, many studies and policies consider how to improve the life quality of citizens and urban environment. This study aims to examine the status of thermal environment variation in the parks and rivers of the city which is the subject of this research and provide foundational data for urban environment plans through research on temperature variation rate and temperature controlling effect.

3D City모델과 CFD 모델을 통합한 대기환경 시각화 연구 (A Study on Atmospheric Environment Visualization by Integrating 3D City Model and CFD Model)

  • 3D City 모델로부터 CFD 모델 입력자료를 생성하여 모델링 품질을 향상시키고 CFD 모델링 결과를 3D 모델과 통합하여 사실적이고 쉽게 인지할 수 있는 시각화 방법에 대하여 연구하였다. 국토지리정보원에서 구축한 3D City 모델을 이용해 세밀도와 정확도가 향상된 CFD 모델 입력자료를 생산하여 적용함으로써 상세한 도시공간구조를 반영하는 향상된 CFD 모델을 생산하였다. CFD_NIMR_SNU 모델로부터 바람의 흐름과 오염물질 확산을 시뮬레이션 하였으며 3D City Model과 CFD 모델을 통합하여 동적으로 시각화 하였다. 본 대기환경 시각화 도구는 도시기후 계획 및 관리에 있어 의사결정을 위한 시각적 소통능력을 향상시켜 비용과 시간을 최소화할 것이다.

부산 지하철 2호선 소음 특성 연구 (The Study of Sound Level Specification of Subway Line II in Busan)

  • The purpose of this paper is to evaluate the noise level and source, reduce the subway line II noise. As a result of measurement of subway line II in Busan the highest value section of uproad line was from Jigegol to Motgol by 89 dB(A). The sections of conversation and listening interference(over 80 dB(A)) were 21 sections(55%) of 38 sections. Among these sections, 15 sections(71%) were produced rolling noise, 3 sections(14%) squeal noise, 2 sections(10%) braking noise and 1 section(5%) fan noise, and then a main noise source was the rolling noise. In case of downroad line, the highest value section was from Busan Metro Art Museum to Centum city, Motgol to Jigegol by 88 dB(A). The sections of conversation and listening interference(over 80 dB(A)) were 18 sections(47%) of 38 sections. Among these sections, 15 sections(83%) produced rolling noise, 2 sections(11%) squeal noise and 1 section(6%) braking noise were investigated in this study. and then a main noise source was the rolling noise.

역사적 도시환경보존을 위한 건축문화재 활용방안 연구 -일본(日本) 카와고에시(川越市) 쿠라츠쿠리(藏造り)를 중심으로- (Use of Architectural Heritage for the Conservation of Historic Urban Environment -Kurazukuri Buildings in the city of Kawagoe, Japan-)

  • Many cities in Japan retain older city structures with buildings in traditional form and style. Visitors are fascinated by charms and tranquility of pre-modern life styles. Architectural features of olden days are well conserved or carefully restored on purpose. In the case of Kawagoe city near Tokyo, local residents are largely replete with visitors without being necessarily aware of slow commodification of historic spaces. Kawagoe is yet socio-culturally sustainable and financially fit. This paper intends to analyse conservation strategies, with focus on architectural heritage, taken for the historic city of Kawagoe. With the population of 300,000 in the suburb of Tokyo, it is one of the best known historic cities in Japan. Kawagoe grew as a merchant city since 1700s, and it once reached to a major port of silk trade en-route to Edo (Tokyo) from northen provinces. Tourism industry is discovered as a key element in the conservation strategies applied to revitalize historic city center, Ichibangai, an important cultural heritage site with special reference to kurazukuri. This paper argues that augmentation of conceptions in architectural heritage is not only advantageous for the conservation of historic urban environment but it also provides opportunities for sustainable urban renewal.

파주출판문화정보산업단지의 이용후평가 (A Study on the Post Occupancy Evaluation of PAJU Book City)

  • PAJU Book City Plan was designed to integrate two concepts of "Publishing Productive City" and "Ecological City" with the goal of "bringing of human being and nature together". This research on the post occupancy evaluation on PAJU Book City has two aims; One is to define how people, mainly working there rather than living, recognize the city and the other is to identify the problems resulted from the first phase of planning process and to suggest feasible solutions at the same time. It would be desirable that the outcome of this research should be used as data sources to improve the second phase of the plan. The evaluation criteria were created by the pre-survey on the basis of previous researches and "Pa Ju publishing city architectural design guide book (1999)". With those criteria, on-site questionnaire was conducted among people who work in PAJU Book City and stratified sampling was used for the proper sampling. The result is as follows; the visual-aesthetic criteria had positive response, which also closely related to a city satisfaction. Considering this result, construction type, circumferential environment of the building and a place of building form were found appropriate. The evaluation on the physical-ecological criteria was relatively low. Respondents were unsatisfied with green space and required better ecological waterway and more roadside tree. As green space was given a lot of weight in satisfaction survey, more green urban areas should be planned in the next phase. The evaluation on the social-behavior criteria got the lowest score. Particularly, the accessibility and convenient facilities were remarkably low. Nonetheless, the self-conceit about PAJU Book City was highly evaluated and played a crucial role in a city satisfaction. In order to achieve the goal, physical beauty, characteristics as a publication city as well as an ecologically sustainable city should be main agenda in the second phase of Paju Book City Plan. Consequently, it is important to strike a balance between the external environment and image media in order that PAJU Book City should be re-born as a culture city.

액체크로마토그래피를 이용한 벌꿀 중 테트라싸이클린계 항생물질의 정량분석 및 잔류조사 (Determination and survey of tetracyclines residue in honey by high performance liquid chromatography)

  • Oxytetracycline, tetracycline, chlortetracycline and doxycycline in honey were separated by solid phase extraction (SPE) and determined with high performance liquid chromatography (HPLC) with UV/Visible detector. Analysis was carried out using following conditions: XTerra $C_8$ column $(3.9\times150mm\;i.d. 5{\mu}m)$, mobile phase composed of 0.01M oxalic acid : methanol : acetonitrile (820 : 80 : 100, v/v/v), isocratic pump at a flow rate of 0.9 ml/min. and $50{\mu}l$ of injection volume, UV/Visible detector with wavelength of 360nm. The calibration curves of four tetracyclines showed linearity $(\gamma^2>0.999)$ at concentration range of $100\~1,000 ng/ml$. The recoveries in fortified honey represented more than $70\%$ with low coefficient of variation $(<10\%)$ for concentration range of four tetracyclines. The detection limits for oxytetracycline, tetracycline, chlortetracycline and doxycycline were 13.8, 14.6, 26.2 and 24.9ng/g in acacia honey. respectively. We also monitored tetracyclines residue in domestic honey [n : 38, acacia (20), wild flower (18) ] and foreign honey [n=22, legally distributed (13), illegally distributed (9)] using modified Charm II screening and HPLC confirmation methods. Seven of the 60 samples $(11.7\%)$ were suspect positive using modified Charm II screening test. Chlortetracycline residue was found in one foreign honey (illegally distributed) tested at concentrations of 0.22 ppm. Conclusively, for more effective control of tetracyclines used in beekeeping should be further survey for residues in honey and also national guidelines (maximum residue limit : MRL) and methods should be obligatory.

The monitoring of heavy metals (lead and cadmium) in honeys consumed in Incheon region

  • This study was conducted to investigate the contents of lead (Pb) and cadmium(Cd) in honeys (n=60) from April to May in 2004 and to provide the scientific basis for heavy metal regulations of Korea Food Code. The samples were digested with acids, then analyzed for the contents of Pb and Cd by an inductively coupled plasma spectrometer (ICP). The contents of Pb and Cd [minimum-maximum (mean)] were 0.189-1.82 mg/kg (0.568) and not detected (ND)-0.016 mg/kg (0.0008) in domestic acacia honeys (n=20), ND-1.702 mg/kg (0.329) and ND-0.243 mg/kg (0.013) in domestic wild flower honeys (n= 18), ND-0.322 mg/kg (0.073), ND-0.027 mg/kg (0.002) in imported honeys (n=13), ND-3.754 mg/kg (0.671) and ND-0.658 mg/kg (0.073) in foreign honeys (n=9) brought by Korean travellers, respectively. According to the results, foreign honeys brought by Korean travellers were detected with the highest level of Pb and Cd. Therefore, we recommend that heavy metals of domestic and foreign honeys should be continuously monitored. It is also thought that these results could be the important references to establish the standard of Pb and Cd in honey.

Prevalence of honeybee diseases in Incheon area in 2011

  • This study investigated the occurrence of honeybee diseases in Incheon area, at the point of great widespread of sacbrood disease in the country. Sixteen resident beekeeping apiaries; 3 native honeybee and 13 European honeybee apiaries were selected for this research. Over 20 adult bees were evenly collected from the most colonies of each apiary three times (March, June, November) within a year. In this work, 13 honeybee diseases including 7 viral diseases, 2 bacterial diseases, 2 fungal diseases, and 2 parasitic diseases were detected by preliminary inspections and PCR. As a result, viral infections were confirmed at 34 among 48 apiaries (70.8%) over the entire examination period. Parasitic diseases showed the highest detection rate of 45.8%, which are detected in 44 among 96 cases. In the seasonal prevalence, 30 cases (15.6%) of 7 pathogens were detected from 14 apiaries in March, 50 cases (24.0%) of 9 pathogens and 56 cases (26.9%) of 9 pathogens were detected from all apiaries in June and November, respectively. Nosema was shown to be the most prevalent pathogen from March to November, followed by sacbrood virus (SBV) and stonebrood. The spread of SBV infection in Incheon would be under-estimated by the increasing of detection rate over the time. Especially, Chinese sacbrood virus was detected from 4 European honybee apiaries, but clinical symptoms were not found. No chalkbrood, acute bee paralysis virus, and chronic bee paralysis virus were detected in this study. The effective therapy and preventive measures should be prepared for beekeeping industry.
