Citrobacter amalonatics와 Citrobacter farmari에 의한 perchlorate 환원

  • The present study reports the novel physiological function of dissimilatory perchlorate reduction by two strains JB101 and JB109 isolated from a sewage treatment facility in Incheon, South Korea. The physiological data of the isolates showed good correspondence with the members of the family Enterobacteriaceae. The partial 16S rRNA and 16S rDNA sequence of strains JB101 and JB109 showed similarity of 99.8% to Citrobacter amalonaticus and 98% to Citrobacter farmari, respectively. The study infers toward possibility of Citrobacter spp. to form an important group of dissimilatory perchlorate reducers within the (equation omitted) subclass of Proteobacteria because the majority of the known. members belong to two monophyletic groups namely Dechloromonas and Dechlorosoma in $\beta$ subclass of Proteobacteria.

Growth Characteristics of Citrobacter sp. MB2, Azo Dyes Decolorizing Bacterium (아조염료 분해균 Citrobacter sp. MB2 생육특성)

  • A Citrobacter sp. MB 2, azo dyes decolorizing bacterium, was isolated from the wastewater and soil and identified as Citrobacter sp.. It was examined that optimum conditions for culture media were 0.5% of sucrose, 1.0% of yeast extract, 0.1% of $K_2HPO_4$, 0.1% of $NaHCO_3$ per distilled water. The best efficient condition of culture was obtained at pH 7.0, $30^{\circ}C$ and aerobic shaking culture. The number of Citrobacter sp. MB2 in optimum medium was increased more than 7 fold compared to basal medium and 50 fold compared to nutrient broth. This strain was exhibited strong resistance against metal salts and antibiotics (ampicillin and penicillin G).

  • The production of antileukemic enzyme methionine ${\gamma}$-lyase (MGL) in distinctly related bacteria, Citrobacter freundii and in their recombinants expressing the Vitresocilla hemoglobin (VHb) has been studied. This study concerns the potential of Citrobacter freundii expressing the Vitreoscilla hemoglobin gene (vgb) for the methionine ${\gamma}$- liyase production. Methionine ${\gamma}$- liyase production by Citrobacter freundii and its $vgb^-$ and $vgb^+$ bearing recombinant strain was studied in shake-flasks under 200 rpm agitation, culture medium and $30^{\circ}C$ in a time-course manner. The $vgb^+$ and especially the carbon type had a dramatic effect on methionine ${\gamma}$- liyase production. The $vgb^+$ strain of C. freundii had about 2-fold and 3.1-fold higher levels of MGL than the host and $vgb^-$ strain, respectively.

Development of a Rapid and Accurate Identification Method for Citrobacter Species Isolated from Pork Products Using a Matrix-Assisted Laser-Desorption Ionization Time-of-Flight Mass Spectrometry (MALDITOF MS)

  • Previous detection methods for Citrobacter are considered time consuming and laborious. In this study, we have developed a rapid and accurate detection method for Citrobacter species in pork products, using matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometry (MS). A total of 35 Citrobacter strains were isolated from 30 pork products and identified by both MALDI-TOF MS and 16S rRNA gene sequencing approaches. All isolates were identified to the species level by the MALDI-TOF MS, while 16S rRNA gene sequencing results could not discriminate them clearly. These results confirmed that MALDITOF MS is a more accurate and rapid detection method for the identification of Citrobacter species.

  • Citrobacter freundii is a facultative anaerobic and a Gram-negative bacterium of Enterobacteriaceae family, and is an opportunistic pathogen. Bacteriophages infecting C. freundii can be an effective treatment for C. freundii infections. Here, the complete genomic sequence is announced for a lytic bacteriophage CF1 infecting C. freundii isolates.

Degradation oof Triphenylmrthane Dyes by Citobacter sp. (Citrobacter sp.에 의한 Triphenylmethane계 색소의 분해)

  • The Optimal condition for degradation of crystal violet and other triphenylmethane dyes by Citrobacter sp. SK-3 isolated from the activated sludge of dye manufacturing factory was investigated. The optimal culture medium for the degradation of triphenylmethane dye was composed of minimum inorganic salt medium supplemented with 0.5% galactose, 0.1% beef extract, with the initial pH of 8.0 to 9.0. Under this condition, Citrobacter sp. SK-3 degraded 200 ppm of crystal violet completely within 24 hours. Citrobactre sp. SK-3 also degraded efficiently malachite green, pararosaniline, brilliant green, methyl violet, basic fuchsin and methyl red. Analysis of the degradation products of crystal violet through this layer chromatography and high performance liquid chromatography indicated that the methyl groups bound to crystal violet backborn were gradually demethylated to pentamethyl-, tetramethyl- and trimethylpararosaniline.

  • The bio-electrode for cytosine has been constructed by immobilizing Proteus mirabilis and Citrobacter freundii on an ammonia gas-sensor. Bacteria containing cytosine deaminase convert one molecule of cytosine into one molecule of ammonia. The Proteus mirabilis bacterial electrode showed linear response to cytosine concentration in the $1.0{\times}10^{-3}\;-\;5.0{\times}10^{-2}$M with a slope of 45-48 mV/decade in 0.2 M phospbate buffer solution at pH 8.4. The Citrobacter freundii bacterial electrode showed linear response to cytosine concentration in the $7.0{\times}10^{-5}\;-\;7.0{\times}10^{-3}$M with a slope of 48 mV/decade in 0.05M phosphate buffer solution at pH 7.6. These electrode were investigated for the effects of pH, temperature, buffer solutions, amounts of bacteria, interferences, inorganic salts and lifetime.

Inhibition Effect of Ginseng Saponin on the Growth of Citrobacterer sp. Isolated from Contaminated Ginseng (오염된 인삼으로부터 분리된 Citrobacter sp.에 대한 인삼사포닌의 생육억제 효과)

  • A bacterium isolated from contaminated white ginseng was identified using API kit and electron microscope. This isolate was determined as rod shaped bacterium having about 1.0 ${\mu}m$ in diameter and 2.0 to 6.0 ${\mu}m$ in length. It had motility by peritrichous flagellum. The isolate had ${\beta}-galactosidase$, arginine dihydrolase and ornithin decarboxylase. It did not have ability not only to use citrate as sole carbon source and but also to produce $H_2S$. However, it could ferment glucose, manitol, sorbitol, rhamnose, arabinose and amygdalin. From these obserbations, the isolate was identified as Citrobacter sp. Ginseng saponin was added to culture of Citrobacter sp. in order to investigate saponin's influence on its growth. The strain was incubated at $38^{\circ}C$ for 3 days after addition of 0.05, 0.5, 2.0 and 4.0% (w/v) of saponin, respectively and the growth rates was investigated. The relative bacterial growth inhibition rates showed 28.6, 66.7, 92.4 and 97.7%, respectively, when compared with saponin non-treated group. These results suggest that the growth of Citrobacter sp. is inhibited by saponin in a concentration-dependent manner.

  • ${\mu}MT$ knockout mice are genetically deficient in the transmembrane domain of mu chain of the immunoglobulin M (IgM) heavy chain, resulting in the absence of mature B cells. ${\mu}MT$ knockout mice is an in vivo model system used to clarify the role of B cells in various diseases. Enteropathogenic Escherichia coli (EPEC) induces acute and chronic diarrheal disease, especially in children of developing countries. The formation of attaching and effacing (A/E) lesion is a prominent pathogenic factor in the intestinal epithelium of EPEC infection. The A/E lesion is modulated by genes located on the pathogenic island locus of enterocyte effacement (LEE) which encode a type III secretion system (T3SS) and A/E lesion-related effector proteins. Citrobacter rodentium is a murine pathogen utilized in studying the pathogenic mechanisms of EPEC in human infections. Citrobacter rodentium produce A/E lesion to attach to intestinal epithelium, thus providing a murine model pathogen to study EPEC. Several studies have investigated the pathogenesis of Citrobacter rodentium in the ${\mu}MT$ knockout mice. In this review, we introduce the ${\mu}MT$ murine model in the context of C. rodentium pathogenesis and describe in detail the role of B cells and antibodies in this disease.

Mass Mortality of Doctor Fish(Garra rufa obtusa) Caused by Citrobacter freundii Infection (Citrobacter freundii 감염에 의한 Doctor fish(Garra rufa obtusa)의 집단 폐사)

  • In this paper, we described a case of mass mortality of doctor fish from a private fish hatchery farm in Korea with a history of abnormal swimming behavior, diffuse bleeding on the skin and fins and sudden death caused by fish pathogenic bacteria, Citrobacter freundii. Twelve moribund fish fingerling samples were submitted to College of Veterinary Medicine, Seoul National University in October 2008 for diagnostic examination. Diagnostic results showed that the morphological and biochemical properties of the bacteria isolated from the moribund fish were C. freundii. The remaining diseased fish from the hatchery farm were given treatment based on our recommendation and successfully recovered.
