국내에 자생하는 큰엉겅퀴와 고려엉겅퀴의 분자유전학적 및 화학적 분석 (Phylogenetic and Chemical Analyses of Cirsium pendulum and Cirsium setidens Inhabiting Korea)

  • 국내에 자생 중인 큰엉겅퀴(Cirsium pendulum)를 강원도의 홍천, 원주, 평창, 양양 및 경기도 가평과 충청북도의 충주에서 채취하고, 고려엉겅퀴(Cirsium setidens)를 강원도의 태백 및 경상북도의 봉화에서 채취하였다. 채취된 식물들의 genomic DNA를 분리하여서 18S rDNA, ITS1, 5.8S rDNA, ITS2 및 28S rDNA의 일부를 증폭하고, 그 염기서열을 분석하였다. 이렇게 얻어진 염기서열을 GenBank에 등록하고, 서로 비교해 보았다. 그 결과 큰엉겅퀴는 6개체 모두에서 18S rDNA 및 5.8S rDNA의 염기서열은 서로 완전히 일치하고 있으나, ITS1과 ITS2의 염기서열에서는 약간의 차이를 보였다. 고려엉겅퀴의 경우에는 18S rDNA, ITS1 및 ITS2에서 2개체가 서로 비교적 큰 차이를 보였다. 일본의 홋카이도에서 채취된 큰엉겅퀴와 중국의 안휘성에서 채취된 엉겅퀴(Cirsium japonicum)를 포함하여서 ITS1과 ITS2 염기서열을 비교한 결과, 엉겅퀴, 큰엉겅퀴, 고려엉겅퀴는 확연하게 서로 다른 그룹을 형성하는 것으로 나타났다. 또한 채취된 큰엉겅퀴 및 고려엉겅퀴들의 silymarin 함량을 분석해 본 결과, 모두에서 그 함량이 2 mg/ml가 넘는 것으로 나타났다. 따라서 엉겅퀴뿐만 아니라 큰엉겅퀴나 고려엉겅퀴도 여러 가지 약리작용을 가진 것으로 보고된 silymarin을 상당히 높은 농도로 함유하고 있는 것으로 나타났다.

엉겅퀴 추출물의 기능 성분 분석 및 TGF-beta에 의한 간 성상 세포 활성 억제 효과 (Analysis of Active Components in Cirsium japonicum var. ussuriense Extracts and Their Effect on TGF-beta Induced Hepatic Stellate Cells Activation)

  • Cirsium japonicum (CJ) leaf (L) alcoholic extracts were investigated for analysis their active components (flavonoids and flavanolignans; silymarins) and inhibitory effect on transforming growth factor (TGF)-${\beta}$ induced hepatic stellate cells (HSCs, LX-2 cells) activation. The CJ root (R) extracts were also analyzed and compared with leaf extracts. Total flavonoid and polyphenol contents of the leaf extracts showed higher than those of the root extracts. The content of each flavonoid compound, which was analyzed by HPLC, in CJ-L extracts was also higher than in CJ-R extracts. The results of flavanolignans content in CJ-L and CJ-R extracts were consistent in flavonoid and polyphenol. We studied inhibitory effect of two extracts against TGF-${\beta}1$ induced HSCs activation. The CJ-L extracts significantly suppressed overexpression of profibrogenic factor, ${\alpha}$-smooth muscle actin and collagen-${\alpha}1$(I). The CJ-R extract also showed inhibitory effect on TGF-${\beta}1$ induced HSCs activation, but the efficacy was lower than in CJ-L extract. These results suggest that CJ-L may contribute to the fibrotic liver treatment.

Protective effect of Cirsium japonicum var. maackii against oxidative stress in C6 glial cells

  • This study was investigated the anti-oxidant property and neuro-protective effect of Cirsium japonicum var. maackii (CJM) against oxidative stress in hydrogen peroxide ($H_2O_2$)-induced C6 glial cells. We measured the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, hydroxyl radical (${\cdot}OH$), and superoxide ($O_2{^-}$) radical scavenging activities of an ethanol extract and four fractions [n-Butanol, ethyl acetate (EtOAc), $CHCl_3$, and n-Hexane] from CJM. The results of this study show that the extract and all fractions from CJM had a dose-dependent DPPH radical scavenging activity. In particular, the EtOAc fraction exhibited the strongest scavenging effect with 88.23% at a concentration of $500{\mu}g/mL$. In addition, the EtOAc fraction from CJM also effectively scavenged ${\cdot}OH$ radicals and $O_2{^-}$ radicals, compared to other extract and fractions. In C6 glial cells, $H_2O_2$ markedly decreased the cell viability as well as increased lactate dehydrogenase (LDH) release and reactive oxygen species (ROS) production. However, the EtOAc fraction of CJM attenuated the cellular damage from the oxidative stress by elevating the cell viability and inhibiting the LDH release and ROS over-production compared with the $H_2O_2$-treated control group. Our findings indicate that the EtOAc fraction from CJM has antioxidant effect and neuro-protective effect against oxidative stress, suggesting that it can be used as a natural antioxidant and therapeutic agent for the prevention of neurodegenerative disorders.

영동지역 5개 초본 수종의 연소특성 (Combustion Characteristics of the 5 Herb Species in Youngdong Areas)

  • 본 연구에서는 영동지역에서 자생하는 초본류 가운데 김의털, 방아풀, 주름조개풀, 칡, 엉겅퀴를 대상으로 발화점시험기, 콘칼로리미터, 연기밀도시험기 등을 이용하여 연소특성을 고찰하였다. 발화온도 범위는 $400{\sim}455^{\circ}C$로 확인되었으며 착화는 일어나지 않았다. 방아풀은 가장 많은 열량을 방출하였고, 김의털은 가장 많은 연기를 방출하였으며, 칡과 엉겅퀴는 가장 많은 CO와 $CO_2$를 방출하였다. 따라서 산불발생 시 방아풀은 열방출량이 많아 산불강도가 클 것으로 생각되며 김의털, 칡, 엉겅퀴는 다량의 연기와 CO, $CO_2$가 방출됨에 따라 피난이 어려울 것으로 사료된다.

엉겅퀴에 함유된 Cirsimaritin 분석을 위한 HPLC 분석법 밸리데이션 (Validation of an HPLC/UV Analysis Method for Cirsimaritin in Cirsium japonicum var. maackii)

  • Cirsium japonicum var. maackii (CM) has been used to treat certain medical conditions such as hemorrhage, hepatitis, and hypertension. Cirsimaritin was previously found as the major flavonoid in CM and is said to contribute to its pharmacological effects. There are currently no reports detailing the qualitative and quantitative detection of phytochemical indicators in the aerial parts of CM. Therefore, we developed a method to rapidly identify and quantify cirsimaritin in CM using HPLC/UV, and we optimized and validated this analytical method. The results showed good linearity in the concentration range tested (0.25-0.015 mg/mL, r2 ≥ 0.9999), accuracy (93.9-111.3%), and precision (RSD ≤ 0.59%). The developed method can therefore be used for the rapid evaluation of cirsimaritin in CM.

Effects of Medicinal herb Extracts and their Components on Steatogenic Hepatotoxicity in Sk-hep1 Cells

  • Herbal medicines are widely used in many countries for the treatment of many diseases. Although the use of herb extracts as alternative medicine is growing, their toxicological properties have not been thoroughly investigated. In this study, we have investigated the effects of water and ethanol extracts of 18 herbs on the hepatic lipid metabolism and steatogenic hepatotoxicity. Ethanol extracts of Cirsium japonicum, Carthamus tinctorius, Rehmanniae glutinosa (preparata), Polygala tenuifolia, Foeniculum vulgare, Polygonum multiflorum, and Acorus gramineus and water extracts of Polygonum multiflorum and Rehmanniae glutinosa induced lipid accumulation in Sk-hep1 human hepatoma cells as determined by Nile red staining. These extracts increased the luciferase activity of sterol regulatory element (SRE) and decreased that of peroxisome proliferator response element (PPRE), indicating the possibilities of enhanced fatty acid synthesis and decreased fatty acid oxidation. To identify the components responsible for the fat accumulation, we tested 50 chemicals isolated from the nine herbs. Apigenin, luteolin, pectolinarin and lupeol from Cirsium japonicum, 8-methoxypsoralen and umbelliferone from Foeniculum vulgare and pomonic acid and jiocerebroside from Rehmanniae glutinosa significantly increased the accumulation of lipid droplets. These results suggest that ethanol extracts of Cirsium japonicum, Carthamus tinctorius, Rehmanniae glutinosa (preparata), Polygala tenuifolia, Foeniculum vulgare, Polygonum multiflorum, and Acorus gramineus and water extracts of Polygonum multiflorum and Rehmanniae glutinosa can cause fatty liver disease by decreasing ${\beta}$-oxidation of fatty acid and increasing lipogenesis.

Cirsium japonicum var. maackii inhibits hydrogen peroxide-induced oxidative stress in SH-SY5Y cells

  • Over-produced reactive oxygen species (ROS) exert oxidative damage on lipids, proteins, and DNA in the human body, which leads to the onset of neurodegenerative diseases such as Alzheimer's disease (AD). In this study, we explored the cellular antioxidant effect of Cirsium japonicum var. maackii (CJM) against hydrogen peroxide (H2O2)-induced oxidative stress in neuronal cells. The antioxidant activity was assessed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, 2',7'-dichlorofluorescin diacetate and nitric oxide (NO) assays, and the molecular mechanisms were examined by Western blot analysis. H2O2 treatment of SH-SY5Y cells decreased cell viability and increased ROS and NO production compared to H2O2-untreated cells. However, CJM increased cell viability and decreased ROS and NO accumulation in the H2O2-treated SH-SY5Y cells compared to H2O2-treated control cells. Especially, the EtOAc fraction from CJM showed the strongest antioxidant effect compared with the other extracts and fractions. Therefore, we further examined the CJM mechanism against oxidative stress using the EtOAc fraction from CJM. The EtOAc fraction up-regulated the expressions of heme oxygenase-1, NAD(P)H quinone oxidoreductase 1, and thioredoxin reductase 1. These results indicate that CJM promotes the activation of antioxidative enzymes, which eliminate ROS and NO, and further leads to an increase in the cell viability. Taken together, our results show that CJM exhibited an antioxidant activity in H2O2-treated SH-SY5Y cells, and it could be a novel antioxidant agent for the prevention or treatment of neurodegenerative disease such as AD.

엉겅퀴 뿌리 물 추출물의 류마티스 관절염 동물 모델에 대한 개선 효과 (Improvement Effect of the Water Extract from the Root of Cirsium japonicum var. ussuriense on Type II Collagen-induced Rheumatoid Arthritis Animal Models)

  • Background: The roots of Cirsium japonicum var. ussuriense (RCJ) have been used as traditional medicine in Korea for hematuria and hematemesis. These extracts exert anti-oxidative and anti-inflammatory effects by scavenging for free radical and regulating the inflammatory response. However, the effect of RCJ on rheumatoid arthritis (RA) has not been elucidated. Thus, we evaluated the water extract of RCJ (WRCJ) using type II collagen-induced RA models. Methods and Results: RA was induced by immunization with type II collagen. All experimental materials were orally administered daily for three weeks. The positive control group was administered with 0.2 mg/kg methotrexate (n = 7), while the experimental group was administered with WRCJ (100 or 500 mg/kg, n = 7). Serum levels of TNF-alpha, Interleukin 6 (IL-6), and type II collagen IgG (CII) were measured using ELISA. Administration of 500 mg/kg WRCJ decreased the levels of TNF-alpha, IL-6, and CII. Moreover, WRCJ treatment diminished swelling of hind legs and infiltration of inflammatory cells in RA models' synovial membrane. Conclusions: These results indicate that WRCJ could improve RA, reduce inflammatory indicators and synovial inflammation. However, further experiments are required to determine how WRCJ can influence the signal transduction pathway in RA.

Antioxidant effects of Cirsium japonicum var. maackii on oxidative stress in C6 glial cells and mice

  • We investigated the effects of Cirsium japonicum var. maackii (CJM) against oxidative stress-induced C6 glial cells and cognitive impairment in mice. To evaluate the anti-oxidative effect of the extract and fractions from CJM, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), reactive oxygen species (ROS), and nitric oxide (NO) assays were conducted in H2O2-treated C6 glial cells. Furthermore, we identified the protective mechanisms of CJM with a scopolamine-treated mice model. The results revealed that H2O2 decreased the cell viability in C6 glial cells, indicating that H2O2 induced oxidative stress in glial cells. However, CJM fractions significantly increased cell viability in H2O2-treated C6 glial cells, which suggested that CJM protected against oxidative stress. CJM extract and fractions also reduced ROS and NO production, which were increased by H2O2 in C6 glial cells. In particular, the EtOAc fraction from CJM (EACJM) effectively protected against oxidative stress by increasing the cell viability and decreasing ROS and NO. Therefore, we carried out further in vivo experiments with EACJM. Scopolamine caused increases of ROS, thiobarbituric acid reactive substances (TBARS), and NO production. However, EACJM effectively alleviated ROS, TBARS, and NO levels compared to scopolamine-injected mice. In addition, EACJM up-regulated protein expressions of superoxide dismutase and glutathione peroxidase, indicating that EACJM enhanced the antioxidative system. Our results demonstrated that CJM had protective effects against oxidative stress in glial cells and memory dysfunction in mice. Based on these results, we propose that CJM could be a potential AD preventive and therapeutic agent.

엉겅퀴 추출물의 항산화성, 항돌연변이원성 및 항암활성 효과 (Antioxidant, Antimutagenicity and Anticancer Activities of Extracts from Cirsium japonicum var. ussuriense KITAMURA)

  • 엉겅퀴 부위별 메탄을 추출 수율을 구한 결과 뿌리 및 잎 추출물은 10% 정도의 수율을 보였으나 줄기 추출물은 2.8%의 낮은 수율을 보였다. 항산화활성 실험 결과 부위별 ethyl acetate 분획물과 butanol분획물에서 강한 항산화 활성을 보였으며, 특히 뿌리의 ethyl acetate 분획물$(RC_{50}:11{\mu}g/m{\ell})$과 butanol 분획물$(RC_{50}:14{\mu}g/m{\ell})$, 잎의 butanol 분획물$(RC_{50}:15{\mu}g/m{\ell})$에서 대조구인 ${\alpha}-tocopherol(RC_{50}:12{\mu}g/m{\ell})$이나 $BHA(RC_{50}:14{\mu}g/m{\ell})$정도의 항산화활성을 보였다. 곰팡이에 대한 포자발아 억제시험에서는 전반적으로 물분획물에서 우수한 활성을 나타냈다. Yeasts에 대한 실험에서는 ethyl acetate 분획물에서 대조구와 유사한 활성을 나타냈다. 박테리아에 대한 항균시험은 역시 ethyl acetate 분획물에서 활성을 나타냈다. Ames test결과 $MNNG(0.4{\mu}g/plate)$의 경우 S. typhimurium TA100 균주에서 시료농도 $1000{\mu}g/plate$에서 핵산 분획물이 88.18%로 가장 높은 억제효과가 나타났다. $4NQO(0.15{\mu}g/plate)$에 대한 S. typhymurium TA98 균주에서 시료농도 $1000{\mu}g/plate$에서는 엉겅퀴 뿌리 추출물 및 에칠아세테이트 분획물이 77, 75.8%의 억제효과를 나타냈으며, TA100 균주에서도 엉겅퀴 뿌리 추출물 및 에칠아세테이트 분획물이 72.5, 74.3%의 억제효과를 나타냈다. $Trp-P-1(0.15{\mu}g/plate)$을 사용한 S. typhimurium TA98 균주에서는 시료농도 $1000{\mu}g/plate$에서 엉겅퀴 뿌리 추출물$(1000{\mu}g/plate)$에서 82.25%의 가장 높은 억제효과를 보였으며, TA100균주의 경우, 시료농도 $1000{\mu}g/plate$에서 부탄을 분획물에서 77.75%의 가장 높은 억제효과를 보였다. 엉겅퀴 뿌리 추출물 및 용매 극성을 이용한 분획물을 대상으로 암세포주에 대한 증식 억제 효과를 실험한 결과, A549, Hep3B, MCF-7 세포주에서 메탄올 추출물이 가장 우수한 억제효과를 나타내었으며 인간유래의 정상 간세포 293에 대하여 시료농도에 따른 세포독성 효과는$1000{\mu}g/ml$ 시료 첨가 시 암세포에 대하여 대부분 60% 전후의 억제율을 보이는데 반해 293에 대해서는 25% 이하의 생육억제율을 보였다. 이는 암세포에 대한 높은 억제효과에 비해 정상세포에 대해서는 비교적 낮은 독성효과를 나타냄을 알 수 있었다.
