Optimal Steaming Condition of Gastrodia elate Blume (Chunma) using Response Surface Methodology (RSM) (반응표면분석법을 이용한 천마의 최적증자조건 설정)

  • The Chunma requires a simple pre-treatment, due to the inconveniences of preservation, toxicities, and pungencies in intact state. Various steaming conditions were examined to establish the anti-oxidative characteristics of Chunma using the Response Surface Methodology (RSM). Total phenolics and electron-donating ability were optimal at 94.43 and $101.67^{\circ}C$ for 42.38 and 41.09 min, respectively. Anti-oxidative (Al) and nitrite-scavenging ability were optimal at $100^{\circ}C$ for 40 min. Optimal steaming conditions were determined as 96{\sim}107{\circ}C$ for $28{\sim}46min$ min, almost identical to the predicted conditions of $100^{\circ}C$, 40 min determined using the superimposed contour map. Anti-oxidative activities (AI) under the optimal conditions were $95.9%{\sim}115.6%$ of those determined under predicted conditions, an indication of the high reliability of the calculated values.

  • The Chunma (Gastrodia elata Blume) requires a simple pre-treatment, due to inconvenience or preservation, toxicities, and pungencies in intact state. Various drying conditions were examined to establish the physicochemical properties of Chunma by the response surface methodology (RSM). Weter activity and total phenolic acid contents of Chunma dried extracts were optimal at $79.56^{\circ}C$ and $79.64^{\circ}C$ for 7.17 hr and 8.74 hr, respectively, for their characteristics. Electron-donating ability and nitrite-scavenging activity were optimal at 79.79 and $7.99^{\circ}C$ for 8.57 and 8.07 hr. We generated a regression equation for each variable and then superimposed the optimal condition in acquiring values for each frying process factor, physicochemical properties, resulting in that the optimal drying condition was at $75{\sim}80^{\circ}C$ for $8{\sim}9 hr$.

  • Gastrodia elata Blume (Chunma) belongs to Orchidaceae, which is a perennial parasitic herbaceous plant and grows in the woods of the central provinces of China, Korea and Japan. Recently, the constituents of the tubers of this plant have been investigated by researchers who have revealed the presence of phenolic compounds including gastrodin as a major constituent, together with 4-hydroxybenzaldehyde, 4-hydroxybenzyl alcohol, parishin, 4,4-dihydroxybenzyl sulfoxide, vanillin, vanillyl alcohol, beta-sitosterol, organic acids and polysaccharides, etc. In this study, we used ultrasonicator and two kinds of enzymes for enhancement of extraction yield. We also used electronic nose for the aroma pattern analysis of Chunma extracts. The concentrations of glucose and functional constituents (gastrodin, vanillin, 4-hydroxybenzaldehyde and 4-hydroxybenzyl alcohol) were measured by biochemistry analyzer and HPLC, respectively. Therefore, we showed that the yield of extraction was increased and discomfortable odor was reduced.

Studies on the Effect of AntiConvulsion and Analgesic by Ukgansan and Ukgansangami (억간산(抑肝散)과 억간산가미방(抑肝散加味方)의 진경(鎭痙) 및 진통(鎭痛) 효과(效果)에 관(關)한 실험적(實驗的) 연구(硏究))

  • In order to prove the effectiveness for anticonvulsion and analgesic of Ukgansan, Ukgansan added Jinphi Banha and Ukgansan added Chunma by experiment, experimental animals(mouse) are injected with strychnine, picrotoxin and caffeine to cause convulsion and ovserved the consumed time from convulsion to death. Comparing data with control group and observation data for frequency of writhing syndrome caused by acetic acid and phenylquinone show the results as follows : 1. Anticonvulsion effect on the convulsion induced by strychnine it was significantly effective in all sample groups. 2. Anticonvulsion effect on the convulsion induced by picrotoxin it was significantly effective in sample A and B. 3. Anticonvulsion effect on the convulsion induced by caffeine it was not recognized in all sample groups. 4. Analgesic effect on the pain induced by acetic acid it was significantly effect in sample A and C. 5. Analgesic effect on the pain induced by phenylquinone it was significantly effective in sample A and C. The results show that Ukgansan can be an effectual cure on anticonvulsion and analgrsic, Ukgansan added Jinphi·Banha on anticonvulsion, and Ukgansan added Chunma on analgesic.

Vasodilation of BCT is Associated with Inhibition of PKC$\alpha$ Translocation and LC20 Phosphorylation

  • We have previously reported that the vasodilatory effect of BanhabackchuIChunma-tang(半夏白朮天麻湯; BCT), a herbal formula, and its mechanism might be associated with at least in part NO pathway. In the present study, we studied the influence of BanhabackchulChenuma-tang (BCT) on the phosphorylation of LC20, in parallel, the distribution of α-protein kinase C(PKCα) by phenylephrine was monitored using laser scan confocal immunofluorescent microscopy in freshly isolated ferret portal vein smooth muscle living single cells. Phenylephrine stimulation induced LC20 phosphorylation and translocation of PKCα. However, BCT dephosphorylated LC20 phosphorylation and inhibited the translocation of PKCα. Our results demonstrate that the mechanism of relaxant effect of BanhabackchulChunma-tang inhibition is associated with inhibition of PKCα activation and LC20 phosphorylation.

  • A method of dynamic analysis is developed for torsional free vibrations of elliptical-box girder type or stiffening truss system suspension bridge. In this study, the method based on a finite element technique using a digital computer, is illustrated by two numerical examples, the Namhae Bridge which is located in Kyungsang nam-do opened on June, 1973, and the Mt. Chunma Bridge is simple span pedestrian's suspension bridge with lateral bracing system in Mt. Chunma, Kyungki-do, are used. In general, dynamic modes of complex suspension bridges are three-dimensional in form, i.e., coupling between vertical and torsional motions. However, introduced that amplitudes of oscillation are infinitesimal for coincidence with the purpose of it's use, thereupon, the torsional vibration analyses are treated without coupling terms. A sufficient number of natural frequencies and mode shapes for torsional free vibration are presented in this paper. In the case of Mt. Chunma Bridge, the natural frequencies and periods are computed with and without reinforcement, respectively, and compared their discrepancies. The influence of the auxiliary reinforcing cables is prevailing in the first few modes, namely, 1st and 2nd in symmetric and 1st, 2nd and 3rd in antisymmetric vibration, and conspicuous in the symmetric compared with the antisymmetric motion, but in the higher modes, this kind of simple accessory elucidates rether converse effects. In the Namhae Bridge, the results are compared with the Manual's obtained by wind tunnel test. It reveals commendable agreement.

Effects of BanhabackchulChunma-tang on the Vascular Relaxation and its mechanism (혈관 긴장성 조절에 미치는 반하백출천마탕)

  • Hypertension is not only a well-established cardiovascular risk factor but also increase the risk of atherosclerosis. Most studies conducted to investigate the effectiveness of treatment for cardiovascular disease such as hypertension have focused primarily on conventional drug and physiotherapeutic treatments. BanhabackchulChunma-tang(半夏白朮天麻湯:BCT) is popular herbal medicine used in clinic for the treatment of various symptoms of drulatory disorders and weakness of digestive system, including anorexa and nausea with vertigo, severe headache, vomiting and so on. However, the mechanisms underlying its efficacy are unknown. This study investigated the effects of BCT as an alternative medication on the contraction induced by phenylephrine and KCI in rat thoratic aorta. BCT revealed siginificant relaxation on phenylephrine-induced arterial contraction, but revealed noncompetitive effect on concentration responses of phenylephrine-induced contraction. Treatment of N-L/sup ω/ -argine methyl ester(L-NAME) and methylene blue(MB)(10/sup -5/M) reduced the relaxation of BCT. BCT also increased in vitro NO production. It suggest that the relaxation effect of BBT is related with NO pathway, becausse the effect of L-NAME and MB are due to inhibition of NO synthesis from endothelial cells. These results indicate that BCT would be effective in hypertension treatment and its mechanism of relaxtion on arterial contraction is likely to be related with NO production, blocking of α-receptor and signal transduction after receptor activation.

Quality Characteristics of Chunma (Gastrodia elata Blume) Jelly with Added Gastrodia elata Blume Concentrate (천마 농축액 첨가량에 따른 젤리의 품질 특성)

  • The purpose of this study was to investigate the effect of Gastrodia elata Blume concentrate on quality characteristics of Gastrodia elata Blume jelly. The Gastrodia elata Blume jelly samples were divided into five groups by adding different Gastrodia elata Blume concentrates. The five groups were divided into GBJ-0, GBJ-5, GBJ-10, GBJ-15, and GBJ-20 (Gastrodia elata Blume jelly with 0, 5, 10, 15, and 20% Gastrodia elata Blume concentrate). The highest contents of crude protein and crude ash, and the brix in the GBJ-20 sample were 0.34 and 0.59%, and 22.28$^{\circ}$ Brix, respectively. The crude fiber and pH change (4.65-4.76) were not significantly observed in any of the GBJ samples. Turbidity, degree of brownness, and total phenolics increased with an increasing the amount of extract. A significant difference was observed between samples with and without the extracts in terms of hardness, springiness, gumminess, and chewiness, as well as K, Mg, Cu, Mn, and Zn contents. Hardness, gumminess, and chewiness increased in all GBJ samples with an increase the amount of extract added, whereas adhesiveness decreased. The highest tartaric acid content in the GBJ-0 sample and malic acid in the GBJ-20 jelly sample were 15.58 mg/100 g and 137.45 mg/100 g, respectively. The levels of Ca, K, and Mg increased with an increase in the amount of added extract. The highest score for overall acceptance (4.13) was in the GBJ-10 sample with 10% Gastrodia elata Blume concentrate.

  • A hydraulic quick coupler is a component used to easily connect or disconnect pipes or hoses that transfer high pressure fluid without leakage in various mechanical devices. In this study, to obtain an optimal design of a 3/8" hydraulic quick coupler, the effect of different shapes of the internal piston on the internal flow characteristics of the coupler was analyzed and evaluated through numerical analysis based on computational fluid dynamics. Subsequently, the optimal shape design of the internal piston was obtained by comparing the flow characteristics results such as velocity distribution, temperature distribution, and the pressure drop of the hydraulic quick coupler.
