Hematological and serum biochemical studies in fresh water fish exposed to acute and chronic copper and mercury toxicity

  • A total number of 668 apparently healthy fish were obtained from farm to study the effect of two heavy metals in a form of (Copper sulfate and Mercuric chloride) on some hematological and biochemical parameters of blood. The $LC_{50}$ /96 hr. of Cu and Hg were estimated and fish exposed to $\text\tiny{^1/_2}$ $LC_{50}$ for 7 days and for $1/_{10}$ $LC_{50}$ for 8 weeks from each product separately. Results showed decrease in RBCs count, PCV% and Hb in acute and chronic mercury while a significant increase was shown in acute and chronic copper toxicity, total leucocytic count showed decrease in acute mercury toxicity and increase in the chronic case, while in copper toxicity non-significant decrease in acute and significant decrease in chronic toxicity was noticed. Elevated serum urea and creatinine in both acute and chronic mercury and copper toxicity was detected. No changes in total bilirubin in the acute mercury and chronic copper toxicity while significant increase in chronic mercury and acute copper. Elevation of serum AST and ALT in some days of acute toxicity of mercury and copper while in chronic mercury toxicity a significant elevation of both serums AST and ALT were detected .while in chronic copper toxicity serum AST was fluctuated and ALT showed no significant changes. CK study revealed significant decrease in acute mercury with fluctuation in the chronic toxicity while in copper toxicity it showed fluctuation in acute and significant decrease in chronic toxicity. Glucose value decreased in acute and chronic mercury toxicity while in copper toxicity it showed significant increase in the acute and increase followed by significant decrease in the chronic copper toxicity.

Toxicity studies on Trigonella foenum-graecum L. seeds used in spices and as a traditional remedy for diabetes

  • Acute (24 h) and chronic (90 days) oral toxicity studies on the ethanol extract of Trigonella foenumgraecum Leguminosae (L.) seeds were carried out. Acute dosages were 0.5, 1.0 and 3 g/kg while chronic dosage was 100 mg/kg per day of the extract. All morphological, biochemical, haematological and spermatogenic changes, in addition to mortality, body weight changes and any change in vital organs were recorded. Histopathological investigations were done on vital organs. Growth arrest in the treated animals was observed. The treated mice gained no significant weight during chronic treatment while there was a significant gain in body weight of the control group mice. Biochemical studies revealed a significant decrease in blood sugar levels of fenugreek treatment groups while haematological parameters remained comparable to the control. In the treatment, male group there was a significant decrease in weight of testes as compared to the control. There was a marginal weight gain in kidney weight of mice after chronic treatment as compared to the control. Fenugreek chronic treatment caused a highly significant spermatotoxic effects in male mice.

만성발암성 흡입독성시험 우선순위 물질 선정 연구 (Prioritization of Chemicals for Chronic/Carcinogenic Inhalation Testing)

  • Objectives: In this study, we seek to perform a priority selection for test substances for chronic inhalation toxicity studies, including acute and subchronic inhalation toxicity studies, which are to be performed after the construction of a chronic/carcinogenicity inhalation toxicity study facility and enactment of pertinent legislation. Methods: Through this study, qualitative and quantitative priority evaluation of test substances according to acute, subchronic and chronic categories were respectively performed and priorities were suggested by expert group review, redundancy and other methods. Meanwhile, a draft on test substance selection criteria, procedures and methods referring to the National Toxicology Program (NTP) system was proposed. Results: This study selected priorities for candidate substances for chronic inhalation toxicity studies to be conducted from 2016. Conclusions: In the future, by assessing in advance the toxicological effects of chemicals to which workers can be potentially exposed in the workplace via long-term inhalation, expected health disturbances among workers will be reduced and it is anticipated that occupational disease induced by chemicals will be effectively prevented.

물벼룩을 이용한 일부 염색폐수의 만성 수질독성 특성 연구 (Chronic Toxicities of Effluents from Dye Industry using Daphnia magna)

  • 본 연구를 통한 결론은 다음과 같다. 5개의 염색폐수배출업소에서 각각 3회씩 채취한 최종 방류수를 대상으로 한 생물독성시험 결과 미생물독성시험에서는 B시료와 E시료에서 독성이 관찰되었고 급성 물벼룩시험에서는 반수치사농도가 C시료에서만 관찰되어 염색폐수의 독성이 시험 생물종별로 다르게 나타날 수 있음을 제시하였다. 동일 생물종을 대상으로 실시한 급성 및 만성시험에서는 급성반수치사가 5개 방류수 중 C시료에서만 비교적 낮은 농도에서 관찰이 된 것에 비해 만성반수치사는 3개 방류수에서 나타났고 $EC_{50}$도 급성영향의 $1/2\~1/5$ 수준에서 결정되었다. 특히 만성노출에 의한 생식 및 성장에 미치는 영향은 치사에 미치는 영향농도보다 더 낮은 농도에서 나타나는 경향을 보이고 있다 따라서 방류수가 연속적으로 수계에 배출되는 특성을 고려하면 만성노출에 의한 영향을 평가하는 것이 중요한 의미를 갖는다. 한편 염색폐수의 경우 동일업종 또는 동일한 시료에서도 독성평가방법에 따라 서로 다른 정도의 독성이 관찰되는 등 독성발현이 매우 다양하게 나타났다. 그러므로 염색폐수가 수중생태계에 미치는 영향을 정확하게 파악하고 제어하기 위해서는 물고기나 조류 등 다른 영양단계에 있는 생물종에 미치는 영향 등의 연구가 요구된다.

Toxicity Studies on Peristrophe paniculata (Forssk) Brummitt;an Ayurveda Drug

  • Chronic oral toxicity studies (90 days) on aqueous and methanol extracts of the whole plant of Peristrophe paniculata (Forssk) Brummitt were carried out in Wistar rats. The dosage was 200 mg/kg/day, p.o. for both the extracts. All external morphological and biochemical changes, in addition to body weight and vital organ weights were recorded. During this investigation, no significant mortality was observed. The results showed that both the extracts were devoid of any toxicity at the dose level studied as compared to the control group.

담수산 물벼룩인 Moina macrocopa를 이용한 과불화화합물 PFOA와 PFOS의 독성평가 (Toxicity Assessment of PFOA and PFOS Using Freshwater Flea Hyalella azteca)

  • Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) are fully flurorinated organic compounds which are highly persistent in environment and accumulated in organism. These chemicals are released to the environment at their manufacture, during their use in industrial and consumer applications and from disposal after their use. The purpose of this study was to determine the effects of PFOA and PFOS on the freshwater flea (Moina macrocopa). Acute toxicity test and chronic toxicity test were performed for 2 days and 10 days, respectively. Acute toxicity was assessed on the basis of mortality, while chronic toxicity was assessed by fecundity as well as mortality. The acute toxicity studies on PFOA and PFOS showed that the values of $LC_{50}$ were $73.9\;mg/L\;and\;27.7\;mg/L$ respectively. In the chronic toxicity test, fecundity was reduced significantly at 24.1 mg/L of PFOA and 9.3 mg/L of PFOS, respectively. Conclusively, the results of this work suggest that Moina macrocopa could be a suitable model organism for screening and assessing of environmental pollutants in water.

Molinate의 물벼룩에 대한 급성 및 만성독성 연구 (The Acute and Chronic Toxicity Studies of Herbicide, Molinate to Waterfleas)

  • 우리나라 하천수 중 검출빈도가 높게 검출되고 있는 제초제 molinate의 담수 무척추동물에 대한 영향을 평가하고자 물벼룩에 대한 급 만성독성시험 및 다세대노출시험을 수행하였다. Molinate는 담수 무척추동물인 물벼룩 Daphnia magna 와 Moina macrocopa,에 대한 $EC_{50}$(48시간)이 각각 11.4와 8.3 mg/L, 번식독성 무영향농도(NOEC)가 2.5와 2.0 mg/L로서 종간 감수성에 있어 큰 차이를 보이지 않았으며 M. macrocopa에 대한 3세대노출시험의 NOEC은 0.16 mg/L이었다. 하천에서 molinate에 의한 물벼룩에 대한 위해 가능성은 낮은 것으로 판단된다.

  • Arsenic is a unique element with distinct physical characteristics and toxicity whose importance in public health is well recognized. The toxicity of arsenic varies across its different forms. While the carcinogenicity of arsenic has been confirmed, the mechanisms behind the diseases occurring after acute or chronic exposure to arsenic are not well understood. Inorganic arsenic has been confirmed as a human carcinogen that can induce skin, lung, and bladder cancer. There are also reports of its significant association to liver, prostate, and bladder cancer. Recent studies have also suggested a relationship with diabetes, neurological effects, cardiac disorders, and reproductive organs, but further studies are required to confirm these associations. The majority of research to date has examined cancer incidence after a high exposure to high concentrations of arsenic. However, numerous studies have reported various health effects caused by chronic exposure to low concentrations of arsenic. An assessment of the health effects to arsenic exposure has never been performed in the South Korean population; thus, objective estimates of exposure levels are needed. Data should be collected on the biological exposure level for the total arsenic concentration, and individual arsenic concentration by species. In South Korea, we believe that biological exposure assessment should be the first step, followed by regular health effect assessments.

단일 한약 복합 처방의 장기간 연용 투여가 신기능에 미치는 영향에 대한 전향적 연구 (A Study Of Effects on Renal Function from Continuous Long-Term Herbal Medication)

  • Background : Due to increased interest in herbal medicines is recent years, medical circles have made studies of toxicity and side effects of herbal medicines. Particularly the kidney is sensitive to toxicity. A few reports concerning the side effects and toxicity of herbal medicine have been presented recently. This has bought on some distrust in herbal medicines among patients and western doctors. Objectives : The purpose of this study is to determine what effects long-term prescription of one herbal medicine may have on renal function. Methods : Nineteen patients took herbal medicine for eight weeks. Tests of their Blood Urea Nitrogen(BUN), Creatinine of blood plasma, and urine (chemical and microscopic) were taken before taking medicine and at the 2nd, 4th, 8th weeks. Results : After taking a herbal medicine, BUN and Creatinine decreased significantly or remained the same in comparison with the prior interval. Chemical and microscopic examination of urine showed no changes. Conclusions : The results suggest that taking this herbal medicine for a long time does not induce nephrotixicity. Further study is needed for investigating safety and toxicity of herbal medicines.

The Chronic and Acute Toxicity of Traditional Medicines Containing Terminalia chebula

  • Objectives: Terminalia chebula, the main ingredient of Altan Arur 5, has been used for many years in traditional medicine. This medicine is more effective than other drugs and is used to treat chronic gastritis and gastrointestinal disorders such as peptic ulcers and esophageal reflux. Other ingredients of Altan Arur 5 are Punica granatum (pomegranate), tulip seeds, black balm, and excreta of Trogopterus xanthipes. The main ingredients of T. chebula are antibacterial and analgesic in traditional medicine. Despite having been used for many years and although many studies have been conducted on the beneficial effects of this medicine and its ingredients, the toxicity of Altan Arur 5 has not yet been elucidated. Therefore, we aimed to study the toxicity of Altan Arur 5 to ensure that it is safe to use. Methods: Acute and chronic toxicity of Altan Arur 5 were assessed in 10 Kunming mice and 8 Sprague-Dawley rats, respectively, in different doses. In the acute toxicity study, Altan Arur 5 was orally administered to Kunming mice in doses of 12 g/kg, 24 g/kg, and 48 g/kg for 14 days. In the chronic toxicity study, it was orally administered to Sprague-Dawley rats in doses of 1.25 g/kg, 2.5 g/kg, and 5 g/kg for 12 weeks. Results: No significant differences were observed in the relative organ weights for mice treated with Altan Arur 5 compared with those in the control group. Furthermore, no macro- or microstructural changes were noted in the organs of any group. Conclusion: Our toxicity testing revealed that the traditional medicine Altan Arur 5 has no toxic effects in vivo.
