In Escherichia coli, L-lysine biosynthetic pathway is composed of nine enzymatic reactions. It has been well established that most of the lysine biosynthetic genes are regulated by the lysine availability, even though they are all scattered around the chromosome without forming any multigenic operon structure. However, no transcriptional regulatory mechanism has been identified except for the activation of lysA gene by the LysR. In this study, changes in transcriptome profiles of wild type cells and lysR deletion mutant cells grown in the absence or presence of lysine were investigated by using DNA microarray technique. Microarray data analysis revealed three groups of genes whose expression varies depending on the availability of lysine or LysR or both. To further examine the regulatory patterns of lysine biosynthetic genes, lacZ operon fusions were constructed and their expression was measured under various conditions. Obtained results strongly suggest that there is an additional regulatory mechanism which senses the lysine availability and coordinates gene expression.
The 63-amino-acid-encoding afsR2 is a global antibiotics-stimulating regulatory gene identified from the chromosome of Streptomyces lividans. To dissect a putative functional domain in afsR2, several afsR2-derivative deletion constructs were generated and screened for the loss of actinorhodin-stimulating capability. The afsR2-derivative construct missing a 50-bp C-terminal region significantly lost its actinorhodin-stimulating capability in S. lividans. In addition, site-directed mutagenesis on amino acid positions of #57-#61 in a 50-bp C-terminal region, some of which are conserved among known Sigma 70 family proteins, significantly changed the AfsR2's activity. These results imply that the C-terminal region of AfsR2 is functionally important for antibiotics-stimulating capability and the regulatory mechanism might be somehow related to the sigma-like domain present in the C-terminal of AfsR2.
The RAD3 gene of Saccharomyces cerevisiae is required for excision repair and is essential for cell viability. The RAD3 encoded protein possesses a single stranded DNA-dependent ATPase and DNA and DNA-RNA helicase activities. To examine the extent of conservation of structure and function of a S. pombe RAD3 during eukaryotic evolution, the RAD3 homolog gene was isolated by screening of genomic DNA library. The isolated gene was designated as HRD3 (homolog of RAD3 gene). Southern blot analysis confirmed that S. pombe chromosome contains the same DNA as HRD3 gene and this gene exists as a single copy in S. pombe. The transcript of 2.8 kb was detected by Northern blot analysis, The level of transcripts increased by ultraviolet (UV) irradiation, indicating that HRD3 is one of the UV-inducible genes in S. pombe. Furthermore, the predicted partial sequence of HRD3 protein has 60% identity to S. cerevisiae RAD3 gene. This homology was particularly striking in the regions identified as being conserved in a group of DNA helicases. Gene deletion experiments indicate that the HRD3 gene is essential for viability and DNA repair function. These observations suggest evolutionary conservation of other protein components with which HRD3 might interact in mediating its DNA repair and viability functions.
This study was carried out to determine the antimutagenic effects of genistein on the somatic mutagenicity induced by aflatoxin B1 (${AFB}_1$), using Drosophila wing spot test system. Mutagen alone or mutagen with genistein were administered to the heterozygous(mwh/+) third instar larvae by feeding, and somatic cell mutations were detected in adult fly wing hairs. Genistein did not show any mutagenicity with the feeding concentrations of 5~15% in the test system. As the feeding concentrations of genistein increased, genistein inhibited the mutagenicity induced by AFB1 (14.6%~62.2% inhibition rate), while as the concentrations of AFB1 increased, small much spots that arise mostly from chromosome deletion and nondisjunction were more strongly suppressed by genistein than the large mwh spots from chromosomal recombination. In each group of different AFB1 concentrations, the rate of inhibition for total mwh spots was dependent on the dose of genistein. These results indicate that genistein have inhibitory effect on the mutagenicity induced by a mtagen, ${AFB}_1$. It seems to suggest that genistein may exert inhibitory effects to mutagenic and/or carcinogenic properties of DNA damaging agents.
Wolff-Hirschorn syndrome is a condition that is caused by a deletion of genetic material near the end of the short (p) arm of chromosome 4. The major features of this disorder include a characteristic facial appearance, delayed growth and development, intellectual disability, and seizures. A 9-year-old girl was brought to the clinic with a chief complaint of dental examination. The child was diagnosed as WHS at Samsung medical center. The child was under Sodium valproate, Atrovastatin medication for epilepsy, hyperlipidemia and had a history of heart surgery. So prophylactic antibiotics were recommended. The child was mentally retarded and had seizure so it was difficult to manage her behavior effectively. Thus dental treatment was carried out under general anesthesia. For prompt sedation induction we used 8% sevoflurane shortly. This report presents the case of a 9-year-old girl with WHS, who has received treatment for extensive caries under general anesthesia.
CATCH 22 syndrome is a one of the most common chromosome microdeletion syndrome with multiple organ anomalies in humans, with an incidence of approximately 1:4,000 to 1:5,000 live births. It is caused by a microdeletion of 1.5 to 3.0 megabases on the long arm of chromosome 22. The phenotypic spectrum of this disorder is wide and various. A 19-year-old patient who showed delayed growth and development (Height; 110 cm, Weight; 18 kg) was referred to our department for the treatment of dental cavities. She was diagnosed as CATCH 22 syndrome in 2004. Physical examination revealed hypertelorism, a short philtrum, thick reflected lips and a small mouth. She underwent cleft palate surgery at 1 year of age and heart valve surgery due to the cardiovascular abnormality at 13 years of age. Convulsive seizures had persisted until 5 years ago but are well controlled at present. Oral examination showed poor oral hygiene, crowding, prolonged retention on #65, 75 and dental cavities on #16, 21, 65, 26, 36, and 46. Cavity treatment and prophylaxis were performed under general anesthesia. Also continuous follow-up checks have been carrying out with the periodic prophylaxis and dental home education. Problems with numerous cavities and gingivitis which can lead to specific risks are common for CATCH 22 syndrome patients. It is therefore of great importance that these patients are referred to foremost physician and dental specialist for the oral care. In addition, preventive treatment targeting the risk of dental cavities and gingivitis is especially important and, as the syndrome involves many different medical problems, the dental treatment should be carried out in collaboration with the patient's physician.
Drosophila의 actin 5C 유전자 promoter에 쥐의 DNA polymerase $\beta$cDNA를 도입시킨 형질전환 초파리가 고감수성 환경성 변이원 검출계로 사용할 수 있는지를 조사하였다. 체세포 염색체 재조환과 체세포 염색체 돌연변이의 검출을 위해서는 geterozygous(mwh/+) 계통을 사용하였다. 염색체상의 결실이나 비분리 등에 의한 small mwh spot의 자연 발생적 빈도는 non-transgenic w 계통과 transgenic p[pol $\beta$]-130 계통에서 각각 0.351 및 0.606 정도였다. 체세포 염색체 재조환에 의한 large mwh spot의 자연 발생적 빈도의 경우는 transgenic p[pol $\beta$]-130 계통(0.063)이 non-transgenic w 계통(0.021)에 비해 약 3배 정도 높게 나타났다. IQ, Glu-P-1 및 {TEX}$AFB_{1}${/TEX} 등의 돌연변이원의 처리에 의한 경우, 두 종류의 mutant clone의 발생 빈도는 쥐의 DNA polymerase $\beta$가 도입된 transgenic p[pol $\beta$]-130 계통이 non-transgenic w 계통에 비하여 모두 약 2-3배 정도 높게 나타났다. 본 연구의 결과는 쥐의 DNA polymerase $\beta$가 최소한 체세포 염색체 돌연변이 유발이나 체세포 염색체 재조환의 생성 과정에 관여함을 의미하며, 형질전환 초파리 계통이 환경성 변이원 검출계로서 충분한 응용가능성이 있음을 보여 주었다.
목 적 : 염색체의 구조 및 유전을 연구하는 학문인 세포유전학은 임상 진단, 생식 문제, 산전 진단, 종양, 유전자의 다형성 및 유전 상담에 있어 중요한 역할을 하고 있다. 이 연구는 단일 기관에서 시행된 말초혈액을 이용한 세포유전학 검사 결과를 검토하여 주요 염색체 이상의 양상과 빈도를 분석하였다. 방 법 : 1981년 5월부터 2005년 10월까지 25년간 경북대학교병원 소아과 염색체 검사실로 각 임상 진료과에서 염색체 이상이 의심되어 의뢰한 말초혈액 검체 4,856례를 대상으로 하여 염색체 핵형을 분석하였다. 결 과 : 총 4,856례 가운데 4,567례를 분석하였다. 이 중 소아는 3,014례(66.0%), 성인은 1,553례(34.0%)였으며, 검사를 의뢰한 가장 흔한 이유는 소아에서는 성장과 발달 장애, 성인에서는 생식 문제였다. 4,567례 중 염색체 이상은 770례(16.9%)에서 발견되었다. 염색체 이상 중 수적 이상은 558례(12.2%), 구조적 이상은 187례(4.1%)였으며, 취약부위나 염색체 파손과 같은 이상이 25례(0.5%)였다. 수적 이상 중 상염색체 이상은 Down 증후군이 294례(6.4%)로 가장 많았으며, Edwards 증후군 7례(0.2 %), Patau 증후군 4례(0.1%) 순이었다. 성염색체의 이상은 Klinefelter 증후군이 131례(2.9%)로 가장 많았고, Turner 증후군 99례(2.2%), XXX 증후군 8례(0.2%), XYY 증후군 3례(0.1%) 순이었다. 구조적 이상은 전위가 84례(1.8%)로 가장 많았다. 결 론 : 본 연구에서 염색체 이상 핵형의 유형과 그 양상을 파악하였으며, 적극적 세포유전학적 연구로 진료와 유전상담에 적용하여야 할 것이다.
저자는 de Grouchy 증후군을 가진 4세 여자 환아에 대한 임상적 및 방사선학적 관찰을 통해 다음과 같은 지견을 얻었다. 1. 환아는 de Grouchy 증후군의 증상인 심장 및 발가락의 기형, 근긴장의 저하, 뇌량의 부분적인 무형성, 시각 장애와 청각 장애, 지적 능력의 저하, 성장 발달의 지연 등을 나타낸다. 2. 환아의 구강 및 두개악안면 소견으로 상악골 열성장과 전치부 반대교합을 보이며, 불량한 구강 위생으로 인한 다발성 치아우식증이 관찰된다. 3. 환아의 구강 위생 관리 및 치료 부위의 유지를 위하여 주기적인 치과 검진이 필요하며, 상악골 열성장과 전치부 반대교합을 개선하기 위해 추후 교정 치료가 필요할 수 있다.
모델 사상성 진균 Aspergillus nidulans는 분화과정을 연구하는 진핵세포 시스템으로 사용되어 왔다. 이러한 분화과정은 매우 다양한 유전자들의 발현을 통하여 조절되며 이와 관련된 다양한 전사요소들의 기능이 연구되어 왔다. 이들 중 forkhead 유전자는 일반적으로 감수분열 및 세포주기 조절에 중요한 역할을 하는 것으로 알려져 왔으며 A. nidulans에서도 유사한 기능을 하리라 예상되어 왔다. 이와 관련된 연구를 위하여 A. nidulans 유전체에 존재하는 6개의 forkhead 유전자를 발견, 확보하였고, 최근에는 효모 및 다른 진균에서는 발견되지 않는 A. nidulans 특이적 forkhead 유전자인 fkhF의 구조와 기능이 분석된 바 있다. 본 연구에서는 fkhF와 매우 유사한 단백질 서열을 가지고 있는 fkhE(AN2025.3) 유전자의 기능을 분석하였다. 본 유전자의 기능을 분석하기 위해 RT-PCR을 통하여 cDNA 서열을 분석한 결과 약 3종류의 서로 다른 mRNA가 존재하는 것이 밝혀졌고 이는 alternative splicing에 의한 것으로 추정되었다. 이들 3종류의 mRNA중 한 종류만 정상적인 ORF를 가지고 있으며 조사한 전체 cDNA 발현의 61%를 차지하였다. fkhE 유전자는 718개의 아미노산을 암호화하는 하나의 ORF를 가지고 있었으며 N 말단에 보존된 forkhead 도메인을 가지고 있었다. fkhE 유전자를 제거한 유전자 제거 돌연변이 균주는 fkhF와 유사하게 고체배지에서는 무성포자의 형성이 저해되었으나 유성분화에는 별다른 영향을 미치지 않았으며 액체 진탕배양에서는 야생형과 다르게 무성포자병(conidiophore)이 형성되었다. 이러한 결과는 fkhE 유전자가 무성분화에 관련되었음을 보여준다.
