Development and Immunochemical Properties of Two Monoclonal Antibodies Specific to Human Chorionic Gonadotropin

  • Using a hybridoma technique, spleen cells of Balb/c mice immunized with human chorionic gonadotropin (hCG) were fused with NS-1 mouse myeloma cells. Two hybrid cell lines, clones KS-8 and KS-19, secreting monoclonal antibodies to hCG, were isolated. KS-8 and KS-19 belong to the immunoglobulin $G_1$ subclass. With the aid of a double-antibody radioimmunoassay, it was established that the KS-8 monoclonal antibody recognizes an immunodeterminant of the $\beta$-subunit of hCG, whereas the KS-19 monoclonal antibody recognizes an epitope present on the $\alpha$-subunit of hCG. The KS-8 monoclonal antibody specifically reacts with human chorionic gonadotropin and shows cross-reactivity of less than 0.3% to other related human glycoprotein hormones. On the other hand, using a hemagglutination test based on antibody-induced agglutination of sheep red blood cells coated with hCG, It was shown that only the KS-19 monoclonal antibody was capable of inducing a positive reaction, although both monoclonal antibodies had similar binding capacity to the coated cells. The results from the dual screening procedures demonstrate that KS-8 and KS-19 monoclonal antibodies show high sensitivity in two different assays, and are hence useful for the qualitative and quantitative determination of hCG by both radioimmunoassay and hemagglutination inhibition tests.

Nitric Oxide Generation from Peritoneal Macrophages by Human Chorionic Gonadotropin (사람 융모 성선 자극 호르몬에 의한 복강 대식세로로부터 산화질소의 발생)

  • Human chorionic gonadotropin (hCG) is a placental hormone and is involved in maintenance of the corpus luteum during pregnancy. In the present study, effect of hCG on nitiric ox ide (NO) generation from peritoneal macrophage was examined. hCG ahd no effect on NO generation by itself, whereas recombinant interferon- ${\gamma}$ (rIFN-${\gamma}$) alone had modest activity. When hCG was used in combination with rIFN-${\gamma}$, there was a marked cooperative induction of NO generation in a dose-dependent manner. The optimal effect of hCG on NO generation was shown at 6 hr after treatment with rIFN-${\gamma}$. Furthermore, northern blot analysis of showed that hCG increased the expression of inducible NO synthase(iNOS) gene. These results suggest that hCG induces NO generation from macrophages by increasing the expression of iNOS gene.

  • Experiments were carried out examine short-term effects of human chorionic gonadotropin (hCG) and long-term effects of luteinising hormone releasing hormone analogue (LHRHa) on milt production of the tongue sole Cynoglossus semilaevis. In the first experiment, each fish was implanted with a blank cholesterol pellet (control), 100 and $200{\mu}g$ LHRHa per kg body weight. In the second experiment, fish were injected with either 100, 200, 400 and 800 IU hCG per kg body weight or same volume of marine fish Ringer's solution. In the first experiment, milt volume was increased in male implanted with $200{\mu}g$ LHRHa pellet compared with other groups at day 10. Injection of 400 and 800 IU hCG resulted in an increase in the milt volume at hour 96 after the treatment. Although statistical difference is unable to confirm because of small milt volume, compared with the control group, hormone pellet-treated groups had a reduction in the mean spermatocrit (Sct) and sperm concentration (Sc). The results suggest that the increase in milt volume is at least partially gonadotropin (GtH)-dependent and increased milt volume has a relationship with milt hydration.

Artificial induction of spawning by human chorionic gonadotropin and carp pituitary extract in marine medaka, Oryzias dancena (인간융모성생식선자극 호르몬과 잉어 뇌하수체에 의한 해산송사리, Oryzias dancena 인공산란 유도)

  • The method of natural spawning is very passive and inconvenient for the study of developmental engineering in marine medaka, Oryzias dancena. The optimum concentration of human chorionic gonadotropin (HCG) and carp pituitary extract (CPE) for ovulation and spawning, and the injection time for the artificial spawning of marine medaka were analyzed in this study. The success rate, survival rate, and hatching rate were highest with 100IU HCG kg-1 BW and 5mg CPE L-1 in both male and female marine medaka (p<0.05). After obtaining unfertilized eggs and sperm by the injection of HCG and CPE into the broodstock of marine medaka, artificial fertilization could be successfully achieved any time fertilized eggs are needed in this species. This result should be useful for developing a study program for marine medaka as an experimental animal.

The relationship between the variants in the 5'-untranslated regions of equine chorionic gonadotropin genes and serum equine chorionic gonadotropin levels

  • Objective: An experiment was conducted to study the association between the single nucleotide polymorphisms (SNPs) in 5'-untranslated regions (5'-UTR) of equine chorionic gonadotropin (eCG) genes and the serum eCG levels. Methods: SNPs in 5'-UTR of eCG genes were screened across 10 horse breeds, including 7 Chinese indigenous breeds and 3 imported breeds using iPLEX chemistry, and the association between the serum eCG levels of 174 pregnant Da'an mares and their serum eCG levels (determined with ELISA) was analyzed. Results: Four SNPs were identified in the 5'-UTR of the $eCG{\alpha}$ gene, and one of them was unique in the indigenous breeds. There were 2 SNPs detected at the 5' end of the $eCG{\beta}$ subunit gene, and one of them was only found in the Chinese breeds. The SNP g.39948246T>C at the 5'-UTR of $eCG{\alpha}$ was associated significantly with eCG levels of 75-day pregnant mare serum (p<0.05) in Da'an mares. Prediction analysis on binding sites of transcription factors showed that the g.39948246T>C mutation causes appearance of the specific binding site of hepatocyte nuclear factor 3 forkhead homolog 2 (HFH-2), which is a transcriptional repressor belonging to the forkhead protein family of transcription factors. Conclusion: The SNP g.39948246T>C at the 5'-UTR of $eCG{\alpha}$ is associated with eCG levels of 75-day pregnant mare serum (p<0.05).

Induction of Artificial Spawning by Human Chorionic Gonadotropin in the Grass Puffer, Takifugu niphobles (태반성 성선 자극 호르몬 처리에 의한 복섬, Takifugu niphobles의 인공 산란유도)

  • Effects of human chorionic gonadotropin (HCG) on ovulation and spawning of the grass puffer, Takifugu niphobles, were investigated. Matured females spawned successfully by all doses of HCG. Spawning usually occurred 24 hours after hormone injection. Body weight of adult, gonadsomatic index (GSI), pseudo-GSI, body weight of spawned egg, success rate of spawn, fertilization rate, hatching rate, and survival rate of juvenile were correlated with increasing HCG doses. However, abnormal rate of juvenile was not correlated with increasing HCG doses. These results should be useful for developing aquaculture program of grass puffer.

  • Equine chorionic gonadotropin (eCG), which consists of highly glycosylated α- and β-subunits, is a unique member of the gonadotropin family because it elicits response characteristics of both follicle-stimulating hormone (FSH) and luteinizing hormone (LH) in other species than the horse. To determine whether α and β subunits can be synthesized as a single polypeptide chain (tethered-eCG) and also display biological activity, the tethered-eCG molecule was constructed and transfected into Chinese hamster ovary (CHO-K1) cells.   • Human chorionic gonadotropin hormone(H.C.G.) is secreted from the villus tissues of the placenta and excreted in lange amount into the urine. Its isolation is chiefly made from the urine of a pregnant woman. Recently, Matsushima attempted isolation of H.C.G. directly from the placenta itself. In order to prepare H.C.G. from human placenta, general method of extractiag and purifying proteins was applied. Its way was as follow: Crude H.C.G. was extracted from placenta with pH 9.0 and pH 5.0 aqua ammonia, and purified with pH 8.0 ammonia and 50% ethanol at pH 4.8. The purified H.C.G. showed two moving bands on the anode by paper electrophoresis. On the other hand, the H.C.G. from pregnancy urine (Standard. Pharm. Co.) showed same two bands but their moving ratio were different. The purified H.C.G. showed gonadotropin effect when it was injected young fomale rats 40r/cc per day for 5 days and weighted the increased ovary weight.
