Influence of the Ceramide(III) and Cholesterol on the Structure of a Non-hydrous Phospholipid-based Lamellar Liquid Crystal : Structural and Thermal Transition Behaviors

  • The effects of the creamide III (CER3) and cholesterol (CHOL) on the structure of a non-hydrous distearoyl phosphatidylcholine (DSPC)-based lamellar liquid crystal (LC) hydrated by only propylene glycol (PG) without water were investigated by differential scanning calorimetry (DSC), X-ray diffractions (XRDs), and polarized microscope (PM). As soon as CER3 was incorporated into the lamellar phase, the characteristic LPP was appeared as well as the characteristic SPP, and the formation of separated CER3 crystalline phase was observed depending upon the increase of CER3 content by XRDs. Also, by DSC, it was shown that the increase of CER3 made the monotectic thermal transition be changed to the eutectic thermal transition which indicates the formation of separated CER3 crystalline phases and the main transition temperatures (Tc1) to be gradually decreased and the enthalpy change (ΔH) to be linearly increased. Incorporating CHOL, the formation of LPP and SPP showed almost similar behaviors to CER3, but incorporating small amounts of CHOL showed the characteristic peaks of CHOL which meant the existence of crystalline CHOL phase due to the immiscibility of CHOL with DSPC swollen by PG differently from CER3, and increasing CHOL made the intensity of the 1st order diffraction for LPP weakened as well as the intensities of the characteristic diffractions for DSPC. Also, in the results of DSC, it showed more complex thermal behaviors having several Tc than CER3 due to its bulky chemical structure. In the present study, the inducement of CER3 and CHOL as other lipids present in human stratum corneum (SC) into a non-hydrous lamellar phase is discussed in terms of the influence on their structural and thermal transition.

콜레스테롤 식이로 사육한 흰쥐의 혈청 및 간지질 성부에 미치는 카페인 및 비타민 E의 영향 (Effect of Caffeine and Vitamin E on the Lipid Composition of serum and Liver in Cholesterol-fed Rats)

  • 카페인과 비타민 E가 체내 지질대사에 미치는 영향을 검토코저, 콜레스테롤 첨가 식이에 카페인(0.25%), 비타민 E (400mg/kg diet) 및 카페인+비타민 E를 각 군에 따라 급여하고 6 주간 사육한 후, 흰쥐의 성장, 각종 장기중량, 혈청 및 간중의 지질성분을 측정하고 아울러 간조직검사를 행한 결과는 다음과 같다. 체중증가량은 CF군만이 유의하게 낮았다. 간 무게는 Basal+Chol.군에 비해 전 실험군에서 유의적인 차이가 없었으며, 신장의 경우 CF군과 CF+VE군에서 유의하게 증가하였다. 혈청중의 총지질은 VE군과 CF+CE군에서 감소하였다. 총 콜레스테롤 및 HDL-Cholesterol은 CF 군에서 증가하였으며, 트리글리세리드(TG)는 Basal+Chol. 군에 비 해 CF군, VE군 및 CF+VE 군에서 유의하게 낮았다. 간중의 조지질은 VE군, CF+VE군에서 감소하였고, TG함량은 VE군, CF+VE군 및 CF군에서 감소하였다. 한편 간조직검사는 Basal+Chol.군이 미세한 과립상의 지방소적으로 팽창된 데 비해 VE군 및 CF+VE군에서 비교적 호전되었으며, CF군의 경우 지방소적의 증가와 동양혈관의 확장이 관찰되었다.

뇌졸중 위험지표로서의 혈중 지질에 대한 환자;대조군 연구 (A Case-Control Study on Blood Lipids as a Risk Factor of Stroke)

  • Background and purpose : Stroke is one of the most deadly diseases and a leading cause of disability. Lipids confer an increased risk for cardiovascular morbidity and mortality. In spite of many studies on the relationship between stroke and lipids, their relationship is not clear. This study was undertaken to determine whether lipids were associated with stroke. Method : We compared the components of blood lipids between stroke patients group (n=217,Cases), non-stroke control group (n=160, Controls) and healthy control group without hypertension, diabetes mellitus, ischemic heart disease, or hyperlipidemia (n=140, Normals). These data were statically analyzed by general linear models and binary logistic regression analysis to get each adjusted odds ratio. Result : The results were as follows. The blood levels of total cholesterol (T-Chol), triglyceride (TG) and low density lipoprotein cholesterol (LDL-Chol) were significantly higher in patients of ischemic stroke, while the blood levels of T-Chol and LDL-Chol were significantly lower, and the blood levels of TG significantly higher in patients of hemorrhagic stroke. The blood level of high density lipoprotein cholesterol (HDL-Chol) was significantly lower in all cases. Conclusion : These results suggest that high TG and low HDL-Chol may be risk factors of hemorrhagic stroke and ischemic stroke, high T-Chol and LDL-Chol may be risk factors of ischemic stroke, and low T-Chol and LDL-Chol may be risk factors of hemorrhagic stroke in Koreans.

Tocopherol 투여가 사람의 HDL Subfractions 와 Lipoprotein Pattern에 미치는 영향 (Effect of Tocopherol Supplementation on Serum HDL Subfractions and Lipoprotein Pattern in College Women)

  • Twenty three healty women on self - selected diet were given 800IU of tocopherol daily for 4 weeks. The levels of tocopherol in plasma and red blood cells( RBC ) and total choles terol, TG, HDL- chol, HDL subfractions and lipoprotein pattern in serum were determined pre-and postregimen at 2 and 4 weeks. No significant change was noted in VLDL, LDL, HDL fraction and LDL/HDL ratio separated by electrophoresis, even though HDL fraction was decreased at 2 wk but slightly increased at 4 wk. There were also no significant changes in the relative amount of HDL-chol and VLDL-chol when cholesterol content of each lipoprotein fraction was assayed. A transient increase in LDL-chol was observed at 2 wk but returned to the pretreatment level. Plasma and RBC tocopherol levels were significantly ( p ^lt;0.05 ) increased and decreased respectively, at both 2 and 4 wk, and LDL-chol was positively correlated to plasma tocopherol level ( p<0.05 ) but not to RBC tocopherol. However HDL-chol fractionated by heparin-Mn was increased at 2 and 4 wk by a significant increase in $HDL_{2}$-chol but no change in $HDL_{3}$-chol, which resulted in a significant reduction of $HDL_{3}$/HDL ratio and increase of $HDL_{2}/HDL$ and $HDL_{2}/HDL_{3}$ ratios. HDL- chol was negatively correlated to the levels of LDL-chol ( p<0.05), VLDL-chol (p<0.01), and T-chol/HDL ratio ( p<0.01 ). Serum TG was significantly decreased ( p <0.05 ) but total cholesterol was decreased only at 4 weeks.

Synthesis of 3β [L-Lysinamide-carbamoyl] Cholesterol Derivatives by Solid-Phase Method and Characteristics of Complexes with Antisense Oligodeoxynucleotides

  • In this report, we describe the synthesis of mono- and di-valent cationic $3\beta$ [L-Lysinamide-carbamoyl] cholesterol (K-Chol) derivatives by solid-phase peptide synthesis method and the characteristics of K-Chol/antisense oligodeoxynucleotide (ODN) complexes. K-Chol was able to interact with antisense ODNs electrostatically and constructed nanometer-sized complexes of 50-100 nm in diameter. The formation of K-Chol/antisense ODN complexes was demonstrated by non-denaturing polyacrylamide gel electrophoresis assay and atomic force microscopy. The cell-associated radioactivity was measured to monitor the cellular uptake of the complexes containing radioactive antisense ODNs using HL 60 cells.

  • This study was performed observe the effect of edible oil on the serum lipid of 0.5% cholesterol-fed rats. In this experiment, male rats of Sprague-Dawley strain were used. The rats were divided into 5groups which were fed differently eigher for 8 weeks : basal diet, 20% sunflower oil diet, 20% soybean oil diet, 20% rapeseed oil diet, 20% coconut oil diet. The followings are the results of this experiment. 1. The total chol., free chol., TG, PL level in the serum were showed tendency of decrease with in creasing of P/S ratios. 2. HDL-chol. level was increased with increasing of P/S ratios but LDL-chol. level was decreased. 3. GOT, GPT, TBA level in the serum were increased with increasing of P/S ratios.

Cholesteryl N-Monomethoxypoly(ethylene glycol)-succinate-L-phenylalanine: Synthesis and Effect on Liposomes

  • Poly(ethylene glycol)-phosphatidylethanolamine conjugate (PEG-PE) has been used in preparing longcirculating liposomes. As a substitute for PEG-PE which can also be used in the long-circulating liposome formualtions, but can be prepared more readily with a lower cost, PEG-Phe-Chol was synthesized from PEG, phenylalanine, and cholesterol. The addition of the PEG derivative to distearoylphosphatidylcholine (DSPC) led to the formation of mixed micelles as well as liposomes when the derivative content was 10 mol% or greater. On the other hand, the addition of just 5 mol% PEG-Phe-Chol to dioleoylphosphatidylethanolamine (DOPE) generated mixed micelles as well as liposomes, but the formation of mixed micelles was completely inhibited by the addition of cholesterol. The leakage of entrapped calcein out of DOPE/cholesterol (7/3) liposomes containing 5 mol% PEG-Phe-Chol was about 45% during the incubation time for 24 h in 50% rabbit plasma, which was similar to that of the same liposomes containing 5 mol% PEG-dipalmitoylphosphatidylethanolamine (DPPE) under the identical conditions.

Radical Scavenging and Antihypercholesterolemic Effects of Red Yeast Rice in Cholesterol Fed Rats

  • This study demonstrates that red yeast rice exhibits radical scavenging and antihypercholesterolemic activities in rats fed cholesterol. Sprague-Dawley male rats were divided to five dietary groups (normal, chol-control; and M-1, M-2 & M-3 administered 150, 300, and 500 mg/kg red yeast rice, respectively) and fed their respective diets for 4 weeks. No significant differences in food efficiency ratio (FER) were found among the five groups. The weight of perirenal fat pads decreased with increasing amounts of red yeast rice supplementation. There was a significant decrease in the levels of cholesterol in M-3 group fed red yeast rice with 500 mg/kg compared to those in the chol-control, M-1 and M-2 groups (p<0.05). Among the rats fed the cholesterol-enriched diet, all groups fed red yeast rice showed higher concentration of the HDL cholesterol, but lower concentration of the LDL cholesterol than those of the chol-control group. The scavenging activity of the methanol extract from red yeast rice was increased with increasing amounts of the extract. The glutathione content in the normal group and in the M-3 group were higher than that in the other groups. The M-3 group showed similar hepatic glutathione contents to those of the normal group. These results suggest that red yeast rice may be safe and effective for lowering serum levels of total and LDL-cholesterol, ratio of non-HDL/HDL, and severity of experimental atherosclerosis.

한국 중년 근로자의 영양소 섭취와 혈청 지질에 관한 연구 (A Study on Nutrients Intake and Serum Lipids Middle-aged Korean Workers)

  • This study aimed at investigating correlation of nutrients intake and serum lipids in middle-aged korean male and female workers. One hundred sixty eight(male ;105, female ;63) for adult aged 30-59yr(average age male ;36.5yr, female ;44.5yr) were selected as subjects during 2 months, from April to May, 1996. Nutrients intake status was investigated by questionnaire, 24-hour recall method. Antropometric assessments, serum lipids and blood pressure of the subjects were investigated. The results as follows: 1. Nutrients intake status ; male was superior to female. Vitamin A intakes of 40-49yr group were inferior to other group but amounts of intake were above Korean RDA(recommended dietary allowances), Intakes of calcium and vitamin C of 30-39yr and over 50yr group in female were below Korean RDA. In overweights group of male and underweight group of female, most nutrients intake were increased than other groups. 2. Antropometric assessments ; female was higher than male in BMI (body mass index). All subjects increasingly with age were increased in BMI. 3. Blood components were above normal range in triglyceride in both sexes and all subjects increasingly with age were increased In level of serum lipids and blood pressure, especially in male's forty and in female's fifty were higher than other groups In serum lipids and blood pressure. In overweights group, serum lipids and blood pressure had a significantly($\alpha$=0.05) increasing tendency in both sexes. 4. Serum lipids and connected factors; atherogenic index correlated positively with triglyceride, total-chol(total cholesterol), LDL-chol(low density-lipoprotein cholesterol) and negatively with HDL-chol(high-density-lipoprotein cholesterol) significantly(P<0.001). Systolic blood pressure correlated positively with total-chol and diastolic blood pressure correlated positively with total-chol, LDL-chol significantly(P<0.05). 5. Serum lipids and nutrients intake status: energy correlated positively(P<0.05) with BMI, dietary fiber correlated positively with LDL-chol(P<0.05), calcium correlated positively with triglyceride, atherogenic index and dietary fiber correlated negatively with BMI in male significantly(P<0.05). Triglyceride correlated positively with vitamin C and phosphorus correlated negatively with BMI in female significantly(P<0.05). Above results, in male's forty and in female's fifty were liable to variation in terms of serum lipids and blood pressure. As increasing triglyceride, total-chol, LDL-chol and decreasing HDL-chol Increased atherogenic index that suggest increasing risk of atherosclerosis. But, this point will be considered more subdivided study.

식이성(食餌性) 섬유소(纖維素)가 콜레스테롤 식이(食餌) 흰쥐의 혈청(血淸) 및 간장지질(肝臟脂質)에 미치는 영향(影響) (Effect of Dietary Fibers on Serum and Liver Lipids of Cholesterol-fed Rats)

  • 최근(最近) 식이성(食餌性) 섬유소(纖維素)가 인간(人間)의 질병발생(疾病發生)에 예방효과(豫防效果)가 있음이 밝혀진 바, 특히 동맥경화증(動脈硬化症)에 관련(關聯)한 cholesterol 저하효과(低下效果)를 조사(調査)하기 위하여 본(本) 실험(實驗)에서는 pectin, agar, pectin+tannic acid, tannic acid 각(各) 5%를 0.5% cholesterol 식이(食餌)에 첨가(添加)하여 Sprague-Dawley계(系) 흰쥐(male)에게 6 주간(週間) 급여(給與)하였다. 이에 흰쥐의 체중증가량(體重增加量), 소화흡수율(消化吸收率), 각종(各種) 장기(臟器) 중량(重量), 혈지(血漬)과 간장(肝臟) 및 분변(糞便)중의 지질(脂質) cholesterol함량(含量)을 측정(測定)한 결과(結果)는 다음과 같다. 1. 체중증가량(體重增加量)과 소화흡수율(消化吸收率)은 5% tannic acid 첨가군(添加群)만이 유의(有意)하게 낮았다. 2. 각종(各種) 장기(臟器)의 중량(重量)에서 간장(肝臟) 비장(脾臟)은 유의성(有意性)이 없었으며 신장(腎臟)은 PEC, AA군(群)에서 감소(減少)하였고, 심장(心臟)은 basal군(群), 폐장(肺臟)은 basal+chol 군(群)에서 유의(有意)하게 증가(增加) 하였다. 3. 혈청(血淸)중 GOT, GPT는 0.5% cholesterol 첨가군(添加群) (basal+chol.)만이 현저하게 높았으며 PEC 군(群)에서 유의(有意)하게 감소(減少)하였다. Total protein은 basal+chol. 만이 유의(有意)하게 높았으나 albumin, A/G ratio는 유의성(有意性)이 없었다. 4. 혈청(血淸)중 tolal lipid와 cholesterol 함량(含量)에는 유의성(有意性)이 나타나지 않은 반면, 간장(肝臟)중 조지질(粗脂質) 및 sterol 함량(含量)은 PEC 군(群)만이 유의(有意)하게 증가(增加)하였다.
