Photosynthetic Responses to Dehydration in Green Pepper(Capsicum annuum L.)Leaves

  • Photosynthetic responses to dehydration were examined by the simulataneous measurement of O2 evolution and chlorophyll (Chl) fluorescence in green pepper leaves. Dehydration was induced by immersing the plant roots directly in the Hoagland solution containing varying concentration (2-30%) of polyethylene glycol(PEG-6000) . Water potential of the leaf was decreased time-and concentation -dependently by PEG-treatment. The decrease in water potential of leaf was correlated with the decrease in both the maximal photosynthesis (Pmax) and quantum yield of O2 evolution, but Pmax dropped more rapidly than quantum yield at all water deficit conditions tested. However, Chl fluorescence parameters were not affected much. Dehydration did not change the initial fluorescence (Fo) and maximum photochemical efficiency(Fv/Fm) of photosystem(PS) II. Both the photochemical quenching (qP) and non-photochemical quenching(NPQ) were not changed by dehydration under low PFR(50 $\mu$mols m-2s-1 ). In contrast, under high PFR(270$\mu$mols m-2s-1)qP was slightly decreased while NPQ was greatly increased. The fast induction kinetics of Chl fluroecence showed no change in Chl fluorescence pattern by dehydration at high PFR (640 $\mu$mols m-2s-1 ), but exhibited a significant drop in peak level(Fp)at low PRFR (70$\mu$mols m-2s-1 ). PS I oxidation and reduction kinetics revealed normal reduction but delayed oxidation to P-700+, suggesting no lesionin electron flow from PSII to PSI , but impaired electron transport to NADP+,These results suggest that water stress caused by PEG-treatment results in the reduction of photosynthesis, promarily due to the reducted electron trasport from PSI to NADP+ or hampered subsequent steps involving Calvin Cycle.

Effect of Calcium Chloride($CaCl_2$) on Chlorophyll Fluorescence Image and Photosynthetic Apparatus in the Leaves of Prunus sargentii (염화칼슘 처리가 산벚나무 엽의 엽록소형광반응과 광합성기구에 미치는 영향)

  • There is a little information on the effect of calcium cloride ($CaCl_2$) which is used as deicing salt in Korea on the physiological responses of the street trees. Prunus sargentii is one of the most widespread tree species of street vegetation in Korea. In this study, the effect of $CaCl_2$ on photosynthetic apparatus such as chlorophyll fluorescence image and light response curve of P. sargentii in relation to their leaf and root collar growth responses were investigated. To study the effect of $CaCl_2$ treatment in the early spring, we irrigated twice in rhizosphere of P. sargentii (3-year-old) planted plastic pots with solution of 0.5%, 1.0%, 3.0% $CaCl_2$ concentration before leaf expansion. Results after treatments, total chlorophyll contents and the chlorophyll a/b, photosynthetic rate, quantum yield, dark respiration decreased with increasing $CaCl_2$ concentration. On the contrary, light compensation point increased with increasing $CaCl_2$ concentration. Through the linear regressions of correlation of photosynthetic rate with photosynthetic parameters (quantum yield, dark respiration and light compensation point), we found a significant relationship (p<0.05) between photosynthetic rate and quantum yield and light compensation point except dark respiration. Calcium cloride ($CaCl_2$) induced inhibition of photochemical efficiency ($F_v/F_M$) and non-photochemical quenching (NPQ) were found in treatments of $CaCl_2$, and these reduction rates between control and CaCl2 treatments were drastically showed at 80 days. We suggest that physiological activities are limited from treatment of $CaCl_2$. These reductions of photosynthetic apparatus ability caused eventually the reduction of leaf and diameter at root collar growth.

  • The study of wave propagation and scattering in biological media has become increasingly important in recent years. The propagation of light within tissues is an important problem that confronts the dosimetry of therapeutic laser delivery and the development of diagnostic spectroscopy. In the clinical application of photodynamic therapy(PDT) and in photobiology, the photon deposition within a tissue determines the spatial distribution of photochemical reactions. Scattered light is measured as a function of the distance (r) between the axis of the incident beam and the detection spot. Consequently, knowledge of the photosensitizer(Chlorophyll-a) function that characterizes a phantom is measured. To obtain the results of scattering coefficients(${\mu}s$) of a turbid material from diffusion described by experimental approach. It was measured the energy fluency of photon radiation at the position of penetration depth. From fluorescence experimental method obtained the analytical expression for the scattered light as the values of $(I/I_o)_{wavelength}$ vs the distance between the center of the incident beam and optical fiber in terms of the condition of "in situ spectroscopy(optically thick)" and real time by fluorometric measurements. The result was compromised with transport of intensities though a random distribution of scatters.

  • The Geostationary Ocean Color Imager (GOCI) will be loaded in Communication, Ocean and Meteorological Satellite (COMS). To efficiently apply the GOCI data in the variety of fields, it is essential to develop the standard algorithm for estimating the concentration of ocean environmental components (, , and ). For developing the empirical algorithm, about 300 water samples and in situ measurements were collected from sea water around the Korean peninsula from 1998 to 2006. Two kinds of chlorophyll algorithms are developed by using statistical regression and fluorescence technique considering the bio-optical properties in Case-II waters. The single band algorithm for is derived by relationship between Rrs (555) and in situ concentration. The CDOM is estimated by absorption coefficient and ratio of Rrs(412)/Rrs(555). These standard algorithms will be programmed as a module of GOCI Data Processing System (GDPS) until 2008.

Development of Ocean Environmental Algorithms for Geostationary Ocean Color Imager (GOCI) (정지궤도 해색탑재체(GOCI) 해수환경분석 알고리즘 개발)

  • Several ocean color algorithms have been developed for GOCI (Geostationary Ocean Color Imager) using in-situ bio-optical data sets. These data sets collected around the Korean Peninsula between 1998 and 2009 include chlorophyll-a concentration (Chl-a), suspended sediment concentration (SS), absorption coefficient of dissolved organic matter ($a_{dom}$), and remote sensing reflectance ($R_{rs}$) obtained from 1348 points. The GOCI Chl-a algorithm was developed using a 4-band remote sensing reflectance ratio that account for the influence of suspended sediment and dissolved organic matter. The GOCI Chl-a algorithm reproduced in-situ chlorophyll concentration better than the other algorithms. In the SeaWiFS images, this algorithm reduced an average error of 46 % in chlorophyll concentration retrieved by standard chlorophyll algorithms of SeaWiFS. For the GOCI SS algorithm, a single band was used (Ahn et al., 2001) instead of a band ratio that is commonly used in chlorophyll algorithms. The GOCI $a_{dom}$ algorithm was derived from the relationship between remote sensing reflectance band ratio ($R_{rs}(412)/R_{rs}(555)$) and $a_{dom}(\lambda)$). The GOCI Chl-a fluorescence and GOCI red tide algorithms were developed by Ahn and Shanmugam (2007) and Ahn and Shanmugam (2006), respectively. If the launch of GOCI in June 2010 is successful, then the developed algorithms will be analyzed in the GOCI CAL/VAL processes, and improved by incorporating more data sets of the ocean optical properties data that will be obtained from waters around the Korean Peninsula.

Effects of Trampling on Growth and Development in Zoysia japonica (답압이 한국잔디의 생육에 미치는 영향)

  • Trampling stress in turfgrass fields caused by traffics often occurs in zoysiagrass (Zoysia japonica) fields including golf course. In order to know the influences of trampling stress on the growth and development of turfgrass, leaf and root growth, chlorophyll fluorescence, chlorophyll content and 2-DE protein analysis were conducted in the turfgrass plants subjected to various levels of trampling stress from 0 to $9,420J\;day^{-1}$ day. Shoot growth was more highly inhibited by trampling stress than root growth. Although root growth was affected by trampling with weak intensity, the highest root length was observed in the turfgrass treated with weak trampling ($1,570J\;day^{-1}$). Chlorophyll fluorescence (Fv $Fm^{-1}$) was drastically lowered by trampling stress with moderate intensity. Leaf number showed similar tendency with leaf greenness. The number was decreased as the trampling intensity was increased. Shoot dry weight was decreased showing a similar tendency with the result of shoot length. The specific protein expressions under weak trampling were related to the functions of stress amelioration. Heat shock 70 kDa protein 10 and oxygen-evolving enhancer protein 1 were the proteins increased positively under trampling stress.

  • Deicing salt is used to melt snow and ice on the road for traffic safety during the winter season, which accumulates in the roadside vegetation and induces visible injuries. The damage may accelerate particularly when it coincides with early spring leaf out. In order to better understand the response mechanisms, C. kousa (3-year-old) was irrigated twice prior to leaf bud in a rhizosphere with solutions of 0.5, 1.0, and 3.0% calcium chloride ($CaCl_2$) concentration, that were made by using an industrial $CaCl_2$ reagent practical deicing material in Seoul. Physiological traits of the mature leaves were progressively reduced by $CaCl_2$ treatment, resulting in reductions of total chlorophyll contents, chlorophyll a:b, photosynthetic rate, quantum yield, stomatal conductance, $F_V/F_M$, and NPQ. On the contrary, light compensation point and dark respiration were increased at high $CaCl_2$ concentration. A decrease in intercellular $CO_2$ concentration by stomatal closure first resulted in a reduced photosynthetic rate and then was accompanied by low substance metabolic rates and photochemical damage. Based on the reduction of physiological activities at all treatments ($CaCl_2$ 0.5%, 1.0%, and 3.0%), C. kousa was determined as one of the sensitive species to $CaCl_2$.

Growth and Physiological Characteristics of Containerized Seedlings of Sageretia thea at Different Fertilization Treatments (시비처리에 따른 상동나무 용기묘의 생장 및 생리특성)

  • This study aims to optimize the appropriate concentration of fertilizers for Sageretia thea by analyzing growth performances (height and root collar diameter) and physiological characteristics (photosynthesis, chlorophyll contents, and chlorophyll fluorescence reaction). As fertilizer concentration was increased to 1.5 g·L-1, growth increased, but it decreased at 2.0 g·L-1 treatment. Root collar diameter growth was reduced because of higher fertilizer concentrations. Photosynthesis reactions showed the highest CO2 reaction curves, maximum photosynthesis rate, and maximum carboxylation rate in the 1.5 g·L-1 fertilizer treatment. The chlorophyll fluorescence reaction and SPAD values revealed that fertilizer treatment improves photosynthesis efficiency and robustness compared with untreated control. Therefore, the appropriate fertilizer concentration for producing good seedling quality of Sageretia thea is 1.0~1.5 g·L-1.

Different Susceptibilities to Low Temperature Photoinhibition in the Photosynthetic Apparatus Among three Cultivars of Cucumber (Cucumis sativus L.)

  • Susceptibility to low temperature photoinhibition in photosynthetic apparatus was compared among three cucumber cultivars, Gahachungjang (GH), Banbaekjijeo (BB) and Gaeryangsymji (GR). By chilling in the light for 6 h, a sustained decrease in the potential quantum yield (Fv/Fm) and the oxidizable P700 contents was observed, and the decrease was less in GH than in BB and GR. Although the difference was small, some $\Phi_{PSII}$ remained in GH after light-chilling for 6 h indicating that a few electrons can flow around photosystem II(PSII). As a consequence, the primary electron acceptor of PSII, $Q_{A}$, was reduced slowly and was not fully reduced after light-chilling for 6 h in GH. Although the amplitude was small, the development of NPQ was also faster in GH, indicating a higher capacity for non-photochemical energy dissipation. The relative fraction of a fast relaxing component of NPQ (qf) was higher in GH. After light-chilling for 5 h, the values of qf in BB and GR became much smaller than that in GH, indicating BB and GR suffered more significant uncoupling of ATPase and/or irreversible damages in PSII. When fluorescence induction transients were recorded after chilling, significant differences in quenching coefficients (qQ and qN) were observed among the three cultivars.
