Antioxidative Activity and Cytotoxicity on Human KB cell of Extracts from Rhododendron mucronulatum Turcz. Flower (진달래꽃 추출물의 항산화 효과 및 인체 KB cell에 대한 세포독성)

  • This study was carried out to investigate the antioxidative activity and cytotoxicity on human KB cell of extract from Rhododendron mucronulatum flower, and also to determine the contents of total polyphenol and flavonoid. The methanol extract of Rhododendron mucronulatum flower was fractionated with various solvents such as hexane, chloroform, ethyl acetate, butanol and water. The DPPH radical scavenging activities of ethyl acetate fraction had stronger activity than other fractions. So, antioxidative substances of ethyl acetate fraction were crude purified by silica gel column chromatography. The DPPH radical scavenging activities of crude purified fraction 2 and 3 were more than 90% at 40 ppm. In the presence of $100{\mu}g/mL$, growth inhibition on human KB cell by WST-1 assay showed 81.2% in chloroform fraction and 74.6% in hexane fraction. The contents of total polyphenol and flavonoid of ethyl acetate traction were 32.70% and 20.30%, respectively, The antioxidative activity showed correlation with total polyphenol and flavonoid contents.

  • Studies on the chemical constituents from a Korean wild mushroom, Phellinus ribis, were carried out. A triterpenoid, two peroxysterols, and a chlorobenzene compound were isolated from the hexane soluble fraction of the methanol extract of dried fruiting bodies of the basidomycetes. Those compounds identifed were 3-hydroxy-20(29)-lupen-28-oic acid (betulinic acid), 5,8-epidioxyergosta-6,22-dien-3-ol(ergosterol peroxide), 5,8-epidioxyergosta-6,9(11),22-trien-3-ol (dehydroperoxyergosterol), and 1,2,4,5-tetrachloro-3,6-dimethoxybenzene. Structural studies were carried out on molecular species of a ceramide and cerebroside isolated from the chloroform soluble fraction of the methanol extract. For ceramide, the major component fatty acids were a-hydroxy fatty acid isomers of $C_{22:00}{\sim}C_{25:00};$ the predominant long-chain bases were trihydroxy sphinganine of $C_{17}{\sim}C_{18}$. The structure of a cerebroside containing mono-sugar was assumed that the long-chain base was $C_{19:2}$ sphingadienine; the major fatty acids were $C_{16}{\sim}C_{15}$ ${\alpha}$-hydroxy fatty acid isomers.

Antioxidant and Antimicrobial Activity of Zostera marina L. Extract

  • Methanol crude extract of the sea grass Zostera marina L. and organic solvent fractions (n-hexane, chloroform, ethyl acetate, n-butanol, and water) were screened for antioxidant activity (total phenolic contents, DPPH scavenging activity, and reducing power) and antimicrobial activity against three human skin pathogens, two bacteria and a yeast; Staphylococcus aureus, Staphylococcus epidermidis, and Candida albicans. Total phenolic contents and 2, 2- diphenyl-1-picrylhydrazyl (DPPH) scavenging activity were highest in the ethyl acetate fraction with 968.50 $\mu$g gallic acid equivalent per milligram of extract, and ca. 95% scavenging activity on the DPPH radicals at 10 mg $ml^{-1}$. In antimicrobial activity tests, MICs (Minimum Inhibitory Concentration) of each Zostera marina extract partitioned ranged from 1mg to 8 mg $ml^{-1}$ (extract/ 10% DMSO) against all three human skin pathogens. The MICs of the ethyl acetate and n-butanol fractions were the same with 1 mg $ml^{-1}$ against S. aureus and C. albicans. The ethyl acetate fraction of Z. marina does protect against free radicals and may be used to inhibit the growth of human skin pathogens.

ACE, α-Glucosidase and Cancer Cell Growth Inhibitory Activities of Extracts and Fractions from Marine Microalgae, Nannochloropsis oculata (해양 미세조류 Nannochloropsis oculata 추출.분획물의 ACE, α-glucosidase 및 암세포 저해 활성)

  • Extracts of the marine microalgae Nannochloropsis oculata were obtained using 80% methanol (MeOH) and water. The 80% MeOH extract was further fractionated with n-hexane, chloroform, ethyl acetate (EtOAc), n-butanol (n-BuOH), and water to isolate the active fraction. Seven samples were prepared and their angiotensin converting enzyme (ACE), $\alpha$-glucosidase, and cancer cell growth inhibitory activities in vitro were determined. The most profound ACE inhibitory activity was observed in the chloroform fraction, while the others had moderate effects. By contrast, greater $\alpha$-glucosidase inhibitory activity was found in the EtOAc fraction, n-hexane fraction, and water extract of N. oculata. The antiproliferative effects of the extracts and fractions against HL-60, U937, CT-26, and SK-Hep1 cancer cells were also determined. The n-BuOH fraction had the strongest antiproliferative effects on CT-26 cells in a time-dependant manner (P<0.05). These results suggest that the extracts and fractions from N. oculata could be used as a potential functional food or as pharmaceutical ingredients.

Isolation and Identification of Antimicrobial Compound from Mokdan Bark (Paeonia suffruticosa ANDR) (목단피로부터 식품부패 미생물에 대한 항균성 물질의 분리 및 동정)

  • Antimicrobial activity of Mokdan bark (Paeonia suffruticosa $A_{NDR}$) was investigated. Methanol extract of dried Mokdan was fractionated to hexane, chloroform, ethylacetate, butanol and aqueous fraction. Ethylacetate fraction among these fractions showed the highest inhibitory effect on the microorganisms such as L. monocytogenes, and E. coli at 500 $\mu\textrm{g}$/disc. Ethylacetate fraction was further fractionated into 3 fractions by silica gel column and thin layer chromatography (TLC). The results showed that ethylacetate fractions No. 1 and 2 had the highest antimicrobial activity. They were mixed again, reseparated, and 3 fractions were obtained. Among them, No. 1 had the highest inhibitory effect on the microorganisms, No. 1 fraction was identified as isobutyl isopentanoate by HPLC, and GC-MS.

Antimicrobial Activities Against Oral Microbes and Growth-inhibitory Effect on Oral Tumor Cell by Extract of Paeonia lactiflora (작약 추출물의 구강병원균에 대한 항균성 및 구강암 세포 증식 억제효과)

  • Paeonia lactiflora was stepwise extracted with hexane, chloroform, ethyl acetate, butanol and water. Anti-microbial activity of each extract was investigated. Methanol extract of P. lactiflora revealed anti-microbial activity against S. mutans, C. albicans, and S. aureus. Also, hexane fraction revealed anti-bacterial activity against S. mutans and ethyl acetate fraction acted as potent anti-microbial agent on C. albicans and S. aureus. The relative growth ratio(RGR) of hexane fraction of P. lactiflora against S. mutans were determined as 77.8% in concentration of 0.125 mg/ml, 98.46% in 0.25 mg/ml and 100% in 0.5 mg/ml. The ethyl acetate fraction of P. lactiflora revealed RGR against C. albicans as 52.5% in concentration of 0.125 mg/ml, 60.83% in 0.25 mg/ml and 78.33% in 0.5 mg/ml. It indicate that increasing concentration increase RGR. The measured minimal inhibitory concentration(MIC) of hexane fraction on S. mutans KCTC 5316 strain was 0.5 mg/ml and MIC of ethyl acetate fraction on C. albicans KCTC 7270 was 2.0 mg/ml. The experiment of inhibition to growth of KB roll(oral squamous cell carcinoma) result 61.9% in butanol, 76.7% in hexane extract of P. lactiflora. The hexane extract exhibit potent inhibition effect to the growth of KB cell. These results suggest that the hexane extract of Paeonia lactiflora has antimicrobial activity against S. mutans and has preventive effect to dental caries in addition to potent inhibition to KB cell growth.

  • This study investigated the anti-bacterial and anti-inflammatory activities of Artemisia iwayomogi Kitamura for medical and food additive applications. A 70% methanol extract of A. iwayomogi Kitam. was fractionated with a series of solvents in order of increasing polarity; these fractionated extracts showed high 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) scavenging activity in the ethyl acetate fractions and high 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity in both the ethyl acetate and chloroform fractions. Levels of total phenolics and flavonoids were highest in the ethyl acetate fraction extracts. Eight bacteria were then treated with the A. iwayomogi Kitam. extract fractions and most showed similar or lower levels of anti-bacterial activity when compared to the control group, except for Staphylococcus aureus. In addition, the anti-oxidant activities of the ethyl acetate fractions of the A. iwayomogi Kitam. extract were higher than those of the other fractions. To confirm the anti-inflammatory activity of the extract fractions, we applied them to RAW 264.7 cells using LPS, and the results indicate that an ethyl acetate fraction at 250 ㎍/ml concentration inhibits nitric oxide production. This study demonstrates that an ethyl acetate fraction of A. iwayomogi Kitam. extract inhibits bacterial and inflammatory activities and could be useful as a potential source of bioactive compounds.

초석잠(Stachys sieboldii MIQ.) 줄기와 뿌리 추출물의 특성분석과 아질산염 소거능에 대한 고찰

  • This study was to search antioxidant constituents of ethyl acetate extract from Stachys siebodlii MIQ. The test of nitrite scavenging abilities were performed on the extracts of methanol, hexane, chloroform, ethyl acetate, butanol, and water, Ethyl acetate extract, The most promising one was fractionated on a silical gel column using elution solvent(chloroform:methanol:water=70:30:5 lower phase) at a flow rate 1.0ml/min. UV-VIS spectral data of each fraction showed adsorption maxima in the range of $284{\sim}330nm$ which is the characteristic range of $210{\sim}290nm$ and $300{\sim}550nm$ phenolic compounds. These results suggested that Stachys siebodlii MIQ. shows natural antioxidant activity. The nitrite scavenging abilities reached the maxium at pH 1.2 and the ethyl acetate fraction of root showed stronger scavenging ability.

Flavonoid Fraction Purified from Rhus verniciflua Stokes Actively Inhibits Cell Growth Via Induction of Apoptosis in Mouse Tumorigenic Hepatocytes

  • Dietary flavonoids are currently receiving considerable attention in developing novel cancer-preventive approaches because of their potential capacities to actively induce apoptosis of cancer cells. In our previous report, a flavonoid fraction, which consisted mainly of protocatechuic acid, fustin, fisetin, sulfuretin, and butein and named RCMF (RVS chloroform-methanol fraction), was prepared from a crude acetone extract of Rhus verniciflua Stokes (RVS) that is traditionally used as food additive and herbal medicine. In this study, we evaluated the effects of the RCMF on cell proliferation and apoptosis using SV40-transformed tumorigenic hepatocytes, BNL SV A.8. Tritium uptake assay showing the proliferative capacity of the cells was strongly suppressed in the presence of RCMF. This anti-proliferative effect was further confirmed through trypan blue exclusion. RCMF-mediated suppression of cell growth was verified to be apoptotic, based on the increase in DNA fragmentation, low fluorescence intensity in nuclei after propidium iodide staining, and the appearance of DNA laddering. Collectively, this study demonstrated that RCMF can be approached as a potential agent that is capable of significantly inhibiting cell growth of hepatic cancer cells.

Antioxidant Activity and Antimicrobial Effect for Foodborne Pathogens from Extract and Fractions of Sanguisorba officinalis L. (지유 추출물 및 분획물의 항산화 활성과 식중독 원인균에 대한 항균활성)

  • Background: This study aimed to investigate the antioxidat and antimicrobial activities of the methanol extract and its fractions prepared from the roots of Sanguisorba officinalis L. Methods and Results: The antioxidant activities were compared by evaluating the DPPH radical and nitric oxide (NO) scavenging ability. Measurement of DPPH radical scavenging ability showed that the $SC_{50}$ values of the ethyl acetate fraction was $3.85{\mu}g/m{\ell}$. The ethyl acetate fraction exhibited the most effective DPPH radical scavenging ability compared with the other samples. As for the NO scavenging ability, at all tested concentrations, the ethyl acetate fraction showed a higher scavenging activity than that of the extract and other fractions. These results are related to the total phenolic compound and flavonoid contents of the ethyl acetate fraction. Antimicrobial activity against foodborne pathogens was investigated using the disc diffusion assay. The ethyl acetate fraction showed the highest antimicrobial activity against gram-positive Staphylococcus aureus and Bacillus cereus. However, the chloroform fraction had a higher antimicrobial activity against gram-negative Vibrio vulnificus than that of the extract and other fractions. Conclusions: The results show that the ethyl acetate fraction had a higher antioxidant as well as antimicrobial activity, than did the other samples. Therefore, the ethyl acetate fraction has potential application in the food industry.
