Antigastritic and Antiulcer Actions of the Extract of Head of Panax ginseng Radix (인삼노두 추출물의 위염 및 위궤양에 대한 효과)

  • In a preliminary screening of plant extracts for the antigastritic and antiulcer actions in rats, the extracts of head of Panax ginseng Radix showed positive activity in HCl ethanol-induced gastric lesion. Among the systematic fractions of hexane, chloroform, butanol and water, the most potent butanol fraction reduced significantly HCl ethanol-induced gastric lesion at the oral dose of 500 mg/kg. In pylorus ligated rats, hexane and butanol fraction showed decreases in the volume of gastric secretion and acid output, of which effects were stronger in butanol fraction. Further assays with butanol fraction disclosed that it significantly suppressed the aspirin-induced and Shay ulcer. The butanol fraction at the intraduodenal dose of 500 mg/kg showed significant stimulation of mucus secretion.

Anti-diabetic and Anti-oxidative Effects of Opuntia humifusa Cladodes (천년초 선인장 줄기의 항당뇨 및 항산화 효과)

  • To investigate the anti-diabetic and antioxidant effects of Opuntia humifusa cladodes, O. humifusa cladodes powder was extracted with 75% ethanol and fractionated with various solvents. Among the extracts fractionated with various solvents, ${\alpha}$-amylase and ${\alpha}$-glucosidase inhibitory activities of O. humifusa cladodes were highest ($89.94{\pm}1.15%$ and $29.01{\pm}3.03%$, respectively) in the ethyl acetate fraction. Further, total phenolic and flavonoid contents of the ethyl acetate fraction were the highest ($196.02{\pm}5.26$ and $114.00{\pm}10.03{\mu}g/mg$, respectively). DPPH and ABTS radical scavenging activities increased according to the concentration of O. humifusa cladodes extract, and those of the ethyl acetate fraction were the highest. Ferric reducing antioxidant powers of chloroform and the ethyl acetate fraction were higher than those of other fractions. Overall, the ethyl acetate fraction of O. humifusa cladodes showed the highest anti-diabetic and antioxidant effects. Results indicate that O. humifusa cladodes powder has potential as a useful ingredient with anti-diabetic and antioxidant effects.

Cytotoxic Effects of Methanol Extract and Fractions from Echinacea angustifolia on Cancer Cells (암세포에 대한 Echinacea angustifolia 순차 용매 추출물의 세포독성 효과)

  • Echinacea is a North American native medicinal herb used traditionally for wounds, burns, snake or insect bites, colds, infections, and inflammation by indigenous Americans. We investigated the effects of the root and stem of fresh Korean-grown Echinacea angustifolia methanol extracts and fractionation extracts on the cytotoxicity against cancer cells (HL60, 3LL). The extracts were prepared by step-wise fractionation of methanol extracts of Echinacea angustifolia using hexane, chloroform, ethyl acetate, buthanol, and water. From the tests, root and stem parts of Echinacea showed the cytotoxic effect on cancer cells. The cytotoxie rate of the ethyl acetate fraction of the root parts showed 79% against HL60 cells at low concentration (0.125 mg/mL), and hexane fraction of the root and stem parts gradually increased as the concentration of samples increased, and the root parts showed 82% at 1.0 mg/mL concentration against HL60 cells, chloroform fraction of the root part showed 78.4% against HL60 cells and 68.4% on 3LL cells at 1.0 mg/mL concentration, water and butanol fraction of these results, it is considered that ethyl acetate fraction of the root and stem parts showed 60.1% to 77.1% against HL60 cells, after testing by MTT assay system. From these results, it is considered that ethyl acetate fraction of the Echinacea angustifolia root parts has stronger anticancer effects than any other fractions in vitro.

  • Methanol extracts of the roots of Atractylodes japonica (alba) (Compositae) and Gentiana scabra (Gentianaceae) were further fractionated with n-butanol, chloroform, n-hexane and water. Then each fraction was p.o. administered to an animal model of hepatitis caused by $CCl_4$ intoxication. Measurements of sleeping time of mice induced by hexobarbital administration indicated that n-butanol and chloroform fractions of Atractylodes japonica (alba), and chloroform and n-hexane fractions of Gentiana scabra showed significant liver protective activities. Measurements of the activities of serum transaminase, S-GOT and S-GPT appeared to support these results.

Antimicrobial Effect of the Fractions Extracted from a Lotus (Nelumbo nucifera) Leaf (연잎 추출물의 항균 효과)

  • In this study the antimicrobial activity of the extract from leaves of lotus (Nelumbo nucifera) was evaluated in comparison with benzoic acid. The $70\%$ ethanol extract was fractionated subsequently by hexane, chloroform, ethyl acetate, butanol and water, and the antimicrobial activity of each dried fraction was tested by paper disk diffusion method. The ethyl acetate fraction exhibited strong antimicrobial activity on the five strains of food born bacteria e.g. Bacillus subtilis, Staphylococcus aureus subsp. aureus, Escherichia coli, Salmonella Typhimurium and Pseudomonas nuorescens. The antimicrobial activity of the fraction was stronger than benzoic acid and showed no appreciable difference between on $Gm^+$ and $Gm^-$ bacteria. Heat treatment of the fraction at $110^{\circ}C$ for 1 hr did not change the degree of inhibitory effect. The ethyl acetate fraction showed almost perfect growth inhibition on the tested strains at over 800 ppm.

ACE, α-Glucosidase and Cancer Cell Growth Inhibitory Activities of Extracts and Fractions from Marine Microalgae, Nannochloropsis oculata (해양 미세조류 Nannochloropsis oculata 추출.분획물의 ACE, α-glucosidase 및 암세포 저해 활성)

  • Extracts of the marine microalgae Nannochloropsis oculata were obtained using 80% methanol (MeOH) and water. The 80% MeOH extract was further fractionated with n-hexane, chloroform, ethyl acetate (EtOAc), n-butanol (n-BuOH), and water to isolate the active fraction. Seven samples were prepared and their angiotensin converting enzyme (ACE), $\alpha$-glucosidase, and cancer cell growth inhibitory activities in vitro were determined. The most profound ACE inhibitory activity was observed in the chloroform fraction, while the others had moderate effects. By contrast, greater $\alpha$-glucosidase inhibitory activity was found in the EtOAc fraction, n-hexane fraction, and water extract of N. oculata. The antiproliferative effects of the extracts and fractions against HL-60, U937, CT-26, and SK-Hep1 cancer cells were also determined. The n-BuOH fraction had the strongest antiproliferative effects on CT-26 cells in a time-dependant manner (P<0.05). These results suggest that the extracts and fractions from N. oculata could be used as a potential functional food or as pharmaceutical ingredients.

Comparative antidiabetic activity of different fractions of methanolic extract of Zingiber officinale Roscoe in streptozotocin induced NIDDM rats

  • Earlier we have reported the antidiabetic activity of fresh juice of rhizomes of Zingiber officinale (Z. officinale) and its correlation with 5-HT receptor antagonism. Since 6-gingerol the marker compound of Z. officinale is reported to posses 5-HT anatgonistic activity, the present investigation, was undertaken to find out the concentration of 6-gingerol present in methanolic extract of Z. officinale and its different fractions (petroleum ether, toluene and chloroform). We also evaluated these fractions for antidiabetic activity in streptozotocin (STZ)-induced neonatal type 2 diabetic rats. Fasting glucose and insulin levels in non insulin dependent diabetes mellitus (NIDDM) rats were found to be significantly (P < 0.05) higher than control rats and these were significantly decreased by treatment with methanolic extract of Z. officinale and its fractions. The results of oral glucose tolerance test (OGTT) showed that methanolic extract and its fractions significantly (P < 0.05) decreased both STZ-induced increase in $AUC_{glucose}$ and $AUC_{insulin}$ values in NIDDM groups. Treatment with petroleum ether fraction produced a greater reduction in elevated glucose and $AUC_{glucose}$ levels as compared to treatment with other fractions. Treatment with methanolic extract of Z. officinale and its fractions also produced significant reduction in the elevated lipid, serum glutamate oxaloacetate transaminase (SGOT) and serum glutamate pyruvate transaminase (SGPT) levels in NIDDM rats. The effect of petroleum ether fraction on elevated lipid, SGOT and SGPT levels was significantly greater as compared to treatment with other fractions. The concentration of 6-gingerol was found to be maximum in petroleum ether fraction (11.430%) and minimum in chloroform fraction (0.973%). The methanolic extract and toluene fraction was found to contain 3.080% and 2.191 %, 6-gingerol respectively. In conclusion, our data suggest that methonolic extract and its fractions possess significant antidiabetic activity in NIDDM rats. The extent of activity appears to be dependent on the concentration of 6-gingerol present in the extract or its fractions.

Antimicrobial Effect of Each Solvent Fraction of Morus alba Linne, Sophora flavescens AITON on Listeria monocytogenes (Listeria monocytogenes의 증식억제에 미치는 뽕나무 및 고삼 에탄올 추출물의 분획별 효과)

  • Ethanol 75% extracts of Morus alba Linne(Ma) and Sophora flavescens AITON(Sf) were fractionated by chloroform($CHCl_{3}$), ethyl acetate(EtOAc), butanol(BuOH) and water. Each fractionate was added to tryptic soy broth varying in extract levels and 5 strains of Listeria monocytogenes(Lm) were inoculated in them for comparing growth inhibition at $32^{\circ}C$. The $CHCl_{3}$ and EtOAc fraction of Ma were superior in growth inhibition of 5 strains but only $CHCl_{3}$ fraction of Sf showed comparatively higher inhibition to Lm ATCC 19113. Lm ATCC 19111 and 19113 were totally inhibited by 500 ppm of EtOAc fraction and Lm ATCC 19112, 19114 and 15313 were also absolutely inhibited by 1000 ppm of $CHCl_{3}$ fraction of Ma 75% ethanol extract.

  • Xanthium sibiricum using extracts antioxidant activity and antimicrobial activity were analyzed and the following results were obtained. Antioxidant activity using DPPH as a VLC column fractionation experiments XS-5 (water fraction of Xanthium sibiricum) concentration in the 500 ${\mu}g/ml$ to 87.53 percent in the highest antioxidant activity was found, VLC fractionation in XS-3-2 (eluted with chloroform/methanol 7~10%) concentration of 500 ${\mu}g/ml$, showed the antioxidant activity of 78.72%. The antioxidant activity measured by PCL XS-1 (n-hexane fraction of Xanthium sibiricum) XS-5 (water fraction of Xanthium sibiricum) and 13.32 nmol open column XS-3-2 (eluted with chloroform/methanol 7~10%) showed antioxidant activity with 14.34 nmol. The antimicrobial activity against Candida albicans XS-2 (methlene chloride fraction fo Xanthium sibiricum) is the concentration of 500 ${\mu}g$/disc 1 mm of the clean zone was formed. XS-3 (ethanol fraction fo Xanthium sibiricum) in the antioxidant activity using DPPH and PCL had higher antibacterial activity was low.

Antibacterial Activity of Some Chrysanthemum spp. (Chrysanthemum속(屬) 식물의 항균성)

  • The yields of methanol extracts of several Chrysanthemum spp.(5 species, 9 parts) and their solvent fractions were investigated. The yields of methanol extracts ranged from 16.9%(for flower of C. indium, Cultivated) to 31.5%(for whole plant of C. indium). In the tests of the antibacterial activity. the methanol extracts from flower of C. zawadskii and C. boreale, whole plant of C. zawadskii, and flower of C. coronarium showed excellent antibacterial activity. Generally, the chloroform fractions exhibited stronger antibacterial activity against all the bacteria tested when compared with other solvent fractions. In case of C. boreale, the chloroform fraction of the whole plant was shown stronger antibacterial activity against all four bacterial strains tested when compared with that of the flower. The chloroform fractions from flower and whole plant of C. boreale and C .zawadskii, and flower of C. coronarium were shown a similar TLC pattern.
