Antibacterial Activity by Chitosan Derivatives with Quaternary Ammonium Salt (4차 암모늄기를 가진 키토산 유도체의 항균성)

  • Chitosan is the deacetylation product of chitin. Chitosan was converted to N-alkyl chitosan derivatives using proper aldehyde having different alkyl chains, then chitosan derivatives with quaternary ammonium salt were producted by quaternization with methyl iodide. Their antibacterial activities aganst S. aureus(ATCC 6538P) and E. coli (ATCC 14339), gram-positive bacteria and gram-negative bacteria, respectively, as introduced alkyl length were evaluated by shake flask method using colony count. The antibacteiral activity was found to be increased as alkyl chains. hydrophobic goups. These antibacterial agents were more active against S. aureus than E. coli. It may be due to the different of structure of cytoplasmic membrane.

  • Non-viral vectors, including lipid- or polymer-based systems, have attracted much attention to date as a gene delivery vehicle, due to safety issues with viral vectors. Chitosan, a naturally existing cationic polymer, has shown great potential as a gene delivery carrier, as it has low immunogenicity and toxicity, excellent transcellular transport ability, and is relatively easy to chemically modify. This review summarizes and discusses the general features of chitosan and its applications as a delivery carrier of DNA and RNA.

Synthesis of (2-pyridyl)-Acetyl Chitosan and Its Antioxidant Activity

  • In this paper, chloracetyl chitosan (CACTS) was prepared at first. In the molecules of CACTS, there are active chlorine groups, which can take part in other reactions. Thus, number of chitosan derivatives will be obtained after chlorine is substituted. Choosing pyridine as the active group, a novel water-soluble chitosan derivative, (2-pyridyl)-acetyl chitosan (PACTS) was obtained and its antioxidant activity against hydroxyl radicals and superoxide radicals was assessed. The results indicated that PACTS had better antioxidant activity than that of chitosan, carboxymethyl chitosan (CMCTS), hydroxypropyl chitosan (HPCTS), and Vitamin C. And the $IC_{50}$ values against hydroxyl radicals and superoxide radicals were 0.31 mg/mL and 0.21 mg/mL, respectively.

  • Chitosan was depolymerized by using nitrous acid. In order to synthesize new fluorinated chitosan oligomer(FCO) derivative, free amine groups of resulting low molucular weight chitosan oligomers were reacted with trifluoroacetic anhydride. The structure changes in the samples were conformed by using FT-IR, $^{1}H\;NMR$, $^{13}C\;NMR$ and $^{19}F\{^{1}H\}NMR$. Antiviral activity of FCO was studied by measuring DAN amounts of the replication viruses at 36 hr after the cells were infected with the viral solution containing FCO of various concentrations. The viral replications in the cells infected with the viral solution containing FCO were proportionally decreased with the FCO does, compared to those of the control groups, indicating that FCO efficiently inhibits viral infection. In particular, viral replication was decreased to 40% in the 1% FCO-treated cells.

  • The polymeric matrices coated with poly(DL-lactide) were prepared using chitosan derivatives such as chitosan, chitosan hydrochloride, and sulfonated chitosan for application of drug delivery systems. The drug release study using prednisolone as a model drug was performed in the hydrochloride solution at pH 1.2. The release rate of drug was decreased according to the increased content of matrices. The release rate of prednisolone according to the kinds of polymeric matrices coated were decreased in the order to chitosan, sulfonated chitosan, and chitosan hydrochloride. Drug release rate of polymeric matrices coated with poly(DL-lactide) was not only two times slower than noncoated one, but also the burst effect of initial period of drug release was decreased in comparison with noncoated one. From these results, it was expected that these formulations based on the chitosan derivative matrices coasted with poly(DL-lactide) were acceptable drug delivery devices for a sustained-release dosage form of drug.

  • Chitin and its derivatives have been introduced to tomato seedlings by direct mixing of chitin, chitosan, or chitooligosaccharides (CO) with soils, watering of chitosan or CO solutions to soils, or chitosan-coating of tomato seeds as a biomodulator of plant growth, and the growth of tomato plants was measured 30- and 45-day after seeding in the pots. The treatment of chitin and its derivatives promoted the growth of tomato plants in plant heights, shoot weight, maximum leaf length and number of leaves. By comparison in the contents of cellular inorganic nutrients in the plants, it was found out that chitin or chitosan treatments accelerated nitrogen and potassium uptake but inhibit calcium uptake, suggesting that chitin and its derivatives modulate absorption of inorganic nutrients through plant roots from environment.

Effect of Antimicrobial Activity by Chitosan Oligosaccharide N-Conjugated with Asparagine

  • Since the number of amino groups which are exposed by deacetylation of acetyl-D-glucosamine influences antimicrobial activity, a chitosan oligosaccharide (COS) derivative by N-conjugation of COS with asparagine, an amino acid with two amino groups, was synthesized and the antimicrobial effect on E. coli growth was compared with other COS derivatives which were N-conjugated with glycine, alanine, aspartic acid, cysteins, an methionine, and unmodified COS. The structure of asparagine N-conjugated COS (Asn-COS) derivative was identified by using a FT-IR, $^{13}C\;FT-NMR$, and an elemental analyzer. The antimicrobial activity of Asn-COS against E. coli growth was significantly improved as compared to the other COS derivatives as well as COS itself. This means that Asn-COS with two positive charges strongly interacts with the carboxyl negative charges on the bacteria cell wall. The results for Asn-COS were as follows: 100% bactericidal activity, 0.002% MIC, and no growth of E. coli during 3 days of culture time, suggesting that Asn-COS may be useful as a new antibiotic agent.

Effect of Chitin Derivatives on Non-steamed Alcohol Fermentation of Tapioca (Chitin 유도체가 타피오카의 무증자 알콜발효에 미치는 영향)

  • Chitin and its derivatives (chitosan and glucosamine) were studied for their effects on ethanol production using YPD (yeast extract 10%, peptone 20%, glucose 20%, agar 20%) medium. All chitin derivatives, particularly chitin, increased ethanol production compared with control. In non-steamed alcohol fermentation of tapioca, addition of 0.9% chitin yielded higher ethanol production (13.6%) with lower acetaldehyde (21.91 ppm) and methanol (65.49 ppm) contents than those (12.7%, 35.05 ppm, 84.31 ppm, respectively) of control after fermentation for 120 hr at $30^{\circ}C$. Results indicate that chitin can be used to increase ethanol production in non-steamed alcohol fermentation of tapioca.

Synthesis and Hydroxyl Radicals Scavenging Activity of 2-Pyridine-acetyl-N-trimethyl Chitosan Chloride

  • A novel chitosan derivative with double quaternary ammonium salt-2-pyridine-acetyl-N-trimethyl chitosan chloride (PATMCS) was synthesized and the antioxidant activity of PATMCS against hydroxyl radicals was assessed. The results indicated that PATMCS had potent hydroxyl scavenging activity. The $IC_{50}$ of PATMCS was 0.13 mg/mL. PATMCS showed 100% scavenging effect at a dose of 0.8 mg/mL which markedly better than that of N-trimethyl chitosan chloride (TMCS). It was confirmed that quaternary chitosan derivatives showed potent antioxidant activity. PATMCS has double quaternary ammonium salt structure in the molecules. Therefore, the antioxidant activity of PATMCS was better than TMCS. The above results are theoretically fundamental for further development and making use of chitosan resources to prepare new antioxidants.
