The Effect of Pulsatile Versus Nonpulsatile Blood Flow on Viscoelasticity and Red Blood Cell Aggregation in Extracorporeal Circulation

  • Background: Extracorporeal circulation (ECC) can induce alterations in blood viscoelasticity and cause red blood cell (RBC) aggregation. In this study, the authors evaluated the effects of pump flow pulsatility on blood viscoelasticity and RBC aggregation. Methods: Mongrel dogs were randomly assigned to two groups: a nonpulsatile pump group (n=6) or a pulsatile pump group (n=6). After ECC was started at a pump flow rate of 80 mL/kg/min, cardiac fibrillation was induced. Blood sampling was performed before and at 1, 2, and 3 hours after ECC commencement. To eliminate bias induced by hematocrit and plasma, all blood samples were adjusted to a hematocrit of 45% using baseline plasma. Blood viscoelasticity, plasma viscosity, hematocrit, arterial blood gas analysis, central venous $O_2$ saturation, and lactate were measured. Results: The blood viscosity and aggregation index decreased abruptly 1 hour after ECC and then remained low during ECC in both groups, but blood elasticity did not change during ECC. Blood viscosity, blood elasticity, plasma viscosity, and the aggregation index were not significantly different in the groups at any time. Hematocrit decreased abruptly 1 hour after ECC in both groups due to dilution by the priming solution used. Conclusion: After ECC, blood viscoelasticity and RBC aggregation were not different in the pulsatile and nonpulsatile groups in the adult dog model. Furthermore, pulsatile flow did not have a more harmful effect on blood viscoelasticity or RBC aggregation than nonpulsatile flow.

An effect of laser irradiation on periodontal pocket tissue (레이저 조사가 치주낭 조직에 미치는 영향)

  • Periodontal pocket is one of the most frequently developed clinical feature on the teeth with periodontal disease. In order to determine the gingival curettage effect of intrapocket irradiation of a pulsed Nd : YAG laser on periodontally involved teeth, bilateral 60 teeth with $4{\sim}6mm$ in probing pocket depth 1 week after supragingival scaling were selected. On half of them the intrapocket irradiation($300{\mu}m$ fiber optic, 1.5W power, for 2min.) of a pulsed-Nd : YAG laser(EL.EN.EN06O, Italy) was applied as the lased group. On the contralateral 30 teeth the subgingival curettage was accomplished by Gracey curettes as the curattage group. The periodontal pocket tissues were surgically excised by the modified Widman flap technique immediately after the intrapocket irradiation or subgingival curettage, subsequently fixed with 10% neutral formalin, sectioned in $4{\sim}6{\mu}m$ thickness, and stained with hematoxylin-eosin. Surface characteristics and incomplete removal of the pocket epithelium were evaluated under light microscope. And the difference between the lased group and the curettage group was statistically analyzed by Chi-square test in Microstat program. The results were as follows ; 1. The plane surface was observed more frequently in the curettage group(73.3%) than in the lased group(23.3%), and the rough surface was observed more frequently in the lased grOoup(63.3%) than in the curettage group(6.7%)(p<0.05). 2. The rate of incomplete removal of the pocket epithelium was relatively high in both the lased group(76.6%) and the curettage group(86.6%), and there was no significant difference between the lased group and the curettage group(p>0.l). The results suggest that the further studies including various power control of laser should be succeeded in order to obtain more favorable results by the intrapocket irradiation of a pulsed Nd:YAG laser than the subgingival curettage with Gracey curettes.

Hazard Analysis for the Cultivation Stage of Strawberry Farms for Securing Preliminary Data to Establish the Good Agricultural Practices (농산물우수관리제도 확립의 기초자료 확보를 위한 딸기농장 재배단계의 위해요소 분석)

  • Physical, chemical and biological hazards of strawberry farms at the cultivation stage were analyzed to establish the GAP(Good Agricultural Practice) system. Samples were collected from the plants, cultivation environments(water, soil and air), and personal hygiene (hand, glove, and clothes) of three strawberry farms(A, B, and C) and were tested to analyze physical, chemical (heavy metals and pesticide residues), and biological(sanitary indications and foodborne pathogens) hazards. Physical hazards such as insects and pieces of metal and glass were found in the strawberry farms and can be potential bow for strawberry products. Heavy metal and pesticide residue as chemical hazards were detected at levels lower than the regulation limit. In case of biological hazards, total bacteria and coliform were detected at the levels of 1.6~7.3 and 1.3~5.6 log CFU/g, leaf, mL, hand or $100cm^2$. However, Escherichia coli was not detected in all samples. Bacillus cereus and Staphylococuus aureus were detected at levels of ${\leq}$ 1.1~6.1 log CFU and 4.7~5.4 log CFU/g, mL, hand or $100cm^2$, whereas Listeria monocytogenes, E. coli O157 and Salmonella spp. were not detected in all samples. This study demonstrates that various harzards were in strawberry farms at the growing stage. Therefore proper management such as GAP is needed to prevent the occurrence of food poisoning associated with the hazards revealed in this study.

Evaluation of Water Quality after Rehabilitation of Cheonggye Stream using AGP Test (조류성장잠재력 조사를 이용한 청계천 복원 이후 수질 평가)

  • Algal growth potential (AGP) test was performed to evaluate the water quality and changes in phytoplankton communities before and after a heavy rain event at six sampling sites in Cheonggye Stream (St. 1 and 2), Jungnang Stream (St. 3 and 4), and Lower part of Han River System (St. 5 and 6) after rehabilitation of Cheonggye Stream, October 2005. To test AGP on each sampling site, cyanobacterium Microcystis aeruginosa was applied as a standard alga. Total nitrogen (TN) showed high values at Jungnang Stream, while St. 4 recorded highest values in this study. However, TN values of Cheonggye Stream and Lower Part of Han River showed similar levels. Total phosphate (TP) also showed high values at Jungnang Stream, while St. 4 recorded highest. However, TP in Cheonggye Stream were extremely low levels. Although chlorophyll-${\alpha}$ (chi-${\alpha}$) contents before the rain event were similar through the sampling sites, chl-${\alpha}$ after the rain increased dramatically at Jungnang Stream and Lower part of Ban River. In particular, after the rain, TP was the highest at St. 4, where Cyclotella sp. dominated the phytoplankton community. When compared with control, AGP values before the rain were comparatively low in all sites, while those after the rain highly increased with the dose-dependently of field water added, due perhaps to the increased nutrients by rainfall. Similar results were observed in Cheonggye Stream. Therefore, for the aesthetic fostering for the citizens, although Cheonggye Stream was Presently being sustained by treated water supply, they have a potential of outbreak of phytoplankton by the increased nutrients supply when a heavy rain comes.

Cell Growth in Suspension-Culture of Populus nigra var. italica and the Efficiency of Micro-Callus Formation according to Cell Plating Method (Populus nigra var. italica현탁배양(懸濁培養) 세포(細胞)의 생장(生長) 및 Cell Plating방법(方法)에 따른 Micro-Callus형성능력(形成能力))

  • In order to know the growth of suspended cells by explant sources, the change of nitrogen contents of cultured cells following the growth periods, capability of micro-callus formation according to cell plating methods, growth of suspended cells on various media, and efficiency of micro-callus formation by using growth regulators and different N strengths were investigated. 1. When suspension culture was tried by using the callus induced from internode and petiole, cell fresh weight and packed cell volume increased with similar way and the growth reached at stationary phase after 12 culture days. 2. N-contents of cultured cells increased upto 3 days and decreased around 6days. But the values increased again upto 9 days, after that they showed gradual decreases. 3. Of cell plating methods, embedding method was the best for micro-callus formation. 4. Growth of suspened cells showed the rest performanoes, when they were cultured on LM medium with 1/2N strengths and BAP 0.01.2.4-D 0.1, and NAA $1.0mg/{\ell}$, after 15 cultured days(upto 76.9 folds). LM medium was better than MS or GD. The combination of auxin and cytokinin was better for cell growing than auxin-treatment only. 5. Micro-callus from single cell and small cell aggregates was formed only on MS and LM media with 2,4-D $1.0mg/{\ell}$.

A Study and Survey on Clinical Nurses concerning the General Items, the Motives of Determining their Profession, the Attitudes toward their Profession and the Desire and Expectation to their Profession and Society (임상간호원에 대한 연구조사)

  • This study of 855 clinical nurses was conducted using a questionnaire that include tour different scales; the motives of determining their profession, the attitudes toward their profession, the general items, and desire and expectation to their profession and society. The data were analyzed by Chi-Square Test and Percentage. The results of this study included Hypothesis are as follows; The respondents were 855 (78.6%) among 1088 clinical nurses who were employed by General of Educational hospitals through the city of Seoul, Pusan, Taegu, Daejun, Kwangju, and Wonju. 1) a. In the Age Distribution, the majority of respondents were under the age of 30yrs(88.2%) and the minority were above 31yrs(11.2%), and the preponderance of the majority to minority(9:1) was noted. In compared with area, a group above 31yrs old in Seoul (6.9%) was lower than other area (16.3%). b. The types of Educational background were 16.3% in Voc.Tr. School, 66.5% in Diploma and 17.1% in Degree.146 clinical nurses were from the Degree course, and 142 (97.3%) CN among those of them were occupied around Seoul and 4(2.7%) around other area. c. In the Marital Status,71,5% were the unmarried and 28.5% were the married. And compared with the area was 20.4% in Seoul and 41.4% in other area. d. Most common Length of Clinical Experience after graduation was under tile 2yrs (55.4%), 3yrs(14.2%, and 4yrs (6.2%). In compared with area, Seoul (15.3%) was lower than other area (38.1%) above 5yrs of clinical experience, and the preponderance of tile other area to Seoul as 2.5: I was noted. 2) a. Hypothesis 1 was significant relation between the types of Educational Background of the CN and their motives for selection of Nursing, P-value was below 0.01. b. There was a significance on hypothesis 2 (P<0.01): that was relation between their motives for selection of clinical nursing field after their graduation and the area which they were employed. c. Hypothesis 4 was accepted as significant relation between the level of satisfaction of their clinical experience after their graduation and the types of educational back ground, P-value was below 0.01. d. There was a significance on hypothesis 5(P<0.01) that was relation between the CN's response about the orientation program and the area which they were employed. e. Hypothesis 6 was retained as significant relation between the area and inservice educational programme of their employed hospital was practising or not. P-value was 0.01. f. Hypothesis 7 was retained as significant relation between the area and the CN's response about the inservice educational programme of their employed. P-value was below 0.01. g. There was a significance on hypothesis 8 (P<0.0l) that was relation between the CN's experience on attending the professional meeting and the area. h. Hypothesis 10 was accepted as significant relation between the response about the present licence system and their educational background. p-value was below 0.01. i. There was a significance on hypothesis 11 (P fO.01) that was relation between the carrying out the regular and delivery vacation and the area. J. Hypothesis 12 was accepted as significant relation between the CN's consideration of the lack of leisure and their marital status. p-value was below 0.01' k. There was a significance on hypothesis 13 (P <0.01) that was relation between the CN's response about their salary and their marital status. l. Hypothesis 14 was significant relation between the most difficulties of CN during their working and the hospital which they were employed. p-value was below 0.01.

Fermentation Properties of Young Radish Kimchi Prepared Using Young Radish Cultivated in the Soil Containing Sulfur and It's Inhibitory Effect on the Growth of AGS Human Gastric Adenocarcinoma Cells (유황처리 열무로 제조한 열무김치의 특성과 인체 위암세포의 성장억제효과)

  • Young radishes (YR, yeolmu in Korean) were cultivated in the soil with and without sulfur. YR-Control (without sulfur) was grown in the normal soil. YR were grown in the soil with $1,818\;g/m^3$ sulfur (YR-A) and $1,818\;g/m^3$ sulfur added lime mortar (YR-B) on it, respectively. Also, we prepared YR kimchis using YR-Control, YR-A and YR-B. The kimchis were fermented at $5^{\circ}C$ for 8 weeks. The growth inhibitory effects of AGS human gastric adenocarcinoma cells of the YR samples and kimchis were investigated. YR kimchis after $4\~5$ weeks at $5^{\circ}C$ showed higher acidity of $0.88\~1.20\%$ with pH $4.3\~4.5$ and the YR kimchis kept approximately pH 4.0 until 8 weeks. The kimchi A and B using YR-A and YR-B showed faster fermentation time, higher level of Leuconostoc sp. and lower level of Lactobacillus sp. during the fermentation, comparing to the control kimchi using YR-Control. Juices from YR-A and YR-B showed higher growth inhibitory effects of AGS human gastric adenocarcinoma cells than the juice from YR-Control at the same concentration. The growth inhibitory effect of YR-A was similar to that of the YR-B. The kimchi A and B juices also exhibited higher inhibitory effects $(74\%)$ on the growth of AGS human gastric adenocarcinoma cells than that of the control kimchi $(57\%)$ at the higher concentration of $20{\mu}L/assay$. Methanol extracts from the YR-kimchis also led to the similar results to the results of the juices. These results suggested that preparing of kimchi using differently cultivated YR especially in the soil with sulfur, which can help to synthesize sulfur-containing compounds, could increase the growth inhibitory effects of AGS human gastric adenocarcinoma cells.

Clinical Characteristics of Paradoxical Response to Chemotherapy in Pulmonary Tuberculosis (항결핵제 사용 중 폐병변의 일시적 악화를 보인 환자의 임상상)

  • Background : The paradoxical response refers to an enlargement of old lesions or unexpected new ones during apparently adequate antituberculous therapy. This response has been reported in cases of intracranial tuberculoma, tuberculous lymphadenopathy, tuberculous pleurisy and pulmonary tuberculosis. However, there are few reports on its frequency and clinical characteristics. Materials and Methods : This study enrolled 205 patients who were treated with first line antituberculous agents for more than 6 months. We retrospectively studied 155 patients with pulmonary tuberculosis and 57 patients with pleural tuberculosis (7 patients had both) from July 1998 to March 2000. The patients were divided into the paradoxical response group and the non-paradoxical group. The clinical characteristics of the paradoxical response group and the non-paradoxical group. The clinical characteristics of the paradoxical group were investigated. Statistical analysis was done with an independent sample T-test and Chi-squared test. Results : 29 of the 205 patients(14.1%) had paradoxical response. Among the 29 patients, there were 19 pulmonary tuberculosis, 8 tuberculous pleurisy(2 patients had both). Paradoxical response appeared 32 days (mean 35 days in pulmonary tuberculosis, mean 25 days in tuberculous pleurisy) after the beginning of chemotherapy. The duration to regress less than half of initial chest lesion was 114 days in pulmonary tuberculosis and 124 days in tuberculous pleurisy, respectively. Most common clinical manifestation of paradoxical response patients was coughing in both pulmonary tuberculosis and tuberculous pleurisy. Male sex, high blood WBC count and high level of pleural fluid LDH were related with paradoxical response. Conclusion : These findings suggest that presponse usually appears 1 month and disappears within 4 months after the beginning of anti-tuberculous chemotherapy. Paradoxical response was relatively correlated with male sex, high blood WBC count and high level of pleural fluid LDH.

Limnological Characteristics and Influences of Free-floating Plants on the Woopo Wetland during the Summer (하계우포습지의 육수학적 특성 및 부유수생식물의 영향)

  • During January 1998-October 1999, the impact of free-floating plants (FFP) on limnology of the wetland ecosystem was evaluated through the investigation of physicochemical characteristics of the Woopo Wetland along with in situ manipulation experiments of aquatic plants. Flooding events occurred in the wetland during the summer period (Jun.-Aug.) and water levels rose to 2-3 m due to precipitation from the catchment and inflow from the main channel of the Nakdong River. Physicochemical parameters and plankton dynamics in the wetland during the summer were greatly influenced by floods and growth of free-floating plants. Dissolved oxygen (Jun.-Sept., 4.5${\pm}$2.5 mg/1; Oct.-May, 8,1 ${\pm}$4.0 mg/1) and pH (Jun.-Sept., 6.9${\pm}$0.4; Oct.-May,7.4${\pm}$0.8) levels were significantly lower during the summer than any other seasons. Three types of enclosure experiments (100 L, treatments with floating plants, screened and opened) were conducted under the presence and absence of sediment for 15 days in the 1999 summer. The treatments with sediment had higher levels of nutrient concentrations than those of the others. Among the treatments with sediment, nutrient concentrations in the treatments with free-floating plants were higher than the others. Zooplankton communities in each treatment showed a similar variation, although the scale of zooplankton densities differed. Rotifer community dominated the zooplankton at the initial phase of the experiment, but decreased drastically along with an increase of cladoceran and copepod communities. In conclusion, low levels of dissolved oxygen and pH in the Woopo Wetland during the summer seemed to be caused by a proliferation of free-floating plants and active decomposition process at the bottom of the sediment.

Effects of Practical Variations in Fasting, Stress and Chilling Regime on Post-slaughter Metabolic Rate and Meat Quality of Pork Loin (절식, 스트레스 및 냉각속도가 돼지도체의 사후 대사속도와 등심근의 육질에 미치는 영향)

  • The study was conducted to investigate the effects of practical variations in feed restriction, pre-slaughter handing and chilling regime on pork quality during ageing. A total of twenty male landraces were allocated into three treatments(i.e., pre-slaughter feeding, stress and chilling regime) in a factorial arrangement. pH, temperature, free calcium ions, WB-shear force, sarcomere length, cooking loss, drip loss and objective color were determined during rigor development and/or 1, 3, 7 d postmortem. Pre-slaughter chasing stress for approximately 15 min had no effects on pH/temperature profile and objective meat quality. There was an interaction(P < 0.05) between the fasting treatment and chi1ling regime for muscle temperature at pH 6.2. Sarcomere length indicated that the current experiment conditions did not induce muscle shortening, with 1.7 to 1.8 ${\mu}m$, in spite of a significant effect of the fasting treatment (P<0.01). Pigs fed until the morning of slaughter showed a low WB-shear force(P < 0.05) until 3 d at I "C. The treatment also resulted in a higher Hunter L* and a*(P < 0.05) at 24 h and 7 d. Fasted pigs showed a significantly(P < 0.05) reduced cooking loss. The current results indicated that feeding upon the morning of slaughter became detrimental on meat color and the negative effect on cooking loss were linearly elevated with increased ageing time. On the other hand, WB-shear force did not distinguishable after 3 d. Collectively, it appeared that feed restriction from a day before slaughter could produce more a desirable meat quality at the time of consuming. However, the limited effect of animal handling and chilling rate on meat quality is not necessarily to extend to that these do not affect pork quality, as that largely depends on experimental design.
