자외선이 조사된 섬유아세포에서 말채나무 잎추출물의 항산화작용 (Antioxidative Activity of Extract of Cornus walteri Wanger Leaves in Human Dermal Fibroblast Irradiated by UVB)

  • We investigated antioxidative activity of the ethanol extracts of leaves of Cornus walteri Wanger (CWE) by treated enzyme in human dermal fibroblast (HDFs) irradiated by UVB. We examined the in vitro chemical and cellular antioxidant activities of CWE in HDFs. We employed scavenging assay for the 1,1-diphenyl-2,5-picrylhydrazyl (DPPH) radicals and cellular antioxidative activity of CWE, and we was investigated in $H_2O_2$-treated or UVB-irradiated HDFs. The CWE effectively scavenged DPPH radicals ($IC_{50}$ $7.03{\pm}0.4{\mu}g/mL$) when compared to the scavenging activities of L-ascorbic acid ($IC_{50}$ $4.69{\pm}0.3{\mu}g/mL$). CWE reduced UVB-induced cellular damage in HS68 cells by MTT assay and inhibited intracellular ROS generation in dose-dependent manner. In addition, CWE also attenuated the elevated levels of 8-isoprostane resulting from UVB-mediated oxidative stress. Collectively, these results suggest that CWE could be a new potential candidate as antioxidant against UVB-induced oxidative stress in HDFs.

식물체에 미치는 연, 아연 ( Pb, Zn ) 의 영향 (Ecological Effects of Zinc and Lead on Plants)

  • This study was carried out to investigate the effects of Zn and Pb concentration on seed germination and plant growth in water and soil culture, and the frequency of chlorosis invegetation and the relationship between plants and soil in the Sambo mine. The inhibition of germination were observed in 1000ppm of Zn, 10ppm of Pb and 5000ppm of Zn + Pb, but germination was more stimulated in 10ppm of Zn than control. The symptoms of chlorosis and abnormality were occurred in plant leaves grown to the soils treated with more than 1000ppm of Pb. Reasons of chlorosis were considered as an antagonistic effect of other metals towards uptake of iron by the plant in Zn treatment. The contents of Zn and Pb in fruits were lower than those of leaves, and that was remarked in case of Pb. With increasing rate of Zn and Pb treatment, chemical components of soils in pot culture were accompanied by slight decrease in pH, total nitorgen and exchangeable K. Chlorotic individuals of 10 species were shown in the areas of the Sambo mine. Chlorotic symptoms were especially extensive and severe in Sophora angustifolia, Populus alba, Spiraea prunifolia, Amorpha fruticota, Lespedeza bicolor and Salix dependens. Plants in the investigated areas grew in soils containing Zn of 311ppm and Pb of 151ppm on an average, and accumulated Zn of 2084ppm and Pb of 49ppm.

Antioxidative Flavonoids from Leaves of Carthamus tinctorius

  • A total of eight flavonoids (1-8), including a novel $quercetin-7-o-(6"-o-acetyl)-{\beta}-D-glucopyranoside$ (6) and seven known flavonoids, luteolin (1), quercetin (2), luteolin $7-o-{\beta}-D-glucopyranoside$ (3), $luteolin-7-o-(6"-Ο-acetyl)-{\beta}-D-glucopyranoside$ (4) quercetin $7-o-{\beta}-D-glucopyranoside$ (5), acacetin 7-o-{\beta}-D-glucuronide (7) and apigenin-6-C-{\beta}-D-glucopyrano $syl-8-C-{\beta}-D-glucopyranoside$ (8), have been isolated from the leaves of the safflower (Carthamus tinctorius L.) and identified on the basis of spectroscopic and chemical studies. The antioxidative activity of these flavonoids was evaluated against 2-deoxyribose degradation and rat liver microsomal lipid peroxidation induced by hydroxyl radicals generated via a Fenton-type reaction. Among these flavonoids, luteolin-acetyl-glucoside (4) and quercetin-acetyl-glucoside (6) showed potent antioxidative activities against 2-deoxyribose degradation and lipid peroxidation in rat liver microsomes. Luteolin (1), quercetin (2), and their corresponding glycosides (3 & 5) also exhibited strong antioxidative activity, while acacetin glucuronide (7) and apigenin-6,8-di-C-glucoside (8) were relatively less active.

NUTRITIONAL EVALUATION OF MAIZE FODDER AT TWO DIFFERENT VEGETATIVE STAGES

  • Different fractions of maize plant and whole mixed fodder were analysed for their chemical composition and dry matter digestibility (DMD). Highest crude protein (CP) values were found in leaves as compared to the other portions. Younger plants contained more CP as compared to the matured ones. The crude fiber (CF) content of various fractions of the plant ranged between 19.12 to 35.60% with maximum values in the bottom portion of the stem. Matured plants contained more CF. The analysis of cell wall constituents indicated that the maximum values for neutral detergent fiber (NDF) were found in the bottom portion and in the whole mixed plant. The highest levels of acid detergent fiber (ADF) were observed in bottom fraction followed by whole mixed plant, whereas the other plant fractions did not show any differences. Variation in acid detergent lignin (ADL) values existed in different fractions of the plant and the lowest were in the top portion of the stem. Although there existed a variation in the mineral composition of different fractions of the plants, the results were non significant. Maximum DMD was found in leaves followed by the whole mixed plant, middle and bottom portion of the stem. The values of DMD were higher in younger plants as compared to the matured ones. It may be concluded that younger plants and the upper portion of the plants have a higher nutritive value as compared to the matured plants and lower portion of the plants.

  • The present study was carried out to obtain some basic informations of the breeding of tobacco varieties. Genetically divergent 8 varieties, 3 flue-cured, 2 burley and 3 sun-cured tobaccos, were used in half diallel cross. In order to analyze the heterosis, combining abilities, modes of inheritance and correlations for some agronomic and chemical characters, 8 parents and 28 $F_2$ were tested. The results obtained were summarized as follows: The percentages of heterosis for yield, stem diameter, internode length and total sugar content in $F_2$ hybrid were positive , where-as those for the other characters were negative. Yield had significant positive genotypic and phenotypic correlations with plant height, days to flower, leaves per plant, leaf length, leaf width and leaf shape index (Leaf length/leaf width). General (GCA) and specific combining abilities (SCA) for all characters of $F_2$ hybrid were significant. In the effect of GCA, yield was greater in combinations of Bulgaria and Xanthi, days to flower and leaves per plant were those of Burley 21 and Va 528, leaf length and width were those of Coker 139, respectively. In the effect of SCA, the major part of characters were greater than those of others in the combinations of Burley 21, Va 528, Hicks.

Chemical Constituents from Leaves of Acanthopanax henryi (II)

  • Nineteen compounds, including one organic acid (1), one anthraquinone (2), one amide (3), and sixteen triterpenoid saponins (4 - 19) were isolated from the leaves of Acanthopanax henryi (Oliv.) Harms (Araliaceae). Their structures were determined on the basis of physicochemical properties and spectral analyses (HR-MS and NMR). Among them, compounds 2, 3, 7, 12 and 19 were new within Araliaceae. Compounds 4, 5, 9 - 11, 13, 14, 16 and 18 were reported for the first time from the Acanthopanax genus. Except for compounds 4 and 9, other compounds were isolated from A. henryi (Oliv.) Harms for the first time. The rare anthraquinone, compound 2, significantly decreased the production of NO and the levels of other inflammatory factors, such as TNF-α and IL-6, in lipopolysaccharide (LPS)-stimulated macrophages in a dose-dependent manner. This is the first time to report anti-inflammatory effect of this compound.

Cytotoxic Components in an Extract from the Leaves and Stems of Stauntonia hexaphylla

  • An investigation was carried out to identify novel anti-cancer compounds from Korean indigenous plant extracts. Bioassay-guided fractionation and chemical investigation of the EtOAc extract from the leaves and stems of Stauntonia hexaphylla resulted in the isolation of two active compounds, hederagenin 3-O-${\alpha}$-L-arabinoside (1) and quercetin (2). The structures of these compounds were elucidated by spectroscopic methods, including UV, IR, MS, NMR techniques and compared with previous spectroscopic data. The cytotoxic effects of fractions and compounds on HCT116 human colon cancer cells were evaluated using the MTT assay. Quercetin showed a stronger anti-cancer effect when compared to hederagenin 3-O-${\alpha}$-L-arabinoside.

Antioxidative flavonoids from the Leaves of Morus alba

  • Nine flavonoids (1-9) were isolated from the leaves of Morus alba (Moraceae). The structures of compounds were determined to be kaempferol-3-O-$\beta$-glucopyranoisde (astragalin, 1) kaempferol-3-O-(6"-O-acetyl)-$\beta$-D-glucopyranoside (2), quercetin-3-O-(6"-O-acetyl)-$\beta$-D-glucopyranoside (3), quercetin-3-O-$\beta$-D-glucopyranoside (4), kaempferol-3-O-$\alpha$-L-rhamnopyranosyl-(1 6)-$\beta$-D-glucopyranoside (5), quercetin-3-O-$\alpha$-L-rhamnopyranosyl-(1 6)-$\beta$-D-glucopyranoside (rutin, 6), quercetin-3-O-$\beta$-D-glucopyranosyl-(1 6)-$\beta$-D-glucopyranoside (7), quercetin-3,7-di-O-$\beta$-D-glucopyranoside (8) and quercetin (9) on th basis of spectroscopic and chemical studies. Compounds 7 and 9 exhibited significant radical scavenging effect on 1,1-diphenyl-2-picryl-hydrazyl radical.

Chemical Constituents of Abies koreana Leaves with Inhibitory Activity against Nitric Oxide Production in BV2 Microglia Cells

  • Eleven compounds were isolated from fresh leaves of Abies koreana (Pinaceae), and structures of these compounds were determined to be 3-hydroxy-2-methyl-4-pyrone (1), maltol-3-O-${\beta}$-D-glucoside (2), (-)-epicatechin (3), naringenin 7-O-${\beta}$-D-glucopyranoside (4), naringenin-7-O-rhamnoglucoside (5), kaempferol 3-O-${\beta}$-D-glucopyranoside (6), (+)-isolariciresinol (7), secoisolariciresinol (8), rhododendrol (9), ferulic acid (10) and 4-(4-hydroxyphenyl)butan-2-one (rheosmin) (11) by comparing $^1H$-, $^{13}C$-NMR and MS spectroscopic data with reference values. Compounds 3, 5, 7, 8, 9, 10, 11 were isolated for the first time from A. koreana. Among eleven isolates, compounds 1, 7 and 11 showed inhibitory activities against lipopolysaccharide (LPS)-induced nitric oxide (NO) production in BV-2 microglia in a concentration dependent manner.

New Cytotoxic Benzopyrans from the Leaves of Mallotus apelta

  • Two new benzopyrans 6-[1'-oxo-3'(R)-hydroxy-butyl]-5,7 -dimethoxy-2,2-dimethyl-2H-1-ben-zopyran (1) and 6-[1'-oxo-3'(R)-methoxy-butyl]-5,7 -dimethoxy-2,2-dimethyl-2H-1-benzopyran (2) were isolated from the leaves of Mallotus apelta Muell.-Arg., (Euphorbiaceae). Their chemical structures were elucidated by spectroscopic analyses, especially by 1D-, 2D-NMR and MS spectra. Compound 1 was found to have strong cytotoxic effect against two human cancer cell lines as human hepatocellular carcinoma (Hep-2, $IC_{50}\;:\;0.49\;{\mu}g/mL$) and rhabdosarcoma (RD, $IC_{50}\;:\;0.54\;{\mu}g/mL$), while compound 2 showed moderate activity against Hep-2 cell line ($IC_{50}\;:\;4.22\;{\mu}g/mL$) by in vitro assay.
