This study was conducted to improve organoleptic quality of gamma-irradiated Tarakjuk at 10 kGy with cheeses (Camembert, Cheddar, Gouda, and Edam) and flavors (Nurungji, cream, and vanilla-cream). Overall acceptability of gamma-irradiated Tarakjuk added each camembert cheese and vanilla-cream flavor was the highest among the all samples. In the effect of sterilization method on the quality of Tarakjuk, the autoclaved samples added with Edam cheese, Gouda cheese, and vanilla-cream flavor, respectively, were showed higher score on the overall acceptability than the irradiated samples, meanwhile the irradiated samples were superior in the other samples. In case of added mixture of Edam cheese and vanilla-cream flavor, irradiated Tarakjuk with mixture was showed the most of high score on overall acceptability. In conclusion, addition of mixture of cheese and flavor to Tarakjuk may help to improve organoleptic quality of sterilized Tarakjuk by gammairradiation.
본 연구에서는 Cheddar 치즈의 숙성기간을 단축시키고 flavor를 증진시키기 위하여 Micrococcus sp. LL3를 치즈 제조시에 첨가할 목적으로 본 균주가 생성하는 aminopeptidase의 특성을 조사하였으며, 또한 본 효소를 정제하였다. L-Leucine-p-nitroanilide를 기질로 사용 하였을 때 본 효소의 최적온도와 pH는 각각 $30^{\circ}C$ 및 7.0이었다. 본 효소는 $50^{\circ}C$까지의 온도에서 10분간 방치하였을 경우에도 안정하였다. $Mg^{++}$ ion에 의해 본 효소의 활성이 촉진되었으나 $Hg^{++}$ ion과 EDTA나 1,10-phenanthroline에 의해서 효소활성이 거의 실활되어 metallopeptidase인 것으로 추정되었다. 본 효소의 기실 특이성은 광범위 하였으나, N-terminal amino acid로서 arginine을 함유한 peptide는 분해하지 못했다. 본 균주가 생성하는 aminopeptidase의 정제를 위하여 DEAE-Sephacel ion chromatography및 gel filtration을 실시하였으며, 이때 분자량 46,500의 효소를 얻을 수 있었다.
본 연구는 전자코를 이용하여 한국에서 시판되는 피자용 모짜렐라 치즈 슈레드 제품과 슈레드 제품 혼합용 원료 치즈에 해당하는 모짜렐라 치즈 블록, 체다 치즈, 흰 곰팡이 숙성 치즈 등 3종류의 국내산 및 수입산 치즈를 대상으로 단기간 저장 및 가열처리에 의한 풍미패턴의 변화를 분석하였고 국내 임실치즈농협에서 새로 개발한 임실 치즈 제품이 국내산 및 수입산 치즈의 향미패턴과의 차이 유무에 대해 접근하여 보고자 하였다. 치즈 종류뿐만 아니라 조리온도 및 저장유무에 따른 향기패턴을 판별함수 분석을 이용하여 통계처리 후 비교 분석한 결과, 피자용 모짜렐라 슈레드 제품의 경우 저장유무보다는 조리온도에 따라 풍미패턴의 차이가 더 구분되었으며 피자용 모짜렐라 슈레드 제품에 혼합용으로 사용되는 치즈의 종류 및 원산지에 따라서 풍미패턴이 각각 다른 것으로 나타났다. 또한 새로 개발된 국내산 임실 치즈는 기타 국내산 치즈와 외국산 치즈와 비교하였을 때 이들의 중간 향에 해당하면서도 독특한 향미를 지니는 것으로 판별되었다.
In this study, we developed kinetic models to predict the growth of pathogenic Escherichia coli on cheeses during storage at constant and changing temperatures. A five-strain mixture of pathogenic E. coli was inoculated onto natural cheeses (Brie and Camembert) and processed cheeses (sliced Mozzarella and sliced Cheddar) at 3 to 4 log CFU/g. The inoculated cheeses were stored at 4, 10, 15, 25, and $30^{\circ}C$ for 1 to 320 h, with a different storage time being used for each temperature. Total bacteria and E. coli cells were enumerated on tryptic soy agar and MacConkey sorbitol agar, respectively. E. coli growth data were fitted to the Baranyi model to calculate the maximum specific growth rate (${\mu}_{max}$; log CFU/g/h), lag phase duration (LPD; h), lower asymptote (log CFU/g), and upper asymptote (log CFU/g). The kinetic parameters were then analyzed as a function of storage temperature, using the square root model, polynomial equation, and linear equation. A dynamic model was also developed for varying temperature. The model performance was evaluated against observed data, and the root mean square error (RMSE) was calculated. At $4^{\circ}C$, E. coli cell growth was not observed on any cheese. However, E. coli growth was observed at $10{\circ}C$ to $30^{\circ}C$C with a ${\mu}_{max}$ of 0.01 to 1.03 log CFU/g/h, depending on the cheese. The ${\mu}_{max}$ values increased as temperature increased, while LPD values decreased, and ${\mu}_{max}$ and LPD values were different among the four types of cheese. The developed models showed adequate performance (RMSE = 0.176-0.337), indicating that these models should be useful for describing the growth kinetics of E. coli on various cheeses.
Twenty-five commercial food grade and alalytical grade lipases were used to study the patterns of release of short-chain free fatty acids (FFA) from milk fat in cheese slurries. Principal component Analysis showed that there were four distinctive groups by the FFA ratios and five groups by the FFA concentrations. However, Average Linkage Cluster Analysis showed that the patterns of FFA released were dependent upon distance defined between groups of lipases. All the lipases tested with both statistical analysis had distinctive specificities in hydrolyzing short-chain FFA from milk fat. Lipases from ruminant-animal origins produced an extremely high ratio (>40%) of butyric acid and a low ratio (<26%) of capric acid to total short chain FFA. Lipases from porcinepancreas and some microbial origins showed balanced production in both bytyric and capric acid. However, most lipases from microbial origins released a high ratio of capric acid but similar ratios to other origin enzymes for short-chain free fatty acids. Ruminant-animal origin lipases produced short-chain FFA much higher in concentration than other lipases. Lipases from porcine pancreas as well as microbial origins showed different concentrations of the fatty acids. Ratios of short-chain FFA in each sample were not significantly changed during incubation periods (4 wk), whereas concentrations of the FFA increased considerably.
This study was performed to examine the levels of biogenic amines (BAs) in foods of animal origin such as egg, ham, sausage, milk, cheese and yoghurt distributed on Korean domestic markets, and to compare the results with those of western countries. Egg yolk had more BAs than egg white. BAs detected in ham samples were the highest level in barbecued tender loin ham. Sausage samples had less BAs than ham samples. However, the delicatessen sausages had significantly higher levels of histamine than the other sausage samples (p<0.001). Cadaverine, spermidine and spermine were detected in market milks and their concentrations were very low compared with other samples. In Cheddar cheese, tyramine (44.46${\pm}$0.83 ${\mu}$g/g) was the major BA. The major BA of stirred yoghurt samples was histamine. These results suggest that BAs of the foods of animal origin distributed on Korean domestic markets were not much different from those of western countries and would not cause any harmful effect to consumers.

LL3를 Cheddar 치즈에 첨가하기 위하여 본 균의 균체생산을 위한 최적배양 온도와 pH 및 배양시간에 따른 casein의 분해형태를 관찰하였다. 본 균주의 생육을 위한 최적 배양온도는 $30^{\circ}C$, 단백분해 효과의 최적 배양온도는 $37^{\circ}C$이었으며, 초기 배양 pH 7.0에서 균의 생육과 단백분해 효과가 가장 좋았다. 배양기간 중 정지기 초기의 세포에서 aminopeptidase의 효소활성이 가장 높았으며, 본 효소의 열 안정성이 높아 $50^{\circ}C$에서 20분간 열처리 후에도 높은 효소활성을 보였다. Casein의 분해속도는 배양 24에서 36시간에서 가장 높았으며, 전기영동을 통한 casein hydrolysis patterns는 ${\alpha}-casein $뿐만 아니라 ${\beta}-casein$도 본 균주에 의해 완전히 분해되었다. 특히 본 균주는 ${\beta}-casein$의 분해력이 더욱 우수하였다.
Cheddar치즈의 숙성기간을 단축시키고 flavor를 증진시킬 목적으로 단백분해력이 있는 Micrococcus sp. LL3를 Cheddar 치즈에 첨가하기 위하여 본 균의 최적배양을 위한 배지조성을 검토하였다. 탄소원으로는 glucose, mannose, fructose 등의 단당류가 양호하였으며, arabinose, xylose는 균의 성장을 심하게 저해하였다. 본 균주에서는 탄소원의 첨가에 의한 catabolite repression은 나타나지 않았다. 또한 질소원으로는 yeast extract 0.2%의 첨가가 균의 성장에 좋았으며, 무기 질소원 첨가시는 균의 성장이 감소하였다. 특히 urea를 첨가시 균의 성장에 심한 저해효과가 있었다. 본 균주는 NaCl 9%첨가시 까지도 균의 생육이 가능했으며, 특히 1%첨가시에는 균체생육 및 단백분해 효과가 다소 증가하였다. 무기염의 경우 $MgSO_4\;0.05%$, 아미노산은 glutamic acid 0.2%첨가시 효과가 좋았다. 그러나 비타민은 $0.1\;{\mu}g/ml$ 수준으로 첨가시에 균의 성장 및 단백분해 효과에 영향을 미치지 않았다.

The distributions and content of geometrical isomers of conjugated linoleic acids (CLA) in dairy foods such as milk, yogurt, and cheese, produced or being sold in the Quebec province of Canada, were investigated by gas chromatographic analysis. The mean contents of total CLA (mg/g fat) were $5.06{\pm}0.74$ in 4 low-fat milk samples, $14.14{\pm}4.95$ in 6 yogurt samples, and $18.22{\pm}7.89$ in 5 natural ripened cheeses. Among the yogurt samples, YY contained the highest content of total CLA ($20.68{\pm}5.17\;mg/g$ fat). Among the cheese samples, Gruyere contained the highest amount of total CLA ($29.86{\pm}0.62$) as well as c-9,t-11 ($22.03{\pm}0.36\;mg/g$ fat), followed by Jarlsberg ($22.76{\pm}0.14$), Provolone ($16.42{\pm}0.52$), Cheddar ($13.83{\pm}0.81$), and Swiss ($8.23{\pm}1.11$). Based on the distribution ratios of CLA isomers in these dairy foods, the c-9,t-11 isomer appeared to be the major CLA isomer in both the low-fat milk ($89.87{\pm}2.39%$) and yogurt ($90.98{\pm}4.42%$). In the cheeses, however, the ratio of c-9,t-11 ($54.86{\pm}13.06%$) was slightly higher than that of c-10,c-12 ($40.81{\pm}13.40%$).
Efforts to have children consume milk continue across the world for the sufficient supply of calcium and protein. This study examines the frequency and amount of dairy milk and the recognition, acceptability, and knowledge of milk and dairy products by considering fifth- and sixth-graders in Hwaseong, Korea. The acceptability of milk and dairy products measured based on a five-point Likert-type scale was assessed to determine its correlations with breakfast frequency, the frequency of milk intake, and milk knowledge. According to descriptive statistics, 47% and 53% of the subjects were fifth- and sixth-graders, respectively, 53% and 47% were male and female, respectively, 45.3% and 51.2% had mothers in their thirties and forties, respectively, and 57% had working mothers. Breakfast frequency was 5.2 and 5.3 times per week, and milk and dairy intake was 4.6 times. The milk knowledge score was 5.3 out of 10 points. The average daily intake of milk was a cup (55.1%), and a vast majority preferred white milk (68.6%). Ice cream bar type (4.49 points) and cone type (4.48 points) showed the highest acceptability, and breakfast frequency was positively correlated with white milk and cheddar cheese (p<0.01). Daily milk intake frequency was positively correlated with white milk and fortified milk. Milk knowledge was positively correlation with white milk, fortified milk, functional yogurt (p <0.05), fermented milk, and cheese (p<0.01). The higher the breakfast frequency, the more likely the intake of white milk (p<0.01). An increase in milk intake increased milk knowledge (p<0.05). The higher the level of knowledge, the more likely the intake of fortified milk and the frequency of milk intake. These results suggest that school lunch milk may increase awareness and milk intake through continuing education in milk knowledge. High acceptability scores for white milk, fortified milk, and fermented milk indicate their suitability for school milk consumption.
