Anti-obese Effects and Signaling Mechanisms of Chaenomeles sinensis extracts in 3T3-L1 Preadipocytes and Obese Mice Fed a High-fat Diet (3T3L-1 지방전구세포와 고지방식이로 유도된 비만 마우스 모델에서 모과 추출물의 항비만 효과와 억제 기전)

  • Obesity is one of the most serious health problem because it induced numerous metabolic syndrome and increases the incidence of various disease, including diabetes, hypertension, dyslipidemia, atherosclerosis, and cancer. In 3T3-L1 adipocytes, increases in reactive oxygens species (ROS) occur with lipid accumulation. NADPH oxidase, producing superoxide anion, may contribute to the development of obesity-associated insulin resistance and type 2 diabetes. In this study, we elucidated the effect of Chaenomeles sinensis koehne extract (CSE) against the development of obesity and the inhibition mechanisms in 3T3-L1 preadiocytes. CSE decreased triglyceride content and inhibited the expression of adipogenic transcription factors including peroxisome proliferator-activated receptor $(PPAR){\gamma}$, CCAT/enhancer binding protein $(C/EBP){\alpha}$ and sterol regulatory element-binding protein (SREBP-1). In addition, CSE highly increased antioxidant activity in a dose-dependent manner. CSE remarkably reduced intracellular ROS increase and NAD(P)H oxidase activity, NOX1, NOX4, Rac1 protein expression, and phosphorylation of p47phox and p67phox We also studied the effect of CSE on weight gain induced by high-fat diet. The oral treatment of CSE (500 mg/kg, body weight) in diet-induced obese (DIO) mice showed decrease in triglyceride and adipocyte size. Therefore, these results indicate that the effect of CSE on anti-obese effects, adipocyte differentiation and reducing triglyceride contents as well as adipocyte size in obese mice, may be associated with inhibition of NAD(P)H oxidase-induced ROS production and adipose transcription factors. These results showed the potential to inhibit the obesity by CSE treatment through controlling the activation of NAD(P)H oxidase in vitro and in vivo obese model.

Inhibitory effect of Chaenomelis Fructus ethanol extract on receptor activator of nuclear factor-kappa B ligand-mediated osteoclastogenesis

  • The fruit of Chaenomeles sinensis (Thouin) Koehne (Chaenomelis Fructus) known as "Mo-Gua" in Korea has been commonly used in traditional medicine to treat inflammatory diseases, such as sore throat. However, its effect on bone metabolism has not been elucidated yet. Here, we examined the effect of Chaenomelis Fructus ethanol extract (CF-E) on receptor activator of nuclear factor (NF)-κB ligand (RANKL)-mediated osteoclast differentiation and formation. CF-E considerably inhibited osteoclast differentiation and tartrate-resistant acid phosphatase-positive multinuclear cell formation from bone marrow-derived macrophages and osteoclast precursor cells in a dose-dependent manner. In addition, the formation of actin rings and resorption pits were significantly suppressed in CF-E-treated osteoclasts as compared with the findings in non-treated control cells. Consistent with these phenotypic inhibitory results, the expressions of osteoclast differentiation marker genes (Acp5, Atp6v0d2, Oscar, CtsK, and Tm7sf4) and Nfatc1, a pivotal transcription factor for osteoclastogenesis, were markedly decreased by CF-E treatment. The inhibitory effect of CF-E on RANKL-induced osteoclastogenesis was associated with the suppression of NFATc1 expression, not by regulation of mitogen-activated protein kinases and NF-κB activation but by the inactivation of phospholipase C gamma 1 and 2. These results indicate that CF-E has an inhibitory effect on osteoclast differentiation and formation, and they suggest the possibility of CF-E as a traditional therapeutic agent against bone-resorptive diseases, such as osteoporosis, rheumatoid arthritis, and periodontitis.

Protective Effects of Chaenomeles sinensis Koehne Extract on Ethanol-induced Liver Damage in Rat (에탄올에 의해 유발된 간독성에 대한 모과 추출물의 보호효과)

  • This study was performed to investigate the effect of ethanol extract of Chaenomeles sinensis Koehne (CS) on alcohol-induced liver damage in rats. Male Sprague-Dawley rats weighing $135{\pm}10g$ were divided into 6 groups for 4 weeks; normal group (ND), alcohol (35%, 10 mL/kg/day) treated group (ET), CS ethanol extract 200 mg/kg/day treated group (ND-CSL), CS ethanol extract 400 mg/kg/day treated group (ND-CSH), CS ethanol extract 200 mg/kg/day and alcohol treated group (ET-CSL), and CS ethanol extract 400 mg/kg/day and alcohol treated group (ET-CSH). The body weight gain and food efficiency ratio were no differences between ND and ET. There were increases in the activities of serum alanine aminotransferase (ALT), asparate aminotransferase (AST), and alkaline phosphatase (ALP) in ET. On the other hand, the administration of CS decreased ALT, AST and ALP activities in serum. It was also observed that the hepatic activities of superoxide dismutase (SOD), catalase, glutathione peroxidase (GSH-Px) and xanthine oxidase (XO) increased by alcohol treatment were also markedly decreased in the CS administered groups as compared with ET. The activities of hepatic SOD, catalase, GSH-Px and XO were riot significantly different among the normal diet groups. Contents of thiobarbituric acid reactive substances (TBARS) were increased by the administration of alcohol, on the other hand, the administration of CS reduced TBARS value in the liver. In addition, the content of glutathione (GSH) in the liver was decreased by alcohol administration, however, GSH increased after administering CS. In conclusion, the administration of alcohol develops the hyperoxidation of liver lipids through tile increase in enzymes activity related to the lipid peroxiation, however, it was decreased after administring CS. Thus, CS may have a possible protective effect on ethanol-induced hepatotoxicity in rat liver.

Nonvolatile Flavor Components in Chinese Quince Fruits, Chaenomeles sinensis koehne (모과의 비휘발성 Flavor 성분에 관한 연구)

  • This study was performed to identify detailed informations on the nonvolatile flavor of Chinese quince fruits, Chaenomeles sinensis koehne. About 72% of the free amino acids were shown to be valine, asparagine, ${\gamma}-aminobutyric\;acid$, aspartic acid and serine. Arginine, tyrosine, methionine and tryptophan were not present. Glutamic acid and glutamine as a amino acid for peptides were the major components, whereas cysteic acid, methionine sulfone and tryptophan were not detected. The nucleotides attained were composed of cytosine, uridine-5'-monophosphate and cytidine-5'-monophosphate, and these were proved to be a very small quantity. Guanosine-5'-monophosphate, inosine-5'-monophosphate and adenosine-5'-monophosphate were not present. The major sugars were shown to be glucose, sorbose, sucrose and fructose. Fructose was the most abundant one among them. A total of 11 organic acids were identified by capillary gas chromatography and capillary gas chromatography-mass spectrometry. The major components identified were tartaric acid and α-ketoglutaric acid. The total content of vitamin C determined was 386.6mg%, and those of ascorbic, dehydroascorbic, and 2, 3-diketo-L-gulonic acid were 28.8mg%, 154.5mg% and 197.3mg%, respectively. Calcium and phosphorus were the major components, while heavy metals such as cadmium, copper and lead were determined to be a small amount. In the result of organoleptic test on the natural and synthetic extract of Chinese quince fruits, the principal taste components consisted of free amino acids, sugars, organic acids, vitamin C and minerals. Five groups mentioned would have a favorable influence upon the taste of fresh Chinese quince fruits.

Toxicity of various fruit tree extracts to five agricultural and four stored-product anthropod pests (다양한 과일나무에서 유래된 추출물의 농업해충 및 저장물해충에 대한 살충활성)

  • Methanol extracts from 43 materials of 17 fruit trees were tested for their insecticidal activities toward five agricultural and four stored-product anthropod pests. Efficacy varied with insect species, plant species, and tissue sampled. At a concentration of 2,500 ppm, strong activity was observed with extracts of Chaenomeles sinensis and Punica granatum seeds against Myzus persicae adult females, Vitis vinifera seeds against Nilaparvata lugens adults, Citrus junos, Citrus paradisi, Cucumis melo Linne var. reticulatus, Diospyros kaki, Malus pumila var. dulcissima, Prunug persica, and V. vinifera seeds against Plutella xylostella, Citrus sinensis seeds against Spedoptera litura larvae, and Ch. sinensis and V. vinifera against Tetranychus urticae adults. Against four stored-product insect pests at 50 ppm, seed extracts of Ch. sinensis and V. vinifera against Sitophilus oryzae adults, and Ch. sinensis, C. junos, D. kaki, and V. vinifera against Callosobruchus chinensis adults gave over 80% mortality. Extracts of all samples exhibited little and no activity against Lasioderma serricorne adults and Plodia interpunctella larvae.

Verification of aecial host ranges of four Gymnosporangium species based on artificial inoculation.

  • Aecial host ranges of four Gymnosporangium species causing cedar-apple rust diseases, G. asiaticum, G. cornutum, 5. japonicum and G. yamadae, were investigated through artificial inoculation. Thirteen species of nine genera among Rosaceous plants, which have been reported as social hosts in Korea, were inoculated with fresh teliospores spores in early days of May of 2000 and of 2001, respectively. In the results, we re-confirmed that there was highly specific relationship between the rust species and aecial hosts and report new aecial hosts of four Gymnosporangium species. Teliospores of G. cornutum collected from Juniperus rigida successively produced spermogonia and aecia only on Sorbus alntifolia, the first report on host alteration of G. cornutum in Korea. Positive responses by teliospores of G. japonicum from J. chinenis of Suwon and from J. chinenis var. horizontalis of Jeju island were obtained only on P. villosa. Crataegus pinnatifida was confirmed as a new aecial host of G. viatium. Until this time, G. ymadae was believed to have Malus as the aecial host. However, teliospores of G. yamadae collected from J. chinensis var. kaizuka successively formed spermogonia and aecia on the leaves of Chaenomeles lagenaria, C. sinensis, Pyrus pyrtifolia var, culta, P. ussuriensis, Malus pumila and M. sileboldii. The date for maturation of spermogonia and aecia, and symptom development varied according to the rust fungi and aecial host plants, respectively.

Screening for Antimicrobial Activity from Korean Plants (국내산 식물의 항균활성 검색)

  • 141 methanol extracts from 125 plant species which populate in Korea were screened for antimicrobial activity against various food-borne pathogens and food spoilage microorganisms. Those plants were selected from 3 different plant groups: traditional herbs, edible plants and flowers. The methanol extracts were tested by using the disk diffusion assay against five bacteria: Bacillus subtilis, Staphylococcus aureus, Pseudomonas aeruginosa, Enterobacter aerogenes, Escherichia coli. From the evaluation of the inhibition zone diameter of microbial growth, the most significant antimicrobial activity against Bacillus subtilis, Staphylococcus auresus, Pseudomonas aeruginosa, Enterobacter aerogenes, Escherichia coli was observed from the extract of Schizandra chinensis (Turcz.) Baill., Rheum officinale Baill., Schizandra chinensis (Turcz.) Baill., Koelreuteria paniculata Lax and Crataegus pinnatifida Bunge, respectively. The extract from many plants - Koelreuteria paniculata Lax, Chaenomeles sinensis Koehne, Scutellaria bacicalensis Georgi, Castanea crenata Sieb. et Zucc., Rosa centifolia L., Allium fistulosum L. var. giganteum Makino, Crataegus pinnatifida Bunge, Schizandra chinensis (Turcz.) Baill., Lonicera japonica - showed antimicrobial activity all four tested bacteria.

  • Stomatal morphological characteristics (shape, density and length) were investigated for 8 ornamental woody species growing in the campus of Sangji College on Usandong, Wonju, Kangwondo. Stomata of Forsythia koreana, Syringa dialata, Zedkova serrata and Chaenomeles sinensis were classified as irregular-celled type (anomocytic), those of Vibrunum sargentii, Liriodendron tulipifera and Magnolia kobus as parallel-celled type (paracytic). For Forsythia koreana, stomatal length was not greatly changed as the leaf blade, growing larger. Number of stomata per unit leaf area was decreased, but number of stomata per single leaf was increased as the leaf blade, growing larger. For Taxus cuspidata, stomatal length was increased, but number of stomata per unit needle area was decreased as the needle, growing larger.

  • This study was carried out to survey and analyze the transition of landscape plants on an apartment complexes in the 1990's and 2000's. The results are summarized as follows; The ratio of landscape area was increased from 30.0% to 34.7%. The number of trees per 100 square meter decreased from 22.5 to 15.7. The number of shrub per 10 square meter increased from 19.3 to 38.7. Species of Evergreen tree were increased 21 to 39. The species that a lot of use continuously were Pinus densiflora, Pinus strobus and Taxus cuspidata. The species that increase use were Abies holophylla and Pinus densiflora for. multicaulis. The species that decrease use exceedingly, were Pinus parviflora, Juniperus chinensis, Pinus koraiensis and Thuja orientalis. The species that do not use were Cedrus deodara. Species of deciduous tree were increased 42 to 68. The species that a lot of use continuously were Zelkova serrata, Acer palmatum, Diospyros kaki, Prunus armeniaca and Prunus yedoensis. The species that increase use were Lagerstroemia indica, Chionanthus retusa, Cornus officinalis, Styrax japonica, Sorbus alnifolia, Prunus mume, Cercidiphyllum japonicum, Acer triflorum, Prunus sargentii, Chaenomeles sinensis and Cornus kousa. The species that decrease use exceedingly, were Acer buergerianum, Sophora japonica, Malus spp., Ginkgo biloba, Zizyphus jujuba var. inermis, Platanus orientalis and Albizzia julibrissin. The species that do not use were Liriodendron tulipifera, Acer saccharinum, Ailanthus altissima and Paulownia coreana. Species of shrub were incresed 39 to 65. The species that a lot of use continuously were Buxus koreana, Rhododendron schlippenbachii for. albiflorum, Rhododendron schlippenbachii, Syringa dilatata and Euonymus japonica. The species that increase use were Rhododendron indicum, Ilex serrata, Spiraea prunifolia var. simpliciflora, Taxus cuspidata var. nana, Kerria japonica, Rhododendron yedoense var. poukhanense, Euonymus alatus, Sorbaria sorbifolia var. stellipila, Nandina domestica, Cornus alba, Hydrangea serrata for. acuminata, Prunus tomentosa, Deutzia parviflora and Pyracantha angustifolia. The species that decrease use exceedingly, were Hibiscus syriacus, Rosa multiflora var. platyphylla, Chaenomeles lagenaria and Rosa spp. The species that do not use were Juniperus chinensis var. sargentii, Jasminum nudiflorum and Sasa borealis.

Effects of Pretreatment on the Field Germination Rate of Some Tree Seed (종자(種子)의 전처리(前處理)가 몇 수종(樹種)의 포장발아율(圃場發芽率)에 미치는 영향(影響))

  • To seek for a proper pretreatment method for increasing field germination rate, field germination rate of 5 tree species including Phellodendron amurense was investigated and was compared among pretreatment methods. The results obtained in this study were as follows ; 1. Field germination rates of Poncirus trifoliata and Fraxinus rhynchophylla among pretreatment methods were significantly different at 1% level, and that of Phellodendron amurense among pretreatment methods were significantly different at 5% level. In thses species, highest field germination rate was observed on the plot of cold moist stratification under the ground (Nochunmaejang ; during 166 days). 2. Field germination rates of Chaenomeles sinensis among pretreatment methods were significantly different at 1% level, and highest field germination rate was observed on the plot of cold moist stratification in the refrigerator (during 110days). 3. Field germination rates of Cudrania tricuspidata among pretreatment methods were not significantly different at 5% level.
