Purification and Characterization of a Cytochrome P-450 from Pravastatin-Producing Streptomyces sp. Y-110.

  • Streptomyces sp. Y-110 cytochrome P-450, induced by the addition of compactin -Na into the culture medium, was purified from the cell extract to apparent homogeniety, mainly by DEAE-Sepharose, hydroxyapatite, and Mono Q column chromatyography. The sepcific activity of purified enzyme on its substrate, compactin-Na, was determined to be 15 nmol of pravastatin per mg protein. The molecular mass of this enzyme on SDS-PAGE was $37{\pm}0.5$ kDa, pI was 4.5, and its CO difference spectrum showed maximum absorption peaks at 452 and 550nm, respectively. The N-terminal amino acid sequence was determined to be Met>Thr>Cys>Thr>Pro>Val>Thr>Val>The>Gly>Ala>Ala>Gly>Gln>Ile>Gly>Tyr>Ala>Leu. Its apparent $K_m$ on compactin-Na was $1.294{\mu}M{\cdot}min^-1,\;and\;V_{max}\;was\;1.028{\mu}M{\cdot}min^-1$. The maximum substrate concentration ($K_s$) for reaction was $270 {\mu}M$and thus $1/[K_s]$ was $3.7{\mu}M$. These physicochemical characteristics and kinetic behavior of this enzyme were compared and shown to be different from those of Streptomyces cytochrome P-450 enzymes reported, suggesting that this enzyme may be an additional member of the Streptomyces cytochrome P-450 family.

  • Walking on split-belt treadmill has been applied to study walking disabilities, such as osteoarthritis (OA), to show asymmetric walking characteristics. In this study, we compared asymmetric walking in OA patients with healthy subjects under split-belt conditions and examined the reproduction of walking asymmetry in OA. Seven OA patients were instructed to walk at four frequencies, while four healthy subjects walked on a treadmill with tied-belt and split-belt conditions. To compare walking asymmetries, kinetic and kinematic measurements were made using force-plates and motion capture cameras, and subsequently center of mass (CoM) velocity, mechanical work and potential energy were calculated. Horizontal velocity change during split-belt walking of healthy subjects was similar to OA patients. Difference of mechanical work during single support phase occurred due to fall of CoM in fast belt. OA walking asymmetry could be reproduced by reducing differences of belt speeds to prevent rapid fall of CoM.

시각정보 유입 특성에 따른 정적 기립균형 분석 (Analysis of the Static Uprighting Balance in the Visual Input Characteristics)

  • Purpose : The current study examines changes of static uprighting balance in the visual input characteristics. Method : Total 50 person(male 16, female 34) were participated in this study. They were tested with 'hole in the card' for identification of dominant eye's side, then they were divided 3 groups(both visual input group, dominant visual input group, and non-dominant visual input group). 3 groups were measured with Romberg test on the force platform device to compare the static uprighting balance characteristics ; moving distance, mean velocity, and sway area of the CoM(center of mass), during 20 seconds. Results : The results by one-way repeated measure ANOVA were as follows. In moving distance and mean velocity of CoM, non-dominant visual input group was unstable than dominant visual group and both visual input group(p<0.05). But, in sway area of CoM, significant difference was not existed statistically. Conclusion : These result can be applied to design the static uprighting balance program using visual input mediation.

Effects of Melatonin on Gene Expression of IVM/IVF Porcine Embryos

  • The effect of melatonin on in vitro embryo development and the expression of antioxidant enzyme gene in preimplantation porcine embryos was determined by modified semi-quantitative single cell RT-PCR. Porcine embryos derived from in vitro maturation /in vitro fertilization were cultured in 5% $CO_2$ and 20% $O_2$ at $37^{\circ}C$ in NCSU23 medium. Melatonin was added to medium at concentration of 1nM, 5 nM, and 10 nM. When treated with 1nM (39.0%) of melatonin, the developmental rate of embryos beyond the morula stage were higher than that of control group (31.0%) (p<0.05). Number of inner cell mass and tropectoderm cell in control (23.0${\pm}$0.5 and 17.3${\pm}$0.8), 1 nM (23.6${\pm}$0.6 and 19.0${\pm}$0.5), and 5 nM (23.3${\pm}$1.1 and 16.3${\pm}$0.8) treated with melatonin were higher than in 10 nM (20.0${\pm}$0.5 and 13.3${\pm}$0.8) treated with melatonin (p<0.05). To develop an mRNA phenotypic map for the expression of catalase, bax and caspase-3, single cell RT-PCR analysis were carried out in porcine IVM/IVF embryo. Catalase was detected in 0, 1 and 5 nM supplemented with melatonin, but bax and caspase-3 were detected in 10 nM treated with melatonin.

Hyperthermal Collision-induced Dissociation of Bromotoluene Radical Cations at Self-Assembled Monolayer Surfaces

  • Hyperthermal ion/surface collisions of bromotoluene radical cations were studied using perfluorinated (F-SAM) and hydroxyl-terminated (OH-SAM) self-assembled monolayer surfaces in a tandem mass spectrometer with BEEQ geometry. The isomers were differentiated by ion abundance ratios taken from surface-induced dissociation (SID). The dissociation rate followed the order of ortho > meta > para isomers. The peak abundance ratio of m/z 51 to m/z 65 showed the best result to discern the isomers. A dissociation channel leading to tolylium ion was suggested to be responsible for the pronounced isomeric differences. The capability of SID to provide high-energy activation with narrow internal energy distribution may have channeled the reaction into the specific dissociation pathway, also facilitating small differences in reaction rates to be effective in the spectral time window of this experiment. All of the molecular ions experiencing reactive collisions with the F-SAM surface undergo transhalogenation, in which a fluorine atom on the surface replaces the bromine in the incoming ions. This reactive collision was dependent on the laboratory collision energy occurring in ca. 40.75 eV range.

How do dense cores embedded in a pc scale filamentary clouds form, by gas flow motions along filamentary clouds and/or contracting motions by themselves?

  • Understanding how the filamentary structure plays a role in the formation of the prestellar cores and stars is a key issue to challenge. We have observed two prestellar cores in surrounding filamentary environments in 13CO, C180 (3-2) and HCO+ (4-3) molecular lines with the Heterodyne Array Receiver Program (HARP) of the James Clerk Maxwell Telescope (JCMT), in order to search for the evidence related to the possible flow motions along the filament and/or the radial accretion (or infalling motions) of gas material toward the dense cores from their surrounding filamentary cloud. In L1544, the velocity gradient of 1.6 km s-1 pc-1 toward the core was measured in a small branch of filament lying on a radial direction of main filament while no velocity gradient along the main axis of filament in both 13CO and C18O lines. In L694-2, we found the velocity gradient of 0.6 km s-1 pc-1 along the filament in only 13CO lines. The projected accretion rate of ~6 M◉ Myr-1 was estimated in both cases. The infall (or radially contracting) velocity of gas material was measured ~0.16 km s-1 in both 13CO and HCO+ lines and in both L1544 and L694-2, which leads to estimate a mass infall rate of ~20 M◉ Myr-1. Our analysis suggests that our targets are at a stage where the gravitational contraction dominates the mass accretion through the surrounding filamentary cloud. This is consistent with the fact that our targets are highly evolved prestellar cores on a verge of star formation. More detailed results will be presented at the meeting.

생장 및 부상성이 향상된 남세균 돌연변이 균주 Arthrospira platensis M20CJK3 (A Mutant Arthrospira platensis M20CJK3 Showing Enhanced Growth Rate and Floatation Activity)

  • 건강보조식품 등으로 이용되는 Arthrospira platensis는 세계적으로 대량생산되고 있으나 생산공정 중 수확단계에서 많은 비용이 소요된다. 본 연구에서는 부상을 이용한 효과적인 수확을 위해 균주의 개량을 시도하였으며, 개량균주의 생리적 물리적 특성을 파악하고자 하였다. Ethyl methanesulfonate (EMS)를 모균주 A. platensis KCTC AG20590에 0.24%의 농도로 10, 20, 30분씩 처리하여, 형태 및 부상성이 우수한 균주 A. platensis M20CJK3 균주를 분리하였다. A. platensis M20CJK3은 느슨한 형태에서 촘촘한 형태로 세포사(trichome)의 길이 및 코일간 간격이 감소하였으며, 생장 및 $CO_2$ 고정능이 각각 15%, 17% 향상되었다. 또한, 개량균주의 부상성은 모균주에 비해 2배 이상 향상되었다. 이차원 전기영동 분석을 통해 모균주와 개량균주의 단백질 발현양상을 비교분석한 결과 광합성 관련 색소의 구조와 광전자전달계에 관련된 단백질의 발현 양상이 차이를 보였다. 본 연구에서 개발된 A. platensis M20CJK3은 고밀도 대량배양 및 수확에 유리하며, A. platensis 유전자 연구의 유용한 모델 균주로 활용될 수 있을 것으로 사료된다.

Hydrophillic Interaction Chromatography-tandem Mass Spectrometry Method for Identification and Quantitation of 5-MeO-DIPT and its Metabolites in Rat Urine

  • 5-Methoxy-N,N-diisopropyltryptamine (5-MeO-DIPT), a psychoactive tryptamine derivative, is a hallucinogenic drug of abuse. In this study, 5-OH-DIPT and its metabolites were identified and the quantitative method was developed and validated by using hydrophilic interaction chromatography-tandem mass spectrometry (HILICMS/MS). Chromatographic separation was achieved on an Atlantis HILIC silica column ($5{\mu}m$, $100{\times}2.1\;mm$). The metabolites of 5-MeO-DIPT in rat urine were characterized via Q1 scanning and product ion scanning. As a consequence, 5-MeO-IPT, 5-OH-DIPT, 6-OH-5-MeO-DIPT and their glucuronide conjugates were detected and identified as the metabolites of 5-MeO-DIPT. Subsequently, a quantitative method for 5-MeO-DIPT and its major metabolites, 5-MeO-IPT and 5-OH-DIPT, was developed in multiple reactions monitoring (MRM) mode. The calibration curves for all analytes evidenced good linearity over the concentration range of 1-1000 ng/mL with linear correlation co-efficients ($r^2$) in excess of 0.99. The intra- and inter-day accuracy and precision were 92.2-110.2% and 1.5-9.9%, respectively.

Large Scale Structures at z~1 in SA22 Field and Environmental Dependence of Galaxy Properties

  • We study galaxy evolution with the large-scale environment with confirmed galaxy clusters from multi-object spectroscopy (MOS) observation. The observation was performed with Inamori Magellan Areal Camera and Spectrograph (IMACS) mounted on the 6.5 m Magellan/Baade telescope in Las Campanas Observatory. With the MOS observation, we spectroscopically confirm 34 galaxy clusters, including three galaxy clusters discovered in Kim et al. (2016) and 11 of them have halo mass of > 1014.5 M. Among the confirmed clusters, 12 galaxy clusters are part of large-scale structure at z ~ 0.9, and their size stretches to 40 Mpc co-moving scale. In this study, we checked the 'web feeding model,' which postulates that more linked (with their environment) galaxy clusters have less quenched populations by investigating the correlation between properties of confirmed galaxy clusters and the large-scale structure environment. Lastly, we found that galaxy clusters that make up the large-scale structure have larger and widely spread values of total star formation density (ΣSFR/Mhalo) than typical clusters at similar redshifts.

Relationship between Center of Pressure and Local Stability of the Lower Joints during Walking in the Elderly Women

  • Objective: The purpose of this study was to determine the relationship between center of pressure (CoP) and local stability of the lower joints, which was calculated based on approximate entropy (ApEn) during walking in elderly women. Method: Eighteen elderly women were recruited (age: $66.4{\pm}1.2yrs$; mass: $55.4{\pm}8.3kg$; height: $1.56{\pm}0.04m$) for this study. Before collecting data, reflective marker triads composed of 3 non-collinear spheres were attached to the lateral surface of the thigh and shank near the mid-segment to measure motion of the thigh and shank segments. To measure foot motion, reflective markers were placed on the shoe at the heel, head of the fifth metatarsal, and lateral malleolus, and were also placed on the right anterior-superior iliac spine, left anterior-superior iliac spine, and sacrum to observe pelvic motion. During treadmill walking, kinematic data were recorded using 6 infrared cameras (Oqus 300, Qualisys, Sweden) with a 100 Hz sampling frequency and kinetic data were collected from a treadmill (Instrumented Treadmill, Bertec, USA) for 20 strides. From kinematic data, 3D angles of the lower extremity's joint were calculated using Cardan technique and then ApEn were computed for their angles to evaluate local stability. Range of CoP was determined from the kinetic data. Pearson product-moment and Spearman rank correlation coefficient were applied to find relationship between CoP and ApEn. The level of significance was determined at p<.05. Results: There was a negative linear correlation between CoP and ApEn of hip joint adduction-abduction motion (p<.05), but ApEn of other joint motion did not affect the CoP. Conclusion: It was conjectured that ApEn, local stability index, for adduction/abduction of the hip joint during walking could be useful as a fall predictor.
