Anti-oxidative Activities of Commercial Edible Plant Extracts Distributed in Korea (국내 유통 중인 식용식물 추출물의 항산화효과)

  • Many plant extracts are known to have antioxidative effects. However, their activities can be reduced or disappeared during mass production process. The purpose of this study is to compare antioxidative effects of edible plant extracts distributed in Korea. forty three kinds of edible plant extracts commercially available in Korea were selected and investigated for their total phenolics contents and antioxidative potentials(DPPH radical and superoxide anion radical scavenging activities). In contents of total phenolics, the commercial plant extracts from Artemisia annua(whole plant), Ilex paraguariensis(leaf, Silybum marianum(fruit and leaf, Ulmus pumila(bark), Coliolus versicolor(fruit), and Curcuma longa(root and stem) contained over 70 mg/g of powder, DPPH radical scavenging activities($SC_{50}$, 50% scavenging concentration) of A. annua, I. paraguariensis, Pinus densiflora(leaf),S. marianum, U. pumila, and C. longa were $53.96{\pm}0.81\;ppm,\;24.61{\pm}2.12\;ppm,\;35.96{\pm}1.11\;ppm,\;57.46{\pm}2.13\;ppm,\;55.25{\pm}1.65\;ppm\;and\;12.99{\pm}1.67ppm$, respectively, while that of positive control(vitamin C) was $3.86{\pm}0.81\;ppm$. $SC_{50}$ values against superoxide anion radical of A. annua, Cinnamomum zeylanicum(bark), I. paraguariensis, Rubus coreanus(fruit and leaf), Morus alba(leaf), P. densiflora, S. marianum, U. pumila, C. versicolor, C. longa, Perilla frutescens var. acuta(leaf), and H. sabdariffa(leaf and newer) were $53.21{\pm}1.83ppm,\;50.12{\pm}2.12ppm,\;5.59{\pm}0.84ppm,\;41.60{\pm}8.93ppm,\;20.19{\pm}0.97ppm,\;15.19{\pm}1.66ppm,\;21.20{\pm}1.88ppm,\;15.71{\pm}0.91ppm,\;55.48{\pm}2.42ppm,\;52.12{\pm}2.44ppm,\;23.80{\pm}1.98ppm\;and\;11.14{\pm}0.51ppm$, respectively($SC_{50}$ value of vitamin C: $9.61{\pm}0.93ppm$). In particular, both 1 paraguariensis and P. densiflora had high content of phenolics as well as high scavenging activities of DPPH radical and superoxide anion radical. Consequently, above two commercial extracts may be useful as a source of antioxidative nutraceutics.

  • Within the framework of DECOVALEX-2019 Task D, full-scale engineered barriers experiment (FEBEX) at Grimsel Test Site was numerically simulated to investigate an applicability of implemented Barcelona basic model (BBM) into TOUGH2-MP/FLAC3D simulator, which was developed for the prediction of the coupled thermo-hydro-mechanical behavior of bentonite buffer. And the calculated heater power, temperature, relative humidity, total stress, saturation, water content and dry density were compared with in situ data monitored in the various sections. In general, the calculated heater power and temperature provided a fairly good agreement with experimental observations, however, the difference between power of heater #1 and that of heater #2 could not captured in the numerical analysis. It is necessary to consider lamprophyre with low thermal conductivity around heater #1 and non-simplified installation progresses of bentonite blocks in the tunnel for better modeling results. The evolutions and distributions of relative humidity were well reproduced, but hydraulic model needs to be modified because the re-saturation process was relatively fast near the heaters. In case of stress evolutions due to the thermal and hydraulic expansions, the computed stress was in good agreement with the data. But, the stress is slightly higher than the measured in situ data at the early stage of the operation, because gap between rock mass and bentonite blocks have not been considered in the numerical simulations. The calculated distribution of saturation, water content, and dry density along the radial distance showed good agreement with the observations after the first and final dismantling. The calculated dry density near the center of the FEBEX tunnel and heaters were overestimated compared with the observations. As a result, the saturation and water content were underestimated with the measurements. Therefore, numerical model of permeability is needed to modify for the production of better numerical results. It will be possible to produce the better analysis results and more realistically predict the coupled THM behavior in the bentonite blocks by performing the additional studies and modifying the numerical model based on the results of this study.

  • In this study, physiological changes in a thermotolerant yeast Saccharomyces cerevisiae KNU5377 cell exposed to 48-hour alcohol fermentation at $40^{\circ}C$ were investigated. After 12 hours of alcohol fermentation at $40^{\circ}C$, the $C_{16:1}$ unsaturated acid of plasma membrane increased to 1.5 times more than the $C_{16:0}$ saturated fatty acid, and to about 2 times more for the $C_{18:1}$ unsaturated fatty acid. Fermentation at both $30^{\circ}C$ and $37^{\circ}C$ fermentation showed the same pattern as that done at $40^{\circ}C$. The pH of the alcohol-fermentation medium was reduced to pH 4.1 from a starting pH of 6.0 through the 12-hr fermentation and then maintained this level during the continuing fermentation. With the process of fermentation, the remaining glucose was reduced, but its amount remaining during the $40^{\circ}C$-fermentation was less reduced than those fermented at $30^{\circ}C$ and $37^{\circ}C$. In the study investigating the changing pattern of cellular proteins in the alcohol-fermenting cells, the SDS-PAGE and 2-D data indicated the most expressed dot was phosphoglycerate kinase, which is one enzyme involved in glycolysis. Why this enzyme was most expressed in the cells exposed to unfavorable conditions such as high temperature, increasing concentration of produced alcohol and long time exposure to other stress factors remains unsolved.

The Characteristics on the Spatial and Temporal Distribution of Phytoplankton in the Western Jinhae Bay, Korea (진해만 서부해역에서 식물플랑크톤의 시.공간적 분포특성)

  • We studied spatial and temporal distributions of the phytoplankton and their relationships to physico-chemical environmental factors in the western Jinhae Bay, Korea from November 2003 to August 2004. In most cases, physico-chemical environmental factors showed homogeneous distribution. The phytoplankton communities were composed of mainly diatoms and dinoflagellates, and their standing crops ranged from $16{\times}10^3\;cells\;l^{-1}\;to\;5,845{\times}10^3\;cells\;l^{-1}$ (with a mean value of $555{\times}10^3\;cells\;l^{-1}$). The bloom of phytoplankton was observed in Gohyun Port in the summer. Seasonal variation of phytoplankton standing crops was higher in winter and summer than in spring and autumn. The dominant species were Skeletonema costatum, Akashiwo sanguinea, Pseudo-nitzschia pungens, Dactyliosolen sp., Leptocylindrus danicus, cryptomonads and etc. Especially, S. costatum was predominant in the summer and A. sanguinea (spring and autumn), Pseudo-nitzschia sp. (summer), Guinardia striata (spring), unidentified flagellates (summer) and cryptomonads (spring) appeared to be an opportunistic species. Concentrations of Chl a ranged from $0.6{\mu}g{\cdot}l^{-1}\;to\;16.7{\mu}g{\cdot}l^{-1}$ (with a mean value of $3.4{\mu}g{\cdot}l^{-1}$). The results of the canonical correspondence analysis implies the study area was grouped into the 2 water masses (inner and outer waters of Gohyun Port) and inner waters had higher abundance and Chl a concentration than outer waters. Also, phytoplankton sanding crops were related with temperature, DO and nutrients ($SiO^2$, TN, TP and etc.) in inner waters. Inner water-mass of Gohyun Port expanded between Gacho Is. and Chilchon Is. during the winter.

The Influence of Breakfast Size to Metabolic Risk Factors (아침식사량이 대사위험요인에 미치는 영향)

  • Skipping breakfast is a risk factor closely related to metabolic syndrome and obesity. We analyzed the relationship between breakfast size, metabolic syndrome and obesity. The study included 5,548 adults who visited a health promotion center at Pusan National University from January to November of 2006. Subjects were divided into four groups according to breakfast size - skipper group (no breakfast), small intake group, medium intake group and large intake group. 959 (17.3%) of the 5548 subjects were included in the Skipper group. Intake of daily calories, proteins, carbohydrates and fats was the lowest in the Skipper group. Breakfast size increased linearly with an increased intake of daily calories, proteins, carbohydrates and fats. Body mass index ($23.4\;kg/m^2$) and waist circumference (79.6 cm) were the lowest in the Small intake group. In the Small intake group, triglycerides, fasting plasma glucose, systolic blood pressure and diastolic blood pressure were the lowest, and high density lipoprotein cholesterol levels were the highest. The number of metabolic risk factors was the lowest in Small intake group. Odds ratio of metabolic syndrome (Odds ratio=0.612) was the lowest in Small intake group. Along with increasing breakfast size, the odds ratio also increased. In this study, breakfast size was found to influence metabolic risk factors. Skipping breakfast worsened metabolic risk factors, while a small breakfast size had a favorable effect on metabolic risk factors.

Spatiotemporal Variations of Seawater Quality due to the Inflow of Discharge from Nakdong River Barrage (낙동강 하구둑 방류수에 의한 하구역 수질의 시공간적 변화)

  • We estimated the pollutant loads for the last 3 years based on the daily discharge at the Nakdong River dam(barrage) and spatiotemporal characteristics of seawater quality in the Nakdong river estuary to investigate the correlation between the pollutant load inflow rate and seawater quality. The main results from this research are summarized as follows. (1) The total discharge at the Nakdong River dam dam the last 11 years has been $224,576.8{\times}10^6m^3/day$. The discharge figures show that the maximum discharge occurs in August with $52,634.2{\times}10^6 m^3/day$ (23.4% of the year's volume), followed by July and Sep. in that order with 23.1 and 17%, respectively. (2) The pollutant load influx from the Nakdong River dam was composed of 307,591.3COD-kg/day, 128.746.1 TN-kg/day, and 107,625.8 TP-kg/day. (3) The surface temperature in the Nakdong River estuary was about $2.137^{\circ}C$ higher than that of the lower layer. The salinity of the lower layer was 2.209%o higher than that of the ocean surface. The salinity of the ocean surface decreased by up to 19.593%o due to the inflow of the discharge at the Nakdong River dam. (4) DO, COD, TN, and SS concentration levels tended to be higher at the ocean surface than in lower layers, whereas the reverse was true for TP. (5) The water mass at the ocean's surface and in the lower layers during the drought and flood seasons tended to be separated by the difference in densities due to the freshwater inflow.

Characterization and Cloning of the Gene Encoding Autoregulator Receptor Protein from Streptomyces longwoodensis (Streptomyces longwoodensis로부터 Autoregulator Receptor Protein 유전자의 클로닝 및 특성)

  • For screening of autoregulator receptor gene from Streptomyces longwoodensis, PCR was performed with primers of receptor gene designed on the basis of amino acid sequences of autoregulator receptor proteins with known function. PCR products were subcloned into the BamHIsite of pUC19 and transformed into the E. coli $DH5{\alpha}$. The isolated plasmid from transformant contained the fragment of 100 bp, which was detected on $2\%$ gel after BamHI treatment. The insert, 100 bp PCR product, was confirmed as the expected internal segment of gene encoding autoregulator receptor protein by sequencing. Southern and colony hybridizations with the 100 bp fragment as a probe allowed to select a genomic clone of S. longwoodensis, pSLT harboring a 4.4 kb SphI fragment. Nucleotide sequencing analyses revealed a 651 bp open reading frame(ORF) were isolated protein showing moderate homology ($35{\sim}46\%$) with the ${\Gamma}$-butyrolactone autoregulator receptors from Streptomyces sp., and this ORF was named sltR The sltR/pET-17b plasmid was constructed to overexpress the recombinant SltR protein (rSltR) in E. coli BL21 (DE3)/pLysS, and the rSltR protein was purified to homogeneity by DEAE-Sephacel column chromatography, and DEAE-5PW chromatography (HPLC). The molecular mass of the purified rSltR protein was 55 kDa by HPLC gel-filtration chromatography and 28 kDa by SDS-PAGE, indicating that the rSltR protein is present as a dimer. A binding assay with tritium-labeled autoregulators revealed that the rSltR has clear binding activity with a A-factor type autoregulator as the most effective ligand.

Optimized Processing Condition of Production of Nannochloropsis oculata under Light-emitting Diode (LED) Condition (LED배양조건에서 미세조류 Nannochloropsis oculata의 생산 효율성을 높이는 공정 최적화)

  • The 100 l culture system was made on the basis of LED light, and Nannochloropsis oculata was cultured in f/2 medium at light intensity ($100{\mu}mol/m^2/s$), culture temperature ($20^{\circ}C{\pm}1^{\circ}C$) and LD cycle (12hr). As a result, the maximum biomass of 1.07 g/l was cultured as a result of 100 l mass culture at $100{\mu}mol/m^2/s$ and 24 mg/l nitrate concentration in LED blue (475 nm). The extraction was carried out using sonicator, homogenizer and chemical method 0.5M HCl shredding method. The contents of chlorophyll a, chlorophyll b and carotenoid were 1.6, 0.5 and 0.3 mg/g cell. When using homogenizer, it was measured at 1.0, 0.6 and 0.2 mg/g cell. The chemical breakdown method of 0.5M HCl, chlorophyll a, b, and carotenoid contents were measured as 0.9, 0.8, 0 mg/g cell. The highest amount of biomass during the distruption time was measured at 3.6 mg/g cell at 15 min disintegration and acetone, 3.6 mg/g cell of acetone, methanol, and ethanol were measured as effective solvents. Concentration was measured by using microfilter, disk type continuous centrifuge and tubular type continuous centrifuge were 16.0, 1.1 and 0.5 g/l, respectively. Four kinds of equipment such as hot air dryer, vacuum dryer, spray dryer and freeze dryer were tested to optimize the drying process. As a result, the recovery rates of spray dryer and freeze dryer were 80% and 60%.

Characterization of a cDNA Encoding Transmembrane Protein 258 from a Two-spotted Cricket Gryllus bimaculatus (쌍별귀뚜라미(Gryllus bimaculatus)의 GbTmem258 cDNA 클로닝과 발현분석)

  • The cDNA that encodes transmembrane protein 258 (Tmem258) was cloned from Gryllus bimaculatus and named GbTmem258. This protein comprises 80 amino acids, has no N-glycosylation site, and contains five potential phosphorylation sites at two serines, two threonines, and one tyrosine. The predicted molecular mass of GbTmem258 is 9.06 kDa, and its theoretical isoelectric point is 5.5. The tertiary structure of GbTmem258 was predicted using the available secondary structure information, which suggests the presence of alpha helices (52.5%), random coils (22.5%), extended strands (16.25%), and beta turns (8.75%). Homology analysis revealed that GbTmem258 exhibits high similarity at the amino-acid level to Tmem258 found in other species. The effect of starvation and refeeding on GbTmem258 mRNA expression was also examined in this study. It was found that GbTmem258 mRNA expression in the hindgut progressively increased throughout the starvation period, peaking at almost 1.5 times the control level after six days of starvation. However, refeeding for one to two days after the six-day starvation period restored GbTmem258 mRNA expression to the control level. In fat body, GbTmem258 mRNA expression was almost 3-fold higher during starvation compared to the control level. Refeeding for one to two days after the six-day fast resulted in a decline in the expression to about a 2.5-fold increase over the control level. Throughout the starving and refeeding periods, no other tissues showed any discernible alterations in GbTmem258 mRNA expression.

Kinematical Analysis of Woman Javelin Throwing (창던지기 동작의 kinematic적 특성분석)

  • The purpose of the study was to provide the fundamental data to instruct athletes through the analysis athletes' movement in javelin. Three athletes in the level of national representative were participated in this study. The study analyzed kinematic variables(lead foot and releasing javelin) through 3-D analysis and obtained the following results. 1. During withdrawal, it is important to maintain of running horizontal velocity. 2. It was showed that throng average height was $84{\pm}3.3%$ and javelin adequative degree, Among the athletes, $S_2$ who had the best record was released the javelin with the fast velocity, but throw the javelin with the less releasing velocity. 3. $S_2$ released after lead foot were completely landed and therefore it is no problem in a kinematic aspect. However, $S_1$ angle was too small. it caused increase of release velocity to be prevented. 4. $S_2$ showing the best result indicated shorter in duration time. Generally, the shorter duration time in release phase showed the longer release distance. Especially $S_1$ and $S_3$ showing the worse result indicated the longer duration time in preparatory phase, causing the breakup of force. Therefore to improve the record, it should be decreased the duration time in preparatory phase. 5. Compared with $S_1$ and $S_3$, $S_2$ showing the best record indicated the higher velocity in center of mass, trunk, upper arm, lower arm and hand That is the higher velocity of upper arm at release leaded the better velocity transfer from upper arm to following lower arm and hand, these action should be considered to be helpful of better record. According to the above conclusion, when the athletic leaders cauch athletes, they should focus on maintaining knee angle, upper body and hip angle in a previous stage of release and throwing angle, throwing height, throwing velocity in a release stage.
