Four Gram-negative, chemoheterotrophic, non-motile, yellow-colored strains were isolated from the East Sea or from deep-sea sediments of Nankai Trough by standard dilution plating. Characterization by polyphasic approaches indicated that the four strains are members of the same species. Phylogenetic analyses based on 16S rRNA gene sequences revealed that the strains formed a coherent and novel genus-level lineage within the family Flavobacteriaceae. The dominant cellular fatty acids were i-C15:0, 3-OH i-C17:0, and 2-OH i-C15:0 and/or C16:1 ${\omega}7c$. Predominance of 2-OH i-C15:0 and/or C16:1 ${\omega}7c$ clearly differentiated the strains from closely related members. The DNA G+C contents ranged 35.1-36.2 mol%. It is proposed, from the polyphasic evidence, that the strains should be placed into a novel genus and species named Sufflavibacter maritimus gen. nov., sp. nov., with strain $IMCC1001^T(=KCCM\;42359^T=NBRC\;102039^T)$ as the type strain.
The objective of this work was to isolate a microorganism, able to produce high lactate dehydrogenase (LDH) activity, for use as a microbial feed additive. The LDH is an important enzyme for lactate conversion in the rumen, thereby possibly overcoming lactic acidosis owing to sudden increases of cereal in the diets of ruminants. In the present study, various bacterial strains were screened from a variety of environments. Among the isolated microorganisms, strain FFy 111-1 isolated from a Korean traditional fermented vegetable food called Kimchi showed the highest enzyme activity, along with retaining strong enzyme activity even in rumen fluid in vitro. Based on morphological and biochemical characteristics as well as compositions of cellular fatty acids plus API analyses, this strain was identified as Lactobacillus sp. The optimum temperature and pH for growth were found to be 30$^{\circ}C$ and pH 6.5, respectively. A maximum cell growth of 2.2 at $A_{650}$ together with LDH activity of 2.08 U per mL was achieved after 24 h of incubation. Initial characterization of FFy 111-1 suggested that it could be a potential candidate for use as a direct-fed microbial in the ruminant animals.
A strictly aerobic, non-motile, ovoid-shaped Alphaproteobacteria, designated strain $JC2049^T$ was isolated from a tidal flat sediment sample. The results of 16S rRNA gene sequence analysis indicated that this isolate belonged to the genus Thalassobius, with a sequence similarity of 96.9-97.3% to other valid Thalassobius spp. The cells required 1-7% NaCl for growth (optimum 2%) and accumulated $poly-\beta-hydroxybutyrate$. Nitrite was reduced to nitrogen, but nitrate was not reduced to nitrite. No genetic potential for aerobic anoxygenic photosynthesis was detected. The primary isoprenoid quinone (Ubiquinone-10), predominant cellular fatty acids $(C_{18:1}{\omega}7c,\;11\;methyl\;C_{18:1}\omega7c\;and\;C_{16:0})$ and DNA G+C content (61 mol %) were all consistent with the assignment of this isolate to the genus Thalassobius. Several phenotypic characteristics clearly distinguished our isolate from other Thalassobius species. The degree of genomic relatedness between strain $JC2049^T$ and other Thalassobius species was in a range of 20-43 %. The polyphasic data presented in this study indicates that our isolate should be classified as a novel species within the genus Thalassobius. The name Thalassobius aestuarii sp. novo is therefore proposed for this isolate; the type strain is $JC2049^T(=IMSNU\;14011^T=KCTC\;12049^T=DSM\;15283^T)$.
Cholesterol assimilated by Lactobacillus acidophilus ATCC 43121 was not metabolically degraded in that most of it was recovered with the cells. Cells grown in the presence of cholesterol micelles and bile salts were more resistant to Iysis by sonication than those grown in their absence, suggesting a possible alteration of cellular membranes. Cholesterol assimilation occurred during growth at pH 6.0, the amount of which was more than that by cells grown without pH control. Cholesterol assimilated by cells was recovered in the membrane fractions of cells both grown at pH 6.0 and without pH control. The effect of unsaturated fatty acids on cholesterol assimilation was not clear, since there was no significant (P> 0.05) difference in the amount taken up from micelles prepared using L-${\alpha}$-phosphatidylcholine, dioleoyl or L-${\alpha}$-phosphatidylcholine, distearoyl. Without Tween 80, little, if any, cell growth or cholesterol uptake was observed. In the presence of 0.05% Tween 80, cholesterol uptake increased dramatically as did growth. However, as the amount of Tween 80 increased beyond 0.05%, cholesterol uptake decreased while the amount of growth remained the same.
An obligate methanol-oxidizing bacterium, Methylobacillus sp. strain SK1, which grows only on methanol was isolated from soil. The isolate was nonmotile Gram-negtive rod. It does not have internal membrane system. The colonies were small, whitish-yellow, and smooth. The guanine plus cytosine content of the DNA was 48 mol%. Cellular fatty acids consisted predominantly of large amounts of straight-chain saturated $C_{16:0}$ acid and unsaturated $C_{16:1}$ acid. The major ubiquinone was Q-8, and Q-10 was present as minor component. The cell was obligately aerobic and exhibited catalase, but no oxidase, activity. Poly-.betha.-hydroxybutyrate, endospores, or cysts were not observed. the isolate could grow only on methanol in mineral medium. Growth factors were not required. The isolate was unable to use methane, formaldehyde, formate, methylamine, and several other organic compounds tested as a sole source of carbon and energy. Growth was optimal at 35.deg.C and pH 7.5. It could not grow at 42.deg.C. The doubling time was 1.2h at 30.deg.C when grown with 1.0%(v/v) methanol. The growth was not affected by antibiotics inhibiting cell wall synthesis and carbon monoxide but was completely suppressed by those inhibiting protein synthesis. Methanol was found to be assimilated through the ribulose monophosphate pathway. Cytochromes of b-, c-, and o- types were found. Cell-free extracts contained a phenazine methosulfate-linked methanol dehydrogenase activity, which required ammonium ions as an activator. Cells harvested after the late exponential phase seemed to contain blue protein.ein.

This paper summarizes and addresses beef-quality characteristics and the beef-grading systems in Korea, Japan, the USA, and Australia. This paper summarizes recent research progresses on the genetic and nutritional factors that affect beef quality. Intramuscular (i.m.) adipose tissue deposition or marbling is a major determinant of beef quality. This paper addresses the mechanisms of i.m. adipose tissue deposition focused on adipogenesis and lipogenesis. We also address selected signaling pathways associated with i.m. adipose tissue deposition. Nutrients contribute to the cellular response and phenotypes through gene expression and metabolism. This paper addresses control of gene expression through several nutrients (carbohydrates, fat/fatty acids, vitamins, etc.) for i.m. adipose tissue deposition. Several transcription factors responsible for gene expression via nutrients are addressed. We introduce the concept of genome-based precision feeding in Korean cattle.

The spectral composition of light can affect the growth and biochemical composition of photosynthetic microalgae. This study examined the use of light filtering through a solution of soluble colored additives, a cost-effective method to alter the light spectrum, on the growth and lipid production of an oleaginous microalga, Nannochloropsis gaditana (N. gaditana). Cells were photoautotrophically cultivated under a white light emitting diode (LED) alone (control) or under a white LED that passed through a solution of red and yellow color additive (4:1 ratio) that blocked light below 600 nm. The specific growth rate was significantly greater under filtered light than white light ($0.2672d^{-1}$ vs. $0.1930d^{-1}$). Growth under filtered light also increased the fatty acid methyl ester (FAME) yield by 22.4% and FAME productivity by 80.0%, relative to the white light control. In addition, the content of saturated fatty acids was greater under filtered light, so the biodiesel products had better stability. These results show that passing white light through an inexpensive color filter can simultaneously enhance cellular growth and lipid productivity of N. gaditana. This approach of optimizing the light spectrum may be applicable to other species of microalgae.

BACKGROUND/OBJECTIVES: Obesity is a risk factor of breast cancer in postmenopausal women. Estrogen deprivation has been suggested to cause alteration of lipid metabolism thereby creating a cellular microenvironment favoring tumor growth. The aim of this study is to investigate the effects of estrogen depletion in combination with excess energy supply on breast tumor development. MATERIALS/METHODS: Ovariectomized (OVX) or sham-operated C3H/HeN mice at 4 wks were provided with either a normal diet or a high-fat diet (HD) for 16 weeks. Breast tumors were induced by administration of 7,12-dimethylbenz(a)anthracene once a week for six consecutive weeks. RESULTS: Study results showed higher serum concentrations of free fatty acids and insulin in the OVX+HD group compared to other groups. The average tumor volume was significantly larger in OVX+HD animals than in other groups. Expressions of mammary tumor insulin receptor and mammalian target of rapamycin proteins as well as the ratio of pAKT/AKT were significantly increased, while pAMPK/AMPK was decreased in OVX+HD animals compared to the sham-operated groups. Higher relative expression of liver fatty acid synthase mRNA was observed in OVX+HD mice compared with other groups. CONCLUSIONS: These results suggest that excess energy supply affects the accelerated mammary tumor growth in estrogen deprived mice.
산림토양의 각 토양층으로부터 분리된 세균 중에는 통상 농도의 NB배지에서는 중식이 현저히 저해되고 희석한 DNB 배지에서만 중식이 가능한 세균이 다수 포 되어 있었으며, 이들 세균은 NB배지를 $10^{-1}$~$10^{-4}$배로 희석한 배지에서의 증식양상에 따라 4가지형으로 구분되었다. 본 연구에서는 저영양세균의 기준에 따라 $10^{-4}$NB(1 mg C/liter)배지에서 양호하게 증식하는 Type II와 IV세균을 저영양세균으로 분류하였고, 60개의 Type IV(편성저영양세균; obligate oligotrophic bacteria)균주를 순수분리 하였다. 이들 저영양성 세균중 질소고정능을 갖는 11균주에 대하여 화학분류 및 계통분류학적 특성을 검토한 결과 모든 균주는 주요 균체지방산으로 $C_{18:1}$)을, 퀴논종은 Q-10을 함유하였으며, G+C함량은 61-64 mol%범위를 나타내었다. 16S rDNA염기서열을 결정한 결과 각 균주는 Proteobacteria $\alpha$-subdivision의 BANA domain (Bradyrhizobium, Agromonas, Nitrobacter 및 Afipia)에 속하였고 Bradyrhizobium japonicum 및 Bradyrhizobium elkanii와 98% 이상의 높은 상동성을 나타내었다. 나타내었다.

The omega-3 fatty acid docosahexaenoic acid (DHA) is known to induce apoptosis and cell cycle arrest via the induction of reactive oxygen species (ROS) production and endoplasmic reticulum (ER) stress in many types of cancers. However, the roles of DHA in drug-resistant cancer cells have not been elucidated. In this study, we investigated the effects of DHA in cisplatin-resistant gastric cancer SNU-601/cis2 cells. DHA was found to induce ROS-dependent apoptosis in these cells. The inositol 1,4,5-triphosphate receptor ($IP_3R$) blocker 2-aminoethyl diphenylboninate (2-APB) reduced DHA-induced ROS production, consequently reducing apoptosis. We also found that G-protein-coupled receptor 120 (GPR120), a receptor of long-chain fatty acids, is expressed in SNU-601/cis2 cells, and the knockdown of GPR120 using specific shRNAs alleviated DHA-mediated ROS production and apoptosis. GPR120 knockdown reduced the expression of ER stress response genes, similar to the case for the pre-treatment of the cells with N-acetyl-L-cysteine (NAC), an ROS scavenger, or 2-APB. Indeed, the knockdown of C/EBP homologous protein (CHOP), a transcription factor that functions under ER stress conditions, markedly reduced DHA-mediated apoptosis, indicating that CHOP plays an essential role in the anti-cancer activity of DHA. These results suggest that GPR120 mediates DHA-induced apoptosis by regulating $IP_3R$, ROS, and ER stress levels in cisplatin-resistant cancer cells, and that GPR120 is an effective chemotherapeutic target for cisplatin resistance.
