$Ca^{2+}$은 다양한 자극과 빛, biotic, abiotic 스트레스, 호르몬 등의 반응에 대한 세포내 2차 신호전달물질로써 중요한 역할을 한다. $Ca^{2+}$의 반응자들은 특정 물질과 경로를 활성화함으로써 신호전달 기능을 한다고 알려져 있는 $Ca^{2+}$ 결합 단백질들이다. 이들 단백질 중, calmidulin (CaM)은 식물과 동물의 특정 단백질의 활성을 조절하는 것으로 잘 알려져 왔다. 특히, 식물은 다양한 CaM 유전자와 특징적인 protein kinase와 전사인자를 포함한 많은 종류의 CaM 관련 단백질들을 가지고 있다. 이로 인해서 식물은 주변의 여러 가지 신호등을 인지할 수 있을 뿐만 아니라 변화된 환경에 적응할 수 있는 것이다. 하지만, 대부분의 CaM이나 이들과 관련된 단백질들의 기능은 최근 활발히 연구되고 있지만 아직 많은 작용 기작이 연구의 대상이 되고 있다. 따라서 CaM의 기능을 좀 더 이해한다면 식물의 환경적 자극에 대한 반응과 식물의 성장과 발달에 있어서 CaM의 역할을 규명하는데 도움을 줄 수 있을 것으로 기대된다. 본 논문은 $Ca^{2+}$-CaM의 신호전달 시스템과, CaM과 관련된 단백질들, 그리고 식물의 biotic, abiotic 스트레스에 대한 외부 자극의 반응에 있어서 CaM의 작용에 대해 기술하였다.
Low-barrier hydrogen bonds (LBHBs) have been proposed to have important influences on the enormous reaction rate increases achieved by many enzymes. ${\Delta}^5$-3-ketosteroi isomerase (KSI) catalyzes the allylic isomerization of ${\Delta}^5$-3-ketosteroid to its conjugated ${\Delta}^4$-isomers at a rate that approache the diffusion limit. Tyr14, a catalytic residue of KSI, has been hypothesized to form an LBHB with the oxyanion of a dienolate steroid intermediate generated during the catalysis. The unusual chemical shift of a proton at 16.8 ppm in the nuclear magnetic resonance spectrum has been attributed to an LBHB between Tyr14 $O{\eta}$ and C3-O of equilenin an intermediate analogue, in the active site of D38N KSI. This shift in the spectrum was not observed in Y30F/Y55F/D38N and Y30F/Y55F/Y115F/D38N mutant KSIs when each mutant was complexed with equilenin, suggesting that Tyr14 could not form LBHB with the intermediate analogue in these mutant KSIs. The crystal structure of Y30F/Y55F/Y115F/D38N-equilenin complex revealed that the distance between Tyr14 $O{\eta}$ and C3-O of the bound steroi was within a direct hydrogen bond. The conversion of LBHB to an ordinary hydrogen bond in the mutant KSI reduced the binding affinity for the steroid inhibitors by a factor of 8.1-11. In addition, the absence of LBHB reduced the catalytic activity by only a factor of 1.7-2. These results suggest that the amount of stabilization energy of the reaction intermediate provided by LBHB is small compared with that provided by an ordinary hydrogen bond in KSI.
Ginsenosides, which are the active materials of ginseng, have biological functions that include anti-osteoporotic effects. Aqueous ginseng extract inhibits osteoclast differentiation induced by receptor activator of NF-${\kappa}B$ ligand (RANKL). Aqueous ginseng extract produces chromatography peaks characteristic of ginsenosides. Among these peaks, ginsenoside Re is a major component. However, the preventive effects of ginsenoside Re against osteoclast differentiation are not known. We studied the effect of ginsenoside Re on osteoclast differentiation, RANKL-induced tartrate-resistant acid phosphatase (TRAP) activity, and formation of multinucleated osteoclasts in vitro. Ginsenoside Re hampered osteoclast differentiation in a dose-dependent manner. In an in vivo zebrafish model, aqueous ginseng extract and ginsenoside Re had anti-osteoclastogenesis effects. These findings suggest that both aqueous ginseng extract and ginsenoside Re prevent bone resorption by inhibiting osteoclast differentiation. Ginsenoside Re could be important for promoting bone health.
Superoxide dismutase (SOD) is physiologically important in regulating cellular homeostasis and apoptotic cell death, and its mutations are the cause of familial amyotrophic lateral sclerosis (FALS). Mitochondrial serine protease HtrA2 has a pro-apoptotic function and has known to be associated with neurodegenerative disorders. To investigate the relationship between genes associated with apoptotic cell death, such as HtrA2 and SOD1, we utilized the pGEX expression system to develop a simple and rapid method for purifying wild-type and ALS-associated mutant SOD1 proteins in a suitable form for biochemical studies. We purified SOD1 and SOD1 (G93A) proteins to approximately 90% purity with relatively high yields (3 mg per liter of culture). Consistent with the result in mammalian cells, SOD1 (G93A) was more insoluble than wild-type SOD1 in E. coli, indicating that research on the aggregate formation of SOD1 may be possible using this pGEX expression system in E. coli. We investigated the HtrA2 serine protease activity on SOD1 to assess the relationship between two proteins. Not only wild-type SOD1 but also ALS-associated mutant SOD1 (G93A) were cleaved by HtrA2, resulting in the production of the 19 kDa and 21 kDa fragments that were specific for anti-SOD1 antibody. Using protein gel electrophoresis and immunoblot assay, we compared the relative molecular masses of thrombin-cleaved GST-SOD1 and HtrA2-cleaved SOD1 fragments and can predict that the HtrA2-cleavage sites within SOD1 are the peptide bonds between leucine 9-lysine 10 (L9-K10) and glutamine 23-lysine 24 (Q23-K24). Our study indicates that SOD1 is one of the substrate for HtrA2, suggesting that both HtrA2 and SOD1 may be important for modulating the HtrA2-SOD1-mediated apopotic cell death that is associated with the pathogenesis of neurodegenerative disorder.
soybean hypocotyl에서 분리한 미토콘드리아로부터 submitochondrial particles와 matrix 단백질 추출액을 준비하고 전기분해법으로 제조한 superoxide를 처리하여 malate dehydrogenase 및 cytochrome C oxidase의 불활성화와 mitochondrial membrane의 과산화를 각각 조사하였다. 막에 결합되어 있는 cytochrome C oxidase나 수용액상태의 malate dehydrogenas는 모두 $O^{-}_{2}$에 대해 매우 민감하게 불활성화되었다. 즉 dismutation 반응이 빠르게 진행되어 실질적으로 효소의 불활성화에 기여하게 될 농도는 대단히 낮을 것으로 추정되는 $O^{-}_{2}$의 명목상 처리농도 1.4mM 전후에서 두 효소는 그 활성을 완전히 상실하였다. 한편 malondialdehyde의 생성을 지표로 하여 측정된 membrane 과산화는 인지질로서만 이루어진 liposome의 경우보다는 다소 낮은 수준이었으나 무시할 수는 없는 정도였다. mitochondrial membrane의 과산화가 상대적으로 억제된 것은 막에 결합되어 있는 항산화제 및 단백질들에 의한 $O^{-}_{2}$소거효과에 기인하였으리라 해석된다. 식물 미토콘드리아의 대표적인 대사과정인 TCA cycle과 호흡전자전달반응의 성분효소들인 malate dehydrogenase와 cytochrome C oxidase가 불활성화되었고 membrane이 과산화 되었다는 사실은, 식물의 냉해와 광피해 발현기작에서 공히공통적인 화학적 인자로 인정되는 $O^{-}_{2}$의 과잉생성 및 축적이 그것의 주 생성처인 미토콘드리아의 생화학적 기능과 구조에 미칠수 있는 파괴적 효과를 적절히 지시하는 것이다.

${\beta}$-lapachone is a naturally occurring quinone that selectively induces apoptotic cell death in a variety of human cancer cells in vitro and in vivo; however, its mechanism of action needs to be further elaborated. In this study, we investigated the effects of ${\beta}$-lapachone on the induction of apoptosis in human gastric carcinoma AGS cells. ${\beta}$-lapachone significantly inhibited cellular proliferation, and some typical apoptotic characteristics such as chromatin condensation and an increase in the population of sub-G1 hypodiploid cells were observed in ${\beta}$-lapachone-treated AGS cells. Treatment with ${\beta}$-lapachone caused mitochondrial transmembrane potential dissipation, stimulated the mitochondria-mediated intrinsic apoptotic pathway, as indicated by caspase-9 activation, cytochrome c release, Bcl-2 downregulation and Bax upregulation, as well as death receptor-mediated extrinsic apoptotic pathway, as indicated by activation of caspase-8 and truncation of Bid. This process was accompanied by activation of caspase-3 and concomitant with cleavage of poly(ADP-ribose) polymerase. The general caspase inhibitor, z-VAD-fmk, significantly abolished ${\beta}$-lapachone-induced cell death and inhibited growth. Further analysis demonstrated that the induction of apoptosis by ${\beta}$-lapachone was accompanied by inactivation of the phosphatidylinositol 3-kinase (PI3K)/Akt signaling pathway. The PI3K inhibitor LY29004 significantly increased ${\beta}$-lapachone-induced apoptosis and growth inhibition. Taken together, these findings indicate that the apoptotic activity of ${\beta}$-lapachone is probably regulated by a caspase-dependent cascade through activation of both intrinsic and extrinsic signaling pathways, and that inhibition of the PI3K/Akt signaling may contribute to ${\beta}$-lapachone-mediated AGS cell growth inhibition and apoptosis induction.
The inhibition/influences of adenine compounds on the heart have been described repeatedly by many investigators, since the first report by Druny and Szent-Gyorgyi [1929]. These studies have shown that adenosine and adenine nucleotides have an over-all effect similar to that of acetylcholine [ACh] by slowing and weakening the heartbeat. The basic cellular and membrane events underlying the inhibitory action of adenosine on sinus rate, however, are not well understood. Furthermore, the physiological role of adenosine in regulation of the heartbeat remains still to be elucidated. Therefore, this study was undertaken in order to examine the response of rabbit SA node to adenosine and to compare the response to that of ACh. Isolated SA node preparation, whole atrial pair, or left atrlal strip was used in each experiment. Action potentials of SA node were recorded through the intracellular glass microelectrodes, which were filled with 3M KCI and had resistance of 30-50 M. All experiments were performed in a bicarbonate-buffered Tyrode solution which was aerated with 3% $CO_2-97%$$O_2$ gas mixture and kept at $35^{\circ}C$. Spontaneous firing rate of SA node at 35C [Mean + SEM, n=16] was 154 + 3.3 beats/min. The parameters of action potentials were: maximum astolic potential [MDP], -731.7mV: overshoot [OS], 9 + 1.4mV; slope of pacemaker potential [SPP], 94 3.0mV/sec.Adenosine suppressed the firing rate of SA node in a dose dependent manner. This inhibitory effect appeared at the concentration of $10^{-6}M$ and was potentiated in parallel with the increase in adenosine concentration. Changes in action potential by adenosine were dose-dependent increase of MDP and decrease of SPP until $10^{-4}$. Above this concentration, however, the amplitude of action potential decreased markedly due to the simultaneous decrease of both MDP and OS. All these effects of adenosine were not affected by pretreatment of atropine [2mg/l] and propranolol [$5{\times}10^{-6}M$]. ACh [$10^{-6}M$] responses on action potential were similar to those of adenosine by increasing MDP and decreasing SPP. These effects of ACh disappeared by pretreatment of atropine [2mg/1]. Inhibition/effects of adenosine and ACh on sinus rate were enhanced synergistically with the simultaneous administration of adenosine and ACh. Marked decrease of overshoot potential was the most prominent feature on action potential. Dipyridamole [DPM], which is known to block the adenosine transport across cell membrane, definitely potentiated the action of adenosine . Adenosine suppressed the sinus rate and atrial contractility in the same dosage range, even in the reserpinized preparation. Above` results suggest that adenosine suppresses pacemaker activity, like ACh, by acting directly on the membrane of SA node, increasing MDP and decreasing SPP.
홍차 열수추출물의 멜라닌 합성 저해능과 작용기전을 알아보기 위해 in vitro 실험으로 항산화능을 확인한 후, melan-a 세포를 사용하여 멜라닌 합성 저해능, tyrosinase 활성 저해능 및 발현저해능 등에 대한 실험을 실시하여 다음과 같은 결과를 얻었다.(1) 항산화능 실험에서 홍차 열수추출물의 총 폴리페놀 및 플라보노이드 함량은 각각 102, 87 mg/g으로 양성대조군인 알부틴(25, 11 mg/g)보다 높았고, 전자공여능 역시 800 ${\mu}g$/mL에서 82%로 알부틴(48%)보다 높았다. (2) Melan-a 세포 실험에서 홍차 열수추출물은 수지상돌기의 발달과 멜라닌 생합성을 효과적으로 저해하였고, 멜라닌 합성 저해능과 tyrosinase 활성 저해능이 알부틴보다 유의하게 (p<0.001) 높았다. (3) Tyrosinase 단백질 발현 저해능 실험에서 알부틴은 발현에 유의한 영향을 미치지 않은 반면, 홍차 열수추출물은 발현을 유의하게 (p<0.001) 저해하였다. (4) 유전자 발현 저해능 실험에서 알부틴과 홍차 열수추출물 모두 tyrosinase, TRP-1과 TRP-2의 mRNA 발현에 영향을 미치지 않았다. 본 연구를 통하여, 홍차 열수추출물은 우수한 항산화능과 함께 멜라닌 합성 저해능을 확인하였으며, 이는 tyrosinase의 생합성을 저해시키기 보다는 tyrosinase 단백질 발현량을 줄이고 tyrosinase 활성을 감소시킴으로써 멜라닌 합성을 저해시키는 것으로 사료된다.
미세조류 유래 mycosporine-like amino acids (MAAs) 혼합물의 항주름활성을 분석하기 위하여 type I procollagen, elastin 및 involucrin 유전자 발현분석을 실시하였다. Chlamydomonas sp.를 80% 에탄올로 추출하고, 컬럼을 통해 분획물을 정제하고, HPLC 분석을 통해 asterina 330+palythine (A+P) 및 shinorine+palythine (S+P) 혼합물을 정제하였다. MTT assay 결과 A+P 및 S+P는 0.1 mg/mL까지 세포독성을 나타내지 않았다. UV에 노출된 섬유아세포에 대한 MAAs 혼합물의 영향을 분석하였을 때, 0.05 mg/mL A+P 및 0.01 mg/mL S+P 농도에서 각각 2.7 및 3.6 배 PCOLCE mRNAs 발현이 증가하였다. Elastin 유전자는 0.01 mg/mL의 A+P 및 S+P를 처리하였을 때 각각 5.59배 및 3.1배 발현이 증가하였다. 특히 0.01 mg/mL의 A+P 및 S+P 농도에서 involucrin mRNAs 발현이 UV 처리된 대조구와 비교하였을 때 5배 및 2.5배 감소하였다. 결론적으로 Chlamydomonas sp. 유래 MAAs 혼합물은 주름개선용 기능성 화장품 개발을 위한 소재로써의 가능성이 충분한 것으로 판단되었다.
해양생물을 포함한 천연물질은 신약개발의 원천 소재로서 매력적이며, 특히 무수한 미지의 해양생물들의 연구가 관심을 받고 있다. 기존의 연구에서 미크로네시아에서 채취한 해면동물 40여종에 대하여 항증식 효과를 다양한 암세포주에서 검색한 바 있다. 본 연구에서는 그 중 Coscinoderma sp.의 작용 및 그 기전을 살펴보았다. 특히, 암 억제유전자 p53의 발현을 억제시킨 세포주(HCT116 p53KO과 RKO-E6)에서의 차이점을 비교하였다. 세포생존률 시험에서 Coscinoderma sp. 추출물은 p53의 유무와 상관없이 암세포의 증식을 억제하였음을 확인하였다. 이 암세포 증식 억제 효과가 p53 존재에 따라 다르게 나타나는지 알아보기 위하여 세포사멸 관련 단백질 발현양을 Coscinoderma sp. 처리한 각 세포주에서 비교하였다. 그 결과, Coscinoderma sp.를 HCT16 세포주에 처리하였을 때, p53과 Noxa의 발현이 증가하는 것을 관찰하였고, caspase-9이 분절되면서 감소하는 것으로부터 apoptosis를 일으킨다고 여겨진다. 반면, p53이 결핍된 HCT116세포주에서는 Coscinoderma sp.에 의하여 p21과 mTOR의 발현이 증가되는 것을 확인하였고, 이는 senescence를 야기할 수 있다고 여겨진다. 본 연구로부터 Coscinoderma sp.는 p53의 존재여부에 따라 상이한 작용기전을 매개하여 대장암 세포주의 증식을 억제한다는 것을 알 수 있었다. 이는 새로운 항암제의 개발 가능성을 제시하는 것으로, Coscinoderma sp.의 활성 성분에 대한 지속적인 연구가 이루어 져야 할 것으로 보인다.

회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
