Different Gene Expression on Human Blood by Administration of OLT-2 (OLT-2의 복용에 의한 인간 혈중 유전자 발현 변화)

  • This study was performed to investigate genes which are differently expressed in human blood by administrating of OLT-2. OLT-2 was medical precipitation composed of three medicinal herbs, Ginseng Radix, Astragali Radix, Glycyrrhizae Radix, and anti-leukemia effect of it was evaluated from Byung Hun Jeon of Wonkwang University this study was approved by Institutional Review Board of Korea Institute of Oriental Medicine (Taejeon, Korea) and four male subjects participated in this study. Gene expressions were evaluated by cDNA chip, in which 24,000 genes were spotted. Hierarchical cluster and biological process against the genes, which expression changes were more than 1.6 fold, were constructed by cluster 3.0 providing Stanford University and EASE(http://apps1 .maid.nih.gov/DAVID). Five groups were clustered according to their expression patterns. Group A contained gene decreased by OLT-2 and increased genes by OLT-2 were involved in Group B, C, D. In biological process, expression of genes involved in cytokine or cell calcium signaling, such as interleukin 18 and G-protein beta 4 were increased, but protein tyrosine phosphatase receptor type c, which function is cell adhesion between antigen-presenting cell and T or B-cell, was decreased by OLT-2. This study provides the most comprehensive available survey of gene expression changes in response to anti-leukemia effect of OLT-2 in human blood.

A Continuous Cell Separator Based on Gravity and Buoyant Forces in Fluids of Dissimilar Density (서로 다른 밀도의 유체 내 바이오 물질이 받는 중력과 부력 차를 이용한 연속적 세포 분리기)

  • We present a continuous cell separator that achieves density-dependent and size-independent cell separation based on the net force of gravity and buoyancy forces on the cells in dissimilar density fluids. Previous cell separators are, based on the size or dielectrophoretic property of the cells and, are suitable for size-dependent and density-independent cell separation. However, these properties can make it difficult to collect the same types of cells with the same density but with size variations. The present separator, however, is capable of collecting the same types of cells based on the cell density in the fluid. Regardless of cell size, the proposed chip isolates low density cells, (white blood cells, or WBCs) at the upper outlet while obtaining high-density cells (red blood cells, or RBCs) from the lower outlet based on density. Efficiency levels for separation of WBCs and RBCs were $90.9{\pm}9.1%$ and $86.4{\pm}1.99%$, respectively. The present separator therefore has the potential for use in the pretreatment of whole blood.

Toxicogenomic Study to Identify Potential New Mechanistic Markers on Direct-Acting Mutagens in Human Hepatocytes (THLE-3)

  • Exposure to DNA-damaging agents can elicit a variety of stress-related responses that may alter the expression of genes associated with numerous biological pathways. We used 19 k whole human genome chip to detect gene expression profiles and potential signature genes in human normal hepatocytes (THLE-3) by treatment of five direct acting mutagens, furylfuramide (AF-2), N-nitroso-N-methylurea (MNU), methylmethanesulfonate (MMS), 4-nitroquinoline-N-oxide (4-NQO) and 2-nitrofluorene (2NF) of the $IC_{20}$ concentration for 3 h. Fifty one up-regulated common genes and 45 down-regulated common genes above 1.5-fold by five direct-acting mutagens were identified by clustering analysis. Many of these changed genes have some association with apoptosis, control of cell cycle, regulation of transcription and signal transduction. Genes related to these functions, as TP73L, E2F5, MST016, SOX5, MAFB, LIF, SII3, TFIIS, EMR1, CYTL1, CX3CR1 and RHOH are up-regulated. Down-regulated genes are ALOX15B, xs155, IFITM1, BATF, VAV2, CD79A, DCDC2, TNFSF8 and KOX8. We suggest that gene expression profiling on mutagens by toxicogenomic analysis affords promising opportunities to reveal potential new mechanistic markers of genotoxicity.

Design of an EEPROM for a MCU with the Wide Voltage Range

  • In this paper, we design a 256 kbits EEPROM for a MCU (Microcontroller unit) with the wide voltage range of 1.8 V to 5.5 V. The memory space of the EEPROM is separated into a program and data region. An option memory region is added for storing user IDs, serial numbers and so forth. By making HPWs (High-voltage P-wells) of EEPROM cell arrays with the same bias voltages in accordance with the operation modes shared in a double word unit, we can reduce the HPW-to-HPW space by a half and hence the area of the EEPROM cell arrays by 9.1 percent. Also, we propose a page buffer circuit reducing a test time, and a write-verify-read mode securing a reliability of the EEPROM. Furthermore, we propose a DC-DC converter that can be applied to a MCU with the wide voltage range. Finally, we come up with a method of obtaining the oscillation period of a charge pump. The layout size of the designed 256 kbits EEPROM IP with MagnaChip's 0.18 ${\mu}m$ EEPROM process is $1581.55{\mu}m{\times}792.00{\mu}m$.

A Unified Potentiostat for Electrochemical Glucose Sensors

  • A unified potentiostat circuit for both $O_2$- and $H_2O_2$- based electrochemical glucose sensors was proposed and its function was verified by circuit simulations and measurement results of a fabricated chip. This circuit consisted of an operational amplifier, a comparator and current mirrors. The proposed circuit was fabricated with a $0.13{\mu}m$ thick oxide CMOS process and an active area of $360{\mu}m{\times}100{\mu}m$. The measurements revealed an input operation range from 0.5 V to 1.6 V in the $H_2O_2$- based bio-sensor and from 1.7 V to 2.6 V in the $O_2$- based bio-sensor with a supply voltage of 3.3 V. The evaluation results showed that the proposed potentiostat circuit is suitable for measuring the electrochemical cell currents of both $O_2$- and $H_2O_2$- based glucose sensors.

Development of the Switching Mode Conversion Type Pulse Charger for the Lead Battery of Solar Cell Generator Equipment by Fly-Back Converter Method (플라이백 컨버터방법에 의한 태양광발전설비의 납축전지 스위칭모드 전환형 펄스충전기 개발)

  • In this paper, the switching mode conversion type pulse charger by fly-back converter method for lead battery of the solar cell generator equipment is proposed. And we propose the control circuit and design method of insulated switching mode convert type pulse charger by fly-back convert method in the lead battery. The proposed system can minimize the current consumption by digital pulse. Also the proposed system can generate the constant 10[KHz] frequency, transmit the signal with main control system in the power control system. And it supervises the state of lead battery using one chip micro processor. The proposed the switching mode conversion type pulse charger by the fly-back converter method can charge fast and stabilize lead battery with nominal value 12[V], 20[AH]. Also we propose the design procedure of the power control circuit for turn ratio of fly-back inductor and determining method of values such as the charging current, bulk current, partial current, over current value and fixed charging voltage. The experiment results for the voltage and current wave for partial, bulk, over and fixed charging period show the good charging effect and performance. And the PCB and internal coupling diagram of the switching mode conversion type pulse charger by fly-back converter method is presented.

Study metal-grade silicon manufacturing and slag refining for the production of silicon solar cell (태양전지용 실리콘 생산을 위한 금속급 실리콘 제조와 슬래그 정련 연구)

  • 야금학적 방법을 통한 태양전지용 실리콘 제조를 위하여 아크로(Arc furnace)에서 제조된 용융 상태의 금속급 실리콘을 슬래그와 직접 반응시켜 불순물을 제거하는 공정에 관한 연구를 수행하였다. 이를 위해 아크로와 고주파 유도용해로(High-frequency induction furnace)를 이용하여 금속급 실리콘을 제조와 정련 특성 실험을 수행하였다. 본 연구에서 금속급 실리콘을 제조하기 위한 장비로 150kW급-DC 아크로와 300kW급-AC 아크로를 사용하였다. 원재료로 규석, 코크스(Cokes), 숯, 그리고 우드칩(Wood chip)을 실험 비율에 맞춰 아크로 내부에 장입하고, 이를 용융환원 방법을 통해 반응을 시켰다. 이때 생산된 금속급 실리콘의 순도는 약 99.2~99.8% 이었으며, 원재료의 순도, 장입 비율 및 아크로 운전 특성에 따라 편차가 있다. 아크로에서 생산된 금속급 실리콘의 경우 인(phosphorus), 붕소(boron)를 다량 함유하고 있고, 이를 제거하기 위하여 50kW급 고주파 유도용해로 장비를 사용하여 슬래그 정련 실험을 수행하였다. 슬래그 정련시 사용한 성분은 SiO2, CaO 그리고 CaF2 이며, 금속급 실리콘과 슬래그의 질량비 및 반응 시간에 따른 실리콘 불순물 특성을 평가하였다. 실험결과 인과 붕소는 각각 1 ppm 이하, 5 ppm 이하 였으며, 칼슘을 제외한 대부분의 금속 불순물의 경우 0.1~0.2% 임을 확인하였다.

  • Coating of individual cells with organic or inorganic materials has drawn a great deal of attention, because it provides the cells with physicochemical durability, which would contribute to the development of bioreactors, biosensor, and lab-on-a-chip, as well as to the fundamental studies in single cell-based biology. Although many strategies have been developed for coating of microbial cells, limited methods are available to coat mammalian cells because most mammalian cells do not have a robust membrane or exoskeleton. Instead, they are enclosed in a lipid bilayer, which is fluidic and vulnerable to changes in its environments. It is more difficult to treat mammalian cells in vitro than microbial cells because the surfaces of mammalian cells are not protected or reinforced by a tough coat. In this work, we report a cytocompatible and degradable nanocoat for mammalian cells. Three types of mammalian cells (HeLa cells, NIH 3T3 fibroblasts, and Jurkat T cells) were individually coated within metal-polyphenol. To maintain the viability of the mammalian cells, we performed the whole processes under strictly physiological culture conditions, and carefully selected nontoxic materials.

  • The main goal of this work is advances in 1.0mm $\times$ 0.5mm light emitting diode using AlInGaN cell structure and display module. In the first place, we proposed $200{\mu}m{\times}200{\mu}m$ cell structure using AlInGaN. Secondly, we describe new type 1.0mm $\times$ 0.5mm blue and white LED fabrication procedure and results of an examination include mobile application.

  • In this paper, the solar cell need the characteristic interpreting because the solar cell changes greatly according to the isolation, temperature and load in the photovoltaic development. Moreover, to get many energy in photovoltaic development need the position tracking of the sun according to the environment change and it is necessary to control the output of solar cells up to the time. Simulation and composed microprocessor and sensor chip an power conversion system with boost converter to experiment results are performed to prove the analysis of the converter operation, and to show the possibility of energy harvesting and photovoltaic development need the position tracking small capacitance, the boost rate of boost converter was similar to 167 percent.
