Effects of Abscisic Acid on the Epidermal Structure and Ontogeny of Stomata in Orostachys malacophyllus Leaves (둥근바위솔(Orostachys malacophyllus) 잎의 표피구조와 기공발생에 미치는 Abscisic Acid의 영향)

  • The effects of abscisic acid(ABA) spraying for 12 weeks on the stomatal types and frequencies of O. malacophyllus leaves were summarized as follows. ABA inhibited the growth of O. malacophyllus. The prominent effect of ABA on the epidermal structure was the promotion of senescence such as thickness of cell walls, smooth sinuosity of cell walls, and large size of epidermal cells. The stomatal frequency was decreased to 23% by 10$\mu\textrm{g}$ ml-1 ABA and to 48% by 100$\mu\textrm{g}$ml-1, and also the stomatal size was more or less smaller than that of control. The developing secondary stomatal mother cell was not found in both 10 and 100$\mu\textrm{g}$ml-1ABA, but the arrested secondary stomatal mother cell was rarely found in 10$\mu\textrm{g}$ ml-1 ABA. The formation of normal stomatal types such as helico-eumesogenous and aniso-eumesogenous was found in both 10 and 100 $\mu\textrm{g}$ ml-1 ABA asin well as control. Also nine abnormal stomatal types were found, and the frequencies were promoted to 6% by 10 $\mu\textrm{g}$ ml-1 ABA and to 17% by 100 $\mu\textrm{g}$ ml-1 ABA. Among these abnomal stomata, four types such as aborted stomata, single-aborted guard cells, arrested stomata, and modified stomatal complexes were found in control as well as in 10 and 100 $\mu\textrm{g}$ ml-1 ABA, but five types such as wrenched stomata, unequal stomata, wavy guard cells, guard cells overlapped by guard cells, and dissolved cell wall stomata were found in both 10 and 100 $\mu\textrm{g}$ ml-1ABA. The modified stomata complexes were abnormal stomatal types which were newly found and also were varied in types.

Treatment of E. coli B with two Antibiotics and their Influence on $T_3$ phage Absorption (대장균(E. coli B)의 항생제처리가 $T_3$ phage의 부착에 미치는 영향)

  • E. coli B was treated with colistin and kanamycin and the influence of these antibiotics on the absorption of $T_3$ phage was studied using the plaque counting method and the electron microscope. E. coli B treated with colistin was sharply inhibited on phage absorption and cell walls were severely damaged showing some spiny appearance around the walls. No influence of kanamycin was noted on phage absorption. Bacterial cells treated with kanamycin showed wave form in the structure of walls and a profound change was noted in the cytoplasm where it was concentrated along the periphery of the inner wall leaving the center of the cell to appear almost empty.

  • Sophorolipid, a biosurfactant produced from Candida bombicola ATCC 22214. showed antimicrobial activity against Bacillus subtilis. Staphylococcus xylosus, Streptococcus mutans, and Propionibacterium acne at 4, 1, 1, 0.5 ppm as MIC(minimum inhibitory concentration). Also 100ppm of sophorolipid inhibited 50% of cell growth of plant pathogenic fungus. Botrytis cinerea. However, sophorolipid showed no effect on the Escherichia coli, indicating its selective antimicrobial activity depending on the cell wall structure. Treatment of B. subtilis with sophorolipid increased the leakage of intracellular enzyme, malate dehydrogenase, indicating the possible interaction of sophorolipid with cellular membrane. Between lactone-type and acid-type sophorolipid, the former showed higher antimicrobial activity.

  • Biosurfactant has antibiological activities. Due to its low toxicity, biosurfactant can be applied to human health care products. Posseing the antibiological activities as well as the surfactant properties implies that biosurfactant can be widely used in the cosmetic industry. In this presentation, applications of biosurfactant have been reviewed including the antimicrobial activities of sophorolipid.   • A strain of Staphylococcus aureus resistant against cadmium was cultivated by using a liquid medium containing 10ppm cadmium ion, and then, it was fractionated into several fractions as described in the previous paper. Content of the metal in each fraction was determined through an atomic absorption spectrometery. The results are as follows; (1) A $690.9{\mu}g$ cadmium was contained in one gram dry cell. (2) A 39.9% of total cadmium was easily extracted by TCA, however a 52.2% was unextractable even by series of extraction with TCA, ethanol-ehter, perchloric acid and ammonium water. (3) Among the fractions prepared along the cellular structure, plasma membrane fraction showed a highest content of the metal by containing a 59.1%. (4) The fraction of cytoplasm and cell wall contained a 26.8 and 14.1%, respectively. (5) More than 90% of the metal contained in the cell wall was detected from the fraction of lipopolysaccharide. It is considered from these results tht at least a 70% of the cadmium up taken by the resistant cell associates with membranous structure in the cell surface.

  • ZnS thin films were prepared on glass substrate at various deposition conditions by a HW apparatus and were systematically investigated the growth characteristics, in terms of deposition edges by a double beam spectro- photometer, and structural analysis by a x-ray diffraction rates were increased with incresing the cell temperature and vapor pressure of sulfur, but were decreased with increasing substrate temperature. The optical characteristics of thin films depends on the deposition rates. The band gap energies of 3.46∼3.52eV measured at room temperature are smaller than the theoretical value of 3.54eV, indicating that impurities exist in the crystal. All ZnS thin films are oriented in the (III) principal direction of a zincblende structure. By introducing the S vapor, optical and crystalline properties have been improved.

Does oral ciprofloxacin affect the structure of thoracic aorta in adult and senile male albino rats? A clue to fluoroquinolones-induced risk of aortic dissection

  • In this study, the effect of oral ciprofloxacin on the structure of the thoracic aorta in rats was investigated. Twenty four male albino rats were divided into 4 groups (6 rats/group): group I (adult control), group II (adult rats treated with ciprofloxacin), group III (senile control), and group IV (senile rats treated with ciprofloxacin). Rats in groups II and IV received ciprofloxacin via oral gavage in a daily dose of 3.5 mg/kg/d for 14 days, while control rats received equivalent amount of distilled water used to dissolve the drug. After 2 weeks, all rats were sacrificed, thoracic aortae were dissected, and half of the specimens were processed for paraffin sections and examined by light microscopy. The other half of the specimens were prepared for scanning electron microscopy. Sections from rats treated with ciprofloxacin showed evident damaging effect on aortic wall particularly in (group IV). Aortic intima showed, focal desquamation of the lining epithelium. Tunica media exhibited loss of the normal concentric arrangement and degeneration of the smooth muscle cells. Immune staining for alpha smooth muscle actin showed muscle damage. Interestingly, some sections in (group IV) showed out-pouch (aneurysm like) of the aortic wall. There was dense collagen fibers deposition. Scanning electron microscopic observations of (group IV) revealed uneven intima, adherent blood cells and fibrin filaments to damaged intima, and out-pouch formation. It was concluded that oral ciprofloxacin caused deleterious structural changes in the thoracic aortic wall of rats explaining clinical observations of fluoroquinolones induced risk of aortic dissection and aneurysm.

  • Lignin in wood cell walls influeced the transformation of the cellulose crystal structure during mercerization. Samples of sound and decayed woods by white rot fungus of Quercus mongolica were treated with 20% aquous NaOH solution, followed by washing and drying, and delignified. The effect of delignification on cellulose structure was investigated by a series of an X-ray diffraction analysis and ultraviolet(UV) microscopy. Delignification of alkali-treated woods did not influence their cellulose crystal structures. It may be concluded that lignin prevents the swelling of wood cellulose during mercerization and restrain the intermingling of cellulose chains.

  • Micro cell structures with high aspect ratio were fabricated by injection molding method. The mold inserts had dimension $1.9cm\times8.3cm$ composed of a lot of micro posts and were fabricated by LIGA process. The size of the micro posts was $157{\mu}m\times157{\mu}m\times500{\mu}m$ and the gaps between two adjacent posts were $50{\mu}m$. Using Polymethylmethacrylate (PMMA) injection molding was performed. The key experimental variables were temperature, pressure, and time. By controlling these, good shaped mim cell structures with $50{\mu}m$ in wall thickness and $500{\mu}m$ in depth were obtained. In order to understand micro molding mechanism, shape changes of molded PMMA were studied with experimental variables. And the durability of mold insert was investigated, too. The results show that the most important factor in molding processes was the mold temperature that is closely related to the filling of the melt into the micro cavity. And the holding time before cooling showed a great effect on the quality of molded PMMA.

Utilization of Putrescine by Streptococcus pneumoniae During Growth in Choline-limited Medium

  • Polyamines such as putrescine are small, ubiquitous polycationic molecules that are required for optimal growth of eukaryotic and prokaryotic cells. These molecules have diverse effects on cell physiology and their intracellular content is regulated by de novo synthesis and uptake from the environment. The studies presented here examined the structure of a putative polyamine transporter (Pot) operon in Streptococcus pneumoniae (pneumococcus) and growth of pneumococci in medium containing putrescine substituted for choline. RT-PCR experiments demonstrated that the four genes encoding the Pot system are co-transcribed with murB, a gene involved in an intermediary step of peptidoglycan synthesis. Pneumococci grown in chemically-defined media (CDM) containing putrescine without choline enter logarithmic phase growth after 36-48 hs. However, culture density at stationary phase eventually reaches that of choline-containing medium. Cells grown in CDM-putrescine formed abnormally elongated chains in which the daughter cells failed to separate and the choline-binding protein PspA was no longer cell-associated. Experiments with CDM containing radiolabeled putrescine demonstrated that pneumococci concentrate this polyamine in cell walls. These data suggest that pneumococci can replicate without choline if putrescine is available and this polyamine may substitute for aminoalcohols in the cell wall teichoic acids.
