The Disruption Yeast Cell Wall by chemical Treatment (화학적 처리방법에 의한 효모의 세포벽 제거)

  • The cell of Kluyveromyces fragilis yeast, which is worthy of an algal substitute, was disrupted by a chemical treatment to increase the digestion of filter-feeders that yeasts are fed to. The optimum conditions of the chemical treatment were obtained by incubating yeasts at 3$0^{\circ}C$ for one hour after treated by 1 M of Na$_{2}$-EDTA that was dissolved in 0.2 M of Tris-buffer and by 0.3 m of 2-mercaptoethanol. The percentage of protop[last production was about 30%. The percentage could be doubled by the pretreatment of three times of 30 seconds sonication.

  • The protoplast formation of Streptomyces mitakaensis was monitored with scanning electron microscopy and transmission electron microscopy. The normal cells formed regular mycelium and spore, and their cell wall and cell membrane appeared to be normal, but the cell wall of the lysozyme treated cells (1 mg/$m\ell$) was damaged, which was finally disappeared from cells to become protoplast in 30 to 60 minutes.

A Comparative Study on Discharge Characteristics of FHD and QFHD AC PDP (FHD와 QFHD 해상도를 가지는 AC PDP의 방전특성 비교연구)

  • We have investigated the luminous efficiency of various cell resolution and structure from 50" FHD to 50" QFHD Plasma Display. The suggested test panels have two different cell array types which are the delta and matrix cell array type. The results showed that, in the case of the suggested QFHDs, the firing and sustain voltage were increased and voltage margin was decreased. These results are caused by the reduced wall voltage and increased charged particle loss, at the side wall. The luminance of the suggested QFHDs was lower from 20% to 40% than that of the suggested FHDs and the power consumption was higher from 42% to 83% than that of the suggested FHDs. In conclusion, the maximum luminous efficiency of the suggested QFHD(D110) has reached about 38%, compared with suggested FHDs($\fallingdotseq$ 2.7 lm/W).

Plant Cell Wall Polysaccharides as Potential Resources for the Development of Novel Prebiotics

  • Prebiotic oligosaccharides, with a degree of polymerization (DP) of mostly less than 10, exhibit diverse biological activities that contribute to human health. Currently available prebiotics are mostly derived from disaccharides and simple polysaccharides found in plants. Subtle differences in the structures of oligosaccharides can cause significant differences in their prebiotic properties. Therefore, alternative substances supplying polysaccharides that have more diverse and complex structures are necessary for the development of novel oligosaccharides that have actions not present in existing prebiotics. In this review, we show that structural polysaccharides found in plant cell walls, such as xylans and pectins, are particularly potential resources supplying broadly diverse polysaccharides to produce new prebiotics.

Value of Porous Titanium Alloy Plates for Chest Wall Reconstruction after Resection of Chest Wall Tumors

  • Objective: To explore the value of porous titanium alloy plates for chest wall reconstruction after resection of chest wall tumors. Materials and Methods: A total of 8 patients with chest wall tumors admitted in our hospital from Jan. 2006 to Jan. 2009 were selected and underwent tumor resection, then chest wall repair and reconstruction with porous titanium alloy plates for massive chest wall defects. Results: All patients completed surgery successfully with tumor resection-induced chest wall defects being $6.5{\times}7cm{\sim}12{\times}15.5$ cm in size. Two weeks after chest wall reconstruction, only 1 patient had subcutaneous fluidify which healed itself after pressure bandaging following fluid drainage. Postoperative pathological reports showed 2 patients with costicartilage tumors, 1 with squamous cell carcinoma of lung, 1 with lung adeno-carcinoma, 1 with malignant lymphoma of chest wall, 2 with chest wall metastasis of breast cancers and 1 with chest wall neurofibrosarcoma. All patients had more than 2~5 years of follow-up, during which time 1 patient with breast cancer had surgical treatment due to local recurrence after 7 months and none had chest wall reconstruction associated complications. The mean survival time of patients with malignant tumors was ($37.3{\pm}5.67$) months. Conclusions: Porous titanium alloy plates are safe and effective in the chest wall reconstruction after resection of chest tumors.

Morphological Alteration of Cell Organelles Affected by UV-B Radiation in Rice Leaf Tissues (자외선에 의한 벼 엽 세포 소기관의 형태적 변화)

  • This experiment was performed to observe morphological changes in rice leaf tissue caused by a successive UV-B radiation. Effect of UV-B radiation on the structural alteration of tissue was not visually found, however, Photosynthate containing phosphate was sharply reduced in proportion with an increase of UV-B radiation. Fundamental components of cuticle layer were being degraded after 6 h of UV-B radiation compared to the control. UV-B-induced mesophyll cell appeared altered because of water stress, the shape of chloroplast appeared to be considerably shrunk and chloroplast thylakoid membranes were severely destructed. Primary cell wall of UV-B-stressed tissue was entirely scattered or disappeared, and the secondary cell wall due to lignin synthesis and deposition resulted in being thickened, almost 2-times, compared with the control.

  • In fast grown softwood, there are very large changes in material properties going outward from the pith to bark such as anatomical, physical and mechanical characteristics. Some of variations in anatomical properties with annual ring were then examined from Pinus koraiensis, Larix leptolepis, and Chamaecyparis obtusa, which are major softwoods of plantation in Korea. The large variations of annual ring width during young age of tree tended to stabilize after 25year through the transitional period in 17~23year. The ring density was 1.5~2.4 in 1~10year period, and 3.5~6.3 in 30~35year period, in which juvenile and mature wood were certainly assumed to be formed, respectively. Variations of tracheid length showed functional relationships with annual rings as logarithm. Demarcation between juvenile wood and mature wood could be 16~19year, which was determined from increase rate of tracheid length of 0.2%. Cell wall thickness increased with increase of annual ring even though large variations were observed as well. Variations of cell wall thickness within species were pronounced in latewood than earlywood. The increase of cell wall thickness from juvenile wood to mature wood was predominant in Larix leptolepis as 2.0times, and least in Chamaecyparis obtusa as 1.1 times. Cell diameters showed trends of increase during young age of 1~15year, and consistent afterward. The variations of cell diameter between radial and tangential direction were greater in latewood, and most pronounced in Chamaecyparis obtusa.

Characterization and distribution of phenolics in carrot cell walls

  • The purpose of this study was to investigate the release of p-hydroxybenzoic acid and other compounds from cell wall materials(CWM) and their cellulose fraction from carrot with chemical and enzymatic hydrolysis. To investigate this effect on cell wall chemistry of carrot, alcohol insoluble residue(AIR) of CWM were prepared and were extracted sequentially with water, imidazole, CDTA(-1, -2), Na$_2$CO$_3$(-1, -2), KOH(0.5, 1.0 and 4M), to leave a residue. These were analysed for their carbohydrate and phenolic acids composition. Arabinose and galactose were the main noncellulosic sugars. Phenolics esterified to cell walls in carrot were found to consist primarily of p-hydroxybenzoic acid with minor contribution from vanillin, ferulic acid and p-hydroxybenzaldehyde. p-Hydroxybenzoic acid was quite strongly bound to the cell wall. The contents of p-hydroxybenzoic acid in 0.5M KOH, Na$_2$CO$_3$-2, IM KOH, and ${\alpha}$-cellulose were 2,097, 1,360, 1,140, and 717 $\mu\textrm{g}$/g AIR from CWM, respectively. Alkali labile unknown aromatic compound(C$\sub$7/H$\sub$10/O$_2$) was found in ${\alpha}$ -cellulose hydrolyzate digested with driselase and cellulase. This compound was also found in hydrolyzate of 2 M trifluoroacetic acid at 120$^{\circ}C$ for 2 hours. Driselase treatment solubilized only 46.6 $\mu\textrm{g}$/g of the p-hydroxybenzoic acid from carrot AIR. These results indicate that p-hydroxybenzoic acid was associated with neutral polysaccharides, long chain galactose and branched arabinan from graded alcohol precipitation.

  • The possibility that DNA could move out of the single cells isolated from geranium (Pelargonium zonale hybrids, 'PintoScarlet') callus was determined by the elechophoretic DNA analysis after treatment of low pH, various concentrations of KNO$_3$, 2,4-D, and GA$_3$ followed by the centrifugal force, all of which are hewn to and the physico-chemical properties of the cell wall. The centrifugal force of l,800 xg was need for DNA migration after the above treatment, but 7k300 xg was required without the treatments. In this experiment the optimum concentration (300 mg/L) of sodium dodecyl sulfate (SDS) used as an anion detergent to collect the negatively charged DNA was very critical not to damage the cell wall It can be concluded that the centrifugal force played a key role for the DNA migration through the cell wall, and the treatments of low pH (4.0), 0.5% KNO$_3$, 1.5 mg/L GA$_3$and 1mg/L 2,4-D further increased the DNA migration.

Changes in Cell Wall Components, and Solubilization and Depolymerization of Pectin and Neutral Sugar Polymers during Softening of 'Tsugaru' Apples ('쓰가루'사과의 연화에 따른 세포벽성분의 변화와 펙틴 및 중성다당류의 가용화와 분해)

  • This study was carried out to investigate changes in cell wall components and solubilization and depolymerization of pectin and neutral sugar polymers during softening of 'Tsugaru' apples. Pectic polysaccharides were solubilized in different solvents, distilled-water, 0.05 M CDTA, 0.05 M $Na_2CO_3$, and 8 M KOH, from cell wall materials during fruit softening. The uronic acid contents in distilled-water fraction rapidly increased along with fruit softening at 4 weeks after ambient storage. In the change of non-cellulosic neutral sugars in the cell wall of ‘Tsugaru’ fruits, the major sugar was galactose and arabinose in distilled-water, 0.05 M CDTA and 0.05 M $Na_2CO_3$ soluble fractions, and it was glucose, galactose and xylose in 8 M KOH fraction. Especially the change of galactose contents in distilled-water fraction was increased greatly along with fruit softening. When uronic acid polymers (UAP) and carbohydrate polymers (CP) in distilled-water fraction were filtered and separated using Sepharose CL-2B column, the high molecular UAP and CP were degraded to the low molecular ones from at harvest to softening fruit. Thus, the amount of high molecular polymers were greatly decreased along with fruit softening.
