A Droplet-Manipulation Method using Opto-thermal Flows on Amorphous Silicon Thin Film (비결정질 실리콘 박막 상에서의 광열 유동을 이용한 액적 조작)

  • We present a droplet-manipulation method using opto-thermal flows in oils. The flows are originated from Marangoni and buoyancy effects due to temperature gradient, generated by the adsorption of light on an amorphous silicon thin film. Using this method, we can transport, merge and mix droplets in an extremely simple system. Since the temperature rise during the operation is small, this method can be used for biological applications without the damage on cell viability.

  • In-plane type micro piezoelectric micro grippers with pneumatic lines for manipulation biological cells and micro parts were designed, fabricated, and characterized. Micro grippers were fabricated through the final micro-sanding process after wafer level bonding between the etched 4' Si wafer with pneumatic channels and 4' glass wafer. Displacements between two jaws of fabricated micro grippers were linearly increased with applying voltages to piezoelectric actuator. In the case of applying 80 V, the displacement between two jaws was $160{\mu}m$. Using fabricated micro grippers, manipulation tests for biological cell and micro parts with the sizes less than $100{\mu}m$ are in process.

Contractile Force Measurements of Cardiac Myocytes Using a Micro-manipulation System

  • In order to develop a cell based robot, we present a micro-mechanical force measurement system for the biological muscle actuators, which utilize glucose as a power source. The proposed measurement system is composed of a micro-manipulator, a force transducer with a glass probe, a signal processor, an inverted microscope and video recording system. Using this measurement system, the contractile force and frequency of the cardiac myocytes were measured in real time and the magnitudes of the contractile force of each cardiac myocyte under different conditions were compared. From the quantitative experimental results, we could estimate that the force of cardiac myocytes is about $20\sim40{\mu}N$, and show that there are differences between the control cells and the micro-patterned cells.

  • This paper proposes a bit manipulation accelerator (BMA) having application specific instructions, which efficiently supports scrambling, convolutional encoding, puncturing, and interleaving. Conventional DSPs cannot effectively perform bit manipulation functions since かey have multiply accumulate (MAC) oriented data paths and word-based functions. However, the proposed accelerator can efficiently process bit manipulation functions using parallel shift and Exclusive-OR (XOR) operations and bit jnsertion/extraction operations on multiple data. The proposed BMA has been modeled by VHDL and synthesized using the SEC $0.18\mu m$ standard cell library and the gate count of the BMA is only about 1,700 gates. Performance comparisons show that the number of clock cycles can be reduced about $40\%\sim80\%$ for scrambling, convolutional encoding and interleaving compared with existing DSPs.

  • This paper describes a cell programming environment that deals with problems associated with programming Flexible Manufacturing Cells(FMCs). The environment consists of the cell programming editor and the automatic generation module. In the cell programming editor, cell programmers can develop cell programs using task level description set which supports task-oriented specifications for manipulation cell activities. This approach to cell programming reduces the amount of details that cell programmers need to consider and allows them to concentrate on the most important aspects of the task at hand. The automatic generation module is used to transform task specifications into executable programs used by cell constituents. This module is based on efficient algorithm and expert systems which can be used for optimal path planning of robot operations and optimal machining parameters of machine tool operations. The development tool in designing the environment is an object-oriented approach which provides a simple to use and intuitive user interface, and allows for an easy development of object models associated with the environment.

Regulatory T Cell Therapy for Autoimmune Disease

  • It has now been well documented in a variety of models that T regulatory T cells (Treg cells) play a pivotal role in the maintenance of self-tolerance, T cell homeostasis, tumor, allergy, autoimmunity, allograft transplantation and control of microbial infection. Recently, Treg cell are isolated and can be expanded in vitro and in vivo, and their role is the subject of intensive investigation, particularly on the possible Treg cell therapy for various immune-mediated diseases. A growing body of evidence has demonstrated that Treg cells can prevent or even cure a wide range of diseases, including tumor, allergic and autoimmune diseases, transplant rejection, graft-versus-host disease. Currently, a large body of data in the literature has been emerging and provided evidence that clear understanding of Treg cell work will present definite opportunities for successful Treg cell immunotherapy for the treatment of a broad spectrum of diseases. In this Review, I briefly discuss the biology of Treg cells, and summarize efforts to exploit Treg cell therapy for autoimmune diseases. This article also explores recent observations on pharmaceutical agents that abrogate or enhance the function of Treg cells for manipulation of Treg cells for therapeutic purpose.

Identification of Gene Locus by the Somatic Cell Hybridization in Chicken (체세포 융합에 의한 닭의 유전인자구명에 관한 연구)

  • This experiment was conducted to improve the performance of chickens by the precise separation and analysis of chromosomes which are integrated genetic materials, and by the use of gene manipulation techniques. Following are the main results obtained. 1. When the chromosomes were separated through the leucocyte culture and analyzed by Giemsa banding techniques (especially by the method in which 20 layers of banding patterns could be found in chromosome #1), the normal Patterns of chromosomes #l-9 and sex chromosomes, and the location of constitutive heterochromatin without any gene activities in all chromosomes were discovered. 2. To utilize the primodial germ cells (PGC) as the genetic vector which is one of the most important gene manipulation techniques, PGC's from triploid were transplanted to normal host embryos. Since the donor PGC's(3n) were found in the gonads of growing host embryos gene manipulation in poultry using PGC's, seemed to be possible.

  • 생물학적인 셀 인젝션 기술은 유전자 주입, 시험관 배양, 인공수정 및 신약개발 분야에서 광범위하게 사용되어 오고 있는 기술이다. 생물공학에서 다루는 셀 인젝션 기술은 크게 착생세포 인젝션과 서스펜디드 셀 인젝션으로 구분할 수 있다. 최근 상용화 장비로 출시되고 있는 것들은 착생세포에 대한 자동 인젝션 시스템이 대부분을 차지하고 있다. 반면, 서스펜디드 셀 인젝션 시스템의 경우는 비교적 최근들어 자동화 장비 및 방법론의 개발에 대한 논의가 이루어지고 있는 실정으로 실제 수많은 연구자들의 노력에 힘입어 서스펜디드 셀을 대상으로 한 통합 자동화 셀 인젝션 시스템의 개발이 가속화되고 있기는 하지만 이에 대한 만족할 만한 성과는 아직 이루어 지지 않고 있는 실정이다. 본 논문은 서스펜디드 셀을 대상으로 한 인젝션 시스템 개발에 관한 것으로 특히 셀 홀딩 시스템과 최적의 인젝션 피펫 궤적을 결정하기 위한 시스템 개발에 관한 것이다. 본 논문에서 다루어지는 서스펜디드 셀은 통합 자동화를 위하여 특별히 고안된 셀홀딩 시스템에 의하여 배열의 형태로 고정되며, 셀 인젝션 시스템은 엠브리오와 인젝션 피켓을 이미지 프로세싱 기술에 의하여 인식하고 피펫의 인젝션 궤적을 결정하는 것을 포함하고 있다.

Single beam acoustic tweezers for biomedical applications (단일 빔 음향 집게를 이용한 바이오메디컬 응용 연구)

  • Acoustic tweezers represent an exceptionally versatile and adaptable collection of instruments that harness the intrinsic power of sound waves to manipulate a wide spectrum of bioparticles, ranging from minuscule extracellular vesicles at the nanoscale to more substantial multicellular organisms measuring in millimeters. This field of research has witnessed remarkable progress over the course of the past few decades, primarily in the domain of Single Beam Acoustic Tweezers (SBAT) which utilizes a single element transducer for its operation. Initially conceived as a method for particle trapping, SBAT has since evolved into an advanced platform capable of achieving precise translation of cells and organisms. Recent groundbreaking advancements have significantly enhanced the capabilities of SBAT, unlocking new functionalities such as particle/cell separation and controlled deformation of single cells. These advancements have propelled SBAT to the forefront of bioparticle/cell manipulation, gathering attention within the scientific community. This review explores the core principles of SBAT and how sound waves affect bioparticles/cells. We aim to build a strong conceptual foundation for understanding advancements in this field by detailing its principles and methodologies.
