Marine Environmental and Phytoplankton Monitoring in Wando Coastal Waters in August during the Period of 2009-2010 (2009-2010년 8월 완도근해 해역의 해양환경 및 식물플랑크톤 모니터링)

  • This study was determined to analysis marine environmental and phytoplankton monitoring in Wando coastal waters in August during the period of 2009-2010 and provided the basic environmental data against Southwest Sea of Korea. In August, 2009, the distribution of water temperature and salinity showed an unique characteristics for all of stations, which was associated with a long duration of rainfall after July, 2009 and a lack of sunshine. In the lower of August, 2010, the stratification of water temperature and salinity was shown, which was caused by an intensive sunshine after the middle of August, 2010. Gangjin Bay and Gogeundo waters showed a highly different water temperature and transparency. It is thought that two regions have a distinct water mass and characteristics. In dissolved oxygen and oxygen saturation were higher according to higher water depth regardless of stations. In August, 2009, different dominant species occurred that Gangjin Bay showed a higher cell density of diatom, but Gonyaulax polygramma played an important role in dominant species in Gogeundo waters. It is associated with the introduction red tide water of G. polygramma occurring to southern coastal water in August, 2009. In August, 2010, diatom was extremely higher cell density compared with that of dinoflagellate that regarded as adaptive microrganism against higher quantity of suspended solid in Wando waters. Although Wando has a record of Cochlodinium red tide, this species may be difficult to grow and persist. Dominant species, diatom, play an important role in food organism for shellfish and forming a ecological lineage.

Effects of Optical Characteristics on the Growth of Benthic Microalga, Nitzschia sp. and Its Growth Kinetics of Phosphate for Bioremediation (생물적 환경정화를 위한 부착미세조류 Nitzschia sp.의 생장에 미치는 광학적 특성과 그에 따른 인산염 성장 동력학)

  • To suggest possible to bioremediation by benthic microalgae Nitzschia sp. isolated from the Jinhae Bay, the studies investigated the effects o flight quality and quantity on the growth of Nitzschia sp. and its growth kinetics for phosphate investigated. The Nitzschia sp. was cultured under blue (450 nm), yellow (590 nm) and red wavelength (650 nm) using light emitting diode (LED) and mixed wavelengths using a fluorescent lamp. The maximum specific growth rate showed the Nitzschia sp. under blue wavelength, although photoinhibition was observed above $100\;{\mu}mol\;m^{-2}\;s^{-1}$. Mixed wavelengths were also observed by decreasing the maximum cell density from high irradiances (>$100\;{\mu}mol$ photons $m^{-2}\;s^{-1}$). The compensation photon flux density ($I_0$) calculated from the mixed wavelengths equated to a depth of 4-10 m in Jinhae Bay, and was lower in the summer season than the depth due to suspended matter (ca. 4 m). Thus, the suitable depth for maximum growth of Nitzschia sp. might be extremely limited. In the growth kinetics for phosphate, half-saturation constant ($K_s$) was similar among different wavelengths, although the maximum growth rate was varied among different wavelengths. Because the $K_s$ was high than that of the phytoplankton, Nitzschia sp. might have adapted to the high nutrient concentrations, and have effective nutrient storage in the cell quota. Thus, Nitzschia sp. may be a useful species for bioremediation of the benthic layer in polluted inner bays by means of irradiated specific wavelength as blue.

Bacteriological Quality of Sea Water at Swimming Beaches in Busan Area (부산시내 해수욕장해수의 세균학적 수질에 관한 연구)

  • This experiment was carried out to evaluate the sanitary quality of sea water at popular swimming beaches such as Haeundae, Kwanganri and Songjeong which are located in the suburbs of Busan Korea. One hundred and seventy five sea water samples were collected from the 25 stations during the both summer seasons in 1979 and in 1980. The sampling stations were established in the above 3 swimming beaches and the estuary of Suyeong river (See Fig.1.). To evaluate the sanitary quality, coliform group, fecal coliform, viable cell count and Vibrio parahaemolyticus were observed. The median value of coliform group MPN of the samples ranged 2,300 to 6,500/100m1 in Kwanganri, 670 to 910/100m1 in Haeundae and 430 to 460/100m1 in Songjeong swimming beach. Geometric mean of fecal coliform MPN was 3,200/100m1 in Kwanganri, 500/100m1 in Haeundae and 360/100m1 in Songjeong swimming beach. Composition of coliform was $23\%$ Escheri-chia coli group, $20\%$ Citrobacter freundii group, $41\%$ Klebsiella aerogenes group and $16\%$others. The density of Vibrio parahaemolyticus in Kwanganri was much higher than those of in other beaches by 5 to 6 times. Plate count of the samples in the estuary of Suyeong river was varied from 300 to $8.5\times10^6/ml$ during the study period. No remarkable difference was observed in bacterial density by tide in swimming beaches but the bacterial density of sea water at ebb tide was obviously higher than those of the samples at flood tide in the estuary of Suyeong river.

Prevention of Power Overshoot and Reduction of Cathodic Overpotential by Increasing Cathode Flow Rate in Microbial Fuel Cells used Stainless Steel Scrubber Electrode (스테인리스강 수세미 전극을 사용한 미생물연료전지의 전력 오버슈트 예방과 환원조 유속 증가에 의한 환원전극 과전압 감소)

  • Power overshoot phenomenon was observed in microbial fuel cells (MFCs) used non-catalyzed graphite felt as cathode. Voltage loss in MFCs was mainly caused by cathode potential loss. Cheap stainless steel scrubber, which has high conductivity, and Pt/C coated graphite felt as cathode were used for overcoming power overshoot and reducing the cathode potential loss in MFCs. The MFCs used stainless steel scrubber showed no power overshoot even slow catholyte flow rate and produced 29% enhanced maximum current density ($23.9A/m^3$) than MFCs used non-catalyzed graphite felt while the power overshoot phenomenon was existed in Pt/C coated MFCs. Increasing catholyte flow rate resulted in disappearing power overshoot of MFCs used non-catalyzed graphite felt. In addition, maximum power density and current density of both MFCs used non-catalyzed graphite felt and stainless steel scrubber increased by 2-3.5 times. Cathode potential losses in all region of activation loss, ohmic loss, and mass transport loss were reduced according to increase of catholyte flow rate. Therefore, stainless steel scrubber has advantages that are economical materials as electrode and prevents power overshoot, leading to enhance electricity generation. In addition, increasing catholyte flux is one of great solution when power overshoot caused by cathodic overpotential is observed in MFCs.

The Effect of Drought Simulated by Discharge Control on Water Quality and Benthic Diatom Community in the Indoor Experimental Channel (인공하천에서 유량감소로 모사한 가뭄효과가 수질 및 부착돌말류 군집에 미치는 영향)

  • We investigated an ecological impact of drought simulated by discharge depletion on the water quality and benthic diatom community in the indoor experimental channel. As artificial substrates slide-glass was installed in acrylic channel for 16 days. Channels were supplied continuously with eutrophic lake water with a discharge rate of 6 L $min^{-1}$ in duplication during the colonized period. And then during the discharge depletion period, three discharge rates were provided: NDF (No depletion of flow rate (Control): 6 L $min^{-1}$), LDF (Low depletion of flow rate: 3 L $min^{-1}$) and HDF (High depletion of flow rate: 1 L $min^{-1}$). Environmental factors in the water, such as suspended solid, Chl-$a$ and nutrients concentration, were measured with periphytic algae including AFDM (ash free dry matter), Chl-$a$ concentration and cell density at 1-day intervals. Light intensity increased significantly with discharge depletion (F=229.5, p= 0.000). $NH_4$-N concentration was highest at HDF. Suspended solid in outflowing water decreased at HDF (88%), LDF (97%) and NDF (99%), compared to inflowing water (100 %). Chl-$a$ in substrates increased more than two times at LDF and HDF than NDF (F= 8.399, p=0.001). Also AFDM and benthic diatom density increased significantly at LDF and HDF than NDF (F=9.390, p=0.001; F=6.088, p=0.007). In all experimental groups, $Aulacoseira$ $ambigua$, $Achnanthes$ $minutissima$ and $Aulacoseira$ $granulata$ were dominant species accounting for greater than 10% of benthic diatom density. The most dominant species, $A.$ $ambigua$ was highest at LDF, followed by HDF and NDF (F=8.551, p=0.001). In conclusion, the effect of drought simulated by discharge depletion in an artificial stream ecosystem caused significant changes on water quality and benthic diatom biomass. This result provides a useful data to understand the effect of draught on stream ecosystem in situ.

Effects of Green Tea Catechin on Bone Disorder in Long-Term Cadmium Treated Rats (만성 카드뮴 처치 쥐에서 골조직 장애에 미치는 녹차 Catechin의 영향)

  • The purpose of this study was to investigate the effects of green tea catechin on change of bone tissue in long-term cadmium treated rats. Sprague-Dawley male rats weighing 100$\pm$10 g were randomly assigned to one normal group and three cadmium treated groups. Cadmium groups were classified to catechin free diet group (Cd-0C group), 0.25% catechin diet group (Cd-0.25C group) and 0.5% catechin diet group (Cd-0.5C group) according to the levels of catechin supplement. Animals were raised for 20 weeks. Cadmium were supplied as drinking water of 50 ppm Cd$^{2+}$. Effects of catechin were analyzed on changes of bony tissue in long-term cadmium treated rats by determining the accumulated cadmium in bone and bone mineral density and micro- photographs of bony tissue. The cadmium accumulation of tibia and femur were higher in Cd-treated groups than in normal group, but they was lowered by catechin supplementation. The bone mineral density (BMD) of tibia and femur in Cd-0C group was significantly lower than in normal group, but it of catechin supplemetation group was similar to normal group. Microphological changers were appeared under a light microscope and an electro microscope reveal no structural changes in bony spicules, marrow cell distribution and cellular morphology in all groups. The bone weight and length tend to decrease in Cd-0C groups. Catechin supplementation in long-term cadmium treated rats depressed the cadmium accumulation in bony tissue that led to improve the bone mineral density in tibia and femur.r.

  • Immunoinhibitory protein extracted from sonicated Treponema denticola have been shown to suppress cell cycle progression of human lymphocytes. To study in detail about the effect of this microorganism on the function of lymphocytes. we investigated the levels of Interleukin-2 (IL-2) and Interleukin-4 (IL-4) production by T lymphocytes before and after the addition of $12.5{\;}\mu\textrm{g}/ml$ T. denticola sonicated extracts. In this study. levels of IL-2 and IL-4 produced from T cells pretreated with sonicated extracts were evaluated by using the quantitative sandwich enzyme immunoassay technique. In response to phytohemagglutinin (PHA) stimulation. T cell produced increased levels of IL-2 and IL-4. However. the expressions of both cytokines were significantly inhibited when PHA activated-T cells were pre-exposed to sonicated T. denticola extracts (p < 0.05). These findings suggest that the T. denticola sonicated extracts induced-immunosuppression in Th1 and Th2 cell functions could be a part of the pathogenic mechanism of the endodontic failure associated with this microorganism.

  • Purpose : The essential triad for nerve regeneration is nerve conduit, supporting cell and neurotrophic factor. In order to improve the peripheral nerve regeneration, we used polyglycolic acid(PGA) tube and brain-derived neurotrophic factor(BDNF) gene transfected Schwann cells in sciatic nerve defects of SD rat. Materials and methods : Nerve conduits were made with PGA sheet and outer surface was coated with poly(lactic-co-glycolic acid) for mechanical strength and control the resorption rate. The diameter of conduit was 1.8mm and the length was 17mm Schwann cells were harvested from dorsal root ganglion(DRG) of SD rat aged 1 day. Schwann cells were cultured on the PGA sheet to test the biocompatibility adhesion of Schwann cell. Human BDNF gene was obtained from cDNA library and amplified using PCR. BDNF gene was inserted into E1 deleted region of adenovirus shuttle vector, pAACCMVpARS. BDNF-adenovirus was multiplied in 293 cells and purified. The BDNF-Adenovirus was then infected to the cultured Schwann cells. Left sciatic nerve of SD rat (250g weighing) was exposed and 14mm defects were made. After bridging the defect with PGA conduit, culture medium(MEM), Schwann cells or BDNF-Adenovirus infected Schwann cells were injected into the lumen of conduit, respectively. 12 weeks after operation, gait analysis for sciatic function index, electrophysiology and histomorphometry was performed. Results : Cultured Schwann cells were well adhered to PGA sheet. Sciatic index of BDNF transfected group was $-53.66{\pm}13.43$ which was the best among three groups. The threshold of compound action potential was between 800 to $1000{\mu}A$ in experimental groups which is about 10 times higher than normal sciatic nerve. Conduction velocity and peak voltage of action potential of BDNF group was the highest among experimental groups. The myelin thickness and axonal density of BDNF group was significantly greater than the other groups. Conclusion : BDNF gene transfected Schwann cells could regenerate the sciatic nerve gap(14mm) of rat successfully.

Chemo-Mechanical Analysis of Bifunctional Linear DGEBA/Linear Amine (DDM, DDS) Resin Casting Systems (DGEBA/방향족 아민(DDM, DDS) 경화제의 벤젠링 사이의 관능기 변화가 물성 변화에 미치는 영향에 대한 연구)

  • To determine the effect of chemical structure of linear amine curing agents on thermal and mechanical properties, standard epoxy resin DGEBA was cured with diaminodiphenyl methane (DDM), diaminodiphenyl sulphone (DDS) in a stoichiometrically equivalent ratio. From this work, the effect of aromatic amine curing agents. In contrast, the results show that the DGEBA/DDS cure system having the sulfone structure between the benzene rings had higher values in the conversion of epoxide, density, shrinkage (%), glass transition temperature, tensile modulus and strength, flexural modulus and strength than the DGEBA/DDM cure system having methylene structure between the benzene rings, whereas the DGEBA/DDM cure system presented higher values in the maximum exothermic temperature, thermal expansion coefficient, and thermal stability. These results are caused by the relative effects of sulfone group having strong electronegativity and methylene group having (+) repulsive property and stem from the effect of the conversion ratio of epoxide group. The result of fractography shows that the each grain size of the DDM/DGEBA system with feather-like structure is larger than that of the DDS/DGEBA system.

EXPRESSION OF PROTEIN KINASE C ISOFORMS IN CHEMICAL CARCINOGEN-INDUCED NEOPLASTIC TRANSFORMATION OF HUMAN EPITHELIAL CELLS (화학적 발암화에 따른 Protein Kinase C의 발현 변화)

  • Protein kinase C (PKC) is known to play a pivotal role in neoplastic transformation cells and its high expression is often found in a variety of types of tumors including oral cancer. While PKC is associated with the altered signal transduction pathway of the tumor cells, it is still unclear which isoform is involved in the carcinogenesis process. Since the cellular distributions and the roles of PKC are isoform-specific, it is very important to identify the specific target molecules to improve our understanding of the carcinogenesis processes. Thus, the present study attempted to perform chemical carcinogen-induced neoplastic transformation of human epithelial cells and analyze the specific isoform of PKCs involved in the cellular transformation. The study analyzed overall PKC responses upon MNNG(N-Methyl-N'-nitro-N-nitroso guanidine) exposure with [$^3H$] PDBu binding assay. PKC translocation was observed at high doses of MNNG treatment in the presence of extracellular calcium. Such effects were not observed in the absence of extracellular calcium. Translocational effects with exposure of MNNG was further enhanced in the presence of hydrocortisone. The result suggests that the type of PKC involved may be $Ca^{2+}$-dependent classical isoform and steroid hormone enhances PKC activation. Among cPKC isoforms examined, only $PKC-{\alpha}$ and r showed significant translocation of protein levels from cytosolic fraction to membrane fraction, as analyzed by immunoblot. $PKC-{\varepsilon}$ in nPKC class showed an inch·eased translocation, but other forms in this class did not show the effect. None of isoforms in aPKC class was affected by MNNG treatment. The study demonstrated that there was a certain specificity in the patterns of isoform induction follwong chemical carcinogen exposure and helped identify all the types of PKC isoforms expressed in human epithelial cells. It was revealed that PKC isoforms were activated in an early resonse to chemical carcinogen, suggesting that PKC be associated with carcinogenesis process from an early stage in this particular cell system. The study will contribute to improving our understanding of chemical-induced carcinogenesis in human cells and may provide a scientific basis to introduce the specific PKC inhibitors as an anticancer drug of epithelial cell-origin cancers including oral cancer.
