Two major endoglucanase genes (cel7B and cel5A) were cloned from Penicillium decumbens 114-2 using the method of modified thermal asymmetric interlaced polymerase chain reaction (TAIL-PCR). The result of Southern blotting suggested that P. decumbens has a single copy of the cel5A gene and a single copy of the cel7B gene in its chromosomal DNA. The expression levels of cel5A and cel7B were determined by means of real-time quantitative PCR, suggesting that the two genes were coordinately expressed, and repressed by glucose and induced by cellulose. Both endoglucanase genes were expressed in Saccharomyces cerevisiae and the recombinant proteins were purified. The recombinant Cel7B and Cel5A were both optimally active at $60^{\circ}C$ and pH 4.0. The recombinant Cel7B showed more than 8-fold, 30-fold, and 5-fold higher enzyme activities toward carboxymethyl cellulose, barley $\beta$-glucan, and PASC, respectively, in comparison with that of Cel5A. However, their activities toward pNPC and Avicel showed minor differences. The results suggested that Cel7B is a strict endoglucanase, whereas Cel5A showed processivity because of its relative higher ability to hydrolyze the crystal cellulose.
To investigate the function of N-glycosylation of Cel5A (endoglucanase II) from Hypocrea jecorina, two N-glycosylation site deletion Cel5A mutants (rN124D and rN124H) were expressed in Saccharomyces cerevisiae. The weights of these recombinant mutants were 54 kDa, which were lower than that of rCel5A. This result was expected to be attributed to deglycosylation. The enzyme activity of rN124H was greatly reduced to 60.6% compared with rCel5A, whereas rN124D showed slightly lower activity (10%) than that of rCel5A. rN124D and rN124H showed different thermal stabilities compared with the glycosylated rCel5A, especially at lower pH value. Thermal stabilities were reduced and improved for rN124D and rN124H, respectively. Circular dichroism spectroscopy showed that the modification of secondary structure by mutation may be the reason for the change in enzymatic activity and thermal stability.

Our previous study on the binding activity between Cel5H and clay minerals showed highest binding efficiency among other cellulase enzymes cloned. Here, based on previous studies, we hypothesized that the positive amino acids on the surface of Cel5H protein may play an important role in binding to clay surfaces. To examine this, protein sequences of Bacillus licheniformis Cel5H (BlCel5H) and Paenibacillus polymyxa Cel5A (PpCel5A) were analyzed and then selected amino acids were mutated. These mutated proteins were investigated for binding activity and force measurement via atomic force microscopy (AFM). A total of seven amino acids which are only present in BlCel5H but not in PpCel5A were selected for mutational studies and the positive residues which are present in both were omitted. Of the seven selected surface lysine residues, only three mutants K196A(M2), K54A(M3) and K157T(M4) showed 12%, 7% and 8% less clay mineral binding ability, respectively compared with wild-type. The probable reason why other mutants did not show altered binding efficiency might be due to relative location of amino acids on the protein surface. Meanwhile, measurement of adhesion forces on mica sheets showed a well-defined maximum at 69±19 pN for wild-type, 58±19 pN for M2, 53±19 pN for M3, and 49±19 pN for M4 proteins. Hence, our results demonstrated that relative location of surface amino acids of Cel5H protein especially positive charged amino acids are important in the process of clay mineral-protein binding interaction through electrostatic exchange of charges.

An endo-${\beta}$-1,4-glucanase gene, cel5L, was cloned using the shot-gun method from Bacillus sp.. The gene, which contained a predicted signal peptide, encoded a protein of 496 amino acid residues, and the molecular mass of the mature Cel5L was estimated to be 51.8 kDa. Cel5L contained a catalytic domain of glycoside hydrolase (GH) family 5 and a carbohydrate-binding module family 3 (CBM_3). Chromatography using HiTrap Q and CHT-II resulted in the isolation of two truncated forms corresponding to 50 (Cel5L-p50) and 35 kDa (Cel5L-p35, CBM_3-deleted form). Both enzymes were optimally active at pH 4.5 and $55^{\circ}C$, but had different half-lives of 4.0 and 22.8 min, respectively, at $70^{\circ}C$. The relative activities of Cel5L-p50 and Cel5L-p35 for barley ${\beta}$-glucan were 377.0 and 246.7%, respectively, compared to those for carboxymethyl-cellulose. The affinity and hydrolysis rate of pNPC by Cel5L-p35 were 1.7 and 3.3 times higher, respectively, than those by Cel5L-p50. Additions of each to a commercial enzyme set increased saccharification of pretreated rice straw powder by 17.5 and 21.0%, respectively. These results suggest CBM_3 is significantly contributing to thermostability, and to affinity and substrate specificity for small substrates, and that these two enzymes could be used as additives to enhance enzymatic saccharification.
A gene encoding thermostable cellulase A (TmCelA) was isolated from Thermotoga maritima. The open reading frame of TmCelA gene was 774 bp long which predicted to encode 257 amino acid residues with a molecular weight of 29,732 Da. To examine the biochemical properties, the TmCelA was overexpressed in E. coli BL21, and expressed protein was purified. The optimum temperature of recombinant TmCelA was 90-95 ℃, and the optimum pH of recombinant TmCelA was approximately pH 5.0. Recombinant TmCelA was stable at temperature below 90 ℃.
A gene encoding thermostable cellulase B (TmCelB) was isolated from Thermotoga maritima. The open reading frame (ORF) of TmCelB gene was 825bp long which predicted to encode 274 amino acid residues with a molecular weight of 31,732 Da. The 17 amino acid residues from N-terminal of the TmCelB was known as signal peptides. To analyze the enzymatic activity and biochemical properties, the ORF of TmCelB gene excluding a putative signal sequence encoding 17 amino acids were introduced into the E. coli expression vector, pRSET-B, and overexpressed in E. coli BL21. The optimum temperature of recombinant TmCelB was around 95 ℃, and the optimum pH of recombinant TmCelB was around pH 4.5. The recombinant TmCelB was stable at temperature below 100 ℃.
A carboxymethyl cellulase gene, cel5B, was cloned, sequenced, and expressed in Escherichia coli. pRCS20 in E. coli was identified from metagenomic cosmid library of cow rumen for cellulase activity on a carboxymethyl cellulose agar plates. Cosmid clone (RCS20) was partially digested with Sau3AI, ligated into BamHI site of pBluescript II SK+ vector, and transformed into E. coli $DH5{\alpha}$. The insert DNA of 1.3 kb was obtained, designated cel5B, which has the activity of hydrolyzation of CMC. The cel5B gene had an open reading frame (ORF) of 1,059 bp encoding 352 amino acids with a signal peptide of 48 amino acids and the conserved region, VIYEIYNEPL, belongs to the glycosyl hydrolase family 5. The molecular mass of Cel5B protein expressed from E. coli $DH5{\alpha}$ exhibited to be about 34 kDa by CMC-SDS-PAGE. The optimal pH was 8.0, and the optimal temperature was about $50^{\circ}C$ for its enzymatic activity.
The conversion of a cellulose-producing cell ($Cel^+$) from Gluconacetobacter hansenii PJK (KCTC 10505 BP) to a non-cellulose-producing cell ($Cel^-$) was investigated by measuring the colony forming unit (CFU). This was achieved in a shaking flask with three slanted baffles, which exerted a strong shear stress. The addition of organic acid, such as glutamic acid and acetic acid, induced the conversion of microbial cells from a wild type to $Cel^-$ mutants in a flask culture. The supplementation of $1\%$ ethanol to the medium containing an organic acid depressed the con-version of the microbial cells to $Cel^-$ mutants in a conventional flask without slanted baffles. The addition of ethanol to the medium containing an organic acid; however, accelerated the conversion of microbial cells in the flask with slanted baffles. The $Cel^+$ cells from the agitated culture were not easily converted into $Cel^-$ mutants on the additions of organic acid and ethanol to a flask without Slanted baffles, but some portion of the $Cel^+$ cells were converted to $Cel^-$ mutants in a flask with slanted baffles. The conversion ratio of $Cel^+$ cells to $Cel^-$ mutants was strongly re-lated to the production of bacterial cellulose independently from the cell growth.

A metagenomic library of cow rumen in the pCC1FOS phage vector was screened in $E.$$coli$ EPI300 for cellulase activity on carboxymethyl cellulose agar plates. One clone was partially digested with $Sau$3AI, ligated into the $Bam$HI site of the pBluescript II SK+ vector, and transformed into $E.$$coli$$DH5{\alpha}$. We obtained a 1.5 kb insert DNA, designated $cel$5C, which hydrolyzes carboxymethyl cellulose. The cel5C gene has an open reading frame (ORF) of 1,125 bp encoding 374 amino acids. It belongs to the glycosyl hydrolase family 5 with the conserved domain LIMEGFNEIN. The molecular mass of the Cel5C protein induced from $E.$$coli$$DH5{\alpha}$, as analyzed by CMC SDS-PAGE, appeared to be approximately 42 kDa. The enzyme showed optimum cellulase activity at pH 4.0, and $50^{\circ}C$. We examined whether the $cel$5C gene comes from the 49 identified cow rumen bacteria using PCR. No PCR bands were identified, suggesting that the $cel$5C gene came from the unidentified cow rumen bacteria.
Benzyladenine (BA, 99% purity), MaxCel$^{(R)}$ (1.9% BA), Fruitone (3.5% NAA), MaxCel$^{(R)}$ + Fruitone, a nd s imazine were applied postbloom as fruitlet thinning agents to mature 'Hongro' and 'Fuji' apple (Malus domestica Borkh.) trees. BA and MaxCel$^{(R)}$ were applied at $100mg{\cdot}L^{-1}$ a.i. while Fruitone at $0.1mg{\cdot}L^{-1}$ a.i. and simazine at $400mg{\cdot}L^{-1}$ a.i. All PGRs were applied at 8 days after full bloom (DAFB, 6 mm fruit diameter) in both cultivars, while simazine was treated twice at 7 and 14 DAFB. In 'Hongro', the number of total fruit set per flower cluster in terminal buds was 1.67, 1.84, and 1.81 in MaxCel$^{(R)}$ + F ruitone, MaxCel$^{(R)}$, and simazine applications, respectively, when compared with 2.35 of water control. These reductions in fruit set were mainly attributed to the increased ratio of defruited clusters by the thinning agents. In 'Fuji' apple, the number of total fruit set per flower cluster in terminal buds was 1.29, 1.60, and 1.76 in MaxCel$^{(R)}$ + Fruitone, Fruitone, and MaxCel$^{(R)}$, respectively, when compared with 2.56 of water control in 'Fuji' apple. The addition of Fruitone to the MaxCel$^{(R)}$ promoted the thinning efficacy in both cultivars, compared to MaxCel$^{(R)}$ only. The thinning efficacies were similarly observed with lateral flowers in both cultivars. A significant increase of fruit weight by the postbloom thinning treatments was observed only in the BA application in 'Hongro', while the effect was observed in BA and MaxCel$^{(R)}$ in 'Fuji'. While the soluble solids content increased in the BA, MaxCel$^{(R)}$ and MaxCel$^{(R)}$+Fruitone treatments in both cultivars, other fruit quality attributes were not affected by the application of post-bloom thinning agents.
