Effect of Phospholipid Zwitterionic Surfactant CDP-W on the Characteristics of Liposome (인지질계 양쪽성 계면활성제 CDP-W 첨가가 리포좀 특성에 미치는 영향)

  • In the present work, the interaction of lipid-based zwitterionic surfactant CDP-W with the vesicle membrane of phospholipids was investigated. For this purpose, interfacial properties such as critical micelle concentration (CMC) and surface tension were measured for the zwitterionic surfactant CDP-W and lecithin S100-3. The zeta potential of 1 wt% aqueous surfactant solutions was also measured as a function of pH to determine the iso-electric point of CDP-W surfactant, where the characteristic of CDP-W surfactant changes from a cationic surfactant to an anionic surfactant. Based on the iso-electric point measurement of CDP-W surfactant, the effects of pH change and CDP-W addition on the stability of S100-3 liposome systems were studied, such as average particle size, polydispersity index (PDI), and zeta potential. The effect of CDP-W on the fluidity characteristics of liposome membranes such as fluorescence anisotropy ratio, deformability, and melting point was investigated at pH 6 where the most stable liposomes were prepared to understand the effect of the fluidity of the liposome membrane on the encapsulation efficiency of active materials and the stability of liposome systems.

Selective Gene Transfer to Hepatocellular Carcinoma Using Homing Peptide-Grafted Cationic Liposomes

  • Gene delivery that provides targeted delivery of therapeutic genes to the cells of a lesion enhances therapeutic efficacy and reduces toxic side effects. This process is especially important in cancer therapy when it is advantageous to avoid unwanted damage to healthy normal cells. Incorporating cancer-specific ligands that recognize receptors overexpressed on cancer cells can increase selective binding and uptake and, as a result, increase targeted transgene expression. In this study, we investigated whether a peptide capable of homing to hepatocellular carcinoma (HCC) could facilitate targeted gene delivery by cationic liposomes. This homing peptide (HBP) exhibited selective binding to a human hepatocarcinoma cell line, HepG2, at a concentration ranging from 5 to 5,000 nM. When conjugated to a cationic liposome, HBP substantially increased cellular internalization of plasmid DNA to increase the transgene expression in HepG2 cells. In addition, there was no significant enhancement in gene transfer detected for other human cell lines tested, including THLE-3, AD293, and MCF-7 cells. Therefore, we demonstrate that HBP provides targeted gene delivery to HCC by cationic liposomes.

  • Gene therapy is a method for the treatment of diseases with introducing the gene-engineered materials into a patient with gene-deficiency disease (e.g. cystic fibrosis) or cancer to produce a therapeutic protein in a patient's cells. Successful gene therapy requires establishing both gene expression systems and delivery systems. Viral and non-viral vectors have been used for gene delivery. Viral vectors have a high transfection efficiency, but are limited in relations to issues of safety, toxicity and immunogenecity. Non-viral vectors are easy to prepare and relatively safe. However, non-viral vectors have a low transfection efficiency. Cationic liposomes are the most available among non-viral vectors. Cationic liposomes have been used to transfect cells both in vitro and in vivo experiments. Besides, several formulations containing cationic lipid are being used in clinical trials in cases of cystic fibrosis or cancer. A crucial subject to the further development of gene delivery vectors will be a long-term gene expression with following characteristics; protecting and deliverying DNA efficiently, non-toxic and non-immunogenic, and easy to produce in large scale.

Liposome-mediated in Vivo Delivery of Transgene by Vein Injection

  • Direct gene transfer to mammalian tissues has significant potential for gene therapy and transgenesis. Liposome-mediated in vivo transfection has begun to gain attention as an alternative to viral vectors, and may also be a good mode of transfection in gene transfer. Interestingly, polymerized cationic liposomes are reported to be very stable in the bloods and efficient for in vivo gene transfer. To examine a possible gene delivery in vivo, we investigated the efficacy and safety of the liposome-mediated gene transfer using vein injection in chick or mouse as model animals. The number of injected pGFP-LacZ using either a commercial or home-made liposomes was 8 and 19 at 16 and 7 day of hatch, respectively. One of injected chick of each experiments was analyzed and the rest is being bred. In mouse, 4/22 showed expression of pGFP-LacZ but 8/22 showed no expression and the remaining animals are also being bred. After injection of liposome/pGFP-LacZ complex into wing vein of 7 or 16 day-old chick, pGFP-LacZ was detected in various tissues isolated from not only young chick but also old chick were turned out to possess. exogenous DNA. Transcripts and proteins of the transgene were also detected by RT-PCR or histochemical analysis, respectively. These results suggest that injected DNA were inserted to genome and produced mRNA and proteins in various tissues and may give an important tools for effective gene delivery in gene therapy or transgenesis.

Immunoliposomes Carrying Plasmid DNA : Preparation and Characterization

  • The objective of this study was to characterize immunoliposomes carrying plasmid DNA with optimal encapsulation efficiency and antibody density. Plasmid DNA was encapsulated by the freezing/thawing method into liposomes composed of POPC (1-palmitoyl-2-oleoyl-sn-glycerol- 3-phosphocholine), DDAB (didodecyl dimethyl ammonium bromide), DSPE-PEG 2000 (distearoyl phosphatidyl ethanolamine polyethylene glycol 2000) and DSPE-PEG 2000-maleimide. The liposomes carrying plasmid DNA were extruded through two stacked polycarbonate filters, of different pore size, to control the liposome size. Then, rat IgG molecules were conjugated to the liposomes. The immunoliposomes containing plasmid DNA were separated from the free plasmid DNA and unconjugated IgG by Sepharose CL-4B column chromatography. The DNA amount encapsulated was affected by DDAB (cationic lipid) concentration, the initial amount of plasmid DNA between 10 ${\mu}g$ and 200 ${\mu}g$, the total lipid amount and plasmid DNA size, but not significantly by liposome size. By varying the ratio of DSPE-PEG 2000-maleimide to IgG, the number of IgG molecules per liposome was changed significantly.

Synthesis and Biological Characterization of Indolicidin Analogues

  • Indolicidin has been known to have a broad spectrum of antimicrobial activities against Gram negative and positive bacteria. Its eight analogues were chemically synthesized. The analogue design was based on the analysis of sequence to elucidate the role of some residues in the antibacterial mechanism of indolicidin. Bactericidal activities were assayed against Escherichia coli and Proteus vulgaris, and the membrane perturbing abilities of the peptides were assayed using a dye containing liposome. Among the eight analogues, $[Gly^4, Gly^6]-Indo,\;[Ile^6,Ile^8]-Indo,\;[Lys^{12}]-Indo$ and $[Thr^2,Tyr^9]-Indo$ showed enhanced antibacterial activities. These results suggest that proline and cationic residues are important in the bactericidal activity of indolicidin. We tried to describe the antimicrobial mechanism of indolicidin with these results.

Physical properties and intracellular uptake of polyethyleneglycol-incorporated cationic liposomes (폴리에틸렌글리콜이 도입된 양이온성 리포솜의 물리적 특성 및 세포이입효과)

  • Liposomes as one of the efficient drug carriers have some shortcomings such as their short circulation time, fast clearance from human body by reticuloendothelial system (RES) and limited intracellular uptake to target cell. In this study, polyethylenglycol (PEG)-incorporated cationic liposomes were prepared by ionic complexation of positively charged liposomes with carboxylated polyethyleneglycol (mPEG-COOH). The cationic liposomes had approximately $98.6{\pm}1.0nm$ of mean particle diameter and $42.8{\pm}0.8mV$ of zeta potential value. The PEG-incorporated cationic liposomes had $110.1{\pm}1.2nm$ of mean particle diameter with an increase of about 10 nm compared to the cationic liposomes. Zeta potential value of them was $12.9{\pm}0.6mV$ indicating 30mV decrease of cationic charge compared to the cationic liposomes. The amount of PEG which was incorporated onto the cationic liposomes was assayed by using picrate assay method and the incorporation efficiency was $58.4{\pm}1.1%$. Loading efficiency of model drug, doxorubicin, into cationic liposomes or PEG-incorporated cationic liposomes was about $96.0{\pm}0.7%$. Results of intracellular uptake which were evaluated by flow cytometry analysis of doxorubicin loaded liposomes showed that intracellular uptake of PEG-incorporated cationic liposomes was higher than the cationic liposomes or DSPE-mPEG liposomes. In addition, cytotoxicity of PEG-incorporated cationic liposomes was comparable to cationic liposomes. Consequently, the PEG-incorporated cationic liposomes of which surface was incorporated with PEG by ionic complex may be applicable as anticancer drug carriers that can increase therapeutic efficacy.

  • Unlike cationic liposome/DNA complexes, neutralliposomes containing plasmid DNA are stable in blood and does not selectively entrapped in the lung. The objective of this study was to construct neutral liposomes containing plasmid DNA with optimal encapsulation efficiency. Plasmid DNA (pGL2 clone 753,-6 kb) was encapsulated by the freeze/thawing method into liposomes composed of 1-palmitoyl-2-oleyl-sn-glycerol-3-phosphocholine(POPC), didodecyldimethylammonium bromide(DDAB), distaroylphosphatidyl-ethanolamine polyethylene glycol 2000(DSPE-PEG 2000) and DSPE-PEG 2000-maleimide. Intracellular delivery and anti-tumor activity of polyethyleneglycol liposomes containing cationic lipid (양이온성 지질이 포함된 PEG 리포솜의 세포내 이입 및 항암효력 평가)

  • Liposomes are spherical vesicles composed of lipid bilayer membranes. However, the conventional liposomes have been found to be plagued by rapid opsonization and taken up by the reticuloendothelial system (RES), resulting in shortened circulation time and limited intracellular uptake to target cell. In this study, polyethyleneglycol-cationic liposomes (PCL) containing cationic lipid and DSPE-mPEG were prepared by thin film cast-hydration method. The PEG liposomes had approximately $97.0{\pm}1.3\;nm$ of mean particle diameter and $-21.7{\pm}1.2\;mV$ of zeta potential value. PCL had $96.4{\pm}1.8\;nm$ of mean particle diameter and $-8.7{\pm}1.1\;mV$ of zeta potential value with a decrease of about 10 mV compared to the PEG liposomes. Loading of model drug, doxorubicin (DOX), in liposomes were carried out by using remote loading method and the loading efficiency of DOX in liposomes was about $95.0{\pm}1.9%$. Intracellular uptake and cytotoxicity of PCL were higher than that of PEG liposomes to murine B16F10 melanoma cells. In addition, anti-tumor activity of PCL was similar to that of PEG liposomes on growth of A549 human lung carcinoma in BALB/c mice. Consequently, PCL modified with cationic lipid may be applicable as anticancer drug carriers that can increase intracellular uptake and therapeutic efficacy.

Effect of Amine Oxide Zwitterionic Surfactant on Characteristics of Liposome (아민 옥사이드 양쪽성 계면활성제 첨가가 리포좀 특성에 미치는 영향에 관한 연구)

  • In this study, zwitterionic surfactants were added to liposome systems at different pH conditions to understand the effect of surfactants on liposome characteristics. For this purpose, amine oxide surfactants having different hydrocarbon chain lengths were synthesized and the structure of the resulting product was elucidated by using $^1H$ NMR, $^{13}C$ NMR, and FT-IR. In addition, the physical properties of newly synthesized surfactants such as critical micelle concentration (CMC), surface tension and isoelectric point were measured. The stability characteristics of liposome systems including average particle sizes and zeta potentials were measured by varying pH and hydrocarbon chain lengths of an amine oxide surfactant. Effects of the pH and hydrocarbon chain length of an amine oxide surfactant on fluidity of a liposome membrane were also examined by measuring the deformability and the binding degree between the surfactant and liposome.
