임상간호사의 감성역량, 학습조직화 및 간호조직문화가 간호업무성과에 미치는 영향 (Effects of Emotional Competence, Learning Organization, and Nursing Organization Culture among Nursing Performance of Clinical Nurses)

  • Objectives : The purpose of this study was to identify the effects of emotional competence, learning organization and nursing organizational culture on performance among clinical nurses. Methods : This study was conducted over a six-month period, with 170 nurses from general hospitals with 400 beds in three regions of Korea. Data were gathered from August 25th to September 10th, 2016, using a self-report questionnaire, and analyzed using the IBM SPSS 19.0 program. Results : The significant predictors of nursing performance were self-awareness (${\beta}=.32$), a master's degree or higher (${\beta}=.20$), relationship management (${\beta}=.26$), personal mastery(${\beta}=.23$), and 5~10 years in a clinical career (${\beta}=.16$). These variables explained 47.3% of the variance in nursing performance. Conclusions : To improve nursing performance, interventions that can enhance nurses' emotional competence and the need for program development to enhance their learning organization, as well as related variables, including academic credentials and clinical career, should be considered.

Residents' Participation and Common Activities in an Intentional Community: The Case of the Mindlre Community

  • A new movement in forming community villages for better living standards is becoming popular. In order to sustain a proper community, it is essential to design proper community programs and facilities, which can encourage the proactive participation of residents. In this study, detailed aspects of the Mindlre (Dandelion) community (including formation process and management of the village) are investigated. The Mindlre community is a successful community village in Korea concerned with residents' participation and community activities. The community is located in Sancheong, Gyungsangnamdo province in Korea. The study methods are references, field trips, and interviews with residents. Six areas were surveyed and investigated: the physical environment of the Mindlre community and eco-friendly elements, respondents' background and the motivation to join, residents composition, residents' participation in the formation process of the community, self-management systems and common activities, and residents's life satisfaction. The mindlre Community was founded on Christian religions faith. The primary idea of this village was building a new hope for rural areas without discrimination. The size of the village was expanded with housing for teachers and students who were linked with the Mindlre School (substitution school). When the community was founded, existing shapes of the village were preserved. The village was physically renovated based on the original environment, in addition nature friendly materials and methods were used. The residents were proactively involved in the community through regular meetings, self-regulation, and community worship. Although the survey results should that most residents were highly satisfied with living standards, there seemed to be no clear division between community facilities and private houses. A systematic and detailed space planning technique was required. In addition, various community facilities are required due to the expansion of the community.

간호조직문화가 간호사의 직무만족과 조직몰입에 미치는 영향 : 중소병원을 대상으로 (Effects of Organizational Cultures on Job Satisfaction and Organizational Commitment : Focused on Small to Medium Sized Hospitals)

  • The purpose of this study was to identify job satisfaction and organizational commitment depends on their organizational cultures. A survey, including the Organizational Culture Scale, Job Satisfaction Scale and Organizational Commitment Scale, was administered to 276 nurses working at 4 small to medium sized hospitals in B city. The dominant organizational culture of nursing organization was relation-oriented culture. The mean score of job satisfaction and organizational commitment was 2.68 and 4.25 respectively. Job satisfaction and organizational commitment were positively correlated with innovation-oriented, task-oriented and relation-oriented culture. Organizational commitment was negatively correlated with rank-oriented culture. Innovation-oriented, clinical career, relation-oriented and rank-oriented culture were variables influencing on job satisfaction and innovation-oriented and rank-oriented culture were major variables influencing organizational commitment. In conclusion, innovation-oriented and rank-oriented culture had a significant influence on nurses' job satisfaction and organizational commitment. Therefore, we have to develop strategies to enhancing the innovation-oriented culture and to reducing the rank-oriented culture.

대장균과 포유류 세포 내에서 parkin의 발현 양상에 관한 연구 (The Expression Patterns of Human Parkin in E. codi and Mammalian Cells)

  • E3 ligase로 알려진 Parkin은 protein quality control에서 중요한 역할을 할 뿐만 아니라, 이런 quality control system의 이상으로 나타나는 퇴행성 뇌질환에도 밀접한 연관성이었다. 이와 같이 생체의 필수적인 업무를 담당하는 Parkin의 기능을 생화학적 측면에서 연구하기 위해서는 고 순도의 단백질을 다량 정제할 수 있는 시스템이 필요하나, 아직까지 Parkin의 발현 양상과 정제법에 관한 연구가 미흡한 상태이다. 본 연구에서는 pCEX system을 이용하여 Parkin을 대장균에서 overexpression시켜 단일 스텝으로 정제할 수 있는 방법을 정립하였다. 저온의 배양조건에서 0.01 mM의 IPTC로 발현을 유도한 결과 $90\%$ 이상의 순도를 가지는 완전한 크기의 Parkin을 정제할 수 있었다. 또한, 여러 tag을 갖는 Parkin plasmid를 제작하였을 뿐만 아니라, 이들을 HEK293 세포에 transfection하여 Parkin의 발현 양상을 비교 분석하였다. 그 결과 Parkin의 N-말단에 pretense에 민감한 절단 부위가 존재한다는 사실을 확인하였다. 본 연구에서 정립한 Parkin 정제법과 포유류 세포에서 Parkin의 발현 양상에 대한 결과는 Parkin의 기질을 탐색하고,그들이 Parkin의 효소 활성 및 기능에 미치는 영향을 조사하기 위한 다양한 연구에 활용할 수 있을 것이다.

  • The human immunodeficiency virus type 1 (HIV-I) Tat protein transduction domain (PTD), which con­tains rich arginine and lysine residues, is responsible for the highly efficient transduction of protein through the plasma membrane. In addition, it can be secreted from infected cells and has the ability to enter neighboring cells. When the PTD of Tat is fused to proteins and exogenously added to cells, the fusion protein can cross plasma membranes. Recent reports indicate that the endogenously expressed Tat fusion protein can demonstrate biodistribution of several proteins. However, intercellular transport and protein transduction have not been observed in some studies. Therefore, this study exam­ined the intercellular transport and protein transduction of the Tat protein. The results showed no evi­dence of intercellular transport (biodistribution) in a cell culture. Instead, the Tat fusion peptides were found to have a significant effect on the transduction and intercellular localization properties. This sug­gests that the HIV-1 PTD passes through the plasma membrane in one direction.

차가버섯의 균사체 및 세포외다당체의 생산조건과 면역활성 (Optimun Conditions for Production of Mycelia and Extracellular Polysaccharide from Inonotus obliquus and Their Immunomodulating Acitivities)

  • 차가버섯의 균사체 생장과 세포외 다당체 생산을 위한 최적의 배양조건을 조사하고, 세포내 외 다당체의 항보체 활성과 대식세포의 lysosomal enzyme 활성을 관찰하였다. 균사체 생장과 세포외 다당체 생산을 위한 최적 pH, 온도, 및 교반속도는 각각 5.5, $25^{\circ}C$, 150 rpm으로 나타났으며, 배양기간은 11일째 최대 균사체 생장(10.89 g/l)과 세포외다당체 생산(1.25 g/l)을 보였다. 항보체 활성은 세포내 외 다당체의 처리 농도가 높을수록 활성이 증가함을 보였고, 대식세포의 lysosomal enzyme 활성은 $100{\mu}g/ml$ 농도에서 음성대조군에 비해 세포내 다당체는 약 2.2배, 세포외 다당체는 약 2배 정도 증가하였다.

BMP-2에 의한 협부 지방 성체 줄기세포의 골형성 (OSTEOGENESIS BY BMP-2 IN ADULT STEM CELL DERIVED FROM BUCCAL FAT PAD)

  • Bone morphogenetic proteins (BMPs) in combination with stem cells gain more significance for their use in bone tissue engineering. The mesenchymal stem cell can be differentiated into osteoblast by the treatment of BMP. The aim of this study is to characterize the osteogenic differentiation process of adult stem cells derived from buccal fat pad according to BMP-2 within culture media and decide the appropriate concentration of BMP-2 to facilitate osteogenesis. The authors procured the stem cell from buccal fat pad and analyzed for presence of stem cell by flow cytomety against CD-34, CD-105 and STRO-1. The buccal fat derived stem cells (BFDC) were treated by application of the different concentration with BMP-2 of 0, 10, 50, 100 and 200ng/ml, respectively. And their ability to differentiate into osteogenic pathway were checked by alkaline phosphatase(ALP) staining, Alizarin red staining and RT-PCR for osteocalcin(OC) gene expression at 7, 14 and 21day of culture. Flow cytometric analysis and biochemical assays demonstrated that BFDC might be a distinguished stem cells, and mineralization was accompanied in proportion to BMP-2 concentration. However, with 100ng/ml concentration of BMP-2, the BFDC demonstrated most efficient staining pattern of ALP and Alizarin red. The feasibility of the osteogenic differentiation in the group of both 50ng/ml and 200ng/ml of BMP-2 showed similar activity and relatively weaker than that of 100ng/ml. These results suggest that the BMP-2 stimulate osteogenesis by BFDC effectively and that bone induction might be controlled through negative regulatory feedback in higher concentration.

Decontamination methods to restore the biocompatibility of contaminated titanium surfaces

  • Purpose: The reaction of cells to a titanium implant depends on the surface characteristics of the implant which are affected by decontamination. The aim of this study was to evaluate the cytocompatibility of titanium disks treated with various decontamination methods, using salivary bacterial contamination with dental pellicle formation as an in vitro model. Methods: Sand-blasted and acid-etched (SA) titanium disks were used. Three control groups (pristine SA disks [SA group]; salivary pellicle-coated SA disks [pellicle group]; and biofilm-coated, untreated SA disks [NT group]) were not subjected to any decontamination treatments. Decontamination of the biofilm-coated disks was performed by 14 methods, including ultrasonic instruments, rotating instruments, an air-powder abrasive system, a laser, and chemical agents. MG63 cells were cultured in the presence of the treated disks. Cell proliferation assays were performed on days 2 and 5 of cell culture, and cell morphology was analyzed by immunofluorescence and scanning electron microscopy (SEM). A vascular endothelial growth factor (VEGF) assay was performed on day 5 of culture. Results: The cell proliferation assay revealed that all decontaminated disks, except for the 2 groups treated using a plastic tip, showed significantly less cell proliferation than the SA group. The immunofluorescence and SEM analyses revealed that most groups showed comparable cell density, with the exception of the NT group, in which the cell density was lower and bacterial residue was observed. Furthermore, the cells grown with tetracycline-treated titanium disks showed significantly lower VEGF production than those in the SA group. Conclusions: None of the decontamination methods resulted in cytocompatibility similar to that of pristine SA titanium. However, many methods caused improvement in the biocompatibility of the titanium disks in comparison with the biofilm-coated, untreated titanium disks. This suggests that decontamination is indispensable for the treatment of peri-implantitis, even if the original biocompatibility cannot be restored.

Improved recovery of active GST-fusion proteins from insoluble aggregates: solubilization and purification conditions using PKM2 and HtrA2 as model proteins

  • The glutathione S-transferase (GST) system is useful for increasing protein solubility and purifying soluble GST fusion proteins. However, purifying half of the GST fusion proteins is still difficult, because they are virtually insoluble under non-denaturing conditions. To optimize a simple and rapid purification condition for GST-pyruvate kinase muscle 2 (GST-PKM2) protein, we used 1% sarkosyl for lysis and a 1 : 200 ratio of sarkosyl to Triton X-100 (S-T) for purification. We purified the GST-PKM2 protein with a high yield, approximately 5 mg/L culture, which was 33 times higher than that prepared using a conventional method. Notably, the GST-high-temperature requirement A2 (GST-HtrA2) protein, used as a model protein for functional activity, fully maintained its proteolytic activity, even when purified under our S-T condition. This method may be useful to apply to other biologically important proteins that become highly insoluble in the prokaryotic expression system.
