The Effects of Onion and Garlic on Copper-Phenanthroline Complex Induced DNA Degradation (Copper-Phenanthroline 복합체에 의해 유도되는 DNA 손상에 대한 양파와 마늘의 억제효과)

  • In the present study, the effects of onion and garlic on copper and 1, 10-phenanthroline com plex induced DNA degradation were investigated by the decreased level of thiobarbiturin acid (TBA) reactive materials. Phenanthroline is specific for copper and the reaction releases TBA reactive material from DNA which can be measured by absorbance at 535nm. The levels of TBA reactive materials were decreased by adding onion or garlic ghomogenate into reaction mixture but the onion had more strong potency and the effect of onion was not changed by boiliing. Superoxide dismutase (SOD) and catalase have no inhibitory effects on copper induced DNa damage but reduced glutathone was more effective. The activities of antioxidant enzymes and the contents of sulfhydryl groups in onion and garlic were also investigated. The activity of SOD was more higher in garlic, but catalase and glutathione peroxidase activities were higher in onion. The contents of induced DNA damage were not by antioxidant enzymes such as SOD, catalase and glutathione peroxidase or sulfhydryl groups, but a substance which is more stable in high temperature.

Effect of Simulated Acid Rain on Fatty Acid Composition and Antioxidant System in Garden Balsam(Impatiens balsamina L.) (인공산성비가 봉선화(Impatiens balsamina L.)의 지방산 구성 및 항산화 작용에 미치는 영향)

  • The effects of simulated acid rain (SAR) on fatty acid composition and biochemical defense responses of plant was investigated. Garden balsam (Impatiens balsamina L.) was exposed to four pH levels (5.6, 4.0, 3.0, 2.0) of SAR and placed in the growth chambers for 2 weeks. SAR drastically inhibited chlorophyll content of garden balsam. The level of $H_2O_2$ was significantly increased by SAR. As pH levels decreased from 5.6 to 2.0, the ratio of unsaturated to saturated fatty acids of garden balsam was increased. Changes of three major polyamines (putrescine, spermidine and spermine) of garden balsam leaves were observed. All of the polyamine contents were increased with SAR. Catalase activities of the plant affected by SAR were increased as the pH decreased. The results indicate that the application of SAR generates oxidative stresses from the garden balsam and retards the plant growth significantly. A biochemical protect mechanism might be activated to neutralize the oxidative stresses generated through SAR.

  • This study investigated the effects of high light intensity (100 KLw) and high temperature (45 ℃, dark) on enzyme (glucose-6-phosphate dehydrogenase, acid phosphatase, catalase, peroxidase, and proteinase) activities and characteristics of Panax ginseng C.A. Meyer leaves. Enzyme activity and protein content decreased rapidly under treatment with high light intensity In P ginseng the thermal stabilities of catalase and peroxidase were high (above 70%), and the coagulation rates of soluble proteins were low (below 17%). Therefore, the decrease in enzyme activity and protein content was not caused by increase in leaf temperature due to the high light intensity, but by increase in proteolytic activities. The photochemical formation rate of superoxide radical (O-2) was higher in the P ginseng leaf extracts than in Solanum nigmm, and was accelerated by addition of crude saponin to the buffer extracts.

Activities of Catalase, Glycolate Oxidase, Hydroxypyruvate and NADPH-Glyoxylate Reductases at Different Growing Stages in the Leaves of Rice Plants (벼의 생육시기에 따른 일부 광호흡효소의 활성변화)

  • Catalase, glycolate oxidase, hydroxypyruvate and NADPH-glyoxylate reductases activities in cell free extracts from leaves of 3 cultivars, Suwon 264, IR 36 and Jin Heung of rice plants were studied at different growing stages. Catalase and glycolate oxidase shows inclining activities toward the maximum vegetative growth whereas declining activities in either the enzymes were noticed during the maturing stage. After the photoperiodic condition exhibit increasing hydroxypyruvate and NADPH-glyoxylate reductases activities with time until maturing stage. No significant differences were found in the enzyme activities, and in analytical data of nitrogen, chlorophyll contents, dry weight and soluble proteins among the 3 cultivars.

Effect of Cassia tora Ethanol Extracts on Carbon Tetrachloride-Induced Liver Injury in Rats (흰쥐에 있어서 사염화탄소 유발 간손상에 미치는 결명자 에탄올 추출물의 영향)

  • This study was conducted to investigate the effect of Cassia tora ethanol extracts on carbon tetrachloride($CCl_4$)-induced hepatotoxicity in rats. Male Sprague-Dawley rats were divided into the following 4 groups: normal group, $CCl_4$, treated group, $CCl_4-0.25%$ Cassia tora ethanol extracts group and $CCl_4-0.5%$ Cassia tora ethanol extracts group. Rats were fed with each experimental diet and water for 5 weeks. Liver weights of rats treated only with $CCl_4$, were significantly increased compared to normal group, but not in rats fed diet containing Cassia tora ethanol extracts. Cholesterol and triglyceride contents in serum and liver were also not influenced by either $CCl_4$, treatment or the supplementation of Cassia tora ethanol extracts. $CCl_4$, treatment significantly increased ALP activities, however the supplementation of Cassia tora ethanol extracts significantly decreased the activities of serum ALT, AST, ${\gamma}-GTP$ in dose-dependent manner. Cassia tora ethanol extracts significantly reduced $CCl_4-induced$ elevation of liver TBARS contents. Activities of superoxide dismutase and catalase were decreased by $CCl_4$, treatment, however by the supplement of Cassia tora ethanol extracts slightly increased activities of SOD and catalase. The activity of glutathione peroxidase in groups fed diets containing Cassia tora ethanol extracts was significantly decreased compared to that of the control group. These results suggest that Cassia tora ethanol extracts may exert protective effect against $CCl_4-induced$ liver injury through the prevention of lipid peroxidation.

  • This study was purposed to compare the activity of alcohol dehydrogenase and antioxidative effects of several plant extracts in the alcohol-treated rat liver. Sprague-Dawley rat weighing about 200 g were divided into the following 6 groups : normal, alcohol group and 4 different plant extracts administrated groups(Soybean sprout, Pine needle, Lentinus edodes, acanthopanacis cortex). Each plant extract was administrated orally by 200mg/kg b.w./day for 8 days before the alcohol treatment (5 g of 30% alcohol /kg b.w. by i.p.injection). All rats were sacrificed at 90 min after the alcohol treatment. The alcohol concentrations in serum of Soybean sprout and pine needle group were significantly lower than the Lentinus edodes and Acanthopanacis cortex group. The activity of alcohol dehydrogenase in the hepatic cytosol of Soybean sprout and Pine needle group was also significantly higher than the alcohol and the other groups However, the activity of catalase seemed not to be affected, although the extract groups showed slightly higher activities of catalase than the alcohol group. These results may indicate that the extracts of Soybean sprout and Pine needle were relatvely effective on the alcohol degradation. the activity of blutathione-peroxidase and lipid peroxidaton of all of the extract groups were significantly lower than the activity of alcohol group. These results can suggest that all of the use plant extracts more or less have an antioxidative effect on the alcohol-induced oxidation and especially, extracts of Soybean sprout and Pine needle have an stimulating effect on the alcohol absorption and degradation.

Effects of Biphenyldimethyl Dicarboxylate(DDB) on the Lipid Peroxidation and Oxygen Free Radical Scavenging Enzyme Activities in Mercuric Chloride-induced Hepntotoxic Rats (Biphenyldimethyl dicarboxylate(DDB)가 염화 제2수은-유발 간독성 흰쥐에서의 지질 과산화와 Oxygen Free Radical 제거효소 활성도에 미치는 영향)

  • In an attempt to define the effects of biphenyldimethyl dicarboxylate (DDB) on the lipid peroxidation and oxygen free radical scavenging enzymes activities in mercuric chloride-induced hepatotoxic rats, we studied malondialdehyde (MDA) level and the activities of catalase and superoxide dismutase (SOD) in liver of the rats at 24 hr after the injection of mercuric chloride. Sprague-Dalwey albino rats were injected subcutaneously with mercuric chloride (5 mg/kg) only and mercuric chloride (5 mg/kg) plus. DDB (200 mg/kg/day, p.o) is administered for 4 days prior to 3 days from the injection of mercuric chloride. The group treated with mercuric chloride showed significantly higher MDA level and lower catalase and SOD activities as compared with that of control group. The group treated with mercuric chloride plus DDB showed significantly lower MDA level and catalase activity and higher SOD activity as compared with that of mercuric chloride-treated group. These results suggest that the excessive oxygen free radicals resulting from the depression of superoxide dismutase activity is an important determinant in the pathogenesis of mercuric chloride-induced hepatotoxicity and DDB has antioxidant effects.

  • The purpose of this study was to investigate the improvement effect of turmeric (Curcuma longa L.) on the hepatic functional enzyme and catalase activity of streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley (SD) male rats were divided into four groups (n=6), and fed experimental diets containing turmeric meal [basal diet+5% turmeric (BT), basal diet+STZ+5% turmeric (ST)], and control (basal diet, BD), BS groups (basal diet+STZ). Serum concentrations of creatinine and blood urea nitrogen (BUN) were significantly decreased (p<0.05) by 5% turmeric supplementation diet. The activities of akaline phosphatase (ALP), lactate dehydrogenase (LDH), aspartate transaminase (AST), alanine transaminase (ALT), amylase and lipase were decreased in the BD, BT and ST group than BS group. The catalase (CAT) activity was significantly increased (p<0.05) in turmeric supplementation diet (BT, ST group) than diabetic group (BS). Furthermore, the activities of amylase and lipase in the sera of turmeric diet group were significantly decreased (p<0.05). In vivo experiments with diabetic rats showed that ingestion of turmeric supplementation diet were effective in creatinine concentration, and hepatic functional enzyme activities.

Activity of Superoxide dismutase(SOD) by fermented soybean (발효 대두 식품의 Superoxide dismutase(SOD) 활성)

  • This study was performed to evaluate the inhibition effects of fermented soybean on lipid perosidation and antioxidative relative enzyme activity. in vivo. Fermented soybean was induced the high SOD activity, while significantly inhibited on the peroxide value of linoleic acid and lipid perxidation from rat microsome induced by Fe$^{2+}$ ascorbate system, Sprague-Dawley(SD) male rats were fed basic diet, and experimental diets group added 200 or 500 mg/kg fermented soybean for 2 weeks. The effect of fermented soybean is also significantly increased catalase and glutathione peroxidase activities, while significantly inhibited the lipid peroxidation of rat liver microsome in a dose dependent manner. Therefore, these results suggest that fermented soybean has antioxidative activity which is related enzyme to prevention of oxidative stress.s.

Antioxidative Effect of Fermented Rhynchosia nulubilis in Obese Rats (비만 흰쥐에서 발효 서목태의 항산화 효과)

  • This study was to examine the antioxidative activity of fermented Rhynchosia nulubilis (FRN) in obese rats. Oxidative stress due to reactive oxygen species (ROS) can cause oxidative damage to cells. Mitochondria are especially important in the oxidative stress as ROS have been found to be constantly generated as an endogen threat. Mitochondrial defense depends mainly on superoxide dismutase whereas microsomal defense depends on catalase, which is an enzyme abundant in microsomes. Seven weeks-aged female Sprague-Dawley rats were divided into four groups and fed high fat diets for 44 days. Also fermented Rhynchosia nulubilis was administered orally for 44 days at 7.5 ml/kg of body weight of rats. The antioxidative activities of fermented Rhynchosia nulubilis were measured by the superoxide dismutase, catalase, malondialdehyde levels in liver homogenate. The levels of malondialdehyde in FRN-treated groups were lower than those in obese groups. Superoxide dismutase and catalase levels were significantly increased. These results demonstrated that fermented Rhynchosia nulubilis had the inhibitive effects of oxidative stress in obese rats, suggesting that fermented Rhynchosia nulubilis would be used as an ingredient of the useful functional products.
