이온화칼슘이 도계과정 중 도체표면의 미생물 교차오염에 미치는 영향 (Effects of ionized calcium on microbial cross-contamination in surface of carcass via slaughter process of chickens)

  • The purpose of this study was to investigate the effects of ionized calcium treatment on total bacterial cross-contamination of chicken carcass surface in the slaughtering process. The growth of Escherichia coli was strongly inhibited in a medium prepared by using a 0.5% ionized calcium solution. The total bacterial cross-contamination of chicken carcass surface and the scalding water was significantly increased as the number of scalding was increased (p<0.05). The total bacterial cross-contamination of chicken carcass surface reached a plateau without a further increase as scalding was performed consecutively for 10 or more times. The total bacterial cross-contamination of the scalding water was significantly increased as the number of scalding was increased (p<0.05). The total bacterial cross-contamination of chicken carcass surface of the chickens raised on a floor type farm was significantly higher than that of the chickens raised in a battery cages (p<0.05). The total bacterial cross-contamination of chicken carcass surface of the chickens raised on a floor type farm was significantly lower in the 0.5% ionized calcium solution treatment group than in the control group (p<0.05).

Effects of Formic Acid Administration in the Drinking Water on Performance, Intestinal Microflora and Carcass Contamination in Male Broilers under High Ambient Temperature

  • In this study, we examined the effects of formic acid administration to the drinking water on performance, intestinal microflora and carcass contamination in male broilers. A total of 312 day-old male broiler chicks were allocated to two groups with three replicates. The first group (control) received normal drinking water (pH 7.4) during the experiment. The second group consumed acidified drinking water (pH 4.5) after 5 d of age. At 43 d of age, twelve birds were randomly selected from the control group to determine the effect of acidified drinking water on carcass contamination. These birds were only given normal or acidified (pH 3) drinking water for 8 h prior to slaughter. The reduction of water pH from 7.4 to 4.5 significantly decreased body weights of male broilers at 21 and 42 d of age. However, no differences were observed between male broilers given normal and acidified drinking water in terms of feed intake, feed conversion ratio and mortality. The pH value of the gizzard contents was not significantly affected by acid water treatment. There were no significant differences in the intestinal population of E. coli, total organism and Salmonella between the groups. The total organism and E. coli counts of the carcass slightly decreased in the acidified group. No Salmonella was identified in carcass samples of any of the treatment groups. The results showed that drinking water acidification did not provide beneficial effects on performance, intestinal microflora and carcass contamination in male broilers.

소규모 돼지도축공정에서 도체오염 미생물의 변화 (Microbial change of pork carcass during processing in small size slaughterhouse)

  • Major hazards existed in slaughterhouse are pathogenic microorganisms originated from intestinal microflora of slaughtered animals. This study was intended for the identification of microbial contamination sources during pork slaughtering in small plants. Total aerobic bacteria, Coliform group, Salmonella spp, Listeria monocytogenes, and Campylobacter jejuni/coli were isolated from the surface sample of pork carcasses. Contamination level among different sampling points of ham, belly and neck did not showed statistical differences. Therefore, the mixed sampling from belly and neck of carcass could be effective for microbiological monitoring. Isolation rates of pathogenic microorganisms showed Salmonella spp 20.9%, Listeria monocytogenes 10.5%, and Campylobacter jejuni/coli 8.1% from 296 sampling points. High prevalence rate of Salmonella spp indicated that the contamination of intestinal microflora occurred due to unsanitary processing control, which required HACCP system in small plants. It was recommended that the prerequisite program should be a key factor for a successful HACCP system implementation especially in small size slaughterhouse.

  • To evaluate potential impacts of shallow groundwater by the leachate from buried carcass, we investigated hydrochemical characteristics of both leachate and shallow groundwater from monitoring wells and surrounding shallow groundwater wells in an area potentially affected by pig carcass burial. The hydrochemical survey was conducted before and after the relocation of a burial pit. The leachate samples and the groundwater affected by leachate showed the hydrochemistry of $Ca-HCO_3$ type with high $NH_4{^+}$ concentrations, while unaffected groundwater was mostly the $Ca(Na)-Cl+NO_3$ type due to pervasive impacts from agrochemicals. The results of factor analysis on hydrochemical data showed the followings: 1) contamination of groundwater from agro-livestock farming and livestock burial are coexisting in the study area, 2) among ionic species, $HCO_3{^-}$, $NH_4{^+}$, $NO_3{^-}$ and Mn are very useful to differentiate the groundwater contamination from leachate, and 3) groundwater contamination by leachate has been recognized around the monitoring wells even after the relocation of a burial pit, likely due to residual contaminants in surrounding soils. Therefore, it is suggested that continued monitoring of groundwater contamination should be conducted after the relocation of carcass burial pits.

Effect of Spinal Cord Removal before or after Splitting and Washing on CNST Decontamination of Beef Carcasses

  • Beef carcasses were examined to explore the effects of spinal cord removal and washing on central nervous system tissue (CNST) decontamination of the surface during the slaughtering process. A total of 15 carcasses were split by sawing centrally down the vertebral column and left sides of split carcasses were used for analysis. Samples were collected by swabbing the surface from 4 defined parts on the interior and 4 on the exterior of carcasses from the abattoir and analyzed using an ELISA-based test. The results showed that automatic and manual spray washing decreased CNST contamination, especially on the interior ventral parts of carcass surfaces (p<0.01), but did not decrease CNST on the interior dorsal parts. Increasing washing time to 60 s did not affect the reduction of CNST contamination. Samples following spinal cord removal prior to splitting showed lower calculated levels of "risk material" than the stated limit of detection (0.1%) of the ELISA kit on interior and exterior carcass parts (p<0.01). Therefore, spinal cord removal prior to splitting could be a very effective way to minimize CNST contamination of beef carcasses.

Influence of pre-slaughter fasting time on weight loss, meat quality and carcass contamination in broilers

  • Objective: An experiment was conducted to determine the appropriate fasting time prior to slaughter for broilers in floor-feed and scatter-feed mode. Methods: On 21 d since hatching, 120 Arbor Acres broilers were divided into floor-feed and scatter-feed groups, chicks from each group were further assigned to feed withdrawal treatments for 0, 4, 6, 8, and 10 h. Some resultant indicators such as carcass contamination, body weight loss, meat quality of 54-day-old broilers were measured. Results: It appears that longer feed withdrawal increased weight loss, lightness, drop loss of meat but reduced pH. A significant higher weight loss and lightness for both floor-feed and scatter-feed chicks coincided after 6 to 10 h feed withdrawal (p<0.05). pH for breast muscle at 45 min postmortem reduced when chicks of scatter-feed were fasted 6 and 10 h, while the reduction of floor-feed group occurred only in 10 h (p<0.05). A noticeable effect of feed withdrawal on drop loss occurred after 10 h fasting in scatter-feed of which drop loss were significantly higher than that for other groups including control (p<0.05). The change of contamination propensity revealed that 6 to 10 h fasting significantly reduced the likelihood of carcass contamination under both floor-feed and scatter-feed (p<0.05). Net weights of intestinal contents for gizzard were significantly reduced after feed deprived for 10 h in floor-feed and 6 and 10 h in scatter-feed (p<0.05). The decrease for whole intestine occurred after floor-feed broilers have been without feed for more than 4 h, scatter-feed broilers for more than 8 h (p<0.05). Conclusion: On the premise that poultry product properties and welfare were not significantly damaged, proper fasting time could reduce carcass contamination. Current data implied that 6 h fasting was recommendable for both floor and scatter feed pre-slaughter broilers.

  • 이 총설은 출하 전 절식에 관한 문헌을 요약 정리한 것이다. 도축 시 도체 오염 방지는 효과적인 절식 프로그램을 실시할 때만이 가능하다. 특별히 법으로 외견 또는 미생물적 오염을 규제하는 나라의 경우 절식은 매우 중요하다. 육계에 대한 절식은 도축 시 장 내용물로부터 도체 오염을 줄일 수 있어 이를 권장하고 있다. 대부분의 연구가 적정한 절식 시간, 도축, 가공의 효율성 유지 및 닭고기 안전성을 확보하기 위한 수단으로서의 절식 효과에 대해 초점을 두고 진행되었다. 실제적인 절식 프로그램은 생축생산 능력과 사육방식, 도축방법, 도축검사 및 도축관련 규정이 달라 나라마다 다르다는 점을 지적한다.

RAPD를 이용한 돈육 가공장의 Listeria 오염양상 분석 (Contamination patterns of Listeria spp. in pork processing plants using random amplified polymorphic DNA)

  • This study was carried out to understand the contamination patterns of Listeria in pork processing plants. A total of 402 samples were collected from carcass, pork during processing, surfaces of equipment and environment, and 238 isolates of Listeria species were identified. L. innocua was found in 64.7% of the isolates, L. monocytogenes in 33.2%, and L. welshimeri in 2.1%. Random amplified polymorphic DNA (RAPD) analysis performed to investigate the origin and routes of Listeria contamination, showed 21 composite types of L. monocytogenes and 26 composite types of L. innocua. It was confirmed that Listeria contamination begins with contaminated incoming carcass and ever-present contaminants in the processing environments. The persistence and dissemination of the same strain of L. monocytogenes and L. innocua throughout the processing line revealed that the sanitation standard operating procedure should be implemented to minimize the risk of colonization in the workplace. Molecular subtyping of L. innocua allowed us to tracing the possibility of cross-contamination during processing.

식육의 처리 단계별 미생물 오염실태와 병원성 미생물의 분포 (Hygienic Quality of Beef and Distribution of Pathogens during Cut-Meat Processing)

  • 위생적인 신선육 처리 방안을 모색하기 위하여 도체처리 과정과 식육처리 과정 중 고기의 표면 및 작업도구의 위생실태를 계절별로 검토하였다. 하절기에 총균수, 저온성 균수, 대장균군수 모두 높게 나타났으며, 특히 도체 세척전보다 세척 후에 미생물 오염도가 높았고, 계절에 관계없이 수송 후의 오염도가 가장 높았다. 칼, 도마와 장갑에서 하절기에 $10^{5}$ $ extrm{cm}^2$을 나타내어 높은 미생물 오염도를 나타내었다. 도체 처리 과정 중 도체 표면에서 Escherichia. coli O157, Pseudomonas aeruginosa, Klebsiella, ornithinolytica, Staphylococcus aureus, E. Tatumella, ptyseos, Serratia odorifera, Aeromonas, Aeromonas sobria, Enterobacter colacae, Flavimonas oryzihabitans oryzihabitans를 분리하였으며, 도마에서는 S. aureus, Listeria grayi, L. monocytogenes, 장갑에서 L. grayi, Erwinia spp., Salmonella spp. S aureus, 골발육에서는 Citrobacter freundii, L. momocytogenes, S. aureus가 분리.동정하였다.

Survey on Geochemical Characteristics of Groundwater Around Carcass Burial Area and Agricultural Area with Livestock Facilities

  • In this study, chemical characteristics of groundwater around carcass burial areas and those in agricultural and livestock-farming complex areas in South Korea were monitored. Groundwater samples were collected from 166 wells around carcass burial sites and 466 wells around the agricultural areas where carcass burial sites are absent. The chemical parameters (pH, electrical conductivity, dissolved oxygen, oxidation reduction potential, $NO_3$-N, $NH_4$-N, and $Cl^-$) in carcass burial areas and agricultural areas were similar. The $NO_3$-N concentrations exhibited minimal seasonal fluctuations below $30mg\;L^{-1}$ in most of the wells, even in the wells located close to the carcass burial sites; and $Cl^-$ concentrations also showed similar patterns. The chemical characteristics of groundwater monitored in this study indicated that groundwater was widely contaminated by agricultural activities and livestock farming, but probably not by leachates derived from nearby carcass burial sites.
