  • 우리는 탄소 전구체로 활성탄 (AC), 활성탄 섬유 (ACF)와 탄소나노튜브 (MWCNT)와 티타늄 전구체로써 TNB를 사용하여 탄소-$TiO_2$ 복합체를 제조하였으며, 이들의 특성을 SEM, TEM, BET, XRD와 EDX를 이용하여 분석하였다. 그리고 이들의 광촉매 활성은 UV 램프 조사에서 일정한 농도의 MB 용액을 이용하여 측정하였다. UV 조사한 후에 MB의 농도는 UV-vis 분광광도기를 이용하여 측정하였다.

Effects of Nitrogen Sources and C/N Ratios on the Lipid-Producing Potential of Chlorella sp. HQ

  • Microalgae are being researched for their potential as attractive biofuel feedstock, particularly for their lipid production. For maximizing biofuel production, it is necessary to explore the effects of environmental factors on algal lipid-producing potential. In this study, the effects of nitrogen (N) sources (NO2-N, NO3-N, urea-N, NH4-N, and N-deficiency) and carbon-to-nitrogen ratios (C/N= 0, 1.0, 3.0, and 5.0) on algal lipid-producing potential of Chlorella sp. HQ were investigated. The results showed that for Chlorella growth and lipid accumulation potential, NO2-N was the best amongst the nitrogen sources, and NO3-N and urea-N also contributed to algal growth and lipid accumulation potential, but NH4-N and N-deficiency instead caused inhibitory effects. Moreover, the results indicated that algal lipid-producing potential was related to C/N ratios. With NO2-N treatment and carbon addition (C/N = 1.0, 3.0, and 5.0), total lipid yield was enhanced by 12.96-20.37%, but triacylglycerol (TAG) yields decreased by 25.52-94.31%. As for NO3-N treatment, carbon addition led to a 17.82-57.43%/25.86-82.67% reduction of total lipid/TAG yields. When NH4-N was used as the nitrogen source, total lipid/TAG yields were increased by 46.67-113.33%/28.99-74.76% with carbon addition. The total lipid/TAG yields of urea-N treatment varied with C/N ratios. Overall, the highest TAG yield (TAG yield: 38.75 ± 5.21 mg/l; TAG content: 44.16 ± 4.35%) was achieved under NO2-N treatment without carbon addition (C/N = 0), the condition that had merit for biofuel production.

  • 섬유소를 분해하여 이용하기 위해서는 섬유소 분해효소를 대량으로 생산하는 것이 중요하며 이를 위해서는 적절한 탄소원을 이용하여 섬유소 분해효소의 대량생산을 유도하는 것이 중요하다. 본 실험에서는 Trichoderma reesei Rut C-30을 섬유소 분해효소의 생산 균주로 solka floc을 탄소원으로 사용하여 섬유소 분해효소의 활성을 증가시키기 위한 배지내 최적 농도가 1%이고 포도당이 0.5%이상 배지에 첨가 될 경우 섬유소 분해효소의 생산이 저해됨을 알 수 있었다. 정제된 섬유소인 solka floc이 목질섬유소인 wheat bran과 함께 탄소원으로 사용되었을 때 각각을 섬유소 분해효소 생산의 유도 물질로 이용했을 때보다 더 높은 CMCase 활성과 FPase 활성을 보였다. Solka floe 1%와 wheat bran 3%을 탄소원으로 사용한 배지의 CMCase 활성이 76 U/ml, FPase 활성이 12.6 U/ml로 가장 높은 값을 나타내었다.

외부탄소원 주입시 영양염류의 생물학적 제거를 위한 자동제어 SBR 공정에 관한 연구 (A Study on Autocontrolled SBR for Biological Nutrient Removal with External Carbon Sources)

  • The purpose of this study is to develop effective operating process in order to achieve more suitable conditions of Anoxic-Oxic-Anoxic-Stripper(AOAS) SBR through real-time control. To improve the removal efficiency, glucose, methanol and synthetic food waste acid fermentant were added as an external carbon source, In the case of glucose and synthetic food waste acid fermentant, TN, TP were removed to average 86.9%, 73.0% respectively. Methanol was removed to average 64.6%, 55.4% respectively. The synthetic food waste acid fermentant proved to be the most efficient and allowed for the substitution of an external carbon source. The removal rate of $COD_{Cr}$, was approximately 90% at all cases. The results of the study that a correlation between ORP (Oxidation-Reduction Potential), pH and DO and nitrification or denitrification when an external carbon source is added and when it isn't was showed that ${\Delta}ORP$ is suitable parameter. ORP reacted properly to denitrification (${\Delta}ORP<-10$) and nitrification (${\Delta}ORP<0$). The use of real-time control saved anywhere between 61 and 67 minutes at the anoxic(1) stage and 26 to 52 minutes at the oxic(1) stage. When the time saved from the anoxic(1) and oxic(1) was added to the anoxic(2) stage for the removal efficiency of TN and TP increased from 0.7 to 13.9% and 12 to 35 % respectively.

Construction of Microbial Fuel Cells Using Thermophilic Microorganisms, Bacillus licheniformis and Bacillus thermoglucosidasius

  • A systematic study of microbial fuel cells comprised of thermophilic Bacillus licheniformis and Bacillus thermoglucosidasius has been carried out under various operating conditions. Substantial amount of electricity was generated when a redox mediator was used. Being affected by operation temperature, the maximum efficiency was obtained at 50$^{\circ}C$ with an open circuit voltage of ca. 0.7 V. While a small change around the optimum temperature did not make much effect on the cell performance, the rapid decrease in performance was observed above 70$^{\circ}C$. It was noticeable that fuel cell efficiency and discharge pattern strongly depended on the kind of carbon sources used in the initial culture medium. In the case of B. thermoglucosidasius, glucose alone was utilized constitutively as a substrate in the microbial fuel cell irrespective of used carbons sources. When B. licheniformis was cultivated with lactose as a carbon source, best charging characteristics were recorded. Trehalose, in particular, showed 41.2% coulombic efficiency when B. thermoglucosidasius was cultured in a starch-containing medium. Relatively good repetitive operation was possible with B. thermoglucosidasius cells up to 12 cycles using glucose as a carbon source, when they were cultured with lactose as an initial carbon source. This study demonstrates that highly efficient thermophilic microbial fuel cells can be constructed by a pertinent modulation of the operating conditions and by carefully selecting carbon sources used in the initial culture medium.

수돗물의 탄소 배출량 평가 (Carbon Emission Evaluation of Tap Water)

  • To evaluate carbon emission in water treatment processes, LCA (life cycle assessment) was applied to 8 multi-regional water treatment plants (WTPs) from intake to supply of tap water. Investigation of 8 WTPs revealed that average carbon emission for 1 $m^3$ of tap water was 221 g. Major carbon emission sources in water supply system were intake and supply processes. Meanwhile, mixing process was the main carbon emission source in unit water treatment processes. Carbon emission was proportional to the turbidity and COD of raw water. Intake of better raw water and minimization of energy consumption in unit processes are needed to reduce carbon emission in the WTPs. In addition, comparison of carbon emission among WTPs can be used as a parameter for optimization of operation and maintenance of water treatment processes.

  • Clostridium butyricum, Lactobacillus amylophillus, Lactobacillus amylovorus, Lactobacillus acidophillus, AI-9 produced hydrogen and /or organic acids using glucose, lactose and starch at the anaerobic culture conditions. Cl. butyricum NCIB 9576 evolved 1,700 ml H2/L-culture broth and accumulated butyric acid, acetic acid, propionic acid and ethanol in its culture broth when lactose was used as a carbon source during 24 hrs of fermentation. L. amylovorus ATCC 33620 accumulated lactic and acetic acids and some reducing sugars when starch was used as a carbon source without hydrogen production. Instead of starch as a carbon source, L. amylovorus ATCC 33620 produced lactic acid from algal biomass during fermentation and the acid-heat or freeze-thaw pretreatment of algal biomass accelerate the lactic acid fermentation.

Mannitol Amendment as a Carbon Source in a Bean-based Formulation Enhances Biocontrol Efficacy of a 2,4-diacetylphloroglucinol-producing Pseudomonas sp. NJ134 Against Tomato Fusarium Wilt

  • Fusarium wilt caused by Fusarium oxysporum has become a serious problem world-wide and relies heavily on chemical fungicides. We selected Pseudomonas sp. NJ134 to develop an effective biocontrol strategy. This strain shows strong antagonistic activity against F. oxysporum. Biochemical analyses of ethyl-acetate extracts of NJ134 culture filtrates showed that 2,4-diacetylphloroglucinol (DAPG) was the major compound inhibiting in vitro growth of F. oxysporum. DAPG production was greatly enhanced in the NJ134 strain by adding mannitol to the growth media, and in vitro antagonistic activity against F. oxysporum increased. Bioformulations developed from growth of NJ134 in sterile bean media with mannitol as the carbon source under plastic bags resulted in effective biocontrol efficacy against Fusarium wilt. The efficacy of the bioformulated product depended on the carbon source and dose. Mannitol amendment in the bean-based formulation showed strong effective biocontrol against tomato Fusarium wilt through increased DAPG levels and a higher cell density compared to that in a glucose-amended formulation. These results suggest that this bioformulated product could be a new effective biocontrol system to control Fusarium wilt in the field.

Cultural Characteristics of Veiled Lady Mushroom, Dictyophora spp.

  • This study was carried out to obtain the basic data for artificial culture of veiled lady mushroom (Dictyophora spp). The optimal conditions for the mycelial growth were $25^{\circ}C$ and pH 5.0 for all isolates except the optimal temperature of $30^{\circ}C$ for D. echinovolvata ASI 32002 and Phallus rugulosus. The optimal medium for Dictyophora spp. was PBA (potato bamboo sawdust extract agar) medium. The strain ASI 32002, D. echinovolvata, grew faster than. D. indusiata ASI 32003 and Phallus rugulosus ASI 25007 on the medium. Carbon sources such as glucose, maltose and inuline were favorable for stimulating a mycelial growth of the two strains of ASI 32002 and ASI 32003. Asparagine and glutamine appeared to be favorable to the strain ASI 32002 and ASI 32003, where as alanine, one of nitrogen source also favorable to the strain ASI 32002. The optimum C/N ratio of the two isolates of ASI 32002 and ASI 32003 was about 25 : 1 when 2% glucose as carbon source was mixed with the basal medium. While, in the case of 4% as carbon source, the optimum C/N ratio was about 30 : 1.

Effect of Salinity, Temperature and Carbon Source on the Growth and Development of Sclerotia of Sclerotinia sclerotiorum Isolated from Semi-arid Environment

  • Studies were conducted to determine the effects of temperature, solute potential and carbon source on the mycelial growth, sclerotia development, and apothecium production of an isolate of Sclerotinia sclerotiorum. Mycelial growth rate was greatest at $25^{\circ}C$ on potato dextrose agar (PDA) medium amended with up to 2% NaCl (${\psi}s{\leq}1.91\;MPa$) and thereafter, growth rate declined. The least number of sclerotia were produced at $20^{\circ}C$on both PDA and malt extract agar (MEA) amended with 8% NaCl (${\psi}s=6.62\;MPa$). With increasing temperature and decreasing solute potential the number and size of sclerotia were significantly reduced. The combined effect of temperature, solute potential and carbon source on sclerotia production were highly significant and had an impact on the development of the rind layer cells of sclerotia. These cells lacked a transparent cell wall which was surrounded by a compact melanized layer, and some of these cells appeared to be devoid of cell contents or were totally vacuolated. The survival of the sclerotia with increase in salinity and temperature appeared to affect melanization and the nature of the rind cells. The observations of this study re-enforces the need for an integrated disease management to control S. sclerotiorum.
