수인성 니켈 급성 노출이 붕어, Carassius carassius의 반수치사농도, 혈액학적 성상 및 혈장 성분에 미치는 독성 영향 (Toxic Effects on Lethal Concentration 50%, Heamatological Parameters and Plasma Components of Crucian carp, Carassius carassius by Acute Waterborne Nickel Exposure)

  • 본 실험은 붕어(Crucian carp, Carassius carassius)(무게 39.7±3.1 g, 전장 14.8±0.5 cm)의 수인성 니켈 0, 10, 20, 40, 80 및 160 mg Ni2+/l 농도로 96시간 급성 노출을 실시하였다. 수인성 니켈에 노출된 붕어의 반수치사농도(LC50)는 117.69 mg Ni2+/l으로 나타났다. 혈액학적 성상에서 RBC count는 수인성 니켈 96시간 급성 노출 중 48시간에서 유의적으로 증가한 반면, 96시간에서 유의적 감소가 나타났다. MCV와 MCH는 96시간에서 80 mg Ni2+/l 농도에서 유의적으로 증가했다. Calcium, magnesium, glucose, cholesterol, total protein, AST, ALT 및 ALP 와 같은 혈장 성분은 수인성 니켈 노출에 의해 유의적 변화가 나타났다. 이 연구의 결과는 수인성 니켈 노출에 따른 붕어의 생존율, 혈액학적 성상 및 혈장 성분의 변화를 확인하고 이는 수인성 니켈의 독성에 의한 것으로 판단했다.

  • The characteristics of the pharyngeal bones and their teeth in Korean Cyprinids fishes, as a rule, are summarized as follow : 1. The materials 13 genera and 17 species from Nak Dong River. 2. Breadth of bone about 2.8(Cyprinus carpio) to 4.9(Culter brevicauda) in bone length. Length of bone about 8.5(Carassius carassius ) to 20.2 (Pseudogobio esocinus ) in body length. Breadth of pitted surface about 66%(Carassius carassius) to 85%(Acanthorhodeus asmussi ) of breadth of bone. 3. The position and distinct ratio of anterior angle correlated. 4. Posterior limb always longer than anteriro, but posterior edentulous process not shorter than anterior's or equal. 5. Zacco platypus with pitted surface on anterior limb , but Z.temmincki without it. 6. Teeth one to three rowed in accordance with the differences of each genus : a. one rowed genera ..... Carassius , Pseudorasbora, Sarcocheilichthys (Sakiyae), Pungtungia, Acheilognathus, Acanthorhodeus. b. two rowed genera ........ Pseudogobio, Gnathopogon, Tribolodon. c. Three rowed genera .... Cyprinus, Hemibarbus, Zacco , Culter 7. Teeth of main (primary ) row with four or five teeth is symmetrized exceptive three genera (Tribolodon, Zacco, Culter). 8. Grinding surface and bill type correlated. 9. In Acheilognathus, A.Yamatsutae have one or two indistinct serration but each individual is unlike.

Population Genetic Structure of Carassius auratus (Pisces: Cypriniformes) in South Korea Inferred from AFLP Markers: Discordance with Mitochondrial Genetic Structure

  • A recent study on the mitochondrial genetic variation of the Carassius auratus population in South Korea suggested that there are 3 distinct mitochondrial lineages in the country, and that they are geographically separated between westward rivers and southward rivers, respectively. In this study, the population genetic structure of amplified fragment length polymorphism (AFLP) of Carassius auratus was investigated. The results of analysis of molecular variance (AMOVA) supported the geographic distinction between westward and southward river populations, but only 3.66% of total genetic variance lies among these populations. The panmicticity of the AFLP genetic variation is backed up by the results of the neighbor-joining dendrogram drawn from a linearized pairwise $F_{ST}$ matrix and Bayesian clustering analysis. The discordance of genetic structure between mitochondrial and AFLP genetic variation may come from difference in effective population size between these markers and/or gene flow between westward and southward river populations through river capture events.

붕어의 Splenocyte 및 Erythrocyte의 Extrachromosomal DNA 관찰 (Electron Microscopic Study on Extrachromosomal DNA from Splenocytes and Erythrocytes of Carassius carassius L.)

  • Extrachromosomal circular DNA complexes from erythrocytes and splenocytes isolated from Carassius carassius were examined by mica-press-absorption method. The method was described that released small polydisperse circular DNA molecules in situ from the erythrocytes and the splenocytes and that allows selective observation of the small circular DNA complexes bound to cellular components. The released polydisperse circular DNA complexes were absorbed preferentially on mica in a divalent cation-free medium then processed for electron microscopy. Small circular DNAs showed a heterogeneous size distribution of $2{\sim}10{\mu}m$ with a mean contour length of $4.3{\mu}m$ for the circulating erythrocytes and that of $0.7{\sim}3.6{\mu}m$ with a mean contour of length $2.04{\mu}m$ for the splencytes. Cells contained $100{\sim}300$ copies and $300{\sim}700$ copies obtained from the erythrocytes and the splenocytes, repectively. Possible biological functional implications for size distribution of extrachromosomal circular DNAs are discussed.

Disc 전기영동법에 의한 수종 담수어 혈장 단백질의 분획 (Fractionation of Plasma Protein on the Several Fresh Water Fishes by Disc Electrophoresis)

  • The plasma proteins of fresh water fishes have been fractionated by disc electrophoresis in acrylamide gels utilized as an electrophoretic supporting medium. The species of fishes examined in this experiment were Anguilla japonica, Misgurnus mizolepis, Parasilurus asotus, Siniperca scherzeri, Pelteobagrus fulvidra, Carassius carassius, Cyprinus carpio, and Hemibarbus labeo, obtained in the Han River. Disc electrophoresis was performed as described by Ornstein and Davis. Gels and buffer solution were prepared by the method developed by W.J.Kim. The separation gels were 7% acrylamide gel. The fractionation of plasma proteins showed 13 bands in Anguilla japonica, 10 in Misgurnus mizolepis, 15 in Parasilurus asotus, 12 in Siniperca scherzeri, 11 in Pelteobagrus fulvidra, 13 in Carassius carassius, 9 in Cyprinus carpio, and 13 in Hemibarbus labeo. The patterns of plasma protein on the each species of fishes were different in the number of bands, ratio of contents, relative mobilities, and forms of fractionation.

카드뮴 축적량과 붕어(Carassius auratus) 아가미 조직 변화의 상관성에 관한 연구 (A Study on Cadmium Accumulations and Histopathological Changes in the Gills of Crucian Carp (Carassius auratus))

  • Histopathological changes and cadmium accumulations in the gills were investigated in crucian carp (Carassius auratus) exposed to 0, 0.01, 0.1, 0.5 mg/1 cadmium (Cd) concentrations for 25 days. After 25 days of exposure. cadmium accumulations increased in each of the different exposure concentrations. Histopathological changes in the gills of curcian carp exposed to cadmium included the acidification of mucous cell, the terminal clubbing of lamellae, the hyperplasia of epitherial cell and the curved of secondary lamellae. The similar histopathological changes were observed once the accumulations of cadmium reached the similar levels found in the present study with the different exposure schemes. Also the acidification of mucous cell in gills appeared when cadmium accumulations reached 5 $\mu\textrm{g}$/g and more. These results suggested that histopathological changes in the gills, an appearance of mucous cell acidification. be used to estimate the relation between toxic effect and cadmium accumulations in the gills.

Clove Oil의 붕어(carassius auratus) 혈액 및 혈액화학치에 대한 영향 (Effects of clove oil on haematology and blood chemistry in crusian carp (Carassius auratus))

  • To determine the effects of anesthesia to clove oil on the secondary stress indices, changes in haematological and blood chemistry were monitored in healthy crucian carps (Carassius auratus). 24 fishes were divided into three groups, and blood was collected before anaesthesia (control group), immediately after anaesthesia at a concentration of 30 mg/l clove oil (anesthetized group), and 24 h after anaesthesia (recovered group). The anesthesia to clove oil significantly increased glucose in anaesthetized group, and constantly decreased lactate dehydrogenase in anaesthetized group and recovered group. However, clove oil had not effect on other biochemical indices. These results suggests that the anesthetic use of clove oil at a concentration of 30 mg/l does not cause irreversible damage in carp.

  • RAPD analysis based on numerous markers have been used to investigate patterns of genetic differentiation ann and within two cultured and wild populations represented by the species crucian carp(Carassius carassius). From RAPD analysis using five primers, a total of 442 polymorphic bands were obtained in two populations and 273 were found to be specific to a wild population. According to RAPD-based estimates, average number of polymorphic bands in wild population was approximately 1.5 times as diverse as that in cultured. The average level of bandsharing values was $0.40 \pm 0.05$ in wild population, but was $0.69 \pm 0.08$ in cultured population, With reference to bandsharing values and banding patterns, wild population was considerably more diverse than cultured population. Knowledge of the genetic diversity of crucian carp should help in formulating more effective strategies for managing this aquacultural fish species.

  • 고기가 자망에 이망될 숙건하에 그물면에 작용하는 붕어(Carassius carassius)의 유영력을 시험수조에서 strain gauge를 써서 즉정한 결과는 다음과 같다. 1. 순간최대유영력 및 지속최대유영력과의 체중과의 관계는 F 하(M) =1.45W, F 하(s) =0.29W 이다. 2. 순관최대유영력 및 지속최대유영력과의 체중과의 관계는 F 하(M) =0.11L 상(2.63), F 하(S) =0.15L 상(1.77)이다. 3. 붕어의 유체저항계수는 C 하(s) ≒0.287이다.

영천호와 대청호에서 담수어류의 microcystin 농축에 따른 인체 건강위해성 평가 (Evaluation of the Potential Human Health Risk Associated with the Microcystin Bioaccumulation in the Freshwater Fish from Lake Yeongcheon and Lake Daecheong)

  • 영천호와 대청호 어류들의 각 조직들(근육, 간, 내장, 아가미)에 농축된 microcystin-LR의 농도 및 TDI(total daily intake)값에 기초하여 인체의 건강위해성을 평가하였다 영천호의 분석결과에서 조식성(藻飾性)인 떡붕어(Carassius cuvieri)는 다른 종들과 비교하여 간을 포함한 대부분의 조직에서 높은 microcystin-LR의 농축양상을 보였다. 대청호에서 잡식성(雜食性)인 누치와 조식성인 떡붕어의 각 조직에서 높은 microcystin-LR 농도가 관찰된 가운데 내장에서는 누치와 떡붕어가 매우 높은 microcystin-LR 농도를 보였으며, 떡붕어는 다른 종들과 비교하여 내장이외에도 간과 아가미에서 높은 microcystin-LR농도를 나타내었다. 인체의 건강위해성 평가를 위해 각 조직에서 도출된 microcystin-LR의 EDI(estimated daily intake) 값들을 TDI 값과 비교하였다. 결과에서, 두 조사지역에서 모두 떡붕어(Carassius cuvieri)는 근육을 제외한 대부분의 조직들에서 EDI값들이 TDI 값을 초과하는 양상을 보였다.
