3D Bioprinted GelMA/PEGDA Hybrid Scaffold for Establishing an In Vitro Model of Melanoma

  • Due to the high incidence of malignant melanoma, the establishment of in vitro models that recapitulate the tumor microenvironment is of great biological and clinical importance for tumor treatment and drug research. In this study, 3D printing technology was used to prepare GelMA/PEGDA composite scaffolds that mimic the microenvironment of human malignant melanoma cell (A375) growth and construct in vitro melanoma micro-models. The GelMA/PEGDA hybrid scaffold was tested by the mechanical property, cell live/dead assay, cell proliferation assay, cytoskeleton staining and drug loading assay. The growth of tumor cells in two- and three-dimensional culture systems and the anti-cancer effect of luteolin were evaluated using the live/dead staining method and the Cell Counting Kit-8 (CCK-8) method. The results showed a high aggregation of tumor cells on the 3D scaffold, which was suitable for long-term culture. Cytoskeleton staining and immunofluorescent protein staining were used to evaluate the degree of differentiation of tumor cells under 2D and 3D culture systems. The results indicated that 3D bioprinted scaffolds were more suitable for tumor cell expansion and differentiation, and the tumor cells were more aggressive. In addition, luteolin was time- and dose-dependent on tumor cells, and tumor cells in the 3D culture system were more resistant to the drug.

  • During image-guided radiation treatment of lung cancer patients, it is necessary to track the tumor motion because it can change during treatment as a consequence of respiratory motion and cardiac motion. In this paper, we propose a method for tracking the motion of the lung tumors based on the three-dimensional image information from planning 4D MDCT and treatment 4D CBCT images. First, to effectively track the tumor motion during treatment, the global motion of the tumor is estimated based on a tumor-specific motion model obtained from planning 4D MDCT images. Second, to increase the accuracy of the tumor motion tracking, the local motion of the tumor is estimated based on the structural information of the tumor from 4D CBCT images. To evaluate the performance of the proposed method, we estimated the tracking results of proposed method using digital phantom. The results show that the tumor localization error of local motion estimation is reduced by 45% as compared with that of global motion estimation.

  • Hazardous air pollution caused by heavy metals in the air is at a serious level. Although manganese(Mn), one of the heavy metals, is a non-carcinogenic substance, it has a harmful influence on the human body. It is partially measured because automatic monitoring technologies have not yet be fully established. We introduced a statistical model for the daily concentration of manganese. Incorporating a linkage between Mn and meteorology, the proposed model is formulated in way to identify meteorological effects and to allow for seasonal trends, enabling not only accurate measurement of manganese concentration, but also information about the evaluation on a Hazard Quotient (non-cancer risk).

  • Single nucleotide polymorphisms (SNPs) are the most abundant form of human genetic variation and are a resource for mapping complex genetic traits. A genome is covered by millions of these markers, and researchers are able to compare which SNPs predominate in people who have a certain disease. The International HapMap Project, launched in October, 2002, motivated us to start the Korean HapMap Project in order to support Korean HapMap infrastructure development and to accelerate the finding of genes that affect health, disease, and individual responses to medications and environmental factors. A Korean SNP and haplotype database system was developed through the Korean HapMap Project to provide Korean researchers with useful data-mining information about disease-associated biomarkers for studies on complex diseases, such as diabetes, cancer, and stroke. Also, we have developed a series of software programs for association studies as well as the comparison and analysis of Korean HapMap data with other populations, such as European, Chinese, Japanese, and African populations. The developed software includes HapMapSNPAnalyzer, SNPflank, HWE Test, FESD, D2GSNP, SNP@Domain, KMSD, KFOD, KFRG, and SNP@WEB. We developed a disease-related SNP retrieval system, in which OMIM, GeneCards, and MeSH information were integrated and analyzed for medical research scientists. The kHapMap Browser system that we developed and integrated provides haplotype retrieval and comparative study tools of human ethnicities for comprehensive disease association studies (http://www.khapmap.org). It is expected that researchers may be able to retrieve useful information from the kHapMap Browser to find useful biomarkers and genes in complex disease association studies and use these biomarkers and genes to study and develop new drugs for personalized medicine.

The Effect of Audiovisual Information with Videotape on Knowledge and Attitude of Early Detection of Gastric Carcinoma (비디오 프로그램을 통한 정보제공이 위암조기발견에 대한 지식과 태도에 미치는 효과)

  • Gastric Carcinoma is very plentiful and significant disease in Korean community. The reason is that Gastric Carcinoma is not a simply finding disease with unclear symptoms for early detection. Early detection and early medical treatment of a cancer patient is an important instrument of reducing the mortility rate. The purpose of this study is to identify the effect of audiovisual information with videotape infulencing on knowledge and attitude of early detection of gastric carcinoma. The research design was a non-equivalent control group, pre-post test. The subjects of this study were 52 members of D-taxi drivers which was located in Pusan. The subjects were twenty six experimental group and twenty six control group. The experimental group was collected from January 15, 1999 to January 26, 1999. The control group was collected from the research was 28 minutes video program which was turning out the form of documentary on the based practical experience. And also the experimental tool was used measurung instrument which measured a dependent, variable throughout the consulatation of an percentage, average, standard deviation, ${\chi}^2-test$ and t-test using spss/pc program. The result of this research were as follows : 1)" The experimental group which was offered the video program, should be higher in knowledge of early detection of the gastric carcinoma than the control group" was supported. (t= -7.754, p=.000) 2) "The experimental group which was offered the video program, should be higher in attitude of the early detection of the gastric carcinoma than the control group" was supported. (t=-4.321, p=.000) Therefore, in conclusion, this study that the audiovisual information with videotape influencing on knowledge and attitude of early detection of gastric carcionma was very representational experience throughout the video of documentary form on the based practical experience was much effected the change of the knowledge and the attitude regarding to the early detection of the gastric carcinoma was verified.

Development of Source Profiles for Asbestos and Non-asbestos Fibers by SEM/EDX (SEM/EDX를 이용한 석면 및 비석명의 오염원분류표 개발)

  • There are many varieties of asbestos: chrysotile, crocidolite, amosite, tremolite, actinolite, and anthophylite. These are widely used in construction materials, brake lining, textile, and so on. Even though non-asbestos fibers such as glassfiber and rockwool have manufactured because asbestos causes asbestosis, lung cancer, mesothelioma, etc., some bad effects of non-asbestos have been also reported. PCM (phase contrast microscopy) and PLM (polarized light microscopy) have been used to qualitatively analyze asbestoses. These techniques have serious drawbacks when identifying and separating various asbestoses. Recently scanning electron microscopy (SEM) equipped with energy dispersive X-ray analysis (EDX) has been known as an useful tool to analyze airborne particle since it provides physical and chemical information simultaneously. The purpose of the study was to classify both asbestos and non-asbestos fibers and finally to develop their source profiles by using the SEM/EDX. The source profiles characterized by 6 different types of asbestos fibers and 2 types of non-asbestos fibers had been developed by analyzing a total of 380 fibers. Analytical parameters used in this study were length, width, aspect ratio, and shape as physical information, and Na, Mg, Al, Si, K, Ca, Cr, Mn, Fe, and Cu as chemical information. All the parameters were intensively reviewed.

A Study on the Consumer Recognition of Food Safety and Food Additives (식품의 안전성 및 식품첨가물에 대한 소비자 인식에 관한 연구)

  • This study was to investigate consumer recognition of food safety and food additives. The results of this study are as follows: 1. When shopping for food, the food safety was rated as foremost concern for the respondents and the details checked most often were manufacture date, packing condition and axpiration date, respectively. When asked their opinion about harmful factors in relation to food safety, the most important consideration was food additives. The highest rating for credibility regarding the sources of information on food was given to scientists in university and institute, lowest one was given to food manufacturer. 2. Government regulation on the use of food additives was known to exist, but control of the uses of food additive was considered inadequate by 60% of the respondents. These results showed significant differences for age (p<0.05), for education level (p<.01) and for income (p<.05) respectively. 3. 47.9% of the respondents indicated that they were willing to purchase the additive free foods, although it was expensive. More than 70% made an effort to eat food with less food additive and were concerned about possible effects of food additives on health, showing significant differences for age (p<0.05), for education level (p<0.05) and for income (p<0.05). 4. Labeling for food additive found on food package was shown to be unsatisfactory, showing significant difference only for income (p<0.05) and more information about food was needed by over 91.3% of the respondents, showing significant difference for education level (p<0.05). 5. Although approximately 80% of the respondents were concerned about artificial preservative, artificial flavor, artificial color and artificial sweetener, the concern about artificial preservative was the highest. Primary causes that respondents felt fear toward food additives were the unknown harms and cancer, and primary sources that respondents got information on food were televison and radio, respectively. From these results, it was shown that consumer considered food safety important and they felt a great deal of concern about food additives.

Recognition of Endocrine-Disrupting Chemicals and Behavior to Reduce Exposure to Endocrine-Disrupting Chemicals in Cooking Staff Working at Child Care Center Located in Gyeonggi Province (경기도 일부 어린이집 조리종사자의 내분비계 장애물질 인식도 및 노출 저감화 행동 조사)

  • Endocrine-disrupting chemicals (EDCs) are chemicals present in the environment that interfere with the normal hormone functions of various organisms and cause genetopathy, deformities, or cancer. This study surveyed the awareness of EDCs with 242 cooking staff at 242 meal service facilities for children located in a part of Gyeonggi Province. To minimize infants' exposure to EDCs, the subjects were provided with information on EDCs for two months. The behavior of reducing exposure to EDCs was analyzed according to the awareness of EDCs and work ethics. In addition, the effects before and after being provided with the information were evaluated. According to the results, the levels of awareness of EDCs and work ethics' scores of the cooking staff were high with 3.95 and 4.39 points, respectively, out of five points. In addition, a higher awareness of EDCs and a higher work ethics' score were associated with an improved behavior of reducing exposure to EDCs (P<0.05). The overall cooking and cleaning behavior for reducing exposure to EDCs showed improvement after providing information (P<0.01). Therefore, these results suggest that it is important to provide continuous education to enhance the awareness of EDCs, work ethics, and behavior to reduce exposure to EDCs.

Differences of Cold-heat Patterns between Healthy and Disease Group (건강군과 질환군의 한열지표 차이에 관한 고찰)

  • The pattern identification of exterior-interior syndrome and cold-heat syndrome is one of the diagnostic methods using most frequently in Oriental medicine. There was no systematic studies analyzing the characteristics of the 'exterior-interior and cold-heat' between healthy and disease group. In this study, cold-heat pattern, blood pressure, pulse rate, height and weight are recorded from 100 healthy subjects and 196 disease subjects with age ranging from 30 to 59 years. To analyze the differences between healthy and disease group, we used the descriptive statistics. And linear regression function, linear support vector machine and bayesian classifier were used for distinguishing healthy group from disease group. The score of both exterior-heat and interior-cold in healthy group is higher than the score in disease group. This means that if one belongs to the disease group, his(or her) exterior gets cold and his interior gets hot. And also, these result have no relevance to age. But, the attempt to classify healthy group from disease group with a exterior-interior and cold-heat and other vital signs did not have good performance. It mean that even though they have a different trend each other, only these kinds of information couldn't classify healthy group and disease group.

A Study on Clinical Decision Support System based on Common Data Model (공통데이터모델 기반의 임상의사결정지원시스템에 관한 연구)

  • Recently, medical IT solutions are being provided on a distributed environment basis. In Korea, the necessity of developing a clinical decision support system that can share medical information in a distributed environment has been recognized and studied. The existing clinical decision support system is being built using only medical information of its own within the hospital. This makes it difficult for existing systems to achieve good results in terms of efficiency and accuracy of decision support. In order to solve these limitations, this paper proposes a design and implementation method of clinical decision support system based on common data model in medical field. To explain the application process of the proposed model, we describe the development scenario of the clinical decision support system for the diagnosis of colorectal cancer. We also propose the essential requirements for the development of successful clinical decision support systems. Through this, it is expected that it will be possible to develop clinical decision support system that can be used in various hospitals and improve the efficiency and accuracy of the system.
