Analysis of Microbial Contamination of Sprouts and Fresh-cut Salads in a Market (유통중인 즉석섭취 새싹채소와 샐러드의 세균오염 분석)

  • Although bacterial outbreaks from ready-to-eat foods have increased, little information is available on microbial quality of sprouts in markets. Fifty sprouts and 30 salads were collected from wholesale markets. Total aerobic count (TAC), coliform, Escherichia coli, and some pathogens were detected. TAC for sprouts was 7.95 log CFU/g and 6.70 for salads, indicating that sprouts were more contaminated by 1 log CFU/g than that of salads. The numbers of coliform were 6.69 log CFU/g for sprouts and 5.42 for salads. E. coli was detected in 16 of 50 sprout samples at 2.38 log CFU/g and eight of 30 salads at 2.21 log CFU/g. Bacillus cereus was detected in 29 of 50 sprout samples and 16 of 30 salads, and the counts were mostly <3 log CFU/g. Salmonella, Staphylococcus aureus, Listeria monocytogenes, Campylobacter jejuni, and Clostridium perfringens were not detected. Therefore, although pathogens may not be a high risk for these foods, the high TAC and E. coli contamination require improved production and distribution methods, particularly for sprouts.

Isolation and Identification of Pathogenic Microorganisms from Soybean Sprouts

  • Raw soybean sprouts were tested for contamination with the following bacteria which have potential for pathogenesis or food spoilage : Salmonella spp., Escherichia coli O157:H7, Yersinia enterocolitica, Vibrio parahae-molyticus, Aeromonas hydrophila, Plesidomonas shigeloides, Pseudomonas aeruginosa, Staphylococcus aureus, Lis-teria monocytogenes, Bacillus cereus, Clostridium perfringens, Campylobacter jejuni, Erwinia spp., and Fusarium spp. Three of the above strains were isolated from the sprouts, and identified by morphological and biochemical methods including an API kit and ATB automated identification system. The isolate cultured in Cereus selective agar, a selective medium, was a Gram-positive, rod shaped, anaerobic spore former. The biochemical and culture tests revealed the following characteristics: catalase-positive, no growth on Simmon's citrate, NO₂ production and requirement of arginine for growth; the ATB automated identification system gave 99.8 % agreement for the identification of Bacillus cereus to the species level. The isolate cultured in Macconkey agar selective medium was Gram-negative, rod shaped and a gas former; the ATB-system gave 99.9% agreement for the identification of Aeromonas hydrophila to the species level. The isolate found in Pseudomonas isolation agar was Gram-negative, rod shaped, cytochrome oxidase-positive, a reducer of nitrates to nitrogen, and pyocyanin producer; the ATB-system gave 99.9 % agreement for the identification of Pseudomonas aeruginosa to the species level. These results indicate that the three bacteria species present in the soybean sprouts were Bacillus cereus, Aero-monas hydrophila, and Pseudomonas aeruginosa. Salmonella spp., Escherichia coli O157:H7, and Yersinia enter-ocolitica, which are associated with serious disease in humans, were not isolated from soybean sprouts examined in this study.

Nucleotide Sequence and Secondary Structure of 16S rRNA from Sphingomonas chungbukensis DJ77 (Sphingomonas chungbukensis DJ77의 16S rRNA 염기서열과 이차구조)

  • A 16S ribosomal RNA gene from S. chungbukensis DJ77 has been sequenced. This sequence had a length of 1,502 bp and was extended for 29 bp at 5' and for 37 bp at 3' from the partial sequence (1,435 bp) registered in 2000 year. Besides, 1 bp was newly added near to the 3' end. We made the secondary structure of the 16S rRNA based on E. coli model and found four specific regions. We found constant and variable regions in genus Sphingomonas as the result of multiple alignment of 16S rRNA gene sequences from Sphingomonas spp. and S. chungbukensis DJ77. We found a stem loop structure in S. chungbukensis DJ77, which was only discovered in C. jejuni to date. It showed the structural agreement despite the difference of the sequences from the both organisms. Finally, S. chungbukensis DJ77 belonged to cluster II (Sphingobium) group, after the classification using phylogenetic analysis and nucleotide signature analysis.

Characteristics of Enteric Pathogens Isolated from Acute Enteritis Patients with Overseas Travel (해외여행 후 발생한 급성장염의 원인 병원체 분석)

  • Background: The risk of imported infectious diseases has been increasing with the annual rise in the number of international travelers. Objectives: This study aims to analyze the distribution and characteristics of intestinal bacteria isolated in 2019 from residents of Chungcheongnam-do Province with experience of travelling overseas. Methods: Twenty-three former overseas travelers with diarrhea were analyzed to detect viruses and bacteria according to the Manual for Detection of Foodborne Pathogens at Outbreaks. Additionally, antibiotic susceptibility tests and 16s rRNA sequencing were performed. Results: Twenty-five strains of ten pathogens were isolated from 18 samples. Pathogenic E. coli was the most common at 57.7%, followed by Clostridium perfringens (15.4%), Campylobacter spp. (7.7%), and Salmonella spp. (7.7%). The serotype of Salmonella was confirmed as Salmonella Braenderup, II 9,46:g,[m],[s],t:[e,n,x]. Conclusions: It was confirmed that the major enteric bacterial pathogens isolated from overseas travelers in Chungcheongnam-do Province were pathogenic E. coli, as found in other studies. The study on Plesiomonas shigelloides is meaningful in that it is reported as a rare case of infection in Korea. Antibiotic resistance and 16s rRNA analysis were performed, which is expected to provide important basic data for the prevention of traveler's diarrhea.

Antioxidant and Antimicrobial Activities of Quinoa (Chenopodium quinoa Willd.) Seeds Cultivated in Korea

  • The objective of this study was to investigate the antioxidant and antimicrobial properties of quinoa cultivated in Korea and to compare it with imported quinoa from the USA and Peru. The highest amount of total flavonoid contents (TFC) with 20.91 mg quercetin equivalents/100 g was measured in quinoa seed extract cultivated in Korea, while the total phenolic contents (TPC) were significantly higher in quinoa from the USA (16.28 mg gallic acid equivalents/100 g). In addition, quinoa extracts cultivated in Korea displayed a superior antioxidant ability in both, ferric reducing antioxidant power and 1,1-diphenyl-2-picrylhydrazyl values. There was a high correlation between TFC and antioxidant activity and a low correlation between TPC and antioxidant activity. The antimicrobial activity of the quinoa extracts was determined using a disc diffusion assay and optical density method. In both assays, the quinoa seed extracts did not have strong antimicrobial activity against foodborne bacteria, including Staphylococcus aureus, Listeria monocytogenes, Bacillus cereus, Escherichia coli, Salmonella Typhimurium, and Campylobacter jejuni.

Backbone 1H, 15N, and 13C Resonance Assignments and Secondary-Structure of the Conserved Hypothetical Protein HP0892 of Helicobacter pylori

  • HP0892 (SwissProt/TrEMBL ID O25552) is a 90-residue conserved hypothetical protein from Helicobacter pylori strain 26695, with a calculated pI of 9.38 and a molecular mass of 10.41 kDa. It belongs to the Plasmid stabilization system protein family (PF05016) in the Pfam database. Proteins with sequence similarity to HP0892 exist in Vibrio choierae, Enterococcus faecalis, Campylobacter jejuni, Streptococcus pneumoniae, Haemophilus influenzae, Escherichia coli O157. Here we report the sequence-specific backbone resonance assignments of HP0892 using multidimentional heteronuclear NMR spectroscopy. About 97.0% (422/435) of the HN, N, CO, $C{\beta}$, $C{\alpha}$ resonances of 90 residues of HP0892 were assigned. On the basis of the resonance assignments, three helical regions and four strand regions were identified using the CSI program. This study is a prerequisite for calculating the solution structure of HP0892, and will be useful for studying its interaction with other molecules.

정책초점2 - 식품자판기 다류.커피.음료류에 대한 미생물 기준 신설

  • 식품자판기에 대한 미생물 기준이 마련되었다. 식품의약품안전청은 식품자판기 다류 커피 음료류에 대한 미생물기준을 담은 "식품의 기준 및 규격"를 일부 개정 고시했다. 이 기준이 마련된 것은 청결상태 등 위생관리가 불량하여 지속적으로 문제가 되고 있는 식품자동판매기 음료 등에 대해 효율적인 관리를 위한 기준이 필요하다는 판단 때문이다. 식품자판기 다류 커피 음료류에 적용되는 세균기준은 3,000/mL 이하(다만, 유가공품, 유산균, 발효제품 및 가열하지 아니한 과일 채소류음료가 함유된 경우는 제외)이어야 하며, 대장균은 음성이어야 한다. 이외 살모넬라(Salmonella spp.), 황색포도상구균(Staphylococcus aureus), 장염비브리오균(Vibrio parahaemolyticus), 클로스트리디움 퍼프린젠스(Clostridium perfringens), 리스테리아 모노사이토제네스(Listeria monocytogenes), 대장균 O157:H7(Escherichia coli O157:H7), 캠필로박터 제주니(Campylobacter jejuni), 여시니아 엔테로콜리티카(Yersinia enterocolitica) 등의 식중독균이 검출되어서는 아니 된다. 또한 가공식품에서 많이 검출되는 바실러스 세레우스(Bacillus cereus)는 g 당 1,000 이하(단 멸균제품은 음성이어야한다)이어야한다. 이번 미생물기준의 마련은 앞으로 법정기준을 위반시 처벌대상이 된다는 점에서 유의해야할 사항이다. 이전까지는 식품자판기에 대한 미생물기준이 마련이 되어 있지 않기 때문에 마땅한 법적인 처벌 기준이 없었던 게 사실이다. 따라서 자판기 운영업체에서는 보다 철저한 위생관리가 요구되고 있다. 위생점검에서 미생물 기준을 지키지 않은 지판기에 대해서는 법적 처벌 근거가 마련이 되는 만큼 식품자판기 위생성 향상에 만전을 기해야 할 것이다. 6월 16일 개정고시된 미생물기준은 7월 1일부터 적용된다.

Survey on the Status of Microbial Contamination of Chicken Meats Collected from Poultry Processing Plants in Nationwide (우리나라 도계장 수거계육의 미생물학적 위생실태 조사)

  • This study was conducted to survey the hygienic status of chicken meats on the microbial levels, which were collected from poultry processing plants located in the local provinces in nationwide including the JeJu island (n=15) in 1997. In particular, Salmonella spp., Campylobacter jejuni, and Listeria monocytogenes, which were retarded as one of the most important entero-pathogens relating to food home illness from poultry, were investigated on their isolation frequency including the other pathogens related on the food-borne illness. A total of 115 processed chickens were submitted on the present study. In general, the bacterial contamination frequency showed more or less lower $(10{\sim}100 cells)$ than those of sold on the retail and super markets and department stores because of lacking of cross-contamination incidences, depending on the total cells, Coliforms and Staphylococcal cells count. While, Salmonella species, Campylobacter jejuni, Listeria monocytogenes, and coagulase positive Staphylococcus aureus isolation frequency of chicken meats from slaughter houses were 58.3%, 37.4%, 43.5%, and 30.4%, in order. But the present microbial isolation data were a little lower levels than those of sold on the retail and super markets and famous department stores in Seoul and GyeongGi province at the same period. It seemed that the cross-contamination problems (including the human, environmental and instrumental factors) during the marketing stage (after the last processing procedure; rinsing step) had the major roles on the increasing of the microbial contamination frequency on the chicken meats after the slaughter houses.

Risk Analysis and Safety Assessment of Microbiological and Chemical Hazards in the Dried Sea Mustard Undaria pinnatifida Distributed in Markets (유통중인 건미역(Undaria pinnatifida)의 미생물학적·이화학적 위해요소 분석 및 안전성 평가)

  • For the safety assessment of microbiological and chemical hazards in dried sea mustard, fifteen samples of dried sea mustards Undaria pinnatifida were purchased from the supermarkets distributed throughout Korea. The contamination levels of total viable bacteria, coliforms, Escherichia coli, and nine pathogenic bacteria [Staphylococcus aureus, Salmonella spp., Listeria monocytogenes, Bacillus cereus, Vibrio spp., Clostridium perfringens, Enterohemorrhagic Escherichia coli (EHEC), Yersinia enterocolitica and Campylobacter jejuni/coli] were quantitatively or qualitatively assessed. Also, the heavy metals (lead, cadmium, total mercury, and inorganic arsenic), and radioactivity (131I, 134CS+137CS) were quantitatively assessed. This microbial and chemical analysis was performed using standard methods in Korean food code. The total viable bacteria ranged from 4.3×102 (5.0×10-1.5×103) CFU/g. Coliforms and E. coli were not detected in all samples (ND, <1 log10 CFU/g). All nine pathogenic bacteria were qualitatively detected as negative. The contamination levels of lead, cadmium, total mercury, and inorganic arsenic were 0.036 (0.015-0.051), 0.117 (0.088-0.156), 0.030 (0.017-0.048), and 0.058 (0.056-0.064) mg/kg, respectively. Radioactivity was also not detected in any sample. The microbial contamination levels determined in the current study may be potentially used as basis for performing microbial risk assessments of dried sea mustards.
