인체 상피세포주와 암세포주에 대한 소목 추출물의 증식억제 효과 (Antiproliferative Effects of Caesalpinia sappan Extract on Human Epithelial Cell Line HaCaT and Cancer Cell Lines)

  • Caesalpinia sappan L. (Leguminosae) is an oriental medicinal herb distributed in China and Taiwan, and its heartwood has been traditionally used as an analgesic, a therapy for thrombosis or tumor. This study was to investigate the proliferation and inhibition effects of Caesalpinia sappan extracts against human epithelial cell and cancer cell lines. The methanol extract of dried C. sappan heartwood was evaporated (KS-6), and then sequentially extracted by hexane (KS-01), chloroform (KS-02), ethyl acetate (KS-03), n-butanol (KS-04), and water (KS-05). After 48 hr of exposure, these fractions at a concentration of $50{\mu}g/ml$ significantly increased, and reduced cell proliferation in both human normal epithelial and cancer cell lines. The ethyl acetate fraction (KS-03) among organic solvent fractions was 120.2% of the most proliferation activity ($50{\mu}g/ml$) against HaCaT human epithelial cell. However, fractions from chloroform, butanolic and methanolic extract had 7.2, 28.7 and 20.8% of antiproliferative effect on HaCaT cell, respectively. In cell proliferation effects of C. sappan extract on HeLa, SiHa and C33A human cervical cancer cells, chloroform fraction (KS-2) was the most antiproliferative activity, its antiproliferative rate (dosage, $50{\mu}g/ml$) relative to control was 25.8, 12.2 and 17.4% for SiHa, HeLa and C33A, respectively. The results indicated that the six extract fractions could induce cell cycle stimulate or arrest in some way. Finally, further investigation is needed to assess the molecular mechanisms mediated anticancer activities of this plant.

Protective Effect of Caesalpinia sappan L. on Hydrogen Peroxide

  • In this study, we demonstrated the antioxidant effect of the Caesalpinia sappan L. extract through the scavenging effect against 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical and the protective effect on protein damage and PC12 cells against cupric ion/hydrogen peroxide. Its IC/sub 50/ value of the scavenging effect against DPPH radical was 7.7 ㎍. Protection of its extract against oxidative bovine serum albumin (BSA)damage induced by hydrogen peroxide was more effective than that of vitamin C. The protective effect on PC12 cells by hydrogen peroxide was shown to be more potent in is extract than in vitamin C. DNA fragmentation analysis also supports this result.

  • Thirteen phenolic compounds, 1,4-dimethoxybenzene (1), 3,4-dihydroxybenzaldehyde (2), (2E)-3-(4-hydroxy-3,5-dimethoxyphenyl)acrylaldehyde (3), 3,7-dihydroxy-4H-chromen-4-one (4), 2,3-dihydroxy-1-(3,4-dihydroxyphenyl)propan-1-one (5), 4-hydroxy-3-methoxybenzoic acid (6), 4-hydroxy-3,5-dimethoxybenzoic acid (7), methyl 3,4-dihydroxybenzoate (8), 4-hydroxy-3,5-dimethoxybenzaldehyde (9), 3,4-dihydroxybenzoic acid (10), 3-hydroxy-1-(4-hydroxy-3-methoxyphenyl)propan-1-one (11), 2,4,6-trihydroxybenzaldehyde (12) and benzene-1,2,4-triol (13) were isolated from the heartwood of Caesalpinia sappan. Their anti-inflammatory activity was evaluated against LPS-induced NO production in macrophage RAW264.7 cells. Among them, compounds 3 and 8 showed strong inhibitory activities toward the LPS-induced NO production in macrophage RAW264.7 cells with $IC_{50}$ values of 14.5 and 21.5 ${\mu}M$, respectively.

소목 천연 염색에 관한 연구 I - 정제 알루미늄 화합물들의 매염효과에 대하여 - (A Study on Natural Dyeing using Caesalpinia sappan -Mordanting Effect of Purified Aluminum Compounds-)

  • Currently, as mordants, heavy metals are being used in dyeing process in order to improve dye uptake and dye fastness. These chemicals, however, are mostly harmful to human body and the environment along with the pollution by effluent. Traditionally, rice straw ash solution, lime, etc. have been used as mordants in order to improve the effects on dye uptake and color fastness of dyestuffs. The study of natural mordants would, therefore, be vital to the development and succession of our traditional dyeing methods. In this study, as a preliminary step toward the use of rice straw ash solution for the nautral dyeing using Caesalpinia sappan, several aluminum compounds were employed as mordants. When the purified aluminum compounds were used, on the basis of unmordanted cotton fabric, the K/S values of the pre-mordanted and simultaneously mordanted fabrics increased by 2 or 3 times, while those of the post-mordanted fabrics decreased.

  • By introducing chitosan treatment prior to dyeing, examination was given to the effect of chitosan coated on the surface of fabrics on the color change of dyed fabric based on th e change of ${\Delta}E$, a*, and b* values. At the same time, the dyeing mechanism of Caesalpinia sappan dyestuff was predicted by the investigation of the change of air-permeability ac cording to the chitosan treatment. The change of elution was investigated by the examination of the elution of metallic ions employed as mordants after soaking dyed fabrics in the solution of alkaline perspiration ba sed upon the fact that chitosan carries excellent absorption ability toward metallic ions.

  • The Caesalpinia sappan is widely used in the traditional oriental herbal medicine for anti-inflammatory, antioxidant effects. The effects of water extract of C. sappan on the cell viability and induction of apoptosis were investigated in human lung cancer cell line A549. The water extract of C. sappan produced apoptotic cell death and DNA fragmentation and nucleus chromatin condensation in A549 cells. The enzyme activity of caspase-3 and protein level of actived caspase-3 were markedly increased in A549 cells treated with the water extract of C. sappan. In addition, the extract of C. sappan induced cleavage of Poly (ADP-ribose) polymerase (PARP), a known substrate for caspase-3, and dropped in cellular ATP levels. These results suggest that the extract of C. sappan exerts anticancer activity by induction of apoptosis via activation of caspase-3, cleavage of PARP protein, and depletion of cellular ATP levels in A549 cells.

소목(Caesalpinia sappan L.)의 에틸아세테이트 분획물이 Streptococcus mutans에 대한 항균활성 및 부착 억제 (Antimicrobial Activities and Adherence Inhibition on Streptococcus mutans by Ethyl Acetate Extract from Caesalpinia sappan L.)

  • 본 연구는 소목(Caesalpinia sappan L.)의 6가지 유기용매 분획물 중 IP-3의 항균활성을 및 부착능을 측정하고 다음과 같은 결과를 얻었다. 1. Methanol에 의해 분리된 소목의 항균 물질 수율은 총 4.8%로 나타났으며, 여러 분획물 가운데 EtOAc 분획물(IP-3)에서 3.94%로 가장 높은 수율을 보였다. 2. 소목의 EtOAc 분획물에서 최종 정제된 활성물질은 UV 288.3 nm에서 최대 흡수피크를 나타내었으며, NMR spectrum을 통해 brazilin(($C_{16}H_{14}O_5$)으로 확인되었다. 3. S. mutans에 대한 소목의 EtOAc 분획물의 항균 활성은 최소억제농도(MIC) 3.1 mg/ml에서 균의 생장을 억제하였다. 4. S. mutans 부착억제효과는 하이드록시아파타이트(HA)를 S. mutans와 24 시간 동안 함께 배양한 대조군은 균 부착률이 높게 관찰되었으나, 소목의 EtOAc 분획물을 처리한 군에서는 HA에 S. mutans의 부착이 매우 적게 나타났다. 이상의 결과를 통해, 소목에서 추출된 EtOAc 분획에 존재하는 brazilin이 구강 내 우식원성세균에 강한 항균력을 가지고 있는 것으로 판단되며, 특히 S. mutans의 부착 억제능이 확인되어 우식원성 균주에 대한 생약치료제로 활용이 가능할 것으로 사료된다.

Color Alteration and Acaricidal Activity of Juglone Isolated from Caesalpinia sappan Heartwoods Against Dermatophagoides spp.

  • Acaricidal effects of materials derived from Caesalpinia sappan heartwoods against Dermatophagoides farinae and D. pteronyssinus were assessed and compared with those evidenced by commercial benzyl benzoate and DEET. The observed responses varied according to dosage and mite species. The $LD_{50}$ values of the methanol extracts derived from C. sappan heartwoods were 6.13 and $5.44{\mu}g/cm^3$ against D. farinae and D. pteronyssinus, respectively. Furthermore, the ethyl acetate fraction derived from the methanol extract was approximately 8.71 more toxic than DEET against D. farinae, and 4.73 times more toxic against D. pteronyssinus. The biologically active constituent from the ethyl acetate fraction of C. sappan heartwood extract was purified via silica gel chromatography and high-performance liquid chromatography. The structure of the acaricidal component was analyzed by $GC-MS,\;^1H-NMR,\;^{13}C-NMR,\;^1H-^{13}C\;COSY-NMR$, and DEPT-NMR spectroscopy, and identified as juglone (5-hydroxy-l,4-naphthoquinone). Based on the $LD_{50}$ values of juglone and its derivatives, the most toxic compound against D. farinae was juglone ($0.076{\mu}g/cm^3$), followed by benzyl benzoate ($9.143{\mu}g/cm^3$) and 2methyl-l,4-naphthoquinone ($40.0{\mu}g/cm^3$). These results indicate that the acaricidal activity of C. sappan heartwoods is likely to be the result of the effects of juglone. Additionally, juglone treatment was shown to effect a change in the color of the cuticles of house dust mites, from colorless-transparent to dark brownish-black. Accordingly, as a naturally occurring acaricidal agent, C. sappan heartwood-derived juglone should prove to be quite 'useful as a potential control agent, lead compound, and house dust mite indicator.

Effect of Ethyl Acetate Extract from Caesalpinia sappan L. on Melanogenesis in Melan-a cells

  • Melanin is a main pigment found in skin. hair and eyes, and tyrosinase plays an important role in the process of melanin polymer biosynthesis. Caesalpinia sappan L.(C. sappan) has been commonly used in Oriental folk medicines to promote blood circulation and as analgesic as well as remedy for thrombosis. Brazilin as a new sunless tanning agent

  • To develop an active material for skin darkening, we examined the effect of 300 plants on tyrosinase activity and found only Caesalpinia sappan has an ability to increase tyrosinase activity highly and melanin contents in B-16 melanoma cells. A compound increasing tyrosinase activity and melanin production was isolated from Caesalpinia sappan Lignum. Brazilin was identified as a new active agent. Brazilin increases the tyrosinase activity and malanin production of B-16 melanoma cells. In conclusion, it seems that brazilin cal be used as a new sunless tanning agent.
