Effects of Different Dietary Cadmium Levels on Growth and Tissue Cadmium Content in Juvenile Parrotfish, Oplegnathus fasciatus

  • This feeding trial was carried out to evaluate the effects of different dietary cadmium levels on growth and tissue cadmium content in juvenile parrotfish, Oplegnathus fasciatus, using cadmium chloride ($CdCl_2$) as the cadmium source. Fifteen fish averaging $5.5{\pm}0.06$ g (mean${\pm}$SD) were randomly distributed into each of twenty one rectangular fiber tanks of 30 L capacity. Each tank was then randomly assigned to one of three replicates of seven diets containing 0.30 ($C_0$), 21.0 ($C_{21}$), 40.7 ($C_{41}$), 83.5 ($C_{83}$), 162 ($C_{162}$), 1,387 ($C_{1,387}$) and 2,743 ($C_{2,743}$) mg cadmium/kg diet. At the end of sixteen weeks of feeding trial, weight gain (WG), specific growth rate (SGR) and feed efficiency (FE) of fish fed $C_{21}$ were significantly higher than those of fish fed $C_{83}$, $C_{162}$, $C_{1,387}$ and $C_{2,743}$ (p<0.05). Weight gain, SGR and FE of fish fed $C_0$, $C_{21}$ and $C_{41}$ were significantly higher than those of fish fed $C_{162}$, $C_{1,387}$ and $C_{2,743}$. Protein efficiency ratio of fish fed $C_0$, $C_{21}$ and $C_{41}$ were significantly higher than those of fish fed $C_{1,387}$ and $C_{2,743}$. Average survival of fish fed $C_0$, $C_{21}$, $C_{41}$ and $C_{162}$ were significantly higher than that of fish fed $C_{2,743}$. Tissue cadmium concentrations increased with cadmium content of diets. Cadmium accumulated the most in liver, followed by gill and then muscle. Muscle, gill and liver cadmium concentrations of fish fed $C_0$, $C_{21}$, $C_{41}$ and $C_{83}$ were significantly lower than those of fish fed $C_{162}$, $C_{1,387}$ and $C_{2,743}$. Based on the ANOVA results of growth performance and tissue cadmium concentrations the safe dietary cadmium level could be lower than 40.7 mg Cd/kg diet while the toxic level could be higher than 162 mg Cd/kg diet.

  • Garlic occupies a special position among the many foods of vegetable origin because it is the sole food for Koreans during the their lives. And vitamin A has been ingested by forms of food or additives. Cadmium has been described as one of the most dangerous trace elements in the food and environment of man and livestocks. Since the de novo synthesis of stress proteins can be detected early after exposure to some agents, analysis of cadmium-induced changes in gene expression , ie. alterations in patterns of protein synthesis, may be useful to develop as biomarkers of exposure and damage for food hygiene. He acute and chronic combine effects of cadmium (Cd, CdCl2 20mg/kg), garlic oil(Dds: diallyl disulfide 50mg/kg, 3 times a week) and vitamin A(Ra: retinol acetate 50,000 IU/kg, 3 times a week) on Wistar male rats were evaluated concerning cadmium contents, tissues enzyme activity, HSP expression histopathological and electron microscopical examinations. The results of the study are as follows ; 1. Less cadmium was absorbed through the digestive tracts, but the ratio of contents in tissue were not changed by the simultaneous adminstration of diallyl disufide or retinol acetate. 2. ALT(alanine aminotransferase) , AST(aspartate aminotransferase), glucose, BUN (blood urea nitrogen), creatinine, the key indices of the clinical changes in hepatic and renal function were significantly hanged by the cadmium treatment after 1 week in liver, after 4 weeks in kidney. 3. Histopathological changes in cadmium treated rats were appeared at 8 weeks age treatment in kidneys. Homogenous eosinophilic material was accumulated in cortical and collecting tubular lumens at 16 weeks. Degenerated or necrotized tubular cells were observed in cortex and medulla. Degenerated seminiferous tubules and homogeneous eosinophilic material was seen in interstitial tissue of rat treated with cadmium for 16 weeks. Calcium deposits were seen in degenerated seminiferous tubules and the tubules showed severe calcification of rat treated with cadmium for 16 weeks. Electron microscope changes in kidney were observed in rats treated with CdCl2 20 mg/kg. Proximal convoluted tubule cells showed selling of cytoplasm and narrow lumen. Capillary endothelial cells showed cytoplasmic vacuoles and swelling. Degenerated epithelial cells were accumulated in tubular lumen of kidney. 4. Enhanced synthesis of 70 KDa relateve molecular mass proteins were detected in 2 hours after cadmium, exposure, with maximum activity occurring at 8~48 hours. Induction of HSP 70 was evident at proximal tubules and glomeruli in kidney. Testicular cells produced enough HSP to be detected normally. From the above results, it could be concluded that HSP70 induction by the cadmium treatment was a rapid reaction to indicated the exposure of xenobiotics, and retinol acetate reduced the cadmium induced nephrotoxicity.

Characterization of a Cadmium-resistant Yeast Strain in Response to Cadmium or Heat Shock Stress

  • A varient strain of budding yeast, Hansenula anomala B-7 which had been identified to be highly resistant to cadmium ions, were observed by transmission electron microscopy. It was shown that the cells accumulated excess amounts of cadmium ions throughout inside the cell rather than on the cell surface. The cell growth in response to cadmium or heat shock stress has also been investigated. It was observed that the cells precultured in the presence of 500 $\mu$ g/ml of Cd ions grew slower than those precultured at 1, 000 $\mu$ g/ml of the metal ions, when they were cultivated in the media containing 1, 000 $\mu$g/ml of the metal ions. Heat shock, however, stimulated the cell growth transiently, when the cells were allowed to grow in the presence of 1, 000 $\mu$g/ml of the metal ions. But the cells given heat shock for more than 100 min received permanent damage to growth. Effects of both stresses on budding rate was also examined. It revealed that the stresses did not change the budding ratio much, which was contradictory to that observed from the same budding yeast, Saccharomyces cerevisiae. Furthermore, the cells treated with 1, 000 $\mu$g/ml of the metal ions not only induced, but also switched off the expression of several new proteins. Some of the cadmium stress-inducible proteins were estimated to be also induced by heat shock stress.

Effect of Dietary Fibers in Rice and Barley on Lipid and Cadmium Metabolism in the Rat (쌀과 보리의 식이 섬유가 흰쥐의 지방 및 Cadmium 대사에 미치는 영향)

  • This study was performed to investigate effect of dietary fibers in rice and barley on glucose, lipid and cadmium (Cd) metabolism in the rat. Fifty-six male Spague-Dawley rats weighing 244.6$\pm$2.7g were blocked into eight groups according to body weight and raised for four weeks with diets containing 0 or 0.04%(w/w) CdCl2 and four different carbohydrate sources, starch, rice flour, barley flour and mixture of rice and barley flour(7 : 3, w/w). Total dietary fibers and $\beta$-glucan contents of barley were about three times higher than those of rice (10.75% vs. 3.94%, 3.11% vs. 1.06%, respectively). Food intake, weight gain, food efficiency ratio, liver and kidney weights were lower in Cd exposed groups, and barley group among Cd exposed animals showed highest weight gain, food efficiency ratio and organ weights. Fasting serum glucose levels were not significantly different among groups, Serum cholesterol level was lowest in Cd exposed barley group. Serum HDL-cholesterol level was higher in none-Cd exposed starch and barley groups, and HDL-cholesterol : total cholesterol ratios were higher in none-Cd rice and mixed flour groups than other groups. Liver total lipid and triglyceride levels were lowest in barley groups regardless of Cd administration. Fecal total lipid, cholesterol and triglyceride excretions were high in barley and mixed flour groups. Liver Cd concentrations were low in Cd exposed barley and mixed flour groups. In Cd exposed barley group, fecal weight and Cd excretion were highest and Cd retention ratio was lowest among groups. Small intestine metallothionein(MT) concentration was highest in Cd exposed rice group, and kidney MT concentration was highest in Cd exposed barley group. In conclusion, cereals showed different effects on lipid and Cd metabolism that might be mediated by dietary fibers in cereals. Especially $\beta$-glucan-rich barley group showed greatest lipid and Cd lowering effects by increasing fecal lipids and Cd excretions.

Effect of Korean Native Plant Diet on Lipid Metabolism, Antioxidative Capacity and Cadmium Detoxification in Rats (한국 고유의 산채류 첨가 식이가 흰쥐의 지방 대사 및 항산화능과 Cadmium 제독에 미치는 영향)

  • This study was performed to investigate the effects of Korean native plant diets on lipid and cadmium (Cd) metabolisms and also antioxidative enzyme activities in rats. Seventy male Sprague-Dawley rats weighing 210.3$\pm$2.7g were blocked into ten groups according to body weight. Rats were raised for four weeks with diets containing 0 or 0.04%(w/w) cadmium chloride and 5%(w/w) plant powder-Ssook(Artemisia princeps var. orientalis HARA), Chamchwi(Aster scaber THUNB), Gomchwi(Lingularia fischeri TUBCZ). or Solbeerum (Portulaca oleracea LINNE). Food intake, weight gain, food efficiency ratio, and weights of liver, kidney, spleen and epididymal fat were significantly lower in Cd-exposed groups, food efficiency ratio, and weights of liver, kidney, spleen and epididymal fat were significanlty lower in Cd-exposed groups. Plasma total lipid level, liver total lipid, cholesterol and triglycerde concentrations, and fecal total lipid, cholesterol and triglyceride excretions were decreased by Cd administration. Total lipid and triglyceride concentration in plasma and liver were lower in Chamchwi groups together with increased fecal total lipid and triglyceride excretions. Cholesterol concentration in plasma and liver were lower in Gomchwi groups with increased fecal cholesterol excretions. Activities of red-blood-cell superoxide dismutase(SOD), and liver catalase, SOD and glutathione peroxidase (GSH-px) were decreased significantly by Cd administration. Thiobarbituric acid reactive substance(TBARS) concentrations of plasma and liver were increased by Cd administration. Thiobarbituric acid reactive substance(TBARS) concentration of plasma and liver were increased by Cd administration. Administration of plant diets decreased plasma and liver catalase, SOD and GSH-px activities in Cd-free diet groups and increased them in Cd-administered groups. Plasma and liver TBARS concentrations were decreased in animals fed plant diets, with Chamchwi showing the most effective antioxidative capacity. The concentration of Cd accumulated in blood and tissue decreased in Cd-exposed groups fed plant diets. Liver Cd concentration were lower in the Chamchwi and Gomchwi groups, and kidney Cd levels were lower in the Ssook and Chamchwi groups. Among them, Chamchwi was the most effective in Cd detoxification. In conclusion, Chamchwi and Gomchwi were effective in lowering tissue lipid levels, with Chamchwi having the greatest antioxidative and detoxifying effects.

  • This study was performed to investigate the effect of pork on the cadmium detoxification in rats. Ninety male Sprague-Dawley strains were divided into five stoups based on cadmium treatment(0, 25, 50, 100, 250 ppm) and cadmium intoxication was observed for 8 weeks. During following 8 weeks, casein was replaced by Pork and the effect of pork was compared with casein. During intoxicating, the growth mte was lessened. Feed efficiency ratio was significantly low, as cadmium intake was high Discontinuing cadmium feeding, the body weights were relieve4 Pork-fed groups seemed to have higher body weight than casein-fed groups. During intoxication, hemoglobin and hematocrit were dependent upon cadmium intake. However, they became normal at detocification stage. Cadmium accumulation of the pour-fed group was lower than that of casein. Especially, the factors which affected the cadmium content in kidney were Cd(p<0.001) and $Cd^*pork(p<0.001)$. Metallothionein(MT) was increased with cadmium, and MT was not likely to be affected by pork. This study may provide scientific background of pork for detoxification from heavy metals such as cadmium.

  • The just molted larvae of the fifth instar were fed on with cadmium trated mulberry leaf and on the third day of the fifth instar the daily change of the total protein, activities of GOT and GPT in larvae haemoymph were sexually analyzed. The result obtained are summarized as follows : 1. The total haemolymph was decreased by cadmium treatment. A decreasing ratio of the total haemolymph protein was higher at the later development stage of the male fifth instar than the female fifth instar. 2. There seemed a slight difference in albumin content of the female blood along with the fifth larvae development between control and cadmium treatment where 7% decrease of blood albumin took place with cadmium treatment comparing to that of cotrol. Contrarily a decrease of blood globulin was made on both sexes. 3. The activities of GOT and GPT were inhibited by cadmium treatment.

Effect of Dietary Calcium, Casein, and Suet on the Accumulation of Cadmium in Mice (생쥐에 있어서 카세인, 칼슘 및 우지가 카드뮴의 조직내 축적에 미치는 영향)

  • This study was carried out to clarify the effect of dietary calcium, casein, and suet on the accumulation of cadmium in mice. It was performed for 30 days, from April 11 to May 10 1988.90 mice were divided into 4 experimental groups and control group with 6 mice each dietary group, and measured survival rate, body weight, and weight ratio of organ to body. The contents of cadmium in liver, kidney, spleen, muscle and skin with hair, and feces were analyzed by atomic absorption spectrophotometer after sacrifice by anesthesia. After 30 days, the survival rate of control group was loot,, but 66.7% in group IV(basal diet+Cd+Ca) with single dose of $100{\mu}g$ cadmium and with free-intake of water containing 50ppm cadmium, and group V(basal diet+Cd+suet) with free-intake of water containing 50ppm cadmium. The rate of weight gain in the case of single dose of $100{\mu}g$ cadmium was highest in group IV as 42.3% and lowest in group V as 26.0%, whereas in the cases of free intake group W was highest as 24.0% and group II(basal diet+Cd) was lowest as 11.6%. The body weight, in the case of single dose of $100{\mu}g$ cadmium showed no increase until 5th day after acute poisoning. But in the case of free intake group, it showed very slight increase through all the breeding period. The weight ratio of organ to body were lowest in the liver of group II in both occasions. The most of cadmium adminstered. were excreted through feces within 2 days after single dose of $100{\mu}g$ cadmium. The contents of cadmium in each group were significantly higher than those of control group. In the liver, kidney, spleen and muscle, group II showed the highest level in both occasions of $100{\mu}g$ single dose and free intake of water containing 50ppm cadmium. In the skin with hair, group ll of the occasion with single dose of $100{\mu}g$ and group V with free intake showed the highest level. And the contents of cadmium in tissue were markedly higher in the occasion of free intake of water containing 50ppm cadmium. From the above results I would conclude that the addition of casein and calcium are effective in the inhibition of intesitnal absorption of cadmium esp. by calcium.

  • Emulsion liquid membrane (ELM) process suffers from emulsion instability problem. So far, emulsion produced by mechanical methods such as stirrer and homogenizer has big size and high emulsion breakage. This paper discussed the application of emulsion produced by sonicator to extract cadmium in a batch ELM system. The emulsions consist of N,N-Dioctyl-1-octanamine (trioctylamine/TOA), nitrogen trihydride (ammonia/NH4OH), sorbitan monooleate (Span 80), and kerosene as carrier, stripping solution, emulsifying agent, and organic diluent, respectively. Effects of comprehensive parameters on extraction efficiency of Cd(II) such as emulsification time, extraction time, stirring speed, surfactant concentration, initial feed phase concentration, carrier concentration, volume ratio of the emulsion to feed phase, and pH of initial feed phase were evaluated. The results showed that extraction efficiencies of Cd(II) greater than 98% could be obtained under the following conditions: 15 minutes of emulsification time, 4 wt.% of Span 80 concentration, 4 wt.% of TOA concentration, 15 minutes of extraction time, 250 rpm of stirring speed, 100 ppm of initial feed concentration, volume ratio of emulsion to feed phase of 1:5, and initial feed pH of 1.53.

Adsorption of Cadmium Ion by Wood Charcoal Prepared with Red oak (Quercus mongolica) (신갈나무 목탄의 카드뮴(Cd)이온 흡착 특성)

  • For investigation of adsoption properties of cadmium elimination by wood charcoal, $25m{\ell}$ aqueous cadmium solutions in various concentrations were treated with 0.2 g wood charcoal of Red Oak (Quercus mongolica) for 280 minutes. Almost 100% of cadmium elimination ratio was obtained in the solutions with initial concentration of 20 and 40 ppm in the treatment, whereas they were 75 and 50% in those of 80 and 160 ppm. In the effect of treatment time, the highest amount of cadmium ions was eliminated during the first ten minutes in each solution so that the elimination ratio of each case was over 70% of the maximum elimination value. From the analysis of adsorptive cadmium adsorption mechanism using the Langmuir adsorption isotherm, it was suggested that cadmium ion molecules were adsorbed at the active sites on the charcoal particle in form of one layer. The Gibbs free energy of the adsorption process was calculated in negative value for each solution. This means the adsorption processes are spontaneous which do not require the extra input energy.
