Circulating Cell-free miRNA Expression and its Association with Clinicopathologic Features in Inflammatory and Non-Inflammatory Breast Cancer

  • Recent discovery showing the presence of microRNAs (miRNAs) in the circulation sparked interest in their use as potential biomarkers. Our previous studies showed the diagnostic potential of miR-451 as a serological marker for inflammatory breast cancer (IBC), miR-337-5p and miR-30b for non-inflammatory breast cancer (non-IBC). The aim of this study is to investigate the prognostic values of circulating miRNAs by comparing the amounts of 12 circulating miRNAs in the serum of IBC and non-IBC from Tunisian breast cancer patients, and by determinating whether correlated pairs of miRNAs could provide useful information in the diagnosis of IBC and non-IBC patients. TaqMan qPCR was performed to detect circulating expression of miRNAs in serum of 20 IBC, 20 non-IBC and 20 healthy controls. Nonparametric rank Spearman rho correlation coefficient was used to examine the prognostic value of miRNAs and to assess the correlation profile between miRNAs expression. Further, a large number of miRNAs were highly correlated (rho>0.5) in both patients groups and controls. Also, the correlations profiles were different between IBC, non-IBC and healthy controls indicating important changes in molecular pathways in cancer cells. Our results showed that miR-335 was significantly overexpressed in premenopausal non-IBC patients; miR-24 was significantly overexpressed in non-IBC postmenopausal patients. Patients with previous parity had higher serum of miR-342-5p levels than those without. Furthermore, patients with HER2+ IBC present lower serum levels of miR-15a than patients with HER2-disease. Together, these results underline the potential of miRNAs to function as diagnostic and prognostic markers for IBC and non-IBC, with links to the menopausal state, Her2 status and parity.

A Study on High-Resolution Seasonal Variations of Major Ionic Species in Recent Snow Near the Antarctic Jang Bogo Station (남극 장보고과학기지 인근에서 채취한 눈시료 내의 주요 이온성분들의 고해상도 계절변동성 연구)

  • A continuous series of 60 snow samples was collected at a 2.5-cm interval from a 1.5-m snow pit at a site on the Styx Glacier Plateau in Victoria Land, Antarctica, during the 2011/2012 austral summer season. Various chemical components (${\delta}D$, ${\delta}^{18}O$, $Na^+$, $K^+$, $Mg^{2+}$, $Ca^{2+}$, $Cl^-$, $SO_4{^2-}$, $NO_3{^-}$, $F^-$, $CH_3SO_3{^-}$, $CH_3CO_2{^-}$ and $HCO_2{^-}$) were determined to understand the highly resolved seasonal variations of these species in the coastal atmosphere near the Antarctic Jang Bogo station. Based on vertical profiles of ${\delta}^{18}O$, $NO_3{^-}$and MSA, which showed prominent seasonal changes in concentrations, the snow samples were dated to cover the time period from 2009 austral winter to 2012 austral summer with a mean accumulation rate of $226kgH_2Om^{-2}yr^{-1}$. Our snow profiles show pronounced seasonal variations for all the measured chemical species with a different pattern between different species. The distinctive feature of the occurrence patterns of the seasonal variations is clearly linked to changes in the relative strength of contributions from various natural sources (sea salt spray, volcanoes, crust-derived dust, and marine biogenic activities) during different short-term periods. The results allow us to understand the transport pathways and input mechanisms for each species and provide valuable information that will be useful for investigating long-term (decades to century scale periods) climate and environmental changes that can be deduced from an ice core to be retrieved from the Styx Glacier Plateau in the near future.

Systematic approaches to identify functional genes using the FOX-hunting system in Chinese cabbage (FOX hunting system을 이용한 배추 기능유전자 탐색)

  • Full-length cDNAs are essential for the correct annotation of genomic sequences and for the functional analysis of genes and their products. To elucidate the functions of a large population of Chinese cabbage (Brassica rapa) genes and to search efficiently for agriculturally useful genes, we have been taking advantage of the full-length cDNA Over-eXpresser (FOX) gene hunting system. With oligo dT column it purify the each mRNA from the flower organs, leaf and stem tissue. And about 120,000 cDNAs from the library were transformed into $\lambda$-pFLCIII-F vector. Of which 115,000 cDNAs from the library were transformed into T-DNA binary vector, pBigs for transformation study. We used normalized full-length cDNA and introduced each cDNA into Arabidopsis by in planta transformation. Full-length Chinese cabbage cDNAs were expressed independently under the CaMV 35S promoter in Arabidopsis. Selfed seeds were harvested from transgenic Arabidopsis. We had selected 2,500 transgenic plants by hygromycin antibiotic tolerant test, and obtained a number of transgenic mutants. Each transgenic Arabidopsis was investigated in morphological changes, fertility and leaf colour. As a result, 285 possible morphological mutants were identified. Introduced cDNA was isolated by PCR amplification of the genomic DNA from the transgenic mutants. Sequencing result and BLAST analysis showed that most of the introduced cDNA were complete cDNAs and functional genes. Also, we examined the effect of Bromelain on enhancing resistance to soft rot in transgenic Chinese cabbage 'Osome'. The bromelain gene identified from FOX hunting system was transformed into Chinese cabbage using Agrobacterium methods. Transformants were screened by PCR, then RT-PCR and real time PCR were performed to analyze gene expression of cysteine protease in the T1 and T2 generations. The anti-bacterial activity of bromelain was tested in Chinese cabbages infected with soft rot bacteria. The results showed that the over-expressed bromelain gene from pineapple conferred enhanced resistance to soft rot in Chinese cabbage.

Effect of Glucosylceramides and Sterols Isolated from Agaricus Blazei Extract on Improvement of Skin Cell (신령버섯에서 분리된 Glucosylceramide 및 Sterol의 피부 세포 개선 효과)

  • Agaricus blazei Murill (Almond mushroom) has many beneficial effects, such as anti-cancer, immuneenhancement, and anti-obesity. Also, its skin benefits have been reported for antioxidant, anti-inflammatory, and whitening. In order to elucidate these effects, many studies have been conducted. In this study, we reconfirmed the skin efficacy of the extract of the mushrooms mushrooms. The Agaricus blazei extract showed inhibition of melanin synthesis, enhancement of collagen synthesis, and up-regulation of gene expression (hyaluronan syntahase-2, 3 and aquaporin-3) at 100 ㎍/mL. and identified the ingredients from the extract. We further investigated them to find an applicability as cosmetic ingredients. The ingredients were confirmed comparison of their spectroscopic data with literature values. They were identified as being ergosterol (1), 5-dihydroergosterol (2), cerevisterol (3), cerebroside B (4), cerebroside D (5), adenosine (6), and benzoic acid (7). Among these compounds, we evaluated skin efficacy for two cerebrosides and three ergosterol derivatives that have not been reported its efficacy. As a result, 5-dihydroergosterol (2) inhibited melanogenesis in B16F10 and promoted collagen biosynthesis in human dermal fibroblast. In addition, cerevisterol (3), cerebroside B (4), and cerebroside D (5) inhibited NO production in RAW 264.7 cell. In particular, cerebroside D (5) increased the expression of hyaluronan synthase-2 and aquaporin-3 genes in HaCaT. These results suggest that Agaricus blazei extract and isolated compounds can be used as cosmetic ingredients.

Crystal Structure of Fully Dehydrated Partially Cs+-Exchanged Zeolite X, Cs52Na40-X (The Highest Cs+-Exchanged Level Achieved by Conventional Method and Confirmation of Special Site Selectivity)

  • The crystal structure of fully dehydrated partially Cs+-exchanged zeolite X, [Cs52Na40Si100Al92O384], a = 24.9765(10) A, has been determined by single-crystal X-ray diffraction techniques in the cubic space group Fd3 at 21 °C. The crystal was prepared by flow method for 5 days using exchange solution in which mole ratio of CsOH and CsNO3 was 1 : 1 with total concentration of 0.05 M. The crystal was then dehydrated at 400 °C and 2 × 10-6 Torr for 2 days. The structure was refined to the final error indices, R1 = 0.051 and wR2 (based on F2) = 0.094 with 247 reflections for which Fo > 4σ (Fo). In this structure, about fifty-two Cs+ ions per unit cell are located at six different crystallographic sites with special selectivity; about one Cs+ ion is located at site I, at the centers of double oxygen-rings (D6Rs), two Cs+ ions are located at site I', and six Cs+ ions are found at site II'. This is contrary to common view that Cs+ ions cannot pass sodalite cavities nor D6Rs because six-ring entrances are too small. Ring-opening by the formation of ?OH groups and ring-flexing make Cs+ ions at sites I, I', and II' enter six-oxygen rings. The defects of zeolite frameworks also give enough mobility to Cs+ ions to enter sodalite cavities and D6Rs. Another six Cs+ ions are found at site II, thirty-six are located at site III, and one is located at site III' in the supercage, respectively. Forty Na+ ions per unit cell are located at two different crystallographic sites; about fourteen are located at site I, the centers of D6Rs and twenty-six are also located at site II in the supercage. Cs+ ions and Na+ ions at site II are recessed ca. 0.34(1) A and 1.91(1) A into the supercage, respectively. In this work, the highest exchange level of Cs+ ions per unit cell was achieved in zeolite X by conventional aqueous solution methods and it was also shown that Cs+ ion could pass through the sixoxygen rings.

Anti-oxidative Activities for the Flavonoids of the Syzygium aqueum Burm.f. Alston Branches from Jeju Island (제주 자생 물사과 가지 유래 Flavonoid 화합물의 항산화 활성)

  • In this study, we investigated the anti-oxidative activities and cell protective effects of the constituents isolated from S. aqueum branches. DPPH and $ABTS^+$ radical scavenging activities were screened for the ethanol extract and solvent fractions, ethyl acetate (EtOAc) and butanol (BuOH) fractions showed potent activities. When HaCaT cells were treated with $H_2O_2$, the ethanol extract and EtOAc fractions ($20{\mu}g/mL$) protected the cells against oxidative damage. Two constituents were isolated from the EtOAc fraction of S. aqueum branches; pinocembrin (1), desmethoxymatteucinol (2). The chemical structures of the isolated compounds were elucidated based on the spectroscopic data including $^1H$ and $^{13}C$ NMR spectra, as well as comparison of the data to the literature values. Anti-oxidative activities and cell protective effects were studied for the isolated compounds. For the anti-oxidative activities, all of the compounds 1 and 2 showed DPPH and $ABTS^+$ radical scavenging activities. Also, from the cell protective effect test, the compounds 1 and 2 protected the cell against oxidative stress by $H_2O_2$. Based on these results, S. aqueum branches extract could be potentially applicable as anti-oxidant ingredients in cosmetic industries.

Mineralogy, Geochemistry, and Evolution of the Mn-Fe Phosphate Minerals within the Pegmatite in Cheolwon, Gyeonggi Massif (경기육괴 철원지역 페그마타이트 내 망간-철 인산염광물의 광물-지화학적 특징 및 진화과정)

  • Mn-Fe phosphate mineral complexes included within the pegmatite are observed at Jurassic Cheolwon two-mica granite in Gyeonggi Massif, South Korea. The genetic evolution between the Cheolwon two-mica granite and pegmatite, and various trend of Mn-Fe phosphate minerals is made by later magmatic, hydrothermal, and weathering process based on mineralogical, geochemical analysis. The Cheolwon two-mica granite is identified as S-type granite, considering its chemical composition (metaluminous ~ peraluminous), post-collisional environment, low magnetic susceptibility, and existence of biotite and muscovite. The K-Ar age (ca. 153 Ma) of pegmatite is well coincident with age of the Cheolwon two-mica granite ($151{\pm}4Ma$). It indicates that these two rocks are originated from the same magma. Pegmatite indicates the LCT geochemical signature, and was classified as muscovite-rare element class / Li subclass / beryl type / beryl-columbite-phosphate subtype pegmatite. The triplite $\{(Fe^{2+}{_{0.4}},Mn_{1.6})(PO_4)(F_{0.9})\}$ is dominant phosphates in later magmatic stage which partly altered to leucophosphite $\{KFe^{3+}{_2}(PO_4)_2OH{\cdot}2H_2O\}$ and jahnsite $\{(Fe^{3+}{_{0.7}},Mn_{2.3})(PO_4)_2OH{\cdot}4H_2O\}$ by hydrothermal alteration. In particular, near fractures, the triplite has been separatelty replaced by the phosphosiderite ($Fe^{3+}PO_4{\cdot}2H_2O$) and Mn-oxide minerals during weathering stage.

3D-QSAR Analysis on the Insecticidal Activities of N-Substituents on Imidazol Ring in Imidacloprid Analogues (Imidacloprid 유도체 중 imidazol 고리상 N-치환체들의 살충활성에 대한 3D-QSAR 분석)

  • CoMFA and CoMSIA model were derived and reviewed on the insecticidal activities of N-substituents (X) on the imidazol ring in imidacloprid analogues at the different alignment condition. Regarding the predictability ($q^2$ or $r_{cv.}^2$) and fitness ($r_{ncv.}^2$) of the two optimized models, the atom based fit (A) alignments were better than that of the field fit (F) alignment and, on the other hand, CoMSIA (A10) model was better than CoMFA (A5) model. Also, from the most optimized CoMSIA (A10) model, the insecticidal activity by N-substituents (X) was dependence on the electrostatic field and H-bond acceptor field. It is predicted that, from the contour maps with optimized CoMSIA (A10) model, H-bond acceptors at ortho- and meta- position will contribute for improving of insecticidal activities and, as the functional groups of carbonyl oxygen atom are charged negatively and positively charged at the ortho- position of benzyl group, insecticidal activities will also be improved.

Removal of Cobalt Ions by Precipitate Foam Flotation (침전 포말부선법에 의한 Cobalt Ion의 제거)

  • Simulated waste liquid containing 50 ppm cobalt ion was t$\xi$sted by precipitate flotation using a sodium lauryl sulfate as a c collector. The effects of initial cobalt ion concentration, pH, surfactant concentration, flotation time, gas flow rate and foreign i ions on removal efficiency of cobalt ion were studied. Pretreatment of the waste liquid with 35% $H_2O_2$, prior to precipitate f flotation made shin of optimal flotation pH from the strong alkalinity to weak alkaline range and made a favorable flotation of c cobalt ion in wide range of pH. For the result of this experiment, 99.8% removal efficiency was obtained on the conditions of initial coball ion concentration 50 ppm, pH 9.5 gas flow rate 70 mllmin, flotation time 30 min. The simulate ion was fanned t to be the most harmful ion against removal of cobalt by precipitate flotation of the species which were tested The presence of 0.1 M of $SO_4^{2-}$ ion decreased remo,때 $\xi$폐iciency of cobalt to 90% while the cobalt were almost entirely removed in the a absence of sulfate ion.

Co-treatment of Sewage Sludge and Cow Manure by Vermistabilization (Vermistabilization에 의한 하수 슬러지와 가축분뇨의 병합처리)

  • The objective of this study was to investigate the effect of cow manure as additive material on the ripening of sewage s sludge for vermistabiJization. The changes of the waste properties by ripening for 50days were observed as a function of the v various mixture ratios of sewage sludge ‘and cow manure. The pH values of the mixture wastes decreased from 7.5-7.67 to 6_ 9~7.2 by the ripening for 50days, and the mixture ratio made dIfferent pH values. The initial value of oxidation-reduction p potential (Eh) of the mixture waste was a negative (-) value indicating an unfavorable condition for earthworm after, but the v values of Eh increased with the opening time. The Increase rate at Eh value was prop$\alpha$rtional to the mixture ratio of cow m manure. The value of alkalinity was also changed into the favorable range for earthworm after 50days except for non-use of 1 the cow manure When the mixture ratio of the cow manure increased from 10% to 30%, the growth of earthworms increased h from 63.7% to 88.3 % tor the survival rate, 265% to 321% for the liveweight increasing rate and 66.7_7% to 91% for hatching f rate of the cocoons. It can be concluded that the proper content of tbe cow manure in the sewage sludge to ensure effective v vermistabilization was over 20%, when the mixture was ripened during 50 days. The quantity of ingestion and 며ectian at 20%-30% was found to be O.15--i.L18g sludge and 0 1l--O.14g solid per capacity earthworm per day, respectively.
