Processing and Taste Compounds of Fish Sauces from Filefish Scrap (말쥐치잔사(殘適)를 이용한 어간장 제조 및 제품의 정미성분)

  • In order to increase the availability of filefish scrap, the ordinary and low salt sauce were prepared, and identified their taste compounds in their products. To process the filefish scrap sauce, chopped filefish scrap was mixed with koji, 25% brine, slat and glucose (25.0 : 65.0 : 12.5 : 7.0, w/w) and fermented at $25{\pm}4^{\circ}C$ for 120 days. The same process was also carried out to process the low salt sauce adding sorbitol, lactic acid and ethyl alcohol (7.0 : 0.7 : 9.0. w/w) instead of salt. While amino nitrogen and volatile basic nitrogen(VBN) of products were decreased, pH and reducing sugar were increased all alone the fermentation period. The major free amino acids of products at final stage of fermentation were glutamic acid, alanine, leucine, lysine and aspartic acid. And the contents of total amino acid in the ordinary and low salt sauce were 4126.6(mg/100m1 sauce), 4519.5(mg/100m1 sauce) after fermentation. Hypoxanthine was revealed as the major constituent among nucleotides and their related compounds through fermentation. Free amino acid-N in the filefish scrap sauces were from 56.3%(ordinary) to 60.7%(low salted) of extractive nitrogen. From the sensory evaluation, the quality of products from filefish scrap sauce were almost equal to sold soy sauce on the market.

Variation of Oil Contents and Fatty acid Compositions in Korean Soybean Germplasms (국내 콩 유전자원의 지방함량 및 지방산 조성변이)

  • The objectives of this study was to determine the oil contents and fatty acid compositions of soybean germplasms including major Korean cultivars, and to provide the basic information of fatty acid composition for development high quality soybean varieties. Among 46 Korean major cultivated soybean samples, the oil contents were varied from 15.8% to 20.3%, and the average oil content was 18.2%. Crude oil content of cv. Saebyolkong (20.3%) was the highest among that of other cultivated soybeans. Likely many other oils of plant origin, most fatty acids in soybean are unsaturated. The highest percentage of fatty acid in soybean was linoleic acid (18:2), followed in a decreasing order by oleic (C18:1), palmitic (C16:0), linolenic (C18:3), and stearic acid (C18:0). The composition of C18:1 and C18:2 fatty acids among 563 soybean germplasms and cultivars which were positively correlated with nutritional quality of edible vegetable oil, were ranged $13.29{\sim}43.30%\;and\;38.21{\sim}61.51%$, respectively, and the C18:3 which were negatively correlated with flavor quality were varied from 5.03% to 11.48%. Also, the composition of C16:0 and C18:0 which were known to raise total cholesterol levels in human serum were ranged $8.50{\sim}14.78%\;and\;1.86{\sim}4.78%$ respectively. On the other hand, the range of fatty acid composition among 46 Korean major soybean cultivars has been investigated to be: C16:0, $8.96{\sim}13.23%$; C18:0, $2.55{\sim}4.20%$; C18:1, $16.33{\sim}36.41%$; C18:2, $42.32{\sim}58.84%$; C18:3, $6.98{\sim}10.72%$. Among the 563 germplasms tested, there are significant correlations among unsaturated fatty acids: negative between C18:1 and C18:2 (r=-0.961**) or C18:3 (r=-0.756**), and positive between C18:2 and C18:3 (r=0.608**). Also, the C18:1 was negatively correlated with saturated fatty acids, C16:0 (r=-0.231**) and C18:0 (r=-0.479**).

Analysis of Nutritional Compounds and Antioxidant Effect of Freeze-Dried powder of the Honey Bee (Apis mellifera L.) Drone (Pupal stage) (서양종 꿀벌(Apis mellifera L.) 수벌번데기 동결건조 분말의 영양학적 성분 및 항산화 효과)

  • In this study, we analyzed the nutritional ingredients of drone pupae (16th to 20th instar old) to evaluate the value of bee products and provide basic data for product diversification, and the extracts prepared using these pupae were tested for physiological activity. According to the analysis of the general ingredients of the freeze-dried powder of these bee pupae, the moisture, crude protein, crude fat, and crude ash was 1.69 ± 0.07%, 48.52 ± 0.20%, 23.41 ± 0.14%, and 4.05 ± 0.02%, respectively. Vitamin C and vitamin E were 14.92 ± 0.52 mg/100 g and 6.06 ± 0.11 mg α-TE/100 g, respectively. Regarding minerals, the highest content of K (1349.13 ± 34.57 mg/100 g) and P (1323.55 ± 43.85 mg/100 g) was observed and Ca and Fe were 55.43 ± 1.51 mg/100 g and 5.49 ± 0.19 mg/100 g, respectively. The fatty acids of the water extracted freeze-dried pupae powder accounted for approximately 59.62 of saturated fatty acids and 40.38 of unsaturated fatty acids, and high-quality fatty acids such as palmitic acid (C16:0) was 35.49 ± 0.08 and oleic acid (C18:1, n-9) was 35.91 ± 0.22 (g/100 g total fatty acids). The total amino acid content was 38.99 ± 2.63 g/100 g and the free amino acid was a total of 5129.04 mg/100 g, of which 1257.68 mg/100 g was proline and 759.12 mg/100 g glutamic acid. The DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity of the drone pupae extract showed values of 0.8 for distilled water extract, 3.2 for 50% EtOH extract, 6.4 for 70% EtOH extract, and approximately 90% for 32 ㎍/mL for 100% EtOH extract. These results suggest that the main compound contributing to the antioxidant activity is a polar compound, and it is highly likely to be a low-molecular protein or a free amino acid. In conclusion, the honey bee drone pupa is excellent as a food resource and can be utilized as a new functional material for food and functional food.

Effect of Dietary Grape Pomace Fermented by Saccharomyces boulardii on the Growth Performance, Nutrient Digestibility and Meat Quality in Finishing Pigs

  • Fifty-six [(Duroc${\times}$Yorkshire)${\times}$Landrace] pigs with an average initial BW of 19.3${\pm}$0.17 kg were used in this 15-wk growth experiment to investigate the effects of grape pomace fermented by Saccharomyces boulardii on pig growth performance, nutrient digestibility and quality attributes of pork. Pigs were allotted to 2 dietary treatments (7 replications) based on their initial BW in a randomized complete block design. The experimental treatments were: i) control (CON; basal diet), ii) FGPP (CON+30 g/kg fermented grape pomace product). Dietary FGPP improved (p<0.05) average daily gain (ADG), coefficient apparent total tract digestibility (CATTD) of dry matter (DM) and nitrogen (N) during 35-70 d of the experiment. Similarly, pigs fed the FGPP supplemented diet had a higher N digestibility (p<0.05) in the finisher phase (day 71-105). Dietary FGPP increased (p<0.05) the marbling score, the redness ($a^*$) and yellowness ($b^*$) values, as well as the anti-oxidative ability (lower TBARS). The inclusion of FGPP reduced palmitic acid (C:16:0), stearic acid (C:18:0), arachidic acid (C:20:0) and SFA levels (p<0.05) in subcutaneous fat. An increased (p<0.05) linoleic acid (C18:2n6), total PUFA and PUFA/SFA ratio were observed in the FGPP group. Dietary FGPP supplementation decreased the arachidic acid (C:20:0) level in longissimus muscle (LM). In conclusion, dietary inclusion of FGPP at the level of 30 g/kg improved the growth performance, nutrients digestibility and altered the fatty acid pattern in the subcutaneous fat as well as some attributes of pork meat.

식물성 유지의 phytosterol, tocopherol 분석 및 CLA를 함유한 재구성 지질의 합성

  • 식물종자 중 참깨, 호두, 통밀, 미강 등을 원료로 하여 8$0^{\circ}C$, soxhlet extractor에서 6시간 동안 추출하여 수율을 알아본 후, 추출된 유지의 지방산 조성을 분석하고, phytosterol 및 tocopherol의 함량을 비교.분석하였다. 추출결과, 호두가 56.11%로 가장 높은 수율을 보였으며, 그 뒤로 참깨, 미강, 통밀 등이 각각 35.65, 14.83, 1.56% 등을 나타내었다. GC에 의한 지방산 조성 분석 결과, 추출된 식물유지 모두 linoleic acid, oleic acid등이 각각 평균 45.19, 32.70% 등으로 가장 높은 비율을 차지하였으며, 그 뒤로 palmitic acid, linolenic acid, stearic acid 등이 각각 평균 14.97, 3.52, 2.95 ㏖% 등의 비율을 나타내었다. GC에 의한 phytosterol 분석결과, 참깨, 호두, 통밀, 미강 등의 총 phytosterol 함량은 평균 1.20%의 비율을 보였고, HPLC에 의한 tocopherol 분석결과, 총 tocopherol 함량은 평균 1.52%의 함량을 보이는 것으로 조사되었다. 한편, 추출된 식물성유지 4종과 Conjugated linoleic acid (CLA)를 각각 1:3 ㏖ 비율로 혼합한 후 고정화 효소인 IM60과 24시간 동안 반응하여 재구성지질을 합성하고 GC에 의해 지방산 조성을 분석하였다. 총 CLA의 함량이 가장 많은 것은 통밀로서 23.75㏖%를 나타내었고, 그 뒤로 미강, 호두, 참깨 등이 각각 19.15, 16.28, 13.46㏖% 등의 함량을 나타내는 것으로 조사되었다.

The Storage Stability of Semi-Salted and Dried Mackerel by Free-Oxygen Absorber (탈산소제에 의한 반염건고등어 저장중의 품질안정성)

  • The preservative effect of modified-atmosphere storage on the shelf-life of packed semi-salted and dried mackerel, Scomber japonicus, was examined. The semi-salted and dried mackerel fillets were packed in laminated plastic film bags (polyester/nylon/casted polypropylene: $12{\mu}m/15{\mu}m/60{\mu}m,\;15{\times}16cm$) filled with air (control, C), nitrogen gas (N), deoxygenized air (O) prepared by using free-oxygen absorber enclosed in the bag, in vacuum(V), and stored at $5^{\circ}C$. The quality of packed sample during the storage were examined in terms of viable cell counts of bacteria, thiobarbituric acid(TBA) value, perozide value(POV), volatile basic nitrogen(VBN), trimethylamine(TMA), adenosine triphosphate(ATP) and its related compounds and sensory evaluation. The results obtained are as follows: The pH of all the samples was in the range of $6.1{\pm}0.2$, and the contents of VBN and amino nitrogen of them increased during storage. In color values, L value(lightness) decreased while a and b values (red and yellow) revealed a tendency to increase during storage. The viable cell counts of the control sample(C) increased to $3.0{\times}10^6/g$ after 15 days storage but those of the other samples(V, N and O)were in the range of $2{\sim}6{\times}10^5/g$ after 20 days storage. The content of TMA increased during storage, but the histamine content showed a little change during storage and its content of all samples were less than 16 mg/100g. The inosinic acid(IMP) was rapidly degraded while inosine and hypoxanthine increased during storage. The TBA value of the control(C) reached a peak in 9 days and then decreased gradually while that of the sample(O) showed a little change during storage. The changes in POV of all the samples during storage showed a similar tendency to the TBA value. Fatty acid composition of raw mackerel consists of $35.6\%$ of saturated acid, $30.3\%$ of monoenoic acid and $34.2\%$ of polyenoic acid. The major fatty acid of the sample products were oleic acid($C_{18:1}$), palmitic acid($C_{16:0}$), docosahexaenoic acid($C_{22:6}$). The contents of polyenoic acid such as $C_{22:6},\;C_{20:5}$ decreased during storage while the other fatty acids showed a little change. From the results of sensory evaluation, the shelf-life of the control sample(C) was about 7 days and that of sample(V), (N) and (O) was about 15 days. It was concluded that deoxygenized atmosphere(free-oxygen absorber enclosed in the bag) was a good condition for preserving the quality of semi-salted and dried mackerel.

Changes of the blood chemistry, lipid and protein components in blood and liver tissue of the rat after oral combined administration of caffeine, iron and vitamin E (Caffeine, 철분 및 vitamin E 혼합투여시 rat의 혈액과 간조직내에서 혈액화학성분과 지질 및 단백질 구성성분의 변화)

  • This study was conducted to identify the effects of caffeine or combinations of caffeine and iron or vitamin E on the lipid and protein components or blood chemistry levels of the serum as well as the total homogenate, mitochondrial and microsomal fraction of the rat(Sprague-Dawley, female) liver. Chronic test were conducted to determine those effects. The chronic test was conducted by dividing rats into 5 groups according to the type of drugs and dosages administrated as follows; the control(group A), and group B was given 25mg/kg caffeine orally once daily for 30 days, group C was given 50mg/kg caffeine orally once daily for 30 days, group D was given 25mg/kg caffeine and orally ferric chloride once daily for 30 days and group E was given 25mg/kg caffeine and 25mg/kg vitamin E once daily for 30 days. The concentrations of glucose, urea nitrogen, uric acid, creatinine, total protein, albumin, A/G ratio, triglyceride, total cholesterol, HDL-cholesterol, free fatty acid, phospholipid as well as the activities of alanine aminotransferase(ALT), aspartate aminotransferase(AST) and alkaline phosphatase(ALP) were measured in the serum of each experimental groups. The concentrations of the carbonyl group and malondiaidehyde(MDA) and the patterns of the SDS-PAGE(Sodium Dodecyl Sulfate - Polyacrylamide Gel Electrophoresis) and fatty acid compositions in free fatty acids and phospholipids were analyzed to determine the oxidative damages and metabolic changes on the lipid and protein components in the serum, and total homogenate, mitochondrial and microsomal fractions of the rat liver. The results obtained from this study were summarized as follows; 1. Body weights of groups B, C, D and E were significantly decreased(p < 0.01) in comparison with that of the control in the chronic test. 2. The concentrations of serum glucose in groups B(124.5mg/dl), C(130.1mg/dl), D(122.1mg/dl), E(119.3mg/dl) were significantly higher(p < 0.01) in comparison to that of the control(101.5mg/dl). But, there were no significant differences in the concentrations of urea nitrogen, uric acid, creatinine, total protein, albumin and A/G ratio in comparison to that of the control. 3. The concentrations of total cholesterol and HDL-cholesterol in serum of groups B(69.6, 53.4mg/dl), C(73.0, 56.3mg/dl), D(68.9, 51.1mg/dl) and E(68.2, 51.3mg/dl) were significantly higher(p < 0.01) in comparison to that of the control(52.6, 38.8mg/dl). On the other hand, the concentrations of triglyceride in serum of groups B(45.0mg/dl), C(40.4mg/dl), D(33.8mg/dl) and E(47.2mg/dl) were significantly lower(p < 0.01) in comparison to that of the control(66.2mg/dl). There were no significant differences in the activities of ALT, AST and ALP in comparison to that of the control. 4. The concentrations of free fatty acid and phospholipid in serum of groups B(45.7, 154.4mg/dl), C(50.0, 167.2mg/dl), D(52.5, 148.4mg/dl) and E(41.1, 159.2mg/dl) were higher(p < 0.01) in comparison to that of the control(35.2, 125.3mg/dl). And the concentrations of the carbonyl group and malondialdehyde in serum of group D(1.82, 0.52nM/mg protein) were significantly higher(p < 0.01) in comparison to the control(1.53nM/mg protein). 5. The concentrations of carbonyl group in total homogenate, mitochondrial and microsomal fraction of group D(1.45, 0.94, 1.67nM/mg protein) were significantly higher (p < 0.01) in comparison to the control(1.16, 0.66, 1.27nM/mg protein). And the concentrations of malondialdehyde in the total homogenate, mitochondrial and microsomal fraction of group D(6.70, 6.10, 1.36nM/mg protein) were significantly higher(p < 0.01) in comparison to the control(5.17, 3.64, 0.68nM/mg protein). 6. As the analytical results of the fatty acid compositions of free fatty acid in serum, the proportions of stearic acid and arachidonic acid of groups B(16.52, 12.62%), C(17.52, 15.18%), D(19.73, 13.47%) and E(17.62, 13.28%) were significantly higher(p < 0.01) in comparison to the control(14.75, 7.88%), but the proportions of oleic acid and linoleic acid of groups B(12.97, 32.59%), C(10.88, 31.23%), D(12.37, 30.66%) and E(11.95, 32.41%) were significantly lower(p < 0.01) in comparison to the control(16.44, 35.12%). Otherwise, as the results of the fatty acid compositions of phospholipid in serum, the proportions of stearic acid and arachidonic acid of groups B(39.37, 16.39%), C(40.63, 17.83%), D(42.73, 15.39%) and E(39.16, 15.70%) were significantly higher(p < 0.01) in comparison to the control(37.74, 14.24%), but the proportions of oleic acid and linoleic acid of groups B(4.03, 14.38%), C(3.54, 12.38%), D(4.52, 11.68%) and E(4.29, 13.64%) were significantly lower(p < 0.01) in comparison to the control(5.53, 16.14%). 7. As the analytical results of the fatty acid compositions of free fatty acid in total homogenate, mitochondrial and microsomal fraction of liver, the proportions of oleic acid of groups B(7.8**, 8.73**, 6.88%) and C(6.89**, 7.75**, 6.58%) were lower(**:p < 0.01) in comparison to the control(8.67, 10.08, 7.81%), but the proportions of arachidonic acid of group C(22.62, 19.79, 23.71%) were significantly higher(p < 0.01) in comparison to the control(20.93, 18.47, 22.24%). And the proportions of palmitic acid of group D(25.95**, 26.16, 26.34**%) were significantly higher(**:p < 0.01) in comparison to the control(24.43, 25.42, 23.34%). In addition, the proportions of linoleic acid of group D(23.43, 25.02, 23.95%) were also significantly higher(p < 0.01) in comparison to the control(22.17, 23.75, 21.26%). The proportions of stearic acid of group D(19.87, 19.76**%) in mitochondrial and microsomal fraction were lower(**:p < 0.01) in comparison to the control(21.01, 24.18%), and the proportions of stearic acid of group E(16.71*, 19.65**%) in mitochondrial and microsomal fraction were significantly lower(**:p < 0.01, *:p < 0.05) in comparison to the control(21.01, 24.18%), and the proportions of linoleic acid of group E(25.04, 29.20, 26.48%) in total homogenate, mitochondria and microsome were significantly higher(p < 0.01) in comparison to the control(22.17, 23.75, 21.26%). 8. As the results of the fatty acid compositions of phospholipid in total homogenate, mitochondrial and microsomal fraction of liver, the proportions of palmitic acid of group D(17.58**, 18.78*, 18.23%**) were significantly higher(**:p < 0.01, *:p < 0.05) in comparison to the control(16.28, 17.22, 16.38%), and the proportions of stearic acid of group D(36.41, 37.23, 39.53%) were also significantly higher(p < 0.01) in comparison to the control(34.18, 34.16, 36.04%). But the proportions of oleic acid(3.41*, 3.11**, 3.12**%) and linoleic acid (18.03**, 15.79**, 14.74**%) of group D were significantly lower(**:p < 0.01, *:p < 0.05) in comparison to the control(oleic : 3.63, 3.72, 3.79%, linoleic : 20.03, 18.71, 18.48%). 9. In order to determine the oxidative damages to the protein in serum, mitochondrial and microsomal fraction of the rat liver, the patterns of the SDS-PAGE were identified, but the results of SDS-PAGE were not significantly different between the control and experimental groups.

Effects of the Mating System on Retail Cut Yield and Meat Quality in Commercial Pigs (비육돈의 교배조합이 부분육 생산수율 및 육질에 미치는 영향)

  • This study was conducted to investigate the effect of the mating system on the retail cut yield and meat quality in pigs. A total of 210 samples were used to estimate meat productivity and meat quality two blocks of two boarlines Duroc (D) and Berkshire (B) were mated to sows of the sowlines Yorkshire×Landrace (YL) and Berkshire (B). Results showed that YLD (YL♀×D♂) and BD (B♀×D♂) had higher percentages of Boston butt and belly than YLB while it has higher percentages of lean meat yield than YLB (YL♀×B♂). There were no significant differences in crude protein and ash content of pork from different breeds (P>0.05). However, YLB and YLD had higher moisture content in loin and Boston butt than BD (P<0.05). The pH of BD was highest in pork loin and belly (P<0.05). There differences in meat yield, pH and moisture content among the genotypes suggested that pork quality might be affected by the lightness and cooking loss. BD was lowest lightness value (CIE L*) and cooking loss in pork loin among the genotypes (P<0.05). The percentage of saturated palmitic acid (C16:0) in Boston butt (P<0.05) of BD had significant lower, while the percentage of unsaturated palmitoleic (C16:1) and oleic acid (C18:1) had higher in pork loin and belly (P<0.05). Drip and marbling score were found significantly different in genotypes and BD had lower drip and higher marbling score in pork loin and belly (P<0.05). The sensory evaluations indicated that the highest overall acceptability in BD was attained. From the results, BD breed had highest meat quality among crossbred pigs.

Analysis of Food Components of Korean Deer Antler Parts (국내산 녹용의 부위별 식품학적 성분 분석)

  • Physicochemical properties of freeze-dried upper and middle parts of Korean raw anther were investigated and analyzed. Moisture contents of upper part and middles part were 51.7 and 63.6%, respectively. Contents of crude protein, crude fat, crude ash, and carbohydrates of upper part of antler were 66.91, 3.52, 22.70, and 6.87%, respectively. Whereas those of middle part were 52.64, 2.50, 34.54, and 6.07, respectively. Phosphorous content was the highest in both upper (2562.0 mg%) and middle (2536.0 mg%) parts. Glycine (7597.9 mg%) and glutamic acid (5769.4 mg%) were major amino acids in upper part, glycine (9612.0 mg%) and proline (5906.3 mg%) in middle part were the major amino acids. Free sugars including fructose, glucose, sucrose, and maltose were not detected. Palmitic acid (upper part 30.83%, middle part 32.23%) was the major fatty acid in crude fat of both upper and middle parts. Vitamin A (retinol) contents of upper and middle parts were 16.40 and 12.32 IU/100 g, respectively. Vitamin C was not detected. Upper part of antler showed the highest redness (8.18), whereas middle part showed the highest lightness (71.72) and yellowness (8.13). The color difference $({\Delta}E\;27.11)$ between upper part and middle part was distinguished remarkably.

Expression Analysis of Anthocyanin Biosynthetic Genes of Tassel and Silks in Gwangpyeongok and Dacheongok (광평옥과 다청옥의 수이삭과 수염에서 안토시아닌 생합성 유전자 발현 분석)

  • Anthocyanins are known to be involved in various functions such as antioxidant and antibacterial activities in plants. Although studies on anthocyanins in corn have been conducted recently, basic research related to anthocyanin biosynthesis is insufficient. In this study, we examined the molecular biological and physicochemical properties related to anthocyanin biosynthesis in the tassel and silks of Gwangpyeongok and Dacheongok cultivars. Anthocyanins were not synthesized in either the tassel or silks in Gwangpyeongok, whereas were synthesized in both in Dacheongok. The total anthocyanin content was approximately 30 times higher in the tassel and silks of Dacheongok than in those of Gwangpyeongok. In addition, C-3-G was measured only in the tassel of Dacheongok, and C-3-G, Pg-3-G, and M-3-G were 45.2 times, 27.3 times, and 37.6 times higher, respectively, in the silks of Dacheongok than of Gwangpyeongok. Expression of F3'H, DFR, and GST genes decreased in the tassel, and that of F3'H and DFR genes decreased in the silks of Gwangpyeongok. It was further confirmed that transcription factor P1 and R1 regulate the expression of anthocyanin biosynthetic genes in the tassel and silks, respectively, in Gwangpyeongok. Linoleic acid (C18:2) decreased by 6.6% and 10.9%, and linolenic acid (C18:3) increased by 8.5% and 8.5%, in the tassel and silks, respectively, of Gwangpyeongok compared to those of Dacheongok. Palmitic acid (C16:0) increased by 4.1% and oleic acid (C18:1) decreased by 2.1% in the silks of Gwangpyeongok compared to that in Dacheongok. In addition, the total fatty acid content in the tassel and silks increased by 10.3% and 30.4%, respectively, in Gwangpyeongok compared to that in Dacheongok. However, no significant results were observed in the analysis of phytosterol components. These results may be utilized as useful resources for the development of functional corn containing a large amount of anthocyanins.
