Blue Mold of Persimmon (Diospyros kaki) Caused by Penicillium crustosum (Penicillium crustosum에 의한 감 푸른곰팡이병 발생)

  • A severe fruit rot of Persimmon (Diospyros kaki cv: Fuyu) was occurred during the storage and transport that infected with blue mold in Sweet Persimmon Experiment Station, Gyeongsangnam-do Agricultural Research and Extension Services, Korea. Fruit surfaces were infected with the fungus first and the colonized fungus formed mycelial mats. From the point of infection, fruits become collapsed and mostly ruptured. The pathogenic fungus from infected fruits was isolated and cultured on PDA. Colony color of the fungus was white at frist than became green on Malt Extract Agar and Czapek Yeast Extract Agar. Conidia were ellipsoid subglobose and 2.6${\sim}$3.8 ${\times}$ 2.4${\sim}$3.8 ${\mu}m$ in size. Stipes were 86${\sim}$320 ${\times}$ 2.8${\sim}$4.3 ${\mu}m$ in size. Rami were 7.5${\sim}$32.6 ${\times}$ 2.6${\sim}$4.2 ${\mu}m$ in size, Ramuli were 12.4${\sim}$14.8 ${\times}$ 3.2${\sim}$3.8 ${\mu}m$ in size, Metulae were 8.9${\sim}$13.6 ${\times}$ 2.8${\sim}$4.6 ${\mu}m$ in size. Phialides were ampulliform, 8.2${\sim}$12.4 ${\times}$ 2.3${\sim}$3.6 ${\mu}m$ in size. Based on the cultural and mycological characteristics and pathogenecity test on host plants, the fungus was identified as s, This is the first report on the blue mold of Persimmon (Diospyros kaki) caused by P. crustosum in Korea.

Effect of Watering Methods on Growth of Soybean Sprout and Culture Temperature (관수방식에 따른 콩나물의 생장과 재배용기 내의 온도 변화)

  • Watering methods for soybean sprouts could be mainly divided into two groups of overspraying and underwatering. The study was carried out to determine the effect of water supplying method on growth, morphological characteristics, colour and cutting resistance of soybean (cv. Junjery) sprouts and culture temperatures. The morphological characters, fresh and dry weights were measured on the 6th day after their culture, but daily mean temperatures inside the plastic culture boxes were measured by data-loggers. Lateral roots were more formed in the underwatering method (UM) than in the overspraying method (OM). Although their total lengths of both methods were nearly same, OM had longer hypocotyl but UM did longer root than the other. Middle and upper parts of hypocotyl were more thickened in UM than in OM. UM showed more hypocotyl fresh and dry weights than OM. There was, however, no significant difference between the two methods in cotyledon, root, total fresh and dry weights although the culture temperature was higher in OM than in UM.

Effects of Storage Conditions on Quality Change after Storage of Deastringencied Persimmons (저장조건이 탈삽감의 저장 후 품질변화에 미치는 영향)

  • Effects of storage method and duration on the post-storage quality changes of deastringencied persimmons were investigated. Astringent persimmons(Diospyros kaki T. cv. Chungdobansi) were treated for 39 hours in 70% CO$_2$+5% O$_2$ at 25 $\^{C}$ for astringency removal, and then stored in CA(5%CO$_2$+3%O$_2$, 8%CO$_2$+3%O$_2$), MAP(0.06 mm LDPE film, 5.2%CO$_2$+6.8%O$_2$) and air at 0$\^{C}$ for 15, 45, 75 arnd 105 days. Subsequent to the storage, persimmons were held in air at 10$\^{C}$ for 4 days. The difference of firmness loss in relations to storage methods at O$\^{C}$ during holding at 10$\^{C}$ observed in fruits stored above 45 days at 0$\^{C}$, CA(8%CO$_2$+3%O$_2$) storage was more retarded the firmness loss than the others. Soluble solids during holding at 10$\^{C}$ trended to decrease in fruits stored above 75 days at 0$\^{C}$, CA and MAP storage were a little effective to keeping soluble solids. Titratable acidity during holding at 10$\^{C}$, decreased in fruits stored until 75 days at 0$\^{C}$ regardless storage methods. Sensory characteristics, color, hardness and overall acceptability of fruits stored in CA were significantly higher than those of fruits stored in MAP and air during shelf-life at 10$\^{C}$ after long-term storage at 0$\^{C}$ . These results show that the storage conditions had residual effect on the quality of deastringencied persimmons during shelf-life after long-term storage.

Role of Mesophyll Morphology in Determination of Leaf Photosynthesis in Field Grown Soybeans (포장생육대두의 엽광합성과정에서 엽육세포 형태의 역할)

  • Photosynthetic variation in field grown soybean [Glycine max (L.) Merr. cv Hodgson78] was studied in relation to leaf anatomical variation. Variations in mesophyll morphology were accentuated by manipulating source and sink size. At R3 stage, two treatments were started: one was thinning and continu-ous debranching(6. 5 plants rather than 26 plants per m of row and remaining plants were debranched weekly), and the other was continuous partial depodding (allowing only one pod to develop at each mainstem node). Gas exchange characteristics, mesophyll cell volume and surface area per unit leaf surface, and microclimatic parameters were measured on the intact terminal leaflet at the 10th node. Observations were made 5 times with 3 to 4 day intervals starting R4 stage. Two models were used to compute leaf photosynthetic rates: one considered no effect of mesophyll morphology on photosynthesis, and the other considered potential effects of variations in mesophyll cell volume and surface area on diffusion and biochemical processes. Seventy nine percent of total photosynthetic variations observed in the experiment was explained by the latter, while 69% of the same variations was explained by the former model. By incorporating the mesophyll morphology concept, the predictability was improved by 14.6% in the field condition. Additional Index Words: photosynthesis model, leaf anatomy, Glycine max (L.)   • This study was conducted in to elucidate the changes of growth characteristics and photosynthesis in three soybean (Glycine max L. cv. Danwonkong, Hwangkeumkong and Kwangankong) 30 day old seedlings to 100mM NaCl concentration containing 1/2 Hoagland's nutrient solution in sand culture. The main stem height and number of main stem node were decreased. thus, leaf area and dry matter were decreased with 100mM NaCl. Growth reduction was less little in Hwangkeumkong than other cultivars. The stem growth rate was affected less than other parts as root or leaf, by NaCl treatment. The specific leaf area (SLA), shoot : root ratio and leaf : root ratio decreased with NaCl solution except for those of Hwangkeumkong. There is no general tendency in leaf thickness by leaf position of three cultivars. The chlorophyll content (SPAD) of the primary and 2nd leaf slightly decreased under NaCl solution but rapidly increased in non-NaCl solution at 15 days after treatments. The photosynthesis, stomatal conductance and transpiration of 2nd leaf positions reduced by NaCl and there were a sigificant correlation between photosynthesis and stomatal conductance or transpiration.

Blue Mold on Melon (Cucumis melo) Caused by Penicillium oxalicum (Penicillium oxalicum에 의한 멜론 푸른곰팡이병)

  • In April of 2002, fruit rot infected with blue mold was found at maturing stage of melon (Cucumis melo cv. Gayabaegja) growing under tunnel cultivation in Daesan-myon, Haman-gun, Gyeongnam Province, Korea. Floral parts were infected first and colonized by fungal mycelial mats. From the point of infection, fruits become collapsed and mostly ruptured. The pathogenic fungus from infected fruits was isolated. Colony color of the fungus was white on MEA and CYA agar, Conidia were ellipsoid and 2.6~7.4$\times$2.6~5.8 ${\mu}{\textrm}{m}$ in size. Stipes were 86~320$\times$2.8~4.3 ${\mu}{\textrm}{m}$ in size. Metulae were 12.4~31.6$\times$2.6~4.2 ${\mu}{\textrm}{m}$ in size. Phialides were ampulliform to cylindroid, and 8.2~15.4$\times$3.6~4.6 ${\mu}{\textrm}{m}$ in size. Rate of infected fruits in the field was 4.3%. Based on the cultural and mycological characteristics and pathogenecity test on host plants, the fungus was identified as Penicillium oxalicum, This is the first report on the blue mold of melon (Cucumis melo) caused by P. oxalicum in Korea.

Occurrence of Blue Mold on Sweet Persimmon(Diospyros kaki) Caused by Penicillium expansum (Penicillium expansum에 의한 감 푸른곰팡이병 발생)

  • A fruit rot of sweet persimmon(Diospyros kaki cv. 'Fuyu') that infected with blue mold was found during the storage and transport in Jinju Gyeongnam Province, Korea. Fruit surfaces that infected with the fungus were formed water soaked lesion at first then gradually colonized with the fungus and formed mycelial mats. From the point of infection, fruits become sunken and mostly ruptured. The pathogenic fungus was isolated from infected fruits and cultured on potato dextrose agar. The colonies of the pathogenic fungi were white at frist then became greyish green on malt extract agar. Conidia were ellipsoidal and $2.6{\sim}3.8{\times}2.4{\sim}3.8{\mu}m$ in size. Phialides were ampulliform, verticilate of 3-7, $8.0{\sim}9.2{\times}2.0{\sim}3.0{\mu}m$ in size. Metulae were verticils of 2-4, smooth, $9.0{\sim}12.6{\times}3.0{\sim}4.6{\mu}m$ in size. Ramuli were groups 1-3, smooth, $11.0{\sim}17.6{\times}2.3{\sim}3.0{\mu}m$ in size. Rami were groups 1-2, $7.5{\sim}32.6{\times}2.6{\sim}4.2{\mu}m$ in size. Stipes were septate, smooth, thin walled, $56{\sim}302{\times}2.8{\sim}4.0{\mu}m$ in size. Penicilli were mostly quaterverticillate. Based on the cultural and mycological characteristics as well as pathogenicity test on host plants, the fungus was identified as Penicillium expansum. This is the first report on the blue mold of sweet persimmon(Diospyros kaki) caused by P. expansum in Korea.

Characteristics of Cucumber mosaic virus-VCH Causing Vein Chlorosis on Red Pepper in Korea (고추에 엽맥퇴록병을 일으키는 오이 모자이크 바이러스(CMV-VCH)의 특징)

  • Cucumber mosaic virus(CMV) was occurred on red pepper showing vein chlorosis or vein necrosis with the incidence rate of 52% from 62 specimens collected in natural fields. Among 32 samples infected with CMV, the specimens of 22 red pepper leaves showing vein chlorosis were infected singly with CMV-VCH. CMV-VCH induced vein chlorosis on the inoculated leaves, and vein banding and vein necrosis on the upper leaves of Nicotiana glutinosa, and then killed after showing bud necrosis. The typical symptoms of vein banding, malformation and blister were produced on the upper leaves of Nicotiana benthamiana and N. tabacum 'Ky-57' without symptoms on the inoculated leaves. The commercial cultivars of 'Bugang', 'Manitta' and 'Gwariput' were shown the typical symptom of vein chlorosis by the mechanical inoculation of CMV-VCH. CMV-VCH was detected specifically by RT-PCR. Virus particles of CMV-VCH were isometric shape having 30 nm diameter. Ultraviolet absorption of purified CMV-VCH was maximum at 260 nm and minimum at 242 nm. The ratio of A260/A280 was 1.71. CMV-VCH had the single nucleo-protein having the molecular weight of 24.5 kDa.

Characterization and Partial Nucleotide Sequence Analysis of Alfalfa Mosaic Alfamoviruses Isolated from Potato and Azuki Bean in Korea

  • Alfalfa mosaic alfamoviruses(AIMV) were isolated from infected potato (Solanum tuberosum) and azuki bean (Paseolus angularis) in Korea. Two AIMV isolated from potatoes were named as strain KR (AIMV-KR1 and KR2) and AIMV isolated from azuki bean was named as strain Az (AIMV-Az). Each isolated AIMV strain was characterized by using their host ranges, symptom developments, serological relations and nucleotide sequence analysis of coat protein (CP) gene. Strains KR1, KR2, and Az were readily transmitted to 20 of 22 inoculated plant species including bean, cowpea, tomato, tobacco, and potato. AIMV-KR1 and KR2 produced the typical symptoms like chlorotic or necrotic spots in Chenopodium quinoa and Solanum tuberosum cv. Superior. AIMV-Az caused bright yellow mosaic symptom and leaf malformation in Nicotiana glauca, which were different from the common mosaic symptom caused by AIMV-KR1 and KR2. Electron microscope observation of purified virus showed bacilliform virions containing a single-stranded plus-strand RNAs of 3.6, 2.6, 2.0 and 0.9 kbp in length, respectively, similar in size and appearance to those of Alfamovirus. In SDS-PAGE, the coat protein of the two viruses formed a consistent band that estimated to be about 24kDa. The CP genes of the AIMV strains, KR1, KR2, and Az have been amplified by RT-PCR using the specific primers designed to amplify CP gene from viral RNA-3, cloned and sequenced. Computer aided analysis of the amplified cDNA fragment sequence revealed the presence of a single open reading frame capable of encoding 221 amino acids. The nucleotide and peptide sequence of viral CP gene showed that strain KR1, KR2, and Az shared highest nucleotide sequence identities with AIMV strain 425-M at 97.7%, 98.2%, and 97.2%, respectively. CP gene sequences of two strains were almost identical compared with each other. Altogether, physical, serological, biological and molecular properties of the purified virus.

  • 최근 많은 산업의 발전으로 인해 환경오염을 유발시키는 폐수가 다량으로 배출되고 있으며, 이러한 폐수 속에는 유기용매, 고분자 물질 및 각종 염 등의 난분해성 물질들이 다량으로 함유되어 있다. 이런 물질들을 분해시키기 위해 물리적, 생물학적 수처리 방법이 많이 이용되고 있지만 이 방법들은 각각 운전비용과 처리비용이 고가인 단점이 있다. 따라서 비용과 효율 측면에서 효과적인 폐수처리를 위해서 전기화학적 폐수처리 방법이 많이 사용되고 있다. 물리적, 생물학적 처리 방법에 비해 비용이 적게 들고, 처리 후 잔류물이 남지 않으며, 독성을 띄는 산화제의 첨가 없이도 높은 폐수처리 능력을 보이기 때문에 친환경적이므로, 전기화학적 폐수산화 처리에 사용되는 불용성 전극에 대한 연구가 많이 진행되어져 오고 있다. 그 중 BDD(Boron-doped diamond) 전극은 표면에서 강력한 산화제인 수산화 라디칼의 높은 발생량으로 인해 뛰어난 폐수처리 능력을 보이므로 불용성 전극 분야에서 활발한 연구가 진행 중이다. 그러나 기존에 BDD 전극의 기판 모재로 이용되던 Si, W, Pb등은 모두 기계적 강도, 폐수처리 능력 및 독성 문제로 인해 한계가 있었고, 특히 Nb기판 위에 형성시킨 BDD 전극은 뛰어난 폐수처리 능력에도 불구하고 비싼 모재 원가로 인해 상용화가 힘든 실정이다. 이런 문제점을 해결하기 위해 높은 기계적 강도와 전기화학적 안정성을 가진 Ti 기판을 사용한 BDD 전극에 대한 연구가 보고되고 있다. 그러나 BDD와 Ti 간의 lattice mismatch, BDD층 형성을 위한 고온 공정시 탄소의 확산으로 인한 기판 표면에서의 TiC층 형성으로 인해 접착력이 감소하여 박리가 생기는 문제점이 있다. BDD와 Ti의 접착력을 향상시키기 위해 융점이 높고, 전기전도성이 우수한 TiN을 diffusion barrier layer로 삽입하면 탄소 확산에 의한 TiC층의 생성을 억제하여, 내부응력에 기인한 접착력 감소를 방지할 수 있다. 또 하나의 방법으로 Ti 기판의 전처리를 통해 BDD층의 접착력을 향상 시킬 수 있다. Sanding과 etching을 통해 기판 표면의 물리, 화학적인 표면조도를 부여하고, seeding을 통해 diamond 결정 성장에 도움을 주는 seed 입자를 분포시킴으로써, 중간층과 BDD층의 접착력을 향상시키고, BDD 결정핵 성장을 촉진시켜 고품질의 BDD박막 증착이 가능하다. 본 연구에서는 기존 Si, Nb 등의 기판 모재를 Ti로 대체함으로써 제조원가를 절감시키고, TiN 중간층을 삽입하여 접착력을 향상 시킴으로써 기존의 BDD 전극과 동등한 수준의 물성 및 수처리 특성을 가진 BDD전극 제작을 목표로 하였다. $25{\times}25mm$의 Ti 기판위에 TiN 중간층을 DC magnetron sputtering을 이용하여 증착 후, BDD 전극 층을 HFCVD로 증착하였다. 전처리를 진행한 기판과 중간층 및 BDD층의 미세구조를 XRD로 분석하였고, 표면 형상을 SEM으로 확인하였다. BDD전극의 접착력 분석을 통해 TiN 중간층의 최적 조성을 도출하고, 최종적으로 BDD/TiN/Ti 전극의 CV특성과 가폐수의 COD분해능력 및 축산폐수, 선박평형수 등의 실제 폐수 처리 능력을 BDD/Si, BDD/Nb 전극과 비교 검토할 것이다.
