v-Crk Induces Rac-dependent Membrane Ruffling and Cell Migration in CAS-deficient Embryonic Fibroblasts

  • Crk-associated substrate (CAS) is a focal adhesion protein that is involved in integrin signaling and cell migration. CAS deficiency reduces the migration and spreading of cells, both of which are processes mediated by Rac activation. We examined the functions of v-Crk, the oncogene product of the CT10 virus p47gag-crk, which affects cell migration and spreading, membrane ruffling, and Rac activation in CAS-deficient mouse embryonic fibroblasts (CAS-/- MEFs). CAS-/- MEFs showed less spreading than did CAS+/+ MEFs, but spreading was recovered in mutant cells that expressed v-Crk (CAS-/-v-Crk MEF). We observed that the reduction in spreading was linked to the formation of membrane ruffles, which were accompanied by Rac activation. In CAS-/- MEFs, Rac activity was significantly reduced, and Rac was not localized to the membrane. In contrast, Rac was active and localized to the membrane in CAS-/-v-Crk MEFs. Lamellipodia protrusion and ruffle retraction velocities were both reduced in CAS-/- MEFs, but not in CAS-/-v-Crk MEFs. We also found that microinjection of anti-gag antibodies inhibited the migration of CAS-/-v-Crk MEFs. These findings indicate that v-Crk controls cell migration and membrane dynamics by activating Rac in CAS-deficient MEFs.

  • This study was attempted to investigate the effects of cromakalim (CRK), $K^{+}$ channel opener, and glibenclamide (GLY), $K^{+}$ channel blocker, on intestinal function of rabbit. CRK supressed the tension and spontaneous movement of intestinal strips. CRK enhanced the tension and spontaneous movement of strips induced by acetylcholine. Also the inhibiting effect of dopamine was potentiated by CRK. GLY augmented the tension, but did not affect the spontaneous movement of strips. GLY inhibited tension and spontaneous movements in intestinal strips induced by acetylcholine, GLY blocked the dopamine-induced attenuation of tension, but not the decrease of spontaneous movements in intestinal strips. The present studies suggest that $K^{+}$ channel opening suppresses intestinal movements, whereas it's blockade enhances intestinal movements in rabbit.abbit.

  • This study was attempted to investigate the effect of cromakalim(CRK). $K^{-}$ channel opener, and glibenclamide(GLY), $K^{-}$ channel blocker, on intestinal function of rabbit. CRK supressed the tension and spontaneous movement of intestinal strips. Such CRK strengthened the tension and spontaneous movement of strips potentiated by acetylcholine, whereas more attenuated those weakened by dopamine.   • Elements that threaten domestic security, such as diplomatic changes in the four major powers surrounding the Korean Peninsula and restrictions on supply and demand of parts due to COVID-19, are constantly increasing. The importance of localizing weapon systems has grown to respond proactively to the threat. As localization is not conducted evenly by the field, it is necessary to change the criteria for selecting localization targets. Establishing standards for selecting targets is difficult due to a lack of research to identify targets. This paper emphasized that the smooth supply of parts from overseas should be examined to identify targets for localization. If suppliers are not distributed evenly in the market, there is a risk of potential supply problems. CRk and HHI, representing the market structure, were proposed as indicators for evaluating the degree of smoothness quantitatively. Among them, CRk is used in Korea to check the market concentration, but there is a limit in the subjectivity of the evaluator, so it cannot be applied without a separate study on the market structure. Therefore, HHI should be applied.

A Pilot Study on Korean Version Development of the Rearing Knowledge and Practice for Infant Parents (부모의 양육지식과 실천 도구 개발 예비연구)

  • Rearing knowledge is a significant factor of relevance for rearing practice, and assessment of rearing knowledge and practice is important for infant parents. The author adopted the Child Rearing Knowledge Scale (CRKS) and Child Rearing Practice Scale (CRPS) that were developed and validated by Saramma & Thomas. According to the international linguistic validation process, pilot testing was done based on 20 infant's mothers in one Oketani massage center and one public health center. Descriptive statistic methods and Wilcoxon Rank Sum Test were employed to evaluate the level and compare the mean score according to general characteristics of subjects of the tools. Reliability and validity were tested by Cronbach's alpha and Spearman Correlations. The mean age of the subjects was 33.94 (${\pm}2.99$) years and the mean age of babies was 3.35 (${\pm}0.58$) months. The understanding level of the Korean version of the CRKS and CRPS was 1.80 (${\pm}0.65$) and 1.33 (${\pm}0.54$), respectively. In addition, the CRKS and CRPS were relatively easy to use. The mean score of the CRKS was 22.50 (${\pm}4.89$), which was a moderate score, while the mean score of the CRPS was 30.75 (${\pm}2.04$), which was high. The Cronbach's alpha values of the CRPS were as follows: feeding, 0.71; growth and development, 0.64; cleaning and protection 0.68; infant stimulation, 0.77. There was a significant correlation between infant stimulation of the CRPS and growth and development of the CRKS (r=0.530, p=0.016). The CRKS score of medical staff was significantly higher than that of non-medical staff (p=0.04). The CRKS and CRPS are expected to be used in clinical or community care practice as easy-to-use tools that are easy to respond to.

Evapotranspiration Measurements using an Eddy Covariance Technique in a Mixed Forest and a rice paddy in Korea (에디 공분산으로 관측된 혼효림과 논에서의 증발산)

  • Evapotranspriation (ET) was measured by eddy covariance method in two key ecosystems in Korea: the Seolmacheon site (a mixed forest in a complex terrain, SMK) and the Cheongmicheon site (a homogeneous rice paddy, CRK). By using the multi-year observations (SMK: Sep. 2007 - Dec. 2009, CRK: Aug. 2008 - Dec. 2009), we quantified ET and analyzed its temporal variations and control mechanisms based on the radiatively coupled combination equation. During the study period, the accumulated precipitation was about 3213 mm for the SMK site, of which about 30% (i.e., 990 mm), returned to the atmosphere as ET. At the CRK site from Jan. - Dec., 2009, the annual ET was 553 mm, which was about 40% of the annual rainfall (of 1401 mm). Both sites showed a characteristic seasonality with mid-season depression in ET that are associated with the reduced amount of available energy during the monsoon season. The decoupling parameter (${\Omega}^*$), which indicates the measure of interaction between vegetation and the atmosphere, averaged about 0.4 for the SMK site and the CRK site during the growing season. The ET from both sites was more influenced by air saturation deficit and surface conductance than available energy.

Biological and Functional Characteristics of Lactic Acid Bacteria in Different Kimchi (김치 종류에 따른 유산균의 생물학적 및 기능적 특성)

  • Biological and functional characteristics of lactic acid bacteria (LAB) were investigated in mustard stem/leaf kimchi (MK), cabbage kimchi (CK), young radish kimchi (YRK), and cubed radish kimchi (CRK). LAB of young radish kimchi were mainly composed of bacilli in contrast to the other kimchi. 89.2% LAB isolated from all kimchi harbored plasmids. However, LAB had an average of $4.1{\pm}0.5$ plasmid bands in YRK, more than MK, CK, and CRK. Exopolysaccharides were produced by 10.9~11.1% of LAB, and were especially by LAB isolated from radish kimchi. A significant percentage of LAB (69.5%) had antibacterial activity against one sensitive strain or more. LAB from CK, YRK and CRK had antimicrobial activities against Bacillus sp., Listeria monocytogenes, and Salmonella Typhimurium, while the LAB from MK had activities against Vibrio parahaemolyticus higher than those from the other kimchi. In YRK and CRK, acid-tolerant LAB were twice as prevalent as those in MK and CK. Bile-tolerant LAB isolated from CRK were more prevalent than other kimchi. When $10^8$ CFU of LAB were added to Caco-2 cells, 12.1% of LAB isolated from all kimchi showed similar adherent activity to Lactobacillus rhamnosus GG. LAB of MK particularly adhered to Caco-2 cells, 2.0~4.1 fold higher than LAB in the other kimchi. From these results, biological and functional characteristics of LAB varied according to the type of kimchi and LAB existing in kimchi were limited to their respective species.

Identification of Genetic Variations in CBL, SORBS1, CRK, and RHOQ, Key Modulators in the CAP/TC10 Pathway of Insulin Signal Transduction, and Their Association with Type 2 Diabetes Mellitus in the Korean Population

  • Recent evidence has strongly suggested that the CAP/TC10 pathway is involved in the trafficking, docking, and fusion of vesicles containing the insulin-responsive glucose transporter Glut4 to the plasma membrane. However, little is known about how the genes employed in the CAP/TC10 pathway are associated with the development of type 2 diabetes mellitus. In this study, we sequenced 4 genes of the CAP/TC10 pathway [SORBS1, CBL, CRK, and RHOQ] in 24 individuals to identify genetic variations in these loci. A total of 48 sequence variants were identified, including 23 novel variations. To investigate the possible association with type 2 diabetes mellitus, 3 single nucleotide polymorphisms from SORBS1, 3 from CBL, and 4 from RHOQ were genotyped in 1122 Korean type 2 diabetic patients and 1138 nondiabetic controls. Using logistic regression analysis, 1 significant association between SNP rs1376405 in RHOQ and type 2 diabetes mellitus [OR = 8.714 (C.I. 1.714-44.29), p = 0.009] was found in the recessive model. Our data demonstrate a positive association of the RHOQ gene in the CAP/TC10 pathway with T2DM in the Korean population.

Efficacy of Lactobacillus fermentum Isolated from the Vagina of a Healthy Woman against Carbapenem-Resistant Klebsiella Infections In Vivo

  • Carbapenem-resistant Enterobacteriaceae (CRE) that produce Klebsiella pneumoniae carbapenemase are increasingly reported worldwide and have become more and more resistant to nearly all antibiotics during the past decade. The emergence of K. pneumoniae strains with decreased susceptibility to carbapenems, which are used as a last resort treatment option, is a significant threat to hospitalized patients worldwide as K. pneumoniae infection is responsible for a high mortality rate in the elderly and immunodeficient individuals. This study used Lactobacillus fermentum as a candidate probiotic for treating CRE-related infections and investigated its effectiveness. We treated mice with L. fermentum originating from the vaginal fluid of a healthy Korean woman and evaluated the Lactobacilli's efficacy in preventive, treatment, nonestablishment, and colonization mouse model experiments. Compared to the control, pre-treatment with L. fermentum significantly reduced body weight loss in the mouse models, and all mice survived until the end of the study. The oral administration of L. fermentum after carbapenem-resistant Klebsiella (CRK) infection decreased mortality and illness severity during a 2-week observation period and showed that it affects other strains of CRK bacteria. Also, the number of Klebsiella bacteria was decreased to below 5.5 log10 CFU/ml following oral administration of L. fermentum in the colonization model. These findings demonstrate L. fermentum's antibacterial activity and its potential to treat CRE infection in the future.

Detection of Mycoplasma Infection in Cultured Cells on the Basis of Molecular Profiling of Host Responses

  • Adaptive responses to diverse microbial pathogens might be limited in relatively few types. Host cell responses to pathogens are believed to be patterned or stereotyped along with species or class. We tried to compose the host response to Mycoplasma in terms of cellular gene expression. Although gene expression profile of two host HeLa and 293 cells were quite different each other, 30 genes were differentially expressed by mycoplasma infection in both of HeLa and 293 cells. Six of them (PR48, MADH4, MKPX, CRK, RBM7, NEK3) were related to cell cycle or proliferation. Another category of genes like IL1 HY1, KLRF1, TNFSF14, GBP1 were host defense to elicit immune responses. With this set of genes, we establish the prediction model for mycoplasma contamination.
