Corydalis Tuber has traditionally been used for the treatment of water retention in the body. Administration of the aqueous extract of Corydalis Tuber has been known to be effective for the control of pain and treatment of arthritis. It was reported that Corydalis Tuber possesses anti-inflammatory activity and modulates the intestinal immune system. The effect of Corydalis Tuber against LPS-stimulated expressions of COX-2, iNOS, and $IL-1{\beta}$ in cells of the murine RAW 264.7 macrophages was investigated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, reverse transcription-polymerase chain reaction (RT-PCR), $PGE_2$ immunoassay, and NO detection. The aqueous extract of Corydalis Tuber was shown to suppress $PGE_2$ production by inhibition on the LPS-stimulated enhancement of COX-2 enzyme activity, $IL-1{\beta}$, and iNOS expression in the RAW 264.7 macrophages. Present results suggest that Corydalis Tuber exerts anti-inflammatory and analgesic effects probably by suppressing of COX-2, iNOS, and $IL-1{\beta}$ expressions, resulting in inhibition of $PGE_2$ synthesis. Objectives : Peripheral nerve injuries are commonly encountered clinical problems and often result in severe functional deficits. Sukjiyanggeun-Tang(shudiyangjin-tang), in oriental medicine, has been used to treat various musculoskeletal disorders. Methods : In the present study, the effects of aqueous extract of Sukjiyanggeun-Tang(shudiyangjin-tang) on functional recovery, severity of pain, and expressions of neurofilament, cycloxygenease-2(COX-2), inducible nitric oxide synthase(iNOS), and tumor necrosis factor-${\alpha}$(TNF-${\alpha}$) following sciatic crushed nerve injury in rats were investigated. For this study, walking tract analysis, plantar test, western blot analysis for COX-2 iNOS, and TNF-${\alpha}$, and Immunofluorescence test for neurofilament were performed. Results : In the present results, sciatic functional index(SFI) in walking tract analysis was significantly decreased following sciatic crushed nerve injury, and pain severity in plantar test was significantly increased. COX-2, iNOS and TNF-${\alpha}$ expressions were increased whereas neurofilament expression was decreased by sciatic crushed nerve injury, In contrast, treatment with Sukjiyanggeun-Tang(shudiyangjin-tang) improved SFI in walking tract analysis and suppressed the pain severity in sciatic crushed nerve injury. Sukjiyanggeun-Tang(shudiyangjin-tang) treatment also suppressed COX-2, iNOS, and TNF-${\alpha}$ expressions and enhanced the neurofilament expression in sciatic crushed nerve injury. Conclusions : In the present study, we have shown that Sukjiyanggeun-Tang(shudiyangjin-tang) is the effective therapeutic modality to ameliorate the symptoms of sciatic crushed nerve injury.
Objectives : The purpose of this study was to investigate the anti-inflammatory and anti-oxidative effects of Osung-tang(OST) extract. Methods : MTT assay was performed to confirm the survival rate of RAW 264.7 cells treated with OST extract(50-400㎍/㎖) and the production of NO from LPS-induced RAW 264.7 cells was confirmed. The mRNA expression of iNOS, COX-2, and NF-κB were measured by real-time PCR. The protein expression of iNOS, COX-2 and p-IκB were measured by western blot and the anti-oxidant activity of OST extract(50-400㎍/㎖) was investigated by measuring DPPH scavenging activity. Results : OST extract showed a cell survival rate of 90% or more at 50-400㎍/㎖. The NO production was inhibited in a dose-dependent manner in RAW 264.7 cells treated with LPS on OST treated group. mRNA expression levels of iNOS, COX-2 and NF-κB decreased in a concentration-dependent manner after treatment with OST(50-400㎍/㎖). Protein expression levels of iNOS, COX-2 and p-IκB decreased in a concentration-dependent manner after treatment with OST(50-400㎍/㎖). It was found that OST has a high free-radical scavenging ability. Conclusions : These results suggest that OST extract can be used as a treatment for various skin diseases by demonstrating its anti-inflammatory and anti-oxidant effects.
본 연구에서는 브로콜리 잎 헥산 분획물의 항염 효과를 평가하여 기능성 식품 및 화장품 소재로의 적용 가능성을 확인하였다. LPS-자극된 RAW264.7 세포에서 전염증성 사이토카인의 생성, iNOS와 COX-2의 발현, MAPK (ERK, JNK, p38) 및 브로콜리 잎 헥산 분획을 사용한 NF-κB의 인산화를 분석하였다. 브로콜리 잎 헥산 분획은 TNF-α, IL-4, IL-6, IL-1β 등의 전염증성 사이토카인의 분비와 iNOS와 COX-2의 발현을 억제했습니다. 또한, 브로콜리 잎 헥산 분획물은 MAPK와 NFκB의 인산화를 감소시켰다. 따라서 브로콜리 잎 헥산 분획물은 식품 및 화장품에서 천연 항염증 소재로 적용 가능성이 있는 것으로 판단된다. 향후 항염증 기전 및 주요 생리활성 물질의 규명에 대한 연구가 필요할 것으로 생각된다.
Objectives : The purpose of this study was to evaluate the anti-inflammatory effect of Tanghwajitongsan(THJTS) through inhibition of NF-κB and MAPK. Methods : We evaluated cell survival rate by MTT assay, NO production by nitrite content in the culture medium. We quantified TNF-α, IL-1β, IL-6 and PGE2 of the cultured supernatant by ELISA. And we evaluated the effect of THJTS on protein expression of NF-κB, MAPK, iNOS and COX-2 by Western blot analysis. THJTS ameliorates LPS-activated alterations in protein expression of NF-κB, p-38, iNOS and COX-2 and production of NO, pro-inflammatory cytokines and PGE2. Also, THJTS ameliorates LOX, PGN and FLA-activated alterations in protein expression of NO, iNOS. THJTS ameliorates only PGN-activated alterations in protein expression of COX-2. Results : THJTS ameliorates LPS-activated alterations in protein expression of NF-κB, p-38, iNOS and COX-2 and production of NO, pro-inflammatory cytokines and PGE2. Also, THJTS ameliorates LOX, PGN and FLA-activated alterations in protein expression of NO, iNOS. THJTS ameliorates only PGN-activated alterations in protein expression of COX-2. Conclusions : This study can provide scientific evidence for the anti-inflammatory effects and underlying mechanisms of THJTS.
배경: Cox-Maze 술식후의 좌심방의 수축력은 우심방에 비하여 회복률이 낮은 것으로 알려져 있다. 술전 좌심방의 크기가 큰 환자군에서의 Cox-Maze 술식은, 좌심방 후벽의 많은 부위를 전기전도계와 분리함으로써 술후 좌심방 기능의 회복을 저하시키는 한 요인이 될 수 있다. 본 연구자들은 좌심방이 심하게 확장된 경우에 좌, 우 폐정맥을 각각 분리하는 변형된 Cox-Maze 술식을 시행함으로써 이러한 술식이 술후 좌심방 기능의 회복에 도움을 주는지 여부에 대해 알아보고자 하였다. 대상 및 방법: 1995년 2월부터 1997년 10월까지 승모판막 질환과 동반된 심방세동으로 Cox-Maze 술식을 시행한 환자들 중, 수술후 6개월 이상의 추적관찰이 이루어지고 동율동 전환이 이루어진 환자를 대상으로 하였다. I군은 좌, 우 폐정맥사이의 거리가 6.5cm미만인 경우(n=30), II군은 좌, 우 폐정맥사이의 거리가 6.5cm 이상인 경우(n=16)로서 I, II군은 좌,우 폐정맥을 함께 분리하는 기존의 Cox-Maze 술식을 시행하였고, III군은 폐정맥사이의 거리가 6.5cm이상인 경우에 좌, 우 폐정맥을 각각 분리하는 변형된 Cox-Maze 술식을 사용하였다(n=9). 결과:각 군사이에는 좌심방 크기를 제외한 검사소견은 차이가 없었으며 III군의 경우 수술시 대동맥차단 시간이 유의하게 증가되었다(p<0.05). 술후 장기 추적 검사상 우심방의 수축력 회복은 I군에서 90%, II군에서 81%, 그리고 III군에서 100%에서 회복됨이 관찰되었다. 좌심방의 수축력회복은 I군에서 63%, II군에서 31%에서 회복되어 좌심방이 확장된 II군에서 유의한 감소가 있었으며 (p<0.04), III군에서는 66%에서 회복되었으나 I, II군과 유의한 차이는 없었다. 결론:이러한 양폐정맥 사이를 분리, 절개하는 변형된 Cox-Maze 술식은 좌심방 수축력의 회복에 긍정적인 영향을 미칠 것으로 추정되며, 향후 더 많은 환자군에서 연구가 시행되어야 하리라 사료된다.
Objective: In this study, we investigated the effects of Rhizoma Cimicifugae (RC) on the induction of nitric oxide (NO) in RAW 264.7 murine macrophages treated with lipopolysaccharide (LPS). Methods: We examined whether RC have an antipyretic effect by inhibition of COX and/or NOS in fever that is evoked by an intraperitoneal injection of LPS in rats. Results: 1. RC inhibited NO metabolites produced by activated murine macrophage in a dose-dependent manner. 2. RC had an antipyretic effect in a dose-dependent manner in LPS-induced fever. 3. RC inhibited levels of both COX-2 protein and iNOS protein increased by the treatment of LPS in brain tissues as well as brain blood vessels. 4. There were no changes in proteins of nNOS/COX-1. Conclusions: RC has an antipyretic effect by attenuating the level of iNOS and COX-2 protein.
A wide arry of naturally occurring substances particularly those present in dietary and medicinal plants, have been reported to possess substantial cancer chemopreventive properties. Certain phytochemicals retain strong antioxidative and anti-inflammatory properties which appear to contribute to their chemopreventive or chemoprotective activities. Inducible cyclooxygenase(COX-2) and nitric oxide synthase (iNOS) are important enzymes that mediate inflammatory processes. There is some evidence that expression of both COX-2 and iNOS is co-regulated by the eukaryotic transcription factor NF-$textsc{k}$B. Increased expression of COX-2 and/or iNOS has been associated with pathophysiology of certain types of human cancers as well as inflammatory diseases. Since inflammation is closely linked to tumor promotion, substances with potent anti-inflammatory activies are anticipated to exert chemopreventive effects on carcinogenesis, particularly in the promotion stage. An example is curcumin, a yellow pigment of turmeric (Curcuma longa L., Zingiberaceae), that strongly occurring diaryl heptanoids structurally related to curcumin have substantial anti-tumor promotional activities in two-stage mouse skin carcinogenesis. Thus, yakuchinone A [1-(4'-hydroxy-3'-methoxyphenyl)-7-phenyl-3heptanone] and yakuchinone B [1-(4'-hydroxy-3'methoxyphenyl)-7-phenylhept-1-en-3-one] present in Alpinia oxyphylla Miquel (Zingiberacease) attenuate phorbol ester-induced inflammation and papilloma formation in female ICR mice. These diarylheptanoids also suppressed phorbol ester-induced activation of epdermal ornithine decarboxylase and its mRNA expression when applied onto shaven backs of mice. Yakuchinone A and B as well as curcumin inhibited phorbol ester-induced expression of COX-2 and iNOS and their mRNA in mouse skin via inactivation of NF-$textsc{k}$B. Capsaicin, a major pungent ingredient of red pepper also attenuated phorbol ester-induced NF-$textsc{k}$B activation. Similar suppression of COX-2 and iNOS and down-regulation of NF-$textsc{k}$B activation for its DNA binding were observed with the ginsenosied Rg3 and the ethanol extract of Artemisia asiatica. We have also found that certain anti-inflammatory phytochemicals exert inhibitory effects on phorbol ester-induced COX-2 expression and NF-$textsc{k}$B activation in immortalized human breast epithelial (MCF-10A) cells in culture. One of the plausible mechanisms undelying inhibition by aforementioned phytochemicals of phorbol ester-induced NF-$textsc{k}$B activation involves interference with degragation of the inhibitory unit, I$textsc{k}$Ba, which blocks subsequent nuclear translocation of the functionally active p65 subunit of NF-$textsc{k}$B. the activation of epidermal NF-$textsc{k}$B by phorbol ester and subsequent induction of COX-2 hence appear to play an important role in intracellular signaling pathwasy leading to tumor promotion and targeted inhibition of NF-$textsc{k}$B may provide a new promising cancer chemopreventive strategy.
In order to understand the mechanism of action of TCDD, we have examined the effect of COX-inhibitors on cyp1a1 activity. We observed the effect of COX-inhibitor on EROD activity in C57BL/6 mouse in vovo. And we also evaluated the effect of COX-inhibitors on cyp1a1 mRNA, mouse cyp1a1 promoter activity and EROD activity in Hepa cell. When Aspirin were pretreated with 3MC in vivo, the EROD activity that was stimulated by 3MC was inhibited. And Pretreatment of Aspirin, Celecoxib, Nimesulide and other several Cox-inhibitors in vitro, inhibited the TCDD stimulated EROD activity and Luciferase acitivity. In case of cyp1a1 mRNA level, Nimesulide and SB100 were able to decrease cyp1a1 mRNA that was stimulated by TCDD, but other tested COX-inhibitors were not decrease. We don't know this different result exactly. For the action of Cox-inhibitors on the Cyp1a1, it seems to be important to do pretreatment of these chemicals as apposed to TCDD. In this study, thus, we have suggested that COX-inhibitors such as aspirin, celecoxib, Nimesulide and other several Cox-inhibitors decrease the TCDD induced Cyp1a1.
서울, 부산, 대구, 대전, 광주 5개 대도시와 그 외 산청과 영천지점의 강우자료를 이용하여 Box-Cox변환기법에 의한 I-D-F곡선을 유도하였다. Box-Cox변환기법의 실용성은 여러 학문분야에서 검증받은 기법으로 분석 자료가 일반 빈도해석을 수행하기에 부족하여 적절한 확률밀도함수를 도출하지 못할 경우에도 사용가능하기 때문에 적용성 측면에서 전통적인 빈도해석 방법보다 확장성이 크다. 전술한 7개 지점의 전체지속기간 강우자료(10분~1440분)와 짧은 지속기간(20분, 30분, 40분, 50분) 강우자료가 누락될 경우의 강우강도를 비교했을 때 100년 빈도 이하 지속기간 10분~60분에서 -23.0~14.7 %의 상대오차가 있었다. 이에 따라 강우분석 시 짧은 지속기간(20분, 30분, 40분, 50분) 강우자료를 포함하여 I-D-F곡선을 유도함이 요구되며 짧은 지속기간(20분, 30분, 40분, 50분) 강우자료가 누락될 경우 소규모 수공구조물 설계 시 원활한 배수를 위해 지점에 따라 기존의 여유율을 높일 필요가 있다.
