Growth Characteristics and Variation in Amino Acids Composition of Quality Protein Maize Lines (고라이신(QPM) 주요 계통과 교잡계의 생육특성 및 아미노산 조성 변이)

  • Maize grain quality can be improved by raising lysine content, which is an essential amino acid present in insufficient quantities in normal maize. Maize varieties with such modifications are known as quality protein maize (QPM). To date, no Korean maize cultivars contain high amounts of lysine. To introduce quality protein maize to Korean cultivars, we crossed QPM CIMMYT maize lines (CML) with KS140, an elite inbred line used as a parent of several cultivars such as 'Gangdaok' and 'Pyeonganok'. We analyzed the phenotypic characteristics of F1 plants as well as the protein contents, amino acids, and fatty acids profiles of the self-pollinated seeds of the F1 hybrids, and evaluated the feasibility of CML as a source of QPM. Days to anthesis of the CML ranged from 78 to 90 days after planting (DAP), whereas a range of 81~87 was recorded for F1 hybrids. The average days to anthesis was 85 for KS140, CML, and the F1 hybrids. The protein content of the CML was measured to be between 9.1 and 12.1%, with the highest and lowest values being recorded in CML153 and 191, respectively. The F1 hybrids had protein contents of 9.1~11.1%, and the highest content was observed in KS141/CML188. The fatty acids profiles were very similar across all analyzed maize samples, and linoleic acid (C18:2) composed the greatest proportion. Glutamic acid made up the largest proportion of amino acids in all maize samples. Lysine composition was highest in CML155 (6.92% of all amino acids), with an average composition of 4.83% across the CML. In contrast, KS140 showed a lysine content of 2.51%. In F1 hybrids, the average lysine composition was 3.46%, and KS140/CML164 (4.18%) and KS140/CML163 (4.99%) contained more lysine than either parent. Taken together, these results indicate that CML could become promising QPM sources to improve grain quality in Korean maize cultivars.

Web-based Chart Generating System Using CML (CML을 이용한 웹 기반 차트출력시스템)

  • Charts can propagate information more effectively by visualizing various types of information. Due to this reason, many web developers often use charts when they display information on the web. However, using a chart requires a special dedicated software to install and it is difficult to share the finished charts since they are usually in raster format. The raster format images are fixed in size, therefore, experience image distortion problem when their sizes are changed. In this paper, we propose a web-based chart generating system using CML(Chart Markup Language) to solve the current compatibility and sharing problems. The proposed chart generating system first analyzes XML, Text, or HTML documents to extract chart information in the documents, and then it converts collected chart information to CML. The converted CML documents are displayed on the web browser using a vector method. The vector method has an advantage of protecting its images from distortion even when image sizes are scaled. The CML, proposed in this paper, is a chart markup language based on XML for modeling charts Information. If CML is used for presenting a chart in a web, it makes easier to share and to convert the chart information.

Hypoxia-inducible factor 1α inhibitor induces cell death via suppression of BCR-ABL1 and Met expression in BCR-ABL1 tyrosine kinase inhibitor sensitive and resistant chronic myeloid leukemia cells

  • Chronic myeloid leukemia (CML) has a markedly improved prognosis with the use of breakpoint cluster region-abelson 1 (BCR-ABL1) tyrosine kinase inhibitors (BCR-ABL1 TKIs). However, approximately 40% of patients are resistant or intolerant to BCR-ABL1 TKIs. Hypoxia-inducible factor 1α (HIF-1α) is a hypoxia response factor that has been reported to be highly expressed in CML patients, making it a therapeutic target for BCR-ABL1 TKI-sensitive CML and BCR-ABL1 TKI-resistant CML. In this study, we examined whether HIF-1α inhibitors induce cell death in CML cells and BCR-ABL1 TKI-resistant CML cells. We found that echinomycin and PX-478 induced cell death in BCR-ABL1 TKIs sensitive and resistant CML cells at similar concentrations while the cell sensitivity was not affected with imatinib or dasatinib in BCR-ABL1 TKIs resistant CML cells. In addition, echinomycin and PX-478 inhibited the c-Jun N-terminal kinase (JNK), Akt, and extracellular-regulated protein kinase 1/2 (ERK1/2) activation via suppression of BCR-ABL1 and Met expression in BCR-ABL1 sensitive and resistant CML cells. Moreover, treatment with HIF-1α siRNA induced cell death by inhibiting BCR-ABL1 and Met expression and activation of JNK, Akt, and ERK1/2 in BCR-ABL1 TKIs sensitive and resistant CML cells. These results indicated that HIF-1α regulates BCR-ABL and Met expression and is involved in cell survival in CML cells, suggesting that HIF-1α inhibitors induce cell death in BCR-ABL1 TKIs sensitive and resistant CML cells and therefore HIF-1α inhibitors are potential candidates for CML treatment.

Evaluation of Drought Tolerance using Anthesis-silking Interval in Maize

  • We screened the drought tolerant maize using seventeen maize genotypes from different sources, nine inbred genotypes from United States Department of Agriculture (USDA) (B73, CML103, CML228, CML277, CML322, CML69, Ki3, Ki11, and NC350), three Southeast Asian genotypes (DK9955, LVN-4, and 333), and five Korean hybrids (Cheongdaok, Gangdaok, Ilmichal, Kwangpyeongok, and Pyeonganok). We evaluated anthesis-silking interval (ASI), leaf senescence (LS), ears per plant (EPP), tassel length (TL), and fresh weight (FW) at silking date. According to ASI and LS examination, CML103 and Kill were drought tolerant genotypes, wheareas Ki3 and 333 were drought susceptible. EPP, TL, and FW differed according to drought resistance. Grain yield was correlated strongly with ASI, but moderately with LS. Difference in ASI between drought-stressed (DS) and well-watered (WW) conditions was less than three days in CML228, CML103, Cheongdaok, NC350, B73, Ki11, CML322, and Kwangpyeongok, whereas that of Ki3, Pyeonganok, and Gangdaok was more than 6.5 days. We concluded that CML228, CML103, Cheongdaok, NC350, B73, Ki11, CML322, and Kwangpyeongok are drought tolerant genotypes, whereas Ki3, Pyeonganok, and Gangdaok are drought susceptible.

Study on the performance indices of low-strength brick walls reinforced with cement mortar layer and steel-meshed cement mortar layer

  • Older brick masonry structures generally suffer from low strength defects. Using a cement mortar layer (CML) or steel-meshed cement mortar layer (S-CML) to reinforce existing low-strength brick masonry structures (LBMs) is still an effective means of increasing seismic performance. However, performance indices such as lateral displacement ratios and skeleton curves for LBMs reinforced with CML or S-CML need to be clarified in performance-based seismic design and evaluation. Therefore, research into the failure mechanisms and seismic performance of LBMs reinforced with CML or S-CML is imperative. In this study, thirty low-strength brick walls (LBWs) with different cross-sectional areas, bonding mortar types, vertical loads, and CML/S-CML thicknesses were constructed. The failure modes, load-carrying capacities, energy dissipation capacity and lateral drift ratio limits in different limits states were acquired via quasi-static tests. The results show that 1) the primary failure modes of UBWs and RBWs are "diagonal shear failure" and "sliding failure through joints." 2) The acceptable drift ratios of Immediate Occupancy (IO), Life Safety (LS), and Collapse Prevention (CP) for UBWs can be 0.04%, 0.08%, and 0.3%, respectively. For 20-RBWs, the acceptable drift ratios of IO, LS, and CP for 20-RBWs can be 0.037%, 0.09%, and 0.41%, respectively. Moreover, the acceptable drift ratios of IO, LS, and CP for 40-RBWs can be 0.048%, 0.09%, and 0.53%, respectively. 3) Reinforcing low-strength brick walls with CML/S-CML can improve brick walls' bearing capacity, deformation, and energy dissipation capacity. Using CML/S-CML reinforcement to improve the seismic performance of old masonry houses is a feasible and practical choice.

Fabrication of Solution-Based Cylindrical Microlens with High Aspect Ratio (고종횡비를 갖는 용액기반 원통형 마이크로렌즈 제조)

  • A cylindrical microlens (CML) has been widely used as an optical element for organic light-emitting diodes (OLEDs), light diffusers, image sensors, 3D imaging, etc. To fabricate high-performance optoelectronic devices, the CML with high aspect ratio is demanded. In this work, we report on facile solution-based processes (i.e., slot-die and needle coatings) to fabricate the CML using poly(methyl methacrylate) (PMMA). It is found that compared with needle coating, slot-die coating provides the CML with lower aspect ratio due to the wide spread of solution along the hydrophilic head lip. Although needle coating provides the CML with high aspect ratio, it requires a high precision needle array module. To demonstrate that the aspect ratio of CML can be enhanced using slot-die coating, we have varied the molecular weight of PMMA. We can achieve the CML with higher aspect ratio using PMMA with lower molecular weight at a fixed viscosity because of the higher concentration of PMMA solute in the solution. We have also shown that the aspect ratio of CML can be further boosted by coating it repeatedly. With this scheme, we have fabricated the CML with the width of 252 ㎛ and the thickness of 5.95 ㎛ (aspect ratio=0.024). To visualize its light diffusion property, we have irradiated a laser beam to the CML and observed that the laser beam spreads widely in the vertical direction of the CML.

Clinical Significance of BCR-ABL Fusion Gene Subtypes in Chronic Myelogenous and Acute Lymphoblastic Leukemias

  • Background: Some reports have suggested that chronic myeloid leukemia (CML) patients have a higher prevalence of M-bcr than acute lymphoblastic leukemia (ALL) patients, which show a higher prevalence of m-bcr. However, the relationship between BCR-ABL subtypes and progression of CML and ALL remains unclear. Materials and Methods: 354 CML chronic phase (CML-CP) patients, 26 CML blastic phase (CML-BP) patients and 72 ALL patients before treatment with BCR-ABL positive were recruited for blood routine examination and bone marrow smear cytology. Some 80 CML-CP and 32 ALL patients after imatinib (IM) treatment were followed-up for BCR-ABL relative concentrations detected after treatment for 3, 6 and 9 months and 1 year. Results: Before treatment, CML-CP patients showed lower BCR-ABL relative concentrations with a higher proportion of M-bcr (42.7%) compared to CML-BP and ALL patients while ALL patients had a higher BCR-ABL relative concentration with high expression of m-bcr (51.4%). Patients with M-bcr demonstrated higher WBC counts than those with m-bcr and the mixed group and higher PLT counts were noted in the CML-CP and ALL groups. After imatinib (IM) treatment, patients with m-bcr showed higher BCR-ABL relative concentrations in both CML-CP and ALL groups. Conclusions: This study identified the BCR-ABL gene as an important factor in CML and ALL cases. The M-bcr subtype was associated more with CML while the m-bcr subtype was associated more with ALL. Patients with m-bcr seem to have a poorer response to IM in either CML or ALL patients compared to M-bcr patients.

Low Level of TERC Gene Amplification between Chronic Myeloid Leukaemia Patients Resistant and Respond to Imatinib Mesylate Treatment

  • The amplification of telomerase component (TERC) gene could play an important role in generation and treatment of haematological malignancies. This present study was aimed to investigate copy number amplification status of TERC gene in chronic myeloid leukaemia (CML) patients who were being treated with imatinib mesylate (IM). Genomic DNA was extracted from peripheral blood of CML-IM Resistant (n=63), CML-IM Respond (n=63) and healthy individuals (n=30). TERC gene copy number predicted (CNP) and copy number calculated (CNC) were determined based on $Taqman^{(R)}$ Copy Number Assay. Fluorescence in situ hybridization (FISH) analysis was performed to confirm the normal signal pattern in C4 (calibrator) for TERC gene. Nine of CML patients showed TERC gene amplification (CNP=3), others had 2 CNP. A total of 17 CML patients expressed CNC>2.31 and the rest had 2.31>CNC>1.5. TERC gene CNP value in healthy individuals was 2 and their CNC value showed in range 1.59-2.31. The average CNC TERC gene copy number was 2.07, 1.99 and 1.94 in CML-IM Resistant patients, CML-IM Respond and healthy groups, respectively. No significant difference of TERC gene amplification observed between CML-IM Resistant and CML-IM Respond patients. Low levels of TERC gene amplification might not have a huge impact in haematological disorders especially in terms of resistance towards IM treatment.

The rs61764370 Functional Variant in the KRAS Oncogene is Associated with Chronic Myeloid Leukemia Risk in Women

  • Background: Chronic myeloid leukemia (CML) is one of the most frequent hematopoietic malignancies in the elderly population; however, knowledge is limited regarding the genetic factors associated with increased risk for CML. Polymorphisms affecting microRNA (miRNA) biogenesis or mRNA:miRNA interactions are important risk factors in the development of different types of cancer. Thus, we carried out a case-control study to test the association with CML susceptibility of gene variants located in the miRNA machinery genes AGO1 (rs636832) and GEMIN4 (rs2740348), as well as in the miRNA binding sites of the genes BRCA1 (rs799917) and KRAS (rs61764370). Materials and Methods: We determined the genotype of 781 Mexican-Mestizo individuals (469 healthy subjects and 312 CML cases) for the four polymorphisms using TaqMan probes to test the association with CML susceptibility. Results: We found a borderline association of the minor homozygote genotype of the KRAS_rs61764370 polymorphism with an increased risk for CML susceptibility (P = 0.06). After gender stratification, this association was significant only for women (odds ratio [OR] = 13.41, P = 0.04). The distribution of the allelic and genotypic frequencies of the four studied SNPs was neither associated with advanced phases of CML nor treatment response. Conclusions: To the best of our knowledge, this study is the first to show a significant association of the KRAS_rs61764370 SNP with CML. To further determine such an association of with CML susceptibility, our results must be replicated in different ethnic groups.

Design Method of Current Mode Logic Gates for High Performance LTPS TFT Digital Circuits (LTPS TFT 논리회로 성능향상을 위한 전류모드 논리게이트의 설계 방법)

  • Development of high performance LTPS TFTs contributed to open up new SOP technology with various digital circuits integrated in display panels. This work introduces the current mode logic(CML) gate design method with which one can replace slow CMOS logic gates. The CML inverter exhibited small logic swing, fast response with high power consumption. But the power consumption became compatible with CMOS gates at higher clock speed. Due to small current values in CML, layout area is smaller than the CMOS counterpart even though CML uses larger number of devices. CML exhibited higher noise immunity thanks to its non-inverting and inverting outputs. Multi-input NAND/AND and NOR/OR gates were implemented by the same circuit architecture with different input confirugation. Same holds for MUX and XNOR/XOR CML gates. We concluded that the CML gates can be designed with few simple circuits and they can improve power consumption, chip area, and speed of operation.
