A Study on the life of Yoon Cho Chang(尹草窓) and Cho-Chang-Kyeul(草窓訣) (윤초창(尹草窓)의 생애(生涯)와 초창결(草窓訣)에 관(關) 연구(硏究))

  • I have studied life of Yoon-Dong-Li(尹東里), who applied Un-Ki(運氣) theory to medicine during Suk-Jong(肅宗) to Joong-Jo(正祖) in Yi-Dynasty and also studied his existing writings named ${\ll}$Cho-Chang-kyeol${\gg}$ (草窓訣). The referenced original work prints were based on 1980's print of Chung-Ku oriental medical society and National Central Library and The academy a korean studies possessed print. Studied with these books, I have concluded as follows. 1. Yoon-Dong-Li(尹東里) courtesy name is Ja-Mi(子美), pen name is Cho-Chang(草窓). He was born in 1705, Suk-Jong(肅宗) 31yrs, and died in 1784, Jeong-Jo(正祖) 8yrs in Yi-Dynasty. He had lineage of third-generationed-doctor and learned medicine from his uncle, Yoon-Woo-Kyo(尹雨敎). His father Yoon-Yi-Kyo(尹이敎) gaved him second influence. 2. Confucianal physician in Ming Dynasty You-Bu(劉溥) respected Ju-Ryeom-Kye(周溓溪), confucianist in Song-Dynasty and You-Bu(劉溥) does not removed grass in front of window (because in chinese, Cho-Charig(草窓) means grass in front of window), and named himself Cho-Chang(草窓). Yoon-Dong-Li(尹東里) followed this suit so called himself Cho-Chang (草窓). 3. The main contents of ${\ll}$Cho-Chang-Kyeol${\gg}$ (草窓訣), (運氣衍論) was written in 1725, when Yoon-Dong-Li(尹東里) was 21yrs old and printed in 1736. The other part of ${\ll}$Cho-Chang-Kyeol${\gg}$ (草窓訣), (用藥篇) was written in 1746. These two parts were not written in the satre time. The (運氣衍論) was written llyears earlier than (用藥). Two parts were combined another day and named (草窓訣). 4. Existing ${\ll}$Cho-Chang-Kyeol${\gg}$ (草窓訣) is transcribed printing with the con tent of "Five elements motion and the six kind of natural factors theory" (五運六氣設). Each edition is generally similar in content, but also has different points each other so naw we hardly finds a complete set of works. 5. ${\ll}$Cho-Chang-Kyeol${\gg}$ (草窓訣) is a first technical book which treats of Un-Ki(運氣) theory in korea. 6. The contents of (運氣衍論) of ${\ll}$Cho-Chang-Kyeol${\gg}$ (草窓訣) is mechanism of diease according to excess and insuffciency of five elements motion and six kinds of nalural faclors.and symploms and priscriptions, includes variated priscriptions. 7. Two parts in (運氣衍論) of ${\ll}$Cho-Chang-Kyeol${\gg}$ (草窓訣), 'Sang-tong' (相通) and 'Kak-Tong' (各通), threats of mochanisim of the disease according to five elements. with the principle of inter-promoting and inter-acting, in the change of ten heavenly stems and five elements motion. 8. In the (用藥篇) of ${\ll}$Cho-Chang-Kyeol${\gg}$ (草窓訣) describes priscription for clinical application according to Un-Ki(運氣) and also records about misuse of medicine. 9. In the ${\ll}$Cho-Chang-Kyeol${\gg}$ (草窓訣), most of priscription are common using one, which also found in ${\ll}$Dong-$\breve{U}$i-Bo-Garm${\gg}$ (東醫寶鑑) and there are few of priscriptions hy Yoon-Dong-Li(尹東里).

Cytogenetic Characteristics of Chinese Hamster Ovarian Cell CHO-K1

  • The Chinese Hamster Ovarian cells CHO-K1 are one of the most extensively used cells for the evaluation of gene expression and toxicology. However, these cells are frequently used for biomedical research without consideration of their cytogenetic characteristics. Therefore, we carried out to investigate the karyologic profiles, the frequency and type of chromosome aberration, and the distribution of telomeric DNA on chromosomes of the CHO-K1 cells. The GTG banding and fluorescence in situ hybridization on CHO-K1 cells were performed to characterize the karyotype and the distribution of telomeric DNA The present study revealed that the chromosome modal number of CHO-K1 cells was 2n=20; eight chromosomes appeared to be identical with those of the normal Chinese hamster, whereas the remaining 12 chromosomes were shown to be translocated, deleted, inversed, or rearranged from Chinese hamster chromosomes. The telomeric DNA on CHO-K1 chromosomes was intensively distributed at the centromeres rather than the ends of chromosomes. In addition, three chromosomes had interstitial telomeres and one marker chromosome entirely consisted of telomeric DNAs. The frequency and type of chromosome aberrations in CHO-K1 cells were examined. Of the 822 metaphase spreads, 68 (8.3%) cells resulted in chromosome aberrations of which the chromosome breakage was the most frequently occurred.

Purification, Characterization, and Gene Cloning of Chitosanase from Bacillus cereus H-l (Bacillus cereus H-1으로부터 Chitosanas리 분리와 특성연구 및 유전자 클로닝)

  • A 1.3-kb of chitosanase gene (choA) encoding 45-kDa polypeptide was cloned, expressed, and characterized from a newly isolated Bacillus cereus H-1. The chitosanase protein (ChoA) of B. cereus H-l was purified to homogeneity by ammonium sulfate precipitation and CM-sephadex column chromatography. Optimum pH was around 7, and stable pH range in the incubation at 50 C was 4-11. Optimum temperature was around 50 C, and enzyme activity was relatively stable below 45 C. ChoA showed the activities toward carboxymethyl cellulose (CMC) in addition to soluble or glycol chitosan. Based on MALDI-TOF MS analysis of purified ChoA, the entire amino acid sequence of ChoA was interpreted by database searching of previously known Bacillus chitosanases. A 1.6 kb of PCR product of corresponding chitosanase gene was obtained and its DNA sequence was determined. The deduced amino acid of choA revealed that ChoA have a 98% homology with those of Bacillus sp. No.7-M strain and Bacillus sp. KCTC0377BP. The recombinant ChoA protein was expressed in E. coli DH5$\alpha$. Deduced amino acid comparison of choA with other chitosanases suggested that it belongs to family 8 microbial endo-chitosanase with chitosanase-cellulase activity.

Isolation of Antifungal Bacterial Strain Bacillus sp. against Gray Mold infected in Kiwi Fruits and its Disease Control (참다래 잿빛곰팡이 병원균에 대한 길항균 Bacillus sp. 분리와 병해 억제 작용)

  • This study was carried out to identity the effects of antifungal bacteria isolated from the soil grown kiwi fruit plants on the growth inhibition of Botrytis cinerea causing gray mold in kiwi fruit plants in the southern districts of Jeonnam. Two hundred and fifty antagonistic microorganisms were isolated and examined into the antifungal activity against Botrytis cinerea. We screened and isolated four bacterial strains which strongly inhibited Botrytis cinerea from the soil grown kiwi fruit plants. And the best antifungal bacterial strain which called CHO 163 was finally selected. Antagonistic microorganism CHO 163 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. Bacillus sp. CHO 163 showed 86.9% of antifungal activity against Botrytis cinerea. By the bacterialization of culture broth and heated filtrates of culture broth, Bacillus sp. CHO 163 showed almost all of antagonistic activity against Botrytis cinerea. And we also confirmed that in vitro the treatment of Bacillus sp. CHO 163 cultured by SD+B+P broth efficiently controled the growth of Botrytis cinerea causing gray mold in kiwi fruit plants.

Effect of Domestic Rose Flower Extracts on the Growth of Chinese Hamster Ovary Cells (국내산 장미꽃 추출물이 Chinese Hamster Ovary 세포 증식에 미치는 영향)

  • The effects of rose flower extracts on the growth of CHO cells were examined. Rose flower extracts were prepared by solvent extraction with hexane, ethylacetate and ether from five domestic rose cultivar, Rosa hybrida L. cv. Mihyang, Noeul, Redqueen, Whitelady and Pinklady, respectively. The effects of rose flower extracts on the growth of CHO cells were measured using MTT colormeteric assay and compared with control. Extracts of rose flowers showed stimulative effect or inhibitory effect on the growth of CHO cells depending on the kinds of solvent and concentration of extracts. Ether extracts of rose flower showed a more effective stimulative effect on the growth of CHO cells at the concentration of 5 ${\mu}g{\cdot}ml^{-1}$. These results suggest that rose flower has the simulating activity on the growth of CHO cells and a potential as new functional food source.

Mechanism of Arsenic-Induced Cytotoxiciht in CHO Cells (CHO 세포에서 비소의 세포독성기전)

  • This study was carried out to examine the mechanism of Arsenic cytotoxicity through several in vitro test systems. Dose-dependent decrease of cell survival by Arsenic was observed by colony forming assay. Arsenic was weak mutagenic in inducing HGPRT point mutation in CHO cells. The frequency of chromosomal aberrations increased in a dose-dependent manner and the most frequent type of chromosomal aberrations induced by Arsenic were chromatid type deletions. U!trafiltrates of culture media from CHO cells treated with Arsenic induced sister chromatid exchanges(SCE) in CHO cells and Arsenic was able to induce lipid peroxidation in CHO cells. The results suggested that the ultrafiltrates of media from CHO cells treated with Arsenic contain clastogenic factor(CF) and Iipid peroxidation might be involved in the formation of CF.

Normal MR Spectroscopic Findings of the Cerebellum and Pons: Comparison of the Results between Intra-Unit and Inter-Unit Studies (소뇌와 뇌교의 정상 자기공명분광 소견: 같은 장비내 및 다른 장비간의 결과 비교)

  • Purpose: The purposes of this study are to evaluate normal MR spectroscopy (MRS) data of the cerebellum and pons, and to evaluate intra-unit and inter-unit differences of the results. Materials and methods: Nine normal volunteers (mean age 23 years) were studied with two 1.5 Tesla MR units (A and B units). A total of four sessions of MRS study were performed with two sessions in ea.ch MR unit in each volunteer. In each session, MRS data were obtained from bilateral cerebellar hemisphere and pons. MRS was performed using spin-echo single voxel technique with repetition time of 2000 ms, echo time of 288ms and 128 averagings. Voxel size was $2{\;}{\times}{\;}2{\;}{\times}{\;}1.5{\;}cm$ in the cerebellum and $1.5{\;}{\times}{\;}1.5{\;}{\times}{\;}1.5{\;}cm$ in the pons. Metabolite ratios of NAA/Cho, NAAcCr, NAAc(Cho+Cr) and Cr/Cho (both peak heights and the peak areas) were compared among the results of four sessions of MRS for evaluation of intra-unit and inter-unit differences. Results: In the cerebellum, mean (SO values of peak height NAA/Cho, NAA/Cr, NAA/(Cho+Cr) and Cr/Cho ratios obtained in the first session of A unit were $1.08{\pm}O.16,{\;}1.44{\pm}O.286,{\;}0.61{\pm}O.09{\;}and{\;}O.76{\pm}0.13$, respectively. Comparing with the results of the second session, intra-unit differences were 3-7%. In B unit intra-unit differences were 1-9%, except for 22% of Cr/Cho ratio. Inter-unit differences between A and B units were 1-26%. In the pons, mean ISD values of peak height NANCho, NANCr, NANICho + Cr) and Cr/Cho ratios obtained in the first session of A unit were $1.51{\pm}O.35,{\;}3.35{\pm}O.78,{\;}1.02{\pm}0.16{\;}and{\;}0.47{\pm}O.14$, respectively. Mean NAAfCho, NANCr and NAAI(Cho+ Cr} ratios of the pons were significantly higher than those of the cerebellum. Intra-unit differences were 2-21 % in A unit and 4-18 % in B unit. Inter-unit differences between A and B units were 1-18%. The mean values obtained by using peak area were generally higher and wider in range than those of peak height in both units. Conclusion: Mean NAAfCho, NAAlCr and NAAf(Cho+Cr) ratios of the cerebellum were significanHy lower than those of the pons. Intra-unit differences of most metabolites ratios were generally not significant, whereas inter-unit differences were greater than intra-unit differences in more metabolite ratios. Therefore, normal MRS data of the cerebellum and the pons should be obtained in every MR unit, before MRS applications in the diseases of the cerebellum and the pons.

  • The present experiment was carried out to examine the mechanism of cytotoxicity of Chromium in CHO cells. Chromium induced chromosomal aberrations in a dose-dependent manner. The most frequent type of aberration was chromatid deletions and chromosome type exchanges were also observed. Ultrafiltrates of culture media from CHO cells treated with Chromium induced sister chromatid exchanges(SCE) in CHO cells and Chromium induced lipid peroxidation. It was suggested that indirect effect through formation of clastogenic factor(CF) as well as direct effect on DNA might contribute to the cytotoxicity of Chromium.

  • Immigrant workers in Korea who have a multi-cultural background are increasing a need is arising to support them with housing that considers their housing culture. The purpose of this study is to gather information to formulate a plan of ChoSun-Tribe Immigrant Workers in Korea. For this purpose, we investigated to the usage of domestic space and the seating style of ChoSun-Tribe members in China and Korea. Ethnographic research with a questionnaire was used to analyze 16 households in China and 17 in Korea. The study results were as follows. 1. Most of the ChoSun-Tribe members in China and Korea thought that the kitchen had to be separated from other areas and also they wanted to have a large kitchen in which they can work comfortably. 2. ChoSun-Tribe members in China used a bathroom as a laundry normally while and ChoSun-Tribe members in China and Korea didn't think that a bathtub was indispensable in the bathroom. 3. The most uncomfortable feature for ChoSun-Tribe members in Korea was having to use a toilet outside so a toilet should be considered inside of the house. 4. ChoSun-Tribe members in China and Korea were accustomed to sitting on the floor for their living usually and ChoSun-Tribe members in China used the living room as a multi-purpose room for the family. 5. A modified Ondol system using water pipes under the floor was gaining popularity in China and was the most desirable heating system for ChoSun-Tribe members in China and Korea also. 6. ChoSun-Tribe members in China and Korea all took off their shoes inside of the house for hygienic reasons so the space for taking on-off shoes was indispensable. 7. Housing for ChoSun-Tribe immigrant workers in Korea needs to be planned with a good environment lots of sunshine and better ventilation.

Serum Levels of Xanthine Oxidase Activities in Cyclohexanone-Treated Rats Pretreated with Carbon Tetrachloride

  • To investigate an effect of cyclohexanone (CHO) treatment on the serum levels of xanthine oxidase (XO) in liver damaged animals, the rats were intraperitoneally pretreated with 50% carbon tetrachloride ($CCl_4$) in olive oil (0.1 mL/ 100 g body weight) 14 times every other day. To the $CCl_4$-pretreated rats, CHO (1.56 g/kg body weight) was injected once and then the animals were sacrificed at 4 hours after CHO treatment. The increasing rate of serum and liver XO activities to the control was higher in CHO-treated animals pretreated with $CCl_4$ than the $CCl_4$-pretreated those. Concomitantly CHO injection to the $CCl_4$-pretreated animals showed somewhat higher Vmax and lower Km value in the kinetics of liver XO enzyme. Furthermore, increasing rate of hepatic malonedialdehyde content to the control was also higher in CHO-treated animals pretreated with $CCl_4$ than $CCl_4$-pretreated those. On the other hand, the injection of CHO to the $CCl_4$-pretreated animals showed the more enhanced liver damage on the basis of liver function finding; liver weight per body weight (%), serum levels of alanine aminotransferase activity and hepatic glucose-6-phosphatase activity. In conclusion, injection of CHO to the $CCl_4$-pretreated rats led to more increased activity of serum XO and it may be caused by acceleration of hepatocyte membrane permeability and induction of enzyme protein.
