Does blastomere biopsy in preimplantation genetic diagnosis affect early serum ${\beta}$-hCG levels?

  • Objective: To determine whether the serum ${\beta}$-human chorionic gonadotropin (hCG) profile following preimplantation genetic diagnosis (PGD) is lower than that of intracytoplasmic sperm injection (ICSI) cycles. Methods: A total of 129 PGD cycles and 1,161 age-matched ICSI cycles, which resulted in pregnancy (serum ${\beta}-hCG{\geq}5$ mIU/mL) on post-ovulation day (POD) 12 were included. We compared the mean serum ${\beta}$-hCG levels on POD 12, 14, 21, and 28, doubling time of serum hCG, and created a cut-off value for predicting a singleton pregnancy in each group. Results: The mean serum ${\beta}$-hCG concentration of the PGD group was significantly lower than that of the control group on POD 12, 14, and 21. The doubling time of serum ${\beta}$-hCG at each time interval showed no significant difference. The cut-off-value of serum ${\beta}$-hCG for predicting a single viable pregnancy was 32.5 mIU/mL on POD 12 and 113.5 mIU/mL on POD 14 for the PGD group, which was lower than that for the control group. Conclusion: Blastomere biopsy may decrease the ${\beta}$-hCG-producing activity of the trophoblasts, especially in early pregnancy. Setting a lower cut-off value of serum ${\beta}$-hCG for predicting pregnancy outcomes in PGD may be needed.

Development and Immunochemical Properties of Two Monoclonal Antibodies Specific to Human Chorionic Gonadotropin

  • Using a hybridoma technique, spleen cells of Balb/c mice immunized with human chorionic gonadotropin (hCG) were fused with NS-1 mouse myeloma cells. Two hybrid cell lines, clones KS-8 and KS-19, secreting monoclonal antibodies to hCG, were isolated. KS-8 and KS-19 belong to the immunoglobulin $G_1$ subclass. With the aid of a double-antibody radioimmunoassay, it was established that the KS-8 monoclonal antibody recognizes an immunodeterminant of the $\beta$-subunit of hCG, whereas the KS-19 monoclonal antibody recognizes an epitope present on the $\alpha$-subunit of hCG. The KS-8 monoclonal antibody specifically reacts with human chorionic gonadotropin and shows cross-reactivity of less than 0.3% to other related human glycoprotein hormones. On the other hand, using a hemagglutination test based on antibody-induced agglutination of sheep red blood cells coated with hCG, It was shown that only the KS-19 monoclonal antibody was capable of inducing a positive reaction, although both monoclonal antibodies had similar binding capacity to the coated cells. The results from the dual screening procedures demonstrate that KS-8 and KS-19 monoclonal antibodies show high sensitivity in two different assays, and are hence useful for the qualitative and quantitative determination of hCG by both radioimmunoassay and hemagglutination inhibition tests.

Characteristic study on the chemical components of Korean curved ginseng products

  • Dried ginseng (DG) is in fact the representing ginseng product in the worldwide market. Although it is made in various packages depending on the processing method, size and age of DG, basic scientific data reporting the chemical components are limited. In this study, 4-year-old curved ginseng (CG), one of the domestic DG products, was selected for further investigation. Eighty-six samples of 30 and 50 piece-grade CG, which are the most widely distributed in the market, were collected for 5 yr. Their major components, such as moisture, total sugar, acidic polysaccharides, total phenolic compounds, and saponins, were analyzed to figure out the standard quality characteristics. The moisture content of all CG samples was less than 15%. The total water-soluble sugar contents were 22.9% to 47.8% and 23.2% to 49.5% in the 30 and 50 piece-grade CG, respectively. The acidic polysaccharide contents were 3.6% to 6.7% and 2.9% to 6.9% in the 30 and 50 piece-grade CG, respectively. The total phenolic compound content was 0.4% to 0.5% in CG, regardless of the piece-grade. The crude saponin content, which represents the active component of ginseng, was over 2% in all samples. In 30 piece-grade CG samples, the contents of major ginsenosides, Rb1, Rf, and Rg1, were 2.2 to 4.7 mg/g, 0.4 to 1.3 mg/g, and 1.6 to 4.0 mg/g, respectively. The ginsenoside contents in 50 piece-grade CG samples were 2.1 to 3.9 mg/g (Rb1), 0.5 to 1.2 mg/g (Rf), and 1.3 to 3.4 mg/g (Rg1). Overall, since there were relatively high standard deviation and coefficient of variation in all the chemical component contents that were assessed, we found some difficulties in showing the CG standard chemical component characteristics by average, standard deviation, and other statistical analysis factors.

무인차량용 단거리 라이다 시스템을 위한 멀티채널 트랜스임피던스 증폭기 어레이 (Multi-channel Transimpedance Amplifier Arrays in Short-Range LADAR Systems for Unmanned Vehicles)

  • 본 논문에서는 0.18um CMOS(1P6M) 공정을 이용하여 무인차량용 단거리 라이다 시스템을 위한 멀티채널 트랜스임피던스 증폭기(TIA) 어레이 회로를 구현하였다. 트랜스임피던스 증폭기 어레이 구조는 전압모드 $4{\times}4$ 채널 Inverter TIA 어레이와 전류모드 $4{\times}4$ 채널 Common-Gate(CG) TIA 어레이 두 가지를 설계했으며, 전체적으로 $4{\times}8$의 32-채널을 갖도록 설계하였다. 먼저, Inverter TIA는 피드백 저항을 가진 Inverter 입력구조와 CML 출력버퍼단으로 구성되어 있으며, 저잡음 및 저전력 특성뿐 아니라, virtual ground를 갖도록 설계함으로써 DC 전류조절이 가능하여 이득과 출력 임피던스 컨트롤이 가능하도록 하였다. 또한, CG-TIA는 on-chip bandgap reference로부터 bias 전압을 이용하고, 소스팔로워 출력버퍼를 사용하여 고주파수 이득을 높였으며, 기본적인 구조 상 CG-TIA는 채널당 칩 면적이 Inverter TIA에 비해 1.26배 작게 설계되었다. 포스트 레이아웃 시뮬레이션 결과, 제안한 Inverter TIA 어레이는 각 채널당 57.5-dB${\Omega}$ 트랜스임피던스 이득, 340-MHz 대역폭, 3.7-pA/sqrt(Hz) 평균 잡음전류 스펙트럼 밀도, 및 2.84-mW (16채널 45.4-mW) 전력소모를 가졌다. CG-TIA 어레이는 채널당 54.5-dB${\Omega}$ 트랜스임피던스 이득, 360-MHz 대역폭, 9.17-pA/sqrt(Hz) 평균 잡음전류 스펙트럼 밀도, 4.24-mW (16채널 67.8-mW) 전력소모를 가졌다. 단, 펄스 시뮬레이션 결과, CG-TIA 어레이가 200-500-Mb/s 동작속도에서 훨씬 깨끗하게 구분 가능한 출력펄스를 보였다.

돼지 수정란의 체외수정 및 발생에 미치는 호르몬 및 Glucose 첨가의 영향에 관한 연구 (Effects of Hormones and Glucose Levels during the In Vitro Culture in Medium on In Vitro Fertilization and Developmental Rates of Porcine Oocytes)

  • The study was conducted to determine the optimal hormone and glucose levels during the in vitro culture of bovine oocytes matured and fertilized in vitro for blastocyst development. Oocytes matured in TCM 199 + 10% FCS + hormones and glucose were fertilized in vitro in a TALP medium with swim up separated and heparin-treated epididymal cauda spermatozoa. Oocytes were cultured for 2~5 days in synthetic oviduct fluid medium (SOFM) supplemented with 10% FGS and with different hormone and glucose levels, and further cultured 5 days same medium in SOFM. The results are summarized as follows : The in vitro maturation and penetration rates of porcine oocytes cultured in TCM 199 media containing PMSG, hCG, PMSG + hCG, hCG + $\beta$ estradiol, PMSG + $\beta$ estradiol 0 to20 hours after insemination were 88.0% and 81.8%, 82.6% and 68.4%, 80.0% and 75.0%, 80.0% and 65.0%, 77.3% and 64.7%, respectively. The in vitro maturation and penetration rates of porcine oocytes cultured in TCM 199 media containing PMSG, hCG, PMSG + hCG, hCG + $\beta$ estradiol, PMSG + $\beta$ estradiol 20 to 40 hours after insemination were 92.0% and 87.0%, 92.0% and 82.6%, 91.3% and 81.0%, 85.2% and 73.9%, 87.5% and 81.0%, respectively. The cleavage and in vitro developmental rates to blastocyst of porcine oocytes cultured in TCM 199 media containing 0.05 mM, 0.10 mM, 0.30 mM, 0.50 mM, 1.00 mM, and 3.00 mM glucose lelvels 0~3 days after insemination were 31.5~48.1% and 10.0~16.7%, respectively. The cleavage and in vitro developmental rates to blastocyst of porcine oocytes cultured in TCM 199 media containing 0.05 mM, 0.10 mM, 0.30 mM, 0.50 mM, 1.00 mM, and 3.00 mM glucose levels 4~8 days after insemination were 30.0~53.8% and 8.7~19.2%, respectively. The cleavage and in vitro developmental rates to blastocyst were higher in TCM 199 media containing various glucose levels 0~3 days after insemination than 4~8 days.

PMSG와 hCG 병용 투여에 의한 Shih-tzu 견에서의 인공 발정 유기 (Effect of Estrus Induction in Shih-tzu Bitches by Administration of PMSG and HCG)

  • 무발정기에 속한 Shih-tzu 견 9두를 대상으로 하여 PMSG(50IU/kg)를 10일 동안 매일 근육주사 후 마지막 10일째에 hCG(1,000IU/head)를 정맥 주사하여 인공적으로 발정을 유기 하였다. 실험견 9두 중 9두(100%)에서 발정 출혈, 회음부 반사, 외음부 종대 및 수컷 허용 등의 임상적인 발정 증상이 관찰되었으며 그중 5두(55.6%)가 임신하였고 4두는 임신되지 않았다(44.4%). 임신견 5두 중 3두(33.3%)가 자연분만하였는데 분만시 산자수는 $1.66{\pm}1.15$$(1{\sim}3)$이었고, 나머지 2두(40.2%) 는 PMSG 첫 투여후 38일과 41일에 조기태아사 하였다. 따라서 Shih-tzu 견에서 PMSG와 hCG 병용 투여에 의한 인공 발정 유기 효과는 발정 발현율 100%, 임신률 55.6%, 분만율이 33.3%이었다.

손바닥 선인장 보충이 고지방식이 흰쥐의 간장, 신장 골격근에 지질괴산화와 SOD단백질 발현에 미치는 영향 (Effects of Opuntia humifusa Supplementation on Lipid Peroxidation and SOD Protein Expression in the Liver, Kidney, and Skeletal Muscle of Rats Fed a High-fat Diet)

  • 본 연구는 안정 시 고지방식이 흰쥐의 신장, 간장 그리고 골격근에서 지질과산화와 SOD 단백질 발현에 손바닥 선인장 보충이 미치는 효과에 대하여 연구하였다. SD계 수컷 흰쥐 16마리를 무작위로 대조군(CG, n=8)과 실험군(EG, n=8)으로 분류하였다. 8주 동안 대조군은 고지방식이를 부하하였으며, 실험군은 5% 손바닥선인장을 보충식 이하였다. 본 실험결과, 간장과 신장의 MDA 농도는 EG군이 CG군에 비해 유의하게 낮게 나타났다(p<0.01). 또한 골격근의 MDA 농도도 EG군이 CG군에 비해 높은 경향이 나타났으나, 통계적으로 유의한 차이는 나타나지 않았다. Cu,Zn-SOD 단백질 발현은 신장에서 EG군이 CG군에 비해 유의하게 증가하였다(p<0.01). Mn-SOD 단백질 발현은 골격근에서 EG군이 CG군에 비해 유의하게 증가하였다(p<0.01). 이상의 결과로부터 손바닥선인장 보충이 고지방식이 흰쥐의 지질과산화 억제작용과 SOD단백질 발현이 기관 특이적인 반응의 항산화 효과가 있는 것으로 나타났다.

CG-5 상대중력계를 이용한 중력관측 및 중력망조정에 관한 연구 (Research for Gravity Measurements Using CG-5 Autograv System and Network Adjustment)

  • 중력측량은 지구중력장 결정, 지각의 수직운동, 지오이드면의 변화, 해수면변화, 기후변화 등 측지학적, 지구동력학적 연구의 기초이다. 국토지리정보원에서는 최근 들어 FG-5 절대중력계를 도입하여 절대중력 기준망의 구축을 위한 틀을 마련하였고 다차원 다기능 기준점인 통합기준점을 전국에 약 1,200점을 설치하여 상대중력측량을 실시함으로써 한국의 고정밀 지오이드모델의 개발에 큰 기여를 할 것으로 판단된다. 본 연구에서는 상대중력측량의 기본적인 이론 및 방법을 상세하게 설명하였고, 최신 상대중력계 Scintrex CG-5를 이용하여 총 21점에 대한 상대중력측량을 수행하였으며, 자유망조정 및 무게제한 망조정 방법을 이용하여 중력성과를 계산 및 비교 분석하였다. 결과, 두 가지 방법 모두 높은 정밀도의 중력성과 계산이 가능하지만 자유망조정이 무게제한 망조정 방법에 비해 상대적인 우위를 보여주는 것으로 나타났다.

돼지 난포란의 체외성숙시 성선자극호르몬의 첨가가 체외성숙, 체외수정 및 배발생에 미치는 영향 (Effect of Gonadotropins added during Maturation of Porcine Oocytes on the In Vitro Maturation, In Vitro Fertilization and Development of Embryos)

  • This study was carried out to investigate the effects of gonadotropins added during maturation of porcine oocytes on the in vitro maturation(IVM), in vitro fertilization(IVF) and developmental potential of embryos. The follicular oocytes were cultured in TCM-199 medium containing different combination of gonadotropins(5$\mu$g /ml FSR or 1OIU /ml PMSG and 1O$\mu$g /ml LH or 1OIU /ml hCG), 10% FCS and 10% PFF for 36~48h in a incubator with 5% $CO_2$ in Air at 39$^{\circ}C$ and then matured oocytes were again cultured to 120h after IVF for 6~7h with heparin(100$\mu$g /m')-treated sperm. When the oocytes were matured for 42brs in the medium containing FSH+LH, FSH+hCG, PMSG+LH or PMSG+hCG, the JVF rate of each treatment was 50.0%, 52.9%, 66.7% and 70.0%, respectively. The highest CEI (cumulus cell expansion index) was obtained from PMSG+hCG-added medium and the highest polyspermic penetration resulted from FSH+LH-added medium. The cleavage of IVF oocytes derived from hormone added IVM was significantly(P<0.05) promoted by PMSG+hCG and the cleavage rate after 36-h, 42-h and 48-h maturation aws 53.0%, 56.7% and 45.6%, respectively. The highest developmental potential resulted from the oocytes derived from PMSG+LH -added IVM.

Construction of Antibodies for Detection and Diagnosis of Cucumber green mottle mosaic virus from Watermelon Plants

  • We immunized BALB/c mice with purified Cucumber green mottle mosaic virus isolate HY1 (CGMMV-HY1). Through the selection of positive clones that were grown on the HAT medium, four sensitive monoclonal clones (CG99-01, CG99-02, CG99-03, and CG99-04) were selected from 500 Hypoxanthine-guanine phosphoribosyltransferase positive hybridoma cells. Four sensitive clones of CGMMV-HYI were determined as IgM type of the subclass of mouse immunoglobulins Ig group. The titer of monoclonal antiserum against CGMMVHY1 was estimated 1:12,800 by the indirect ELISA. Although monoclonal antibodies (MAbs) from CG99-01 and from CG99-04 cross-reacted with Zucchini green mottle mosaic virus and Kyuri green mottle mosaic virus, MAb from the cell line CG99-03 was highly specific to CGMMV. No MAbs cross-reacted with Cucumber mosaic virus-Fny. Only CG99-04 reacted with Pepper mild mottle virus weakly and CG99-02 reacted with both CGMMV and KGMMV. CGMMV was detected from the rind of watermelon fruit by DAS-ELISA of CGMMV-HY1, but not from the flesh of watermelon. Average seed transmission rate of CGMMV in watermelon was $24\%$ from symptomatic watermelon collected from 5 regions of Gyeongnam province. CGMMV was detected by DAS-ELISA with specific MAb of CGMMVHY1 periodically from root stock, during the sequential process for nursery seedling in Haman. Necrotic spots on cotyledons of root stock seedling progressed to reveal the typical symptomatology on the primary leaves of scion upon grafting. Here, we have established MAb based ELISA system, which could accurately detect CGMMV from watermelon seeds, nursery seedlings, transplants and field samples from greenhouse or open out door field as well.
