Optical absorption of filter glasses colored by CdS, CdSe, ZnS, and ZnSe microcrystallites (CdS, CdSe, ZnS 및 ZnSe 미세결정을 이용한 filter용 유리의 광흡수특성)

  • The optical absorption characteristics of glasses colored by CdS. CdSe, $CdS_{1-x}Se_x$,ZnS, and ZnSe microcrystallites have been studied with emphasis on changes of the cut-off wavelength. The sharpcut filters with various cut-off wavelengths were fabricated by controlling the composition and the heat-treatment temperature. The cut-off wavelength shifts due to the different heat-treatment temperatures for CdS. CdSe, ZnS, and ZnSe doped SK-16 glasses were found to be relatively small(~30 nm). However, by treating the CdS1,Se, doped SK-16 glasses with different x values at the same heattreatment temperature, it was possible to obtain the filters with various cut-off wavelengths(~130 nm) without losing the high extinction coefficients of the filters. The filter glasses with various cut-off wavelngths(~100 nm) and high extinction coefficiencts were also produced by treating the CdSe and $CdS_{0.5}Se_{0.5}$ doped ZK-1 glasses at different temperatures.

The Study on Absorption of $Cd^{2+}$ and $Pb^{2+}$ by Persicaria thunbergii and Rumex crispus, and Their Phytochelatin (고마리와 소리쟁이에 의한 $Cd^{2+}$$Pb^{2+}$의 흡수 및 phytochelatin에 관한 연구)

  • When Persicaria thunbergii and Rumex crispus were treated with Cd($NO_3$)$_2$ and Pb($NO_3$)$_2$ of 5 or 10 mM for 5 days, the amount of bioaccumulation of $Pb^{2+}$ in the leaf of P. thunbergii was 2.87-8.08$\mu\textrm{g}$/g and that of $Cd^{2+}$ was 0.82-2.79$\mu\textrm{g}$/g. In the case of P. thunbergii, the concentration of $Pb^{2+}$ in the leaf was higher than that of $Cd^{2+}$. On the other hand, in R. crispus, the concentration of $Cd^{2+}$ and $Pb^{2+}$ were similar as follows ; 1.49$\mu\textrm{g}$/g in $Cd^{2+}$ 5mM, 2.90$\mu\textrm{g}$/g in Cd2+ 10mM, 1.83$\mu\textrm{g}$/g in $Pb^{2+}$ 5mM and 2.73$\mu\textrm{g}$/g in $Pb^{2+}$ 10mM. The remaining rate of heavy metals and the variation of pH in the cultured soil decreased as compared with control (100 % and pH 6.48) after 5 days as follows; to 77.l% and pH 6.39 in $Cd^{2+}$ 5mM, 90.2% and pH 5.79 in $Cd^{2+}$ 10 mM, 81.1% and pH 6.00 in $Pb^{2+}$ 5mM, and 85.7% and pH 5.80 in $Pb^{2+}$ 10 mM. The result of size exclusion chromatography, several phytochelatins were seperated from the extract of the leaf of both plants treated with heavy metals. The molecular mass of these phytochelatins were estimated as follows; in the case of P. thunbergii, about 4,300-8,600 da by $Cd^{2+}$ and about 3,200-9,700 da by $Pb^{2+}$, and in R. crispus, about 4,300 da by $Cd^{2+}$ and about 3,200-7,500 da by $Pb^{2+}$. Effect of Dietary β-Cyclodextrin on Egg Quality and Cholesterol Content of Egg Yolks (난황 콜레스테롤 및 계란 품질에 관한 식이내 베타-사이클로덱스트린의 효과)

  • This study was to determine the effect of dietary $\beta$-cyclodextrin ($\beta$CD) on egg quality and cholesterol content of egg yolk. One-hundred 28-week-old laying hens were randomly distributed into five groups of twenty hens each and maintained in individual laying cages for 10 weeks of the experiment. They were assigned to the five treatments of experimental layer diets containing 0, 3, 5, 7 or 10% pure $\beta$CD of diets. Food intake, egg production and egg weight varied slightly and insignificantly in the hens fed with 0, 3, 5 or 7% $\beta$CD-added diets. But food intake, egg production, and egg weight were uniformly lower (p<0.05) in the hens fed with 10% $\beta$CD-added diets. Food conversion rate, when expressed per number egg, was impaired (p<0.05) without having an obvious relation to the dietary $\beta$CD level. No difference in haugh unit, egg yolk color, and egg shell thickness was found among treatment. The cholesterol content of egg yolks was significantly decreased (p<0.05) to 3.76, 5.12, 5.49 and 6.48 mg/g yolk in $\beta$CD 3, 5, 7 and 10% treatments respectively when compared to that of the $\beta$CD 0% group. The cholesterol content of eggs, when expressed in mg/60 g egg, was greatly (p<0.05) decreased to 54, 71, 77 and 86 mg in $\beta$CD 3, 5, 7 and 10% treatments when compared to that of the $\beta$CD 0% group. The most striking effects (p<0.05) were observed in $\beta$CD 5% and 7% treatments, whose cholesterol content decreased from 28.19% to 30.23%. In conclusion, $\beta$CD added-diets fed to laying hens can reduced the cholesterol content of egg yolk, and this new type of egg yolk with lower cholesterol can be considered a functional livestock food.

Influence of Toxic Heavy Metals on Germination of Rice Seeds and Growth of Rice Seedling (수도생육(水稻生育)에 대한 유해(有害) 중금속(重金屬)의 영향(影響) - 발아 및 묘대기(苗垈期) 생육(生育)에 대하여 -)

  • Rice seeds, suweon 264, were germinated under 5 levels of toxic heavy metals, Cd(0, 0.05, 0.5, 5, 20ppm), Cu(0, 0.05, 0.5, 5, 20, ppm), Cr(0, 0.5, 1, 5, 10ppm), Ni(0, 0.5, 1, 5, 10ppm), Co(0, 0.5, 1, 5, 10ppm), Zn(0, 0.5, 5, 20, 40, ppm), Pb(0, 0.5, 5, 20, 40ppm) and Mn(0, 1, 10, 25, 50, ppm) in culture solution, and then grown with supplying culture solution contained respective concentrations. Germination and growth response to the toxic heavy metals were studied. Results obtained are as follows : 1) The germination injury of rice seeds by excess concentration of toxic heave metal in culture solution occured in Cd and Cu; below 0.05 ppm, Ni; below 0.5 ppm, Mn; below 1.0 ppm, Co and Cr; 0.5-1.0 ppm, and 0.5-5 ppm, Zn and Pb. Thereby, in the order of degrees of the elements toxicity to germination, they were arranged as follows : Cd>Cu>Ni>Co>Cr>Mn>Zn$$\geq_-$$Pb. 2) Toxic heavy metal concentrations in culture solution, which result in decreasing dry weight due to the injury of excess concentration of treated elements, were Cd: below 0.05 ppm, Ni, Cr and Co; below 0.5 ppm, Cu and Zn; 0.5-5 ppm, Pb; 5-20 ppm and Mn; 10-25 ppm. The order was Ni>Cd>Cr>Co>Cu$$\geq_-$$Zn>Pb>Mn. 3) The critical contents of Cd, Ni, Pb, Cu, Zn, Mn, and Co in dry matter, Which result in decreasing dry weight, were considered to be 0.05-15.5, 1.50-25.0, 24.0-28.0, 26.5-62.5, 470-645.0, 231.0-500.0 and below 15.0 ppm, respectively. 4) The contents of Cr, Co, Cd, Pb, Ni, Cu and Zn in dry matter by 0.5 ppm treatment concentration of each heavy metals was trace, 15.0, 17.5, 24.0, 25.0, 84.5 and 470.0 ppm, respectively. Thereby, in the order of each element to uptaked by rice seedlings, they were arranged as follow; Zn>Cu>Ni>Pb>Cd>Co>Cr. 5) The hazardous concentrations of root activity by toxic heavy metals in culture solution were Cd; below 0.05, Cu; 0.05-0.5, Cr; below 0.5, Ni; 0.5-1.0, Co; 0.5-1.0, Zn; above 0.5, Pb; 0.5-5.0 and Mn; 1.0-10.0 ppm. The hazardous degree of root activity by toxic heave metals was in the order of Cd>Cu>Cr>Zn>Ni>Co>Pb>Mn.

The Initial Stages of the Oxidation of the CdTe surfaces (CdTe 표면의 산화과정의 초기단계)

  • By means of X-ray photoelectron spectroscopy(XPS), the initial stages of the oxidation of the cleaved CdTe (110) surface and the sputtered CdTe surface with oxygen exposure are invetigated. From the analyses of the spectra of Te 3dsn, Cd 3d5/2, 0 ls and Cd MNN Auger lines, it is shown that two oxygen atoms bond to one Te atom at the initial stages.

Effect of CdTe Deposition Conditions by Close spaced Sublimation on Photovoltaic Properties of CdS/CdTe Solar Cells (CdTe박막의 근접승화 제조조건에 따른 CdS/CdTe 태양전지의 광전압 특성)

  • CdTe films were deposited by close spaced sublimation with various substrate temperatures, cell areas, and thicknesses of CdTe and ITO layers and their effects on the CdS/CdTe solar cells were investigated. The resistivity of CdTe layers employed in this study was 3$\times$ $10^{4}$$\Omega$cm For constant substrate temperature the optimum substrate ternperature for CdTe deposition was $600^{\circ}C$. To obtain larger grain size and more compact microstructure, CdTe film was initially deposited at 62$0^{\circ}C$, and then deposited at 54$0^{\circ}C$. The CdTe film was annealed at 62$0^{\circ}C$ and $600^{\circ}C$ sequentially to maintain the CdTe film quality. The photovoitaic cell efficiency improved by the "two-wave" process. For constant substrate temperature, the optimum thickness for CdTe was 5-6$\mu m$. Above 6$\mu m$ CdTe thickness, the bulk resistance of CdTe film degraded the cell performance. As the cell area increased the $V_{oc}$ remained almost constant, while $J_{sc}$ and FF strongly decreased because of the increase of lateral resistance of the ITO layer. The optimum thickness of the ITa layer in this study was 300~450nm. In this experiment we obtained the efficiency of 9.4% in the O.5cm' cells. The series resistance of the cell should be further reduced to increase the fill factor and improve the efficiency.

The increased expression of CD99 in a differentiated neuroblastoma cell line (신경모세종의 분화에 따른 인체 CD99의 표현의 증가)

  • Background: The human mic2 gene is a pseudoautosomal gene that encodes a cell surface antigen, CD99. High levels of CD99 constitute a tumor marker in Ewing s sarcoma (ES). We have recently demonstrated that CD99-induced apoptosis occurs only in undifferentiated ES cells, not in differentiated ES cells, raising the possibility of the involvement of CD99 in neural ontogeny. Methods: To elucidate the relations between the expression of CD99 and the differentiation of neural cells and the mechanism by which the expression of CD99 is regulated, we analyzed the differential patterns of CD99 expression in SH-SY5Y by treatment of 12-O-tetradecanoyl-13-phorbol acetate (TPA) and retinoic acid. In addition, to explore the transcriptional activity of CD 99 during neural cell differentiation, SH-SY5Y cells were transiently transfected with a CD99 promoter-driven luciferase construct, and treated with the inducers. Results: In immunoblotting and flow cytometry, the expression level of CD99 was increased on differentiated SH-SY5Y cells induced by TPA and retinoic acid. The luciferase activity was elevated by the treatment with TPA, known to mature SH-SY5Y cells toward a sympathetic neuronal lineage, whereas retinoic acid inducing a sympathetic chromaffin lineage displayed little effect. Conclusion: The result indicates that CD99 might be expressed only on cells maturing toward a neuronal lineage among differentiating primitive neuronal cells. In addition, the expression of CD99 seems to be regulated at the transcriptional level during the differentiation.

A Study on the Cd Accumulation and Tolerance of Pisolithus Tinctorius in Vitro (배양기내(培養器內)에서 모래밭버섯균(菌)의 Cd 축적(蓄積)과 내성(耐性)에 관(關)한 연구(硏究))

  • This study was conducted to test the Cd accumulation and Cd-tolerance of Pisotithus tinctorius(Pt). Pt was isolated from Pinus thunbergii forest in Muan, Chonnam Province in 1997. Pt was cultured on MMN medium supplemented with $CdSO_4{\cdot}5H_2O$ at the final concentration of 0, 0.2, 0.5, 2, and $10{\mu}g/m{\ell}$ for 40 days. Growth rate and tolerance index of the fungus were measured every week, while Cd concentration, superoxide dismutase(SOD), and glutathione reductase(GR) of the fungus were analyzed at the end of the culturing, Pt showed growth reduction in vitro at $2{\mu}g/m{\ell}$ Cd in the medium and almost stopped growth at $10{\mu}g/m{\ell}$ Cd. Tolerance index of Pt decreased with increasing Cd concentration. Cd concentration of Pt was the highest at $10{\mu}g/m{\ell}$ Cd. Activities of SOD did not show significant difference between Cd concentrations, but GR of Pt increased at $0.5{\mu}g/m{\ell}$ Cd, and decreased at $2{\mu}g/m{\ell}$ Cd. Consequently Pt could be called Cd accumulator with a tolerance mechanism to Cd. Their tolerance to Cd were expressed through the higher production of antioxidants such as GR. Pt may be used for revegetation and decontamination of soil polluted by heavy metals.

CD43 Expression Regulated by IL-12 Signaling Is Associated with Survival of CD8 T Cells

  • Background: In addition to TCR and costimulatory signals, cytokine signals are required for the differentiation of activated CD8 T cells into memory T cells and their survival. Previously, we have shown that IL-12 priming during initial antigenic stimulation significantly enhanced the survival of activated CD8 T cells and increased the memory cell population. In the present study, we analyzed the mechanisms by which IL-12 priming contributes to activation and survival of CD8 T cells. Methods: We observed dramatically decreased expression of CD43 in activated CD8 T cells by IL-12 priming. We purified $CD43^{lo}$ and $CD43^{hi}$ cells after IL-12 priming and analyzed the function and survival of each population both in vivo and in vitro. Results: Compared to $CD43^{hi}$ effector cells, $CD43^{lo}$ effector CD8 T cells exhibited reduced cytolytic activity and lower granzyme B expression but showed increased survival. $CD43^{lo}$ effector CD8 T cells also showed increased in vivo expansion after adoptive transfer and antigen challenge. The enhanced survival of $CD43^{lo}$ CD8 T cells was also partly associated with CD62L expression. Conclusion: We suggest that CD43 expression regulated by IL-12 priming plays an important role in differentiation and survival of CD8 T cells.

Effects of Chitosanoligosaccharide on the Mouse Hepatotoxicity Induced by Cadmium (카드뮴으로 유발된 생쥐 간독성에 대한 키토산올리고당의 효과)

  • This research was conducted to determine the effects of chitosanoligosaccharide on liver poisoning induced by cadmium (Cd). Three groups of mice were used in this research. The group was only injected with cadmium (5.0 mg/kg; i.p.) (group Cd) and the other group was injected with cadmium and chitosanoligosaccharide (0.5% solution) at the same time (group Cd+Chi). In order to investigate the inhibitory action of chitosanoligosaccharide on liver damage, cadmium concentration in liver tissues and metallothionein (MT) concentration were relatively measured. In addition, histological observations were made to determine the morphologic injury of liver tissues. Cadmium concentration in liver tissues was drastically lower in groups Cd+Chi than in group Cd. MT concentration in liver tissues was lower in group Cd than in groups Cd+Chi. As the result of electron microscopic observation, mitochondria in group Cd showed a severe swelling phenomenon, RER fragment and ribosome dropout. However, in groups Cd+Chi, mitochondria with high electron density were distributed and RER forming a typical lamellae with ribosome was observed. From these results, cadmium toxicity on rat liver tissues could be lessened by chitosanoligosaccharide.
