Helicobacter pylori 감염 소아에서 위점막 면역반응 (Gastric mucosal immune response of Helicobacter pylori-infected children)

  • 목 적 : H. pylori의 감염은 세균의 병독 인자, 숙주 인자, 환경 인자가 복합적으로 작용하여 다양한 위장관 병변을 일으킨다. H. pylori의 병리 기전으로 지금까지는 주로 세균의 병독성에 초점을 맞추었으나 최근에는 숙주의 위점막 면역반응이 주목을 받고 있다. 소아는 술, 담배, 약물과 같은 환경 인자의 영향이 적어 연구에 적합한 대상이며 감염 초기를 반영한다는 측면에서 중요한 의의가 있음에도 불구하고 소아에서 위점막 면역반응에 관한 연구가 거의 없다. 이에 H. pylori 감염 소아에서 위점막 림프구를 분석하여 소아에서 H. pylori 감염으로 인한 국소 면역반응의 특징을 알아보고자 하였다. 방 법 : H. pylori 양성 소화궤양군 10명, H. pylori 양성 위염군 15명, H. pylori 음성 대조군 20명에서 얻은 위전정부 생검조직에서 hematoxylin-eosin 염색과 modified Giemsa 염색을 시행하여 개정된 시드니 체계에 따라 위염의 정도를 점수화하였다. 위점막에서 림프구 면역표현형을 알기 위해 CD3, CD4, CD8 T세포와 CD20 B세포에 대한 면역조직화학염색을 시행하였으며 점막 고유층에서는 400배 고배율 현미경 하에서 $0.0625m^2$ 당 양성 림프구 수를 기록하였고, 상피세포 내에서는 100개 상피세포 당 양성 림프구 수를 기록하였다. 결 과 : 림프구 침윤은 점막 고유층에서 상피세포 내보다 확연히 많았다. 점막 고유층에서 H. pylori 양성군에서 대조군에 비해 CD3, CD4, CD8 T세포와 CD20 B세포가 유의하게 증가하였고(P<0.01) H. pylori 양성 소화궤양군은 위염군과 달리 대조군에 비해 CD8 T세포가 유의하게 증가하였다(P<0.01). 한편, 상피세포 내에서는 H. pylori 양성 소화궤양군과 위염군 모두 대조군에 비해 CD4 T세포가 유의하게 증가하였다(P<0.01). H. pylori의 밀도, 다핵형 중성구의 활동성, 만성 염증 정도와 림프구 수는 점막 고유층에서 상피세포 내보다 더 밀접한 상관관계가 있었다. 결 론: H. pylori에 감염된 소아의 국소 면역반응은 주로 위점막 고유층에서 일어나며 T세포와 B세포가 함께 관여하였다. H. pylori 양성 소화궤양군에서 점막 고유층의 CD8 T세포가 유의하게 증가하여 임상질환과 점막면역의 연관성을 추정할 수 있었다. 향후 H. pylori 감염에서 국소 면역반응으로 야기되는 점막 손상, 연령과 인종에 따른 차이, 임상질환과의 연관성 등에 대하여 더 많은 연구가 이루어져야 할 것이다.

Phosphatase Ssu72 Is Essential for Homeostatic Balance Between CD4+ T Cell Lineages

  • Ssu72, a dual-specificity protein phosphatase, not only participates in transcription biogenesis, but also affects pathophysiological functions in a tissue-specific manner. Recently, it has been shown that Ssu72 is required for T cell differentiation and function by controlling multiple immune receptor-mediated signals, including TCR and several cytokine receptor signaling pathways. Ssu72 deficiency in T cells is associated with impaired fine-tuning of receptor-mediated signaling and a defect in CD4+ T cell homeostasis, resulting in immune-mediated diseases. However, the mechanism by which Ssu72 in T cells integrates the pathophysiology of multiple immune-mediated diseases is still poorly elucidated. In this review, we will focus on the immunoregulatory mechanism of Ssu72 phosphatase in CD4+ T cell differentiation, activation, and phenotypic function. We will also discuss the current understanding of the correlation between Ssu72 in T cells and pathological functions which suggests that Ssu72 might be a therapeutic target in autoimmune disorders and other diseases.

Expression of Epstein-Barr Virus Gene and Clonality of Infiltrated T Lymphocytes in Epstein-Barr Virus-associated Gastric Carcinoma

  • Background: Epstein-Barr virus associated gastric lymphoepithelioma-like carcinoma (LELC) is characterized by the intensive infiltration of lymphoid cells, the presence of EBV, and the better prognosis over typical adenocarcinoma. Thus, it was assumable that viral latent proteins may be responsible for the recruitment of a certain T cell repertoire to EBV-associated gastric carcinoma. Methods: To examine above possibility, EBV gene expression in gastric carcinoma tissues and usage of TCR among the tumor infiltrating lymphocytes were analyzed. Results: EBV specific DNA and EBERs RNA were detected in 4 out of 30 patients. RT-PCR analysis revealed that all 4 of EBV-positive tumor tissues expressed EBNA1 mRNA and BARTs and LMP2a was detected only one sample out of 4. However, the EBNA2 and LMP-1 transcripts were not detected in these tissues. $CD8^+$ T cells were the predominant population of infiltrating lymphocytes in the EBV-positive gastric carcinoma. According to spectra type analysis of infiltrating T cells, 10 predominant bands were detected by TCR $V{\beta}$ CDR3 specific RT-PCR from 4 EBV-positive tumor tissues. Sequence analysis of these bands revealed oligoclonal expansion of T cells. Conclusion: These findings suggest that clonally expanded T cells in vivo might be a population of cytotoxic T cells reactive to EBV-associated gastric carcinoma.

Natural Killer and CD8 T Cells Contribute to Protection by Formalin Inactivated Respiratory Syncytial Virus Vaccination under a CD4-Deficient Condition

  • Respiratory syncytial virus (RSV) causes severe pulmonary disease in infants, young children, and the elderly. Formalin inactivated RSV (FI-RSV) vaccine trials failed due to vaccine enhanced respiratory disease, but the underlying immune mechanisms remain not fully understood. In this study, we have used wild type C57BL/6 and CD4 knockout (CD4KO) mouse models to better understand the roles of the CD4 T cells and cellular mechanisms responsible for enhanced respiratory disease after FI-RSV vaccination and RSV infection. Less eosinophil infiltration and lower pro-inflammatory cytokine production were observed in FI-RSV vaccinated CD4KO mice after RSV infection compared to FI-RSV vaccinated C57BL/6 mice. NK cells and cytokine-producing CD8 T cells were recruited at high levels in the airways of CD4KO mice, correlating with reduced respiratory disease. Depletion studies provided evidence that virus control was primarily mediated by NK cells whereas CD8 T cells contributed to IFN-γ production and less eosinophilic lung inflammation. This study demonstrated the differential roles of effector CD4 and CD8 T cells as well as NK cells, in networking with other inflammatory infiltrates in RSV disease in immune competent and CD4-deficient condition.

Different Pattern of p27kip1 and p21cip1 Expression Following Ex Vivo Activation of CD8+ T Lymphocytes

  • T cell proliferation is a pivotal to an effective immune response. Cyclin-dependent kinase (cdk) inhibitor, $p27^{kip1}$ is degraded to initiate T cell expansion. In this study, we show that although the expression of $p27^{kip1}$ protein was down-regulated, that of $p21^{cip1}$, another cdk inhibitor, was up-regulated in $CD8^+$ T cells following in vitro stimulation. Ex vivo gB antigen-stimulation following HSV immunization increased $p21^{cip1}$ positive cells that co-expressed IFN-$\gamma$. Moreover, $p21^{cip1}$ was co-expressed with IFN-${\gamma}$ in E7 antigen-stimulated $CD8^+$ T cells, whereas $p27^{kip1}$ was not. Our findings imply a role of $p21^{cip1}$ proteins in antigen-induced effector $CD8^+$ T cells differentiation in vivo.

은교산의 면역계에 미치는 효과 (Effects of Eunkyo-San on the Immune System)

  • The purpose of this research was to investigate the effect of Eunkyo-San(EKS) on the immune system. Administration of EKS(500 mg/kg) enhanced viability of splenocytes and thymocytes in BALB/c mice, and also EKS increased of splenic B, T lymphocytes and thymic T lymphocytes, significantly increased CD4 positive TH cells. EKS markedly enhanced the production of -interferon in mice serum. EKS accelerated the production of nitric oxide in peritoneal macrophages. These results suggest that EKS have an immune-enhancing activity.

  • Granulomatous meningoencephalomyelitis (GME) is a sporadic, idiopathic, non suppurative inflammatory disease of the canine central nervous system. GME appears to have a worldwide distribution and to occur mostly in young to middle-age dogs of small breeds. A 6-year-old female mixed dog with wry neck, ataxia and rolling was submitted to the Cheju National University for diagnosis. Grossly, in the brain, cerebrospinal fluid was mildly increased and dilatation of the subarachnoid blood vessels was observed. Histopathologically, the lesions were characterized by perivascular cuffs of lymphocytes, various numbers of macrophages and plasma cells in the brainstem and cerebral white matter. Numerous granuloma composed of lymphocytes and histiocytes were scattered throughout the brainstem. Two malacic foci characterized by axonal swelling and gitter cell infiltration with hemorrhage were noted in the medulla oblongata and cerebellum. Special stains failed to demonstrate any infectious agents. Immunohistochemically, the infiltrated cells demonstrated strong positive reactions for CD3, a marker for T lymphocytes origin. Based on the clinical signs, histopathology, and immunohistochemistry, this case was diagnosed as GME in a mixed dog.

녹용 에탄올 분획이 생쥐의 T-Lymphocyte에 미치는 영향 (Effect of Ethyl Alcohol Fraction of Cervus nippon on Mouse T-Lymphocyte)

  • In this study, the effect of 70% ethyl alcohol fraction of Cervus nippon(CN-E) on mouse T-lymphocyte was investigated in vivo. The administration of CN-E(100 mg/kg) enhanced the proliferation of thymocytes, the population of $CD4^+CD8^-$ single-positive cells and the production of $interferon-{\gamma}$ in thymocytes and splenocytes. The administration of CN-E did not induce DNA fragmentation and reduce mitochondrial transmembrane potential in thymocytes. These results indicate that the CN-E contams a stimulative component on the proliferation of thymocytes, the population of $T_H$ cells and the production of $interferon-{\gamma}$ in T-lymphocytes.

  • 와송 (OJB)의 면역 및 항암 효과를 관찰한 결과, 와송은 생쥐의 비장 및 흉선세포의 생존율을 증가시켰으며, 임파구 아집단분석 결과 비장 T세포를 유의성있게 증가시켰는데 비장 T세포 중의 $T_H$세포 및 Tc세포가 모두 증가되었고, 혈청 중 ${\gamma}-interferon$의 생성을 현저하게 증가시켰다. 또한 와송은 L1210세포 및 U937세포 등의 백혈병세포의 아폽토시스를 촉진시키는 항암능력을 보유하고 있었다.

Deletion Timing of Cic Alleles during Hematopoiesis Determines the Degree of Peripheral CD4+ T Cell Activation and Proliferation

  • Capicua (CIC) is a transcriptional repressor that regulates several developmental processes. CIC deficiency results in lymphoproliferative autoimmunity accompanied by expansion of CD44hiCD62Llo effector/memory and follicular Th cell populations. Deletion of Cic alleles in hematopoietic stem cells (Vav1-Cre-mediated knockout of Cic) causes more severe autoimmunity than that caused by the knockout of Cic in CD4+CD8+ double positive thymocytes (Cd4-Cre-mediated knockout of Cic). In this study, we compared splenic CD4+ T cell activation and proliferation between whole immune cell-specific Cic-null (Cicf/f;Vav1-Cre) and T cell-specific Cic-null (Cicf/f;Cd4-Cre) mice. Hyperactivation and hyperproliferation of CD4+ T cells were more apparent in Cicf/f;Vav1-Cre mice than in Cicf/f;Cd4-Cre mice. Cicf/f;Vav1-Cre CD4+ T cells more rapidly proliferated and secreted larger amounts of IL-2 upon TCR stimulation than did Cicf/f;Cd4-Cre CD4+ T cells, while the TCR stimulation-induced activation of the TCR signaling cascade and calcium flux were comparable between them. Mixed wild-type and Cicf/f;Vav1-Cre bone marrow chimeras also exhibited more apparent hyperactivation and hyperproliferation of Cic-deficient CD4+ T cells than did mixed wild-type and Cicf/f;Cd4-Cre bone marrow chimeras. Taken together, our data demonstrate that CIC deficiency at the beginning of T cell development endows peripheral CD4+ T cells with enhanced T cell activation and proliferative capability.
