Some of endocrine disruptors with sexual hormone-like effects have been increasingly reported to be immunotoxic in many species in recent several years. Phthalate esters have possible effects on the endocrine system. Prenatal exposure to di(n-butyl) phthalate (DBP) has been reported to impair the androgen-dependent development of the male reproductive tract in rat. Therefore, the immunomodulatory effect of DBP was investigated in the developing immune system of fetal and neonatal Sprague-Dawley rats. Timed-bred pregnant SD rats were given to the doses of 0, 250, 500, and 750 mg DBP/kg$\cdot$ body weight /day by gavage once a day from gestational day (GD) 5 to 18. On GD19 or GD22/postnatal day one (PD1), the dams were euthanized, and the changes in organ weights and thymus phenotypes were examined for their offsprings. At 750 mg DBP/kg$\cdot$b.w./day in maternal exposure group, GD19 fetuses showed decreases in body weight. The spleen/body weight ratios were reduced in GD 19 fetuses from the dams exposed to 500 and 750 mg DBP/kg$\cdot$b.w./day. There were no significant changes in thymus and spleen cellularities though these cellularities showed a tendency to decrease in a dose dependent way. In the DBP-exsposed GD22/PD1 offsprings, the body weights, the relative organ weights and the cellularities did not exhibit alteration. Additionally, the percentages of CD3$^{+}$(CD4$^{+}$CD8$^{+}$, CD4$^{+}$CD8$^{-}$, CD4$^{-}$CD8$^{+}$, and CD4$^{-}$CD8$^{-}$) and CD3$^{-}$(CD4$^{+}$CD8$^{+}$, CD4$^{+}$CD8$^{-}$, CD4$^{-}$CD8$^{+}$, and CD4$^{-}$CD8$^{-}$) thymocyte subsets were not changed in any DBP-treated group. The proliferative responses of splenic T cells to Con A and B cells to LPS were decreased in all DBP-exposed GD22/PD1 offsprings.
Objective: The purpose of this research is to examine the effects of Mahaenggamseok-tang-gagambang (MGTG) on CD3+ T cells, CD4+ T cells and CD8+ T cells in ovalbumin (OVA)-induced asthmatic mice. Methods: C57BL/6 mice were injected, inhaled and sprayed with OVA for 12 weeks (four a week) for asthma induction. Two experimental groups were treated with different concentrations of MGTG (400 mg/kg and 200 mg/kg) extract and cyclosporin A (10 mg/kg) for the later 8 weeks. At the end of the experiment, the mice lung was removed and analyzed CD3+ T cells, CD4+ T cells and CD8+ T cells by flow cytometer. Results: Numbers of CD3+ T cells in lung of the MGTG groups (200, 400 mg/kg) were significantly decreased compared with that of control group. Numbers of CD4+ T cells in lung of the MGTG groups (200, 400 mg/kg) were significantly decreased compared with that of control group. Numbers of CD8+ T cells in lung of the MGTG groups (200, 400 mg/kg) were significantly decreased compared with that of control group. Conclusion: The results of this study suggest that MGTG alleviated asthmatic hyperreactiviry through CD4+ and CD8+ T cells. Further study of relative cytokines is expected.
Background: To explore the prevalence of lymphocyte subgroups $CD3^+$$CD4^+$ and $CD3^+$$CD8^+$ and their surface receptors NKG2D and NKG2A in patients with non-small cell lung cancer (NSCLC). Materials and Methods: A total of 40 patients with NSCLC were divided into different groups according to different clinical factors (TNM staging, pathological patterns and genders) for assessment of relations with $CD3^+$$CD4^+$ and $CD3^+$$CD8^+$ and the surface receptors NKG2D and NKG2A of T lymphocytes in peripheral blood by flow cytometry. Results: Patients in the advanced group had evidently lower levels of $CD3^+$$CD4^+$ but markedly higher levels of $CD3^+$$CD8^+$ in peripheral blood than those with early lesions (p<0.05). In addition, NSCLC patients in the advanced group had obviously higher $CD3^+$$CD4^+$ NKG2D and $CD3^+$$CD8^+$ NKG2A expression rates but lower $CD3^+$$CD4^+$ NKG2A and $CD3^+$$CD8^+$ NKG2D expression rates (p<0.05). However, there were no significant differences between NSCLC patients with different genders and pathological patterns in expression levels of lymphocyte subgroups $CD3^+$$CD4^+$ and $CD3^+$$CD8^+$ and their surface receptors NKG2D and NKG2A. Conclusions: Unbalanced expression of surface receptors NKG2D and NKG2A in $CD3^+$$CD4^+$ and $CD3^+$$CD8^+$ lymphocytes may be associated with a poor prognosis, greater malignancy and immunological evasion by advanced cancers, related to progression of lung cancer.
악성종양의 발생과 진행에 면역 방어기능이 중요한 역할을 하리라는 가설은 모두가 공감하는 사실이다. 이중 T-lymphocyte와 Natural killer cell (이하 NK cell이라함)은 종양 면역학에 특히 중요한 임파구로 이런 임파구의 혈액분포양상은 면역방어기능을 짐작할 수 있는 간접적인 자료가 될 수 있다. 저자들은 치료전 두경부 악성종양환자에서 혈액을 채취하여 T-lymphocyte와NK cell의 분포양상을 검사하고, 방사선치료 환자에서는 NK cell activity를 측정하였기에 다음과 같은 결과를 보고하는 바이다. 1) 두경부 악성 종양 환자군에서 CD3+ cell은 감소하고 NK cell은 증가하며 CD4/CD8 비율은 변화가 없었다. 2) 병변이 진행되면서 CD3+ cell과 CD4+ cell은 감소하고 NK cell은 증가하였으며 CD4/CD8 비율의 변화는 없었다. 3) 방사선치료에 의해 CD3+ cell과 CD4+ cell, CD4/CD8 비율은 감소하였고, NK cell과CD8+cell은 증가하였다. 4) 방사선치료에 의한 CD4/CD8 비율의 감소와, CD8+ cell의 증가는 NK cell의 증가에 의한 것이라 추정되고, NK cell을 제외하면 CB4/CD8 비율의 변화는 없었다. 5) 방사선치료 환자에서 NK cell activity는 증가하였고, 이런 증가가 T-lymphocyte기능의 감소를 보상해 주고 있었다.
고려홍삼이 HIV-1 감염자에게서 바이러스의 활발한 증식으로 유발된 면역계의 과활성화를 장기간에 걸쳐 억제하는지 살펴보기 위해 면역 활성화 지표인 soluble CD8항원(sCD8)을 측정하였다. HIV-1 감염 대조군 49명, zidovudine (ZDV) 복용군 22명, KRG 복용군 48명, KRG와 ZDV 병용군(49명)을 대상으로 하였다. HIV-1 감염 대조군에서는 혈청 sCD8 농도가 31-48개월 동안 33% 증가하였으며(P < 0.05), sCD8/CD8+T 림프구 비도 $21{\pm}13$개월동안 54% 증가하였다(P < 0.001). ZDV 복용군에서는 혈청 sCD8농도가 초기 6개월 동안은 감소하였으나 그 이후로는 서서히 증가하였으며, sCD8/CD8+T 림프구 비도 증가하였다. KRG 복용군은 sCD8 농도가 31-48개월 동안 45% 감소하였으며(P < 0.01), sCD8/CD8+T 림프구 비도 $19\;{\pm}\;11$개월 동안 19% 감소하였다(P < 0.05). 병용군에서는 sCD8 농도가 31-48개월 동안 29%(P < 0.01) 감소하였다. 추적관찰 기간 동안 각 군간의 sCD8 농도 변화가 명확한 차이를 보였으며, KRG 복용군에서는 ZDV 복용군에서 관찰된 'rebound현상'(sCD8이 초기에 감소하다가 다시 증가하는 경향)이 없었다. 결론적으로 본 연구에서는 HIV-1 감염환자에서 KRG를 장기간 복용시켰을 경우 sCD8 농도가 지속적으로 감소하여, KRG가 면역계의 소모적인 과활성화 상태를 방지함을 알 수 있었다.
Background: Toll-like receptors (TLRs) play a fundamental role in innate immunity through their capacity to recognize pathogen-associated molecular patterns. Also, TLRs that are expressed in T cells are reported to function as co-stimulatory receptors. However, the functional capacity of TLRs on CD4 T and CD8 T cells has not been directly compared. Here we compared CD4 and CD8 T cell responses to TLR2 ligand plus TCR-mediated stimulation. Methods: TLR2 expression was analyzed on T cell subsets under naive and alloantigen-primed conditions. We analyzed the effects of TLR2 co-stimulation on proliferation and survival of T cell subsets in vitro when stimulated with soluble anti-CD3 in the presence or absence of synthetic ligand $Pam_3CSK_4$. Results: TLR2 expression on CD8 T cells was induced following activation; this expression was much higher than on CD4 T cells. Thus, the molecule was constitutively expressed on Listeriaspecific memory CD8 T cells. Based on these expression levels, proliferation and survival were markedly elevated in CD8 T cells in response to the TLR2 co-stimulation by $Pam_3CSK_4$ compared with those in CD4 T cells. Conclusion: Our data show that TLR2 co-stimulation is more responsible for proliferation and survival of CD8 T cells than for that of CD4 T cells.
Breast cancer is the most common cancer among the population as a whole and among females, yet it is highly curable if diagnosed at an early stage. Different methods are used to diagnose breast cancer. One of these methods features immunological tests using flow cytometry to determine T-lymphocyte (CD4/CD8) ratios in peripheral blood. One hundred patients with breast cancer (50 from Basra, Iraq, and 50 from Baku, Azerbaijan) confirmed to have breast cancer by histopathology were studied. Blood samples were collected from all patients before initiation of treatment and were used for analysis. The mean age of women from Basra was $51.2{\pm}10.7years$ and that of women from Baku was slightly higher at $54.8{\pm}12.2$. The mean CD4/CD8 ratio in Basra was 1.4 and in Baku was 1.8 with P value < 0.05. The percentage of Basra patients who have CD4/CD8 value less than 1 was 50%, while the percentage for Baku patients was 24 % (p < 0.05). While the CD4/CD8 T-lymphocyte ratio might be useful for early diagnosis in patients with breast cancer parallel with other confirmed tests factors involved in explaining variation between countries such as that observed here need to be taken into account.
Objectives : In order to investigate the effect of Bobitang on SD rats with deteriorated immunity caused by methotrexate. Methods : The test sample were dosed once a day for 14 days by gastric gavage at a dosage 1,000, 500 and $250mg/kg/10m{\ell}$ from 2 days after last MTX-dosing, and the changes on body weight and gains, spleen weight and total blood leukocyte numbers, total lymphocyte numbers, the percentage of B-cell, T-cell, CD3+CD4+ T-cell, CD3+CD8+ T-cell and CD4+/CD8+ T-cell ratios in the blood and spleen were observed. Results : The changes on body weight gains, the spleen weight, the total blood leukocyte numbers, the total lymphocyte numbers in the blood and spleen, the ratio of T-cell in the blood and spleen, the ratio of CD3+CD4+ T-cell in the blood and spleen were increased significantly in BBT Extracts groups as compared with control group. The ratio of B-cell in the blood and spleen was not increased significantly in BBT Extracts groups as compared with control group. The percentage of CD3+CD8+ T-cell in the blood and spleen was decreased significantly in BBT Extracts groups as compared with control group. The ratio of CD4+/CD8+ T-cell in blood and spleen was increased significantly in BBT Extracts group as compared with control group. Conclusions : According to the above results, Bobi-Tang has an effect of increasing immune responses on SD rats with deteriorated immunity caused by methotrexate.
Objective: Cluster of differentiation 4 protein (CD4) gene is an important immune related gene which plays a significant role in T cell development and host resistance during viral infection. Methods: In order to unravel the relationship of CpG island methylation level of CD4 gene with its gene expression and T lymphocyte subpopulation traits, we used one typical Chinese indigenous breed (Dapulian, DP) and one commercial breed (Landrace), then predicted the CpG island of CD4 gene, determined the methylation status of CpG sites by bisulfite sequencing polymerase chain reaction (BSP), and carried out the correlation analyses of methylation frequencies of CpG sites with mRNA expression and T lymphocyte subpopulation traits. Results: There was one CpG island predicted in the upstream -2 kb region and exon one of porcine CD4 gene, which located 333 bp upstream from the start site of gene and contained nine CpG sites. The correlation analysis results indicated that the methylation frequency of CpG_2 significantly correlated with CD4 mRNA expression in the DP and Landrace combined population, though it did not reach significance level in DP and Landrace separately. Additionally, 15 potential binding transcription factors (TFs) were predicted within the CpG island, and one of them (Jumonji) contained CpG_2 site, suggesting that it may influence the CD4 gene expression through the potential binding TFs. We also found methylation frequency of CpG_2 negatively correlated with T lymphocyte subpopulation traits CD4+CD8-CD3-, CD4-CD8+CD3- and CD4+/CD8+, and positively correlated with CD4-CD8+CD3+ and CD4+CD8+CD3+ (for all correlation, p<0.01) in DP and Landrace combined population. Thus, the CpG_2 was a critical methylation site for porcine CD4 gene expression and T lymphocyte subpopulation traits. Conclusion: We speculated that increased methylation frequency of CpG_2 may lead to the decreased expression of CD4, which may have some kind of influence on T lymphocyte subpopulation traits and the immunity of DP population.
