Protective Effect of Ethanol Extract of Artemisiae vulgaris L. on hepatic injury Induced by Carbon tetrachloride In Rat. (애엽 에탄올 추출물이 사염화탄소로 유발된 흰쥐의 간 손상 보호효과)

  • This study was done to investigate the protective effects of ethanol extract Artemisiae vulgaris L(Av) on carbon tetrachloride(CCl4)intoxicated rats. Male sprague Dawley rats(200~210g)was used. experimental groups were divided into normal group, CCl4-control group, and ethanol extract CCl4-treated group. CCl4-treated groups were injected with CCl4 0.6mg/kg.b.w(i.p). The activities of Alanine aminotransferase(ALT), Aspartate aminotransferase(AST), Alkaline phosphatase(ALP), Glutamyl transpeptidase(γ-GT), Lactate dehydrogenase(LDH) in extract pretrated group was significantly decreased(p<0.05) compared to the CCl4-control group. The contents of triglyceride, cholesterol and lipid peroxide were significantly decreased(p<0.05). whereas the contents of HDL-cholesterol and glutathione(GSH) were significantly increased(p<0.05). These results suggest that extract of Artemisiae vulgaris L(Av) has hepatoprotective effect in the CCl4-intoxicated rats.

The Measurement of Flash Point for Unflammable-Flammable Binary Mixtures(CCl4+o-Xylene and CCl4+p-Xylene) Using Open Cup Tester (개방식 장치를 이용한 난연성-가연성 이성분계 혼합물(CCl4+o-Xylene and CCl4+p-Xylene)의 인화점 측정)

  • The flash point is used to categorize inflammable liquids according to their relative flammability. The flash point is important for the safe handling, storage, and transportation of inflammable liquids. The flash point temperature of two binary liquid mixtures($CCl_4+o-xylene$ and $CCl_4+p-xylene$) has been measured for the entire concentration range using Tag open cup tester. The flash point temperature was estimated using Raoult's law, UNIQUAC model and empirical equation. The experimentally derived flash point was also compared with the predicted flash point. The empirical equation is able to estimate the flash point fairly well for $CCl_4+o-xylene$ and $CCl_4+p-xylene$ mixture.

Healing and preventive effects of low-esterified pectin on liver injury induced by carbon tetrachloride in rats

  • The purpose of this study was to investigate the pharmacological effects of low-esterified pectin on carbon tetrachloride $(CCL_4)-induced$ hepatotoxicity in rats. The study included two experiments. In the first experiment the animals were given daily $CCL_4$ through gavage for 7 days and then 10, 50, or 250 mg/kg b.w. of pectin for 21 days. At the end of experiment rats were killed within 24 hours. The increased bilirubin level, enhanced alanine aminotransferase and aspartate aminotransferase activity in plasma induced by $CCL_4$ were partly normalized by pectin administration in a dose-dependent manner. The pectin treatment also resulted in significant recovery of $CCL_4-induced$ decrease of the liver glycogen content. In addition, pectin significantly improved $CCL_4-induced$ alterations of pro-oxidant and antioxidant biochemical parameters in liver and plasma compared to those of rats administered $CCL_4$. In the second experiment the animals were given daily 10, 50 or 250 mg/ kg b.w. of pectin for 21 days before a 7-day administration of $CCL_4$. Rats were killed 24 hours after the end of experiment. Pretreatment with pectin before $CCL_4$ administration resulted in significantly inhibited increase of the blood enzymatic activities of alanine and aspartate aminotransferases and bilirubin level in a dose-dependent manner. Also, preliminary administration of pectin prevented elevation of malondialdehyde and conjugated diene levels in liver and plasma as well as a reduction of glutathione content in liver of rats given $CCL_4$. These results suggest that low-esterified pectin exert healing and preventive effects on $CCL_4-induced$ hepatotoxicity in rats.

Effect of A Combined Prescription on Liver Injury of Rats by $CCl_4$ (사역산합평위산가인진택사방(四逆散合平胃散加茵蔯澤瀉方)이 $CCl_4$에 의한 흰쥐의 간(肝) 손상에 미치는 영향)

  • Objectives : This study was undertaken to determine if a combined(SPe) has a protective effect against functional failure induced by $CCl_4$ in rat liver. Methods : Acute liver injury which initiated from free radical induced by $CCl_4$, were applied to rats and data were obtained. Liver injury was estimated by measuring aspartate aminotransferase(AST) and alanine aminotransferase(ALT) activity in serum. Lipid peroxidation was examined by measuring malondialdehyde, a product of lipid peroxidation. GSH activities in liver tissues were also measured. Results : When rats were treated intraperitoneally with $CCl_4$, serum AST and ALT were increased compared with the control, which was significantly inhibited by pretreatment of SPe. SPe also prevented reduction in GSH induced by $CCl_4$. Conclusion : Above results suggest that SPe exerts protective effect against $CCl_4$ by its antioxidant action in liver tissues. Thus, SPe may be used in prevention and treatment of drug-induced liver cell injury. However, the precise mechanisms of SPe protection remain to be determined.

  • Solvent fractions were prepared from traditional herbal drugs which of methanol extracts inhibited $CCl_4$-induced cytotoxicity in primary cultured rat hepatocytes and c ontinuously assayed their effects. Ethylacetate and n-buthanol fractions from Cibotii Rhizoma and chloroform fraction from Gelatina Nigra inhibited the release of LDH and GPT from $CCl_4$-treated hepatocytes, respectively. Water fraction (WAR) among solvent fractions from Astragali Radix showed the most potent inhibitory effect on the release of GOT or GPT by treatment with $CCl_4$. All of solvent fractions prepared from Eucommiae Cortex had no effect on $CCl_4$-induced cytotoxicity. Chloroform and ethylacetate fractions from Rehmanniae Radix Preparata increased the release of GPT from $CCl_4$-treated hepatocytes. n-Hexan, chloroform or ethylacetate fraction from 5 herbal drugs increased the release of LDH, GOT or GPT from normal hepatocytes at the dose of 1.Omg/ml. Administration of WAR suppressed the elevation of GOT, ALP activities and MDA contents in the serum as well as in the liver tissue of $CCl_4$-intoxicated rats. Based on these results, isolation of antihepatotoxic substances from WAR is under the process.

  • Artemisia Iwayomogi Compositae) has been used clinically for jaundice, hepatitis, liver cirrhosis etc. The purposes of present study were to examine pharmacological effects of Artemisia Iwayomogi water extract(AIWE) on biochemical parameters (activities of ALP and LAP, contents of glucose, total bilirubin, total protein and albumin in serum, A/G ratio, and levels of hepatic glycogen) against hepatic injury by carbon tetrachloride($CCl_4$) in rats. The results were as follows ; 1. Increased ALP activities by $CCl_4$ were very significantly(p<0.001) decreased in AIWE posttreatment groups at 72 hours and significantly(p<0.05) decreased in AIWE pretreatment groups at 72 hours. Increased LAP activities by $CCl_4$ were significantly (p<0.05) decreased in AIWE posttreatment groups at 72 hours. A little increased total bilirubin contents by $CCl_4$ were very significantly (p<0.001) decreased in AIWE posttreatment groups at 24, 48 and 72 hours. 2. Increased glucose contents by $CCl_4$ were decreased in AIWE posttreatment groups. Decreased hepatic glycogen levels by $CCl_4$, were significantly (p<0.05) increased in AIWE posttreatment groups at 48 and 72 hours. 3. Decreased total protein contents by $CCl_4$ were significantly (p<0.05) increased in AIWE posttreatment groups at 48, 72 hours. Decreased albumin contents by $CCl_4$ were increased in proportion to numbers of AIWE treatments in AIWE pre- and posttreatement groups. Decreased A/G ratios by $CCl_4$ were significantly (p<0.05) increased in AIWE posttreatment groups at 48 hours. In conclusion, AIWE did not affect normal liver function and had hepatoprotective effects rather than direct preventive effects to $CCl_4$-induced cholestasis, damages in metabolisms of glucose, protein and bilirubin.

Protective Effects of Extracts of Diospyrus kaki Folium against Hepatotoxicity in Carbon Tetrachloride Intoxicated Rats (감잎 추출물이 사염화탄소로 유발된 흰쥐의 간손상에 미치는 효과)

  • This study was done to investigate the protective effects of Diospyrus kaki Folium on carbon tetrachloride intoxicated rats. Male Sprague-Dawley rats (200~230g) was used. Experimental groups were divided into normal group and $CCl_{4}$-treated group and then $CCl_{4}$-treated groups were divided into 7 groups: only $CCl_{4}$-treated group, methanol extract pretreated group, butanol fraction pretreated group and $H_{2}O$ fraction pretreated group, $CCl_{4}$ treated groups were injected with $CCl_{4}$ 0.6 mg/kg (ip). The activities of ananine aminotransferasein hexane, chloroform and $H_{2}O$ fraction pretreated groups were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group and also activity of aspartate aminotransferase and hepatic malondialdehyde content in $H_{2}O$ fraction pretreated group were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group. The activity of alkaline phosphatase in chloroform and $H_{2}O$ fraction pretreated groups were decreased compared to the only $CCl_{4}$-treated group but the difference was not statistically significant. The activity of ${\gamma}$-glutamyltranspeptidase in hexane, chloroform and ethylacetate fraction pretreated groups were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group. The contents of cholesterol and triglyceride were not influenced in any fraction pretreated groups, but HDL-cholesterol content in hexane and chloroform fraction pretreated groups were significantly increased (p<0.05) compared to the only $CCl_{4}$-treated group. Therefore, these results demonstrate a possible hepatoprotective role of $H_{2}O$ fraction of Diospyrus kaki Folium methanol extract in carbon tetrachloride intoxicated rats.

Protective Effects of Extracts of Hovenia dulcis Thunb on Hepatotoxicity in Carbon Tetrachloride Intoxicated Rats (지구자나무 추출물이 사염화탄소로 유발된 흰쥐의 간손상에 미치는 영향)

  • This study was performed to investigate the protective effects of Hovenia dulcis Thunb on hepatotoxicity in carbon tetrachloride-intoxicated rats. Male Sprague-Dawley rats (220~240 g) were used as experimental groups, which were divided into 7 groups; Control group, $CCl_4$-treated group, hexane fraction pretreated and $CCl_4$-treated group, chloroform fraction pretreated and $CCl_4$-treated group, ethylacetate fraction pretreated and $CCl_4$-treated group, butanol fraction pretreated and $CCl_4$-treated group, $H_2O$ fraction pretreated and $CCl_4$-treated group. After 6 days, the activities of aminotransferase, contents of cholesterol, TG and hepatic lipid peroxide content in chloroform fraction pretreated and $CCl_4$-treated group were significantly decreased (p<0.05) compared to the only $CCl_4$-treated group. The content of glutathione and activities of GST in chloroform fraction pretreated and $CCl_4$-treated group were also significantly increased (p<0.05) compared to the only $CCl_4$-treated group. In addition, activities of SOD, catalase and GSH-Px in chloroform fraction pretreated and $CCl_4$-treated group were significantly decreased (p<0.05) compared to the only $CCl_4$-treated group. These results indicated that the chloroform fraction of Hovenia dulcis Thunb methanol extract showed hepatoprotective effect in carbon tetrachloride-intoxicated rats.

  • To investigate effects of A. gmelini on the 14-day CCl$_4$induced hepatotoxicity, extracts were prepared in 3 ml saline at the dose of 50, 500, 5,000 mg/kg b.w. to administer orally everyday and same concentration (1 : 9, CCl$_4$: olive oil v/v) of CCl$_4$administered intraperitoneally with 2.5 ml/kg b.w. On the 7th day, hemanalysis showed following recovery values; AST 9.9∼64.6%, ALT 36.9∼71.9%, ALP 75.3∼93.7%, BUN 53.8∼59.7%, TBIL 60.4∼100.0< %, TCHO 77.7∼100.0< %, and TG 60.4∼100.0< %. Even if, 14-day CCl$_4$induced hepatotoxicity recovery was found to depend on doses of extract, and recovery values of each treatment were AST 13.8∼56.4%, ALT 15.7∼68.0%, ALP 53.4∼84.4%, BUN 76.9∼100.0< %, TBIL 60.4 ∼ 100.0< %, TCHO 82.6∼99.3< %, and TG 56.7∼99.7%. By histological examination of liver, hydropic degeneration, fatty change, lipid accumulation and necrosis were also recovered.

  • In oriental medicine, Artemisia Iwayomogi(Compositae) has been used clinically for jaundice, hepatitis, liver cirrhosis etc. The purposes of present study were to examine pharmacological effects of Artemisia lwayomogi water extract(AIWE) on weights of body, liver, kidney, spleen and adrenal, and on biochemical parameters (activities of AST, ALT and LDH, contents of cholesterol and triacylglycerol, and levels of hepatic lipid peroxide) against hepatic injury by carbon tetrachloride($CCl_4$) in rats. The results were as follow; 1. Body weights were reduced by $CCl_4$. In AIWE pretreatment groups, reduction of body weights was inhibited at 48 hours. Increased liver weights by $CCl_4$ were reduced in proportion to numbers of treatment of AIWE in AIWE pre- and posttreatment groups. Increased kidney weights by $CCl_4$ were reduced in AIWE pretreatment groups at 72 hours. Increased weights of spleen and adrenal by $CCl_4$ were not affected by AIWE treament. 2. Increased AST activities by $CCl_4$ were significantly (p<0.05) decreased in AIWE posttreatment groups at 48 and 72 hours. Increased ALT activities by $CCl_4$ were significantly(p<0.05) decreased in AIWE posttreatment groups at 48 hours. Increased LDH activities by $CCl_4$ were very significantly (p<0.01, p<0.001) decreased in AIWE posttreatment groups at 48 and 72 hours, respectively. 3. Increased cholesterol contents by $CCl_4$ were significantly (p<0.05) decreased in AIWE posttreatment groups at 24 and 48 hours. Decreased triacylglycerol contents by $CCl_4$ were significantly (p<0.05) increased in AIWE posttreatment at 48 and 72 hours. 4. Increased hepatic lipid peroxide levels by $CCl_4$ were significantly (p<0.05, p<0.01) decreased in AIWE posttreatment groups at 48 and 72 hours, respectively. In conclusion, AIWE did not affect normal liver function and had property of antioxidant, due to reduced lipid peroxidation by $CCl_4$. AIWE seems to have hepatoprotective effects rather than direct preventive effects to $CCl_4$-induced necrotic degeneration of liver cell, cholestasis and damages in metabolism of lipid.
