The effects of various sedative cyclopeptides and peptide alkaloids from the Zizyphus species on calmodulin-dependent $Ca^{2+}$ -ATPase and phosphodiesterase were Investigated. Calmodulin-induced activation of $Ca^{2+}$-ATPase was strongly inhibited by sanjoinine-A dialdehyde (IC_{50}$, 2.3$\mu\textrm{m}$), -Ah1 (IC_{50}$, 4.0$\mu\textrm{m}$), -A (IC, 4.6$\mu\textrm{m}$), and -G2 (IC_{50}$, 7.2$\mu\textrm{m}$), while calmodulin-induced activation of phosphodiesterase was strongly inhibited by both deachuine- S10 (IC_{50}$, 4.9$\mu\textrm{m}$) and sanjoinine-D (IC_{50}$, 9.0$\mu\textrm{m}$). The inhibitory activity of the various cyclopeptides and peptide alkaloids on $Ca^{2+}$-ATPase was found to correlate well with their Sedative activity.
CA저장 중 저장기체 조성에 따른 사과 Fuji의 증산속도를 측정하고, 같은 조건에서 증산속도를 예측하기 위한 수학적 모델을 설정하여 증산속도를 예측하였다. 온도 $0^{\circ}C$, 상대습도 98%, 공기 유속 0.25m/s의 저장조건에서 6주 동안 CA저장하였을 때 사과 Fuji의 호흡속도는 일반저온저장에 비하여 50%이하로 낮출 수 있었다. 같은 저장조건에서 일반저온저장에서의 사과의 증산속도가 CA저장에 비하여 50~70 % 높았으며, 일정한 산소농도의 CA저장에서는 저장기체 중 이산화탄소농도가 높을수록 증산속도는 감소하는 것으로 나타났다. 본 연구에서 채택한 모델로 예측한 증산속도는 실측치와 유사한 값을 나타내어 본 연구에서 채택한 모델로 CA 저장 중 저장기체 조성에 따른 사과 Fuji의 증산속도를 잘 예측할 수 있었다. 사과의 증산속도는 호흡열량에 비례하여 증가하는 경향을 나타내었으나 증산속도의 증가폭은 호흡열량의 증가폭에 미치지 못하였다. 이는 호흡열량이 증가하면 사과의 증발표면의 온도가 높아져서 증산속도가 커질 수 있게 되지만, 증산속도의 증가에 따른 증발잠열의 증가가 증발표면의 온도를 미세하게 낮추어 주므로 일어나는 현상으로 볼 수 있다.

Background: Citric acid (CA) and sodium hypochlorite (NaOCl) have been used to disinfect animals to protect them against avian influenza and foot-and-mouth disease. Objectives: We performed a good laboratory practice (GLP)-compliant animal toxicity study to assess the acute toxic effects of CA and NaOCl aerosol exposure in Sprague-Dawley rats. Methods: Groups of five rats per sex were exposed for 4 h to four concentrations of the two chemicals, i.e., 0.00, 0.22, 0.67, and 2.00 mg/L, using a nose-only exposure. After a single exposure to the chemicals, clinical signs, body weight, and mortality was observed during the observation period. On day 15, an autopsy, and then gross findings, and histopathological analysis were performed. Results: After exposure to CA and NaOCl, body weight loss was observed but recovered. Two males died in the CA 2.00 mg/L group and, two males and one female died in the 2.00 mg/L NaOCl group. In the gross findings and histopathological analysis, discoloration of the lungs was observed in the CA exposed group and inflammatory lesions with discoloration of the lungs were observed in the NaOCl exposed group. These results suggest that the lethal concentration 50 (LC50) of CA is 1.73390 mg/L for males and > 1.70 mg/L for females. For NaOCl, the LC50 was 2.22222 mg/L for males and 2.39456 mg/L for females. Conclusions: The Globally Harmonized System is category 4 for both CA and NaOCl. In this study, the LC50 results were obtained through a GLP-based acute inhalation toxicity assessment. These results provide useful data to reset safety standards for CA and NaOCl use.
Effects of lead(Pb) and calcium(Ca) on growth responses, accumulation of heavy metals and nitrogen fixation activities of Melilotus suaveolens seedlings were quantitatively analyzed during growing period. Pb contents of the root treated with 30, 100 ppm Pb and 100 ppm Pb added 100 ppm Ca were 54.1, 90.9 and 26.1 folds higher than that of the control, respectively, at pH 4.2 in 28th days, and heavy metal content of plant increased with increasing of pH and Pb concentration. The melilot plant was classified as a Pb accumulator by higher accumulation of Pb in shoot than that of root. Pb treatments resulted in inhibiton of height and chlorophyll contet, and Ca treatment increased height and chlorophyll content insignificantly at pH 4.2 in 28 days. The plant biomass reduced 49, 60 and 54% at pH 4.2 and 47,53 and 50% at pH 6.5, respectively, by 30, 100 ppm Pb and 100 ppm Pb added 100 ppm Ca treatment. Specific nitrogen fixation of nodules reduced 68.4% and 46.6% by 100 ppm Pb treatment and 3.7% and 24.9% by 100 ppm Pb added 100 ppm Ca at pH 4.2 and pH 6.5, respectively, so Ca inhibited significantly Pb activity and toxicity in acdic pH. Nodule formation were reduced to 33, 33 and 50% at pH 4.2 and 50, 33 and 38% at pH 6.5 by 30, 100 ppm Pb and 100 ppm Pb added 100 ppm Ca, respectively.
본 연구에서는 담배식물의 생장과 glutathione reductase와 catalase의 활성에 미치는 p-CA, BA 및 SA의 영향을 연구하였다. 담배식물의 생장에 미치는 p-CA, BA 및 SA의 영향을 알아보기 위하여, $10^{-5}$ mM p-CA, BA 및 SA가 각각 함유된 MS배지에서 배양한 후 20일부터 50일까지 10일 간격으로 담배의 생장을 비교하였다. 3가지 물질 모두에 의해 50일에서 생장이 가장 잘 되였다. Glutathione reductase의 활성에 미치는 효과를 측정한 결과, BA의 활성은 대조구보다 감소하였으며, p-CA 역시 활성이 감소하였고, SA의 활성은 증가하였다. 또한 모두 40일에서 가장 활성이 높았다. Catalase의 활성에 미치는 효과를 측정한 결과, SA의 활성은 증가하였으며, BA와 p-CA의 활성은 감소하였다. 또한 모든 실험구에서 40일에 활성이 가장 양호하였다. 결론적으로 p-CA와 BA는 담배식물의 생장을 촉진시켰으며, 50일에서 생장이 가장 양호하였으나 항산화 효소의 활성을 억제하였다. SA는 담배식물의 생장을 억제시켰으나 항산화 효소의 활성은 촉진시켰으며, 40일에서 가장 활성이 양호하였다.
Chrysosplenol C [5,6-dihydroxy-2-(4-hydroxy-3-methoxyphenyl)-3,7-dimethoxychromen-4-one] is a flavonoid found in Miliusa balansae and Pterocaulon sphacelatum. We have recently shown that chrysosplenol C has positive inotropic effect in isolated rat ventricular myocytes. In the present study, we explored a possible mechanism for the positive inotropic effect of chrysosplenol C by examining intracellular $Ca^{2+}$ transients during action potentials. The intracellular $Ca^{2+}$ transients were measured by confocal $Ca^{2+}$ imaging in field-stimulated single rat ventricular myocytes. Chrysosplenol C (50 ${\mu}M$) significantly increased the magnitudes (${\Delta}F/F_0$) of $Ca^{2+}$ transients (control, $1.08{\pm}0.05$; chrysosplenol C, $1.25{\pm}0.03$; n=8, P<0.01). Half decay time of the action potential-induced $Ca^{2+}$ transient was not altered by chrysosplenol C (50 ${\mu}M$) (control, $154{\pm}6$ ms; chrysosplenol C, $167{\pm}11$ ms; n=21). The $Ca^{2+}$ content in the sarcoplasmic reticulum (SR), measured as caffeine (10 mM)-induced $Ca^{2+}$ transient, was significantly decreased by chrysosplenol C (50 ${\mu}M$). These results indicate that chrysosplenol C increases $Ca^{2+}$ transients without altering $Ca^{2+}$ removal kinetics in ventricular myocytes, providing a possible mechanism for its positive inotropic effect.
The magnesium-base AZ61 alloy was cast while adding 1% CaO powder into the melt. It was hot extruded, and oxidized at $550-650^{\circ}C$ in air in order to study its microstructure and oxidation behavior. Initially added CaO powder reacted with Al in the melt to $Al_2Ca$ particles that aligned along the extrusion direction. The formed $Al_2Ca$ particles increased the oxidation resistance through forming the superficial CaO scale at the upper part of the thin MgO oxide scale.
A biomimetic approach was applied for the chemical deposition of calcium phosphate (CaP) coatings on 3 mole% yttrium-stabilized zirconia [3YSZ] powders. The solid-state reaction of 3YSZ powders with the addition of CaP was investigated for the development of biodegradable zirconia ceramics. The solid-state interaction between the 3YSZ matrix powders and the CaP additives differed from the behavior of commercial zirconia matrix powders. The 3YSZ powders were chemically reacted with precursors for the CaP formation. 3YSZ powders were mixed in an aqueous solution of Ca-P precursors and the CaP was precipitated on the surface of the 3YSZ matrix powders. The CaP-doped YSZ powders were calcined at $1100^{\circ}C$ and shaped powder blocks were then fired at $1600^{\circ}C$ for 2 h. The CaP phase formation was investigated using FE-SEM and XRD analysis.
We investigated the effect of ${\alpha}-adrenergic$ and cholinergic receptor agonists on $Ca^{2+}$ current in adult rat trigeminal ganglion neurons using whole-cell patch clamp methods. The application of acetylcholine, carbachol, and oxotremorine ($50\;{\mu}M\;each$) produced a rapid and reversible reduction of the $Ca^{2+}$ current by $17{\pm}6%,\;19{\pm}3%,\;and\;18{\pm}4%$, respectively. Atropine, a muscarinic antagonist, blocked carbachol- induced $Ca^{2+}$ current inhibition to $3{\pm}1%$. Norepinephrine ($50\;{\mu}M$) reduced $Ca^{2+}$ current by $18{\pm}2%$, while clonidine ($50\;{\mu}M$), an ${\alpha}2-adrenergic$ receptor agonist, inhibited $Ca^{2+}$ current by only $4{\pm}1%$. Yohimbine, an ${\alpha}2-adrenergic$ receptor antagonist, did not block the inhibitory effect of norepinephrine on $Ca^{2+}$ current, whereas prazosin, an ${\alpha}1-adrenergic$ receptor antagonist, attenuated the inhibitory effect of norepinephrine on $Ca^{2+}$ current to $6{\pm}1%$. This pharmacology contrasts with ${\alpha}2-adrenergic$ receptor modulation of $Ca^{2+}$ channels in rat sympathetic neurons, which is sensitive to clonidine and blocked by yohimbine. Our data suggest that the modulation of voltage dependent $Ca^{2+}$ channel by norepinephrine is mediated via an α1-adrenergic receptor. Pretreatment with pertussis toxin (250 ng/ml) for 16 h greatly reduced norepinephrine- and carbachol-induced $Ca^{2+}$ current inhibition from $17{\pm}3%\;and\;18{\pm}3%\;to\;2{\pm}1%\;and\;2{\pm}1%$, respectively. These results demonstrate that norepinephrine, through an ${\alpha}1-adrenergic$ receptor, and carbachol, through a muscarinic receptor, inhibit $Ca^{2+}$ currents in adult rat trigeminal ganglion neurons via pertussis toxin sensitive GTP-binding proteins.
